| 1 | Cell sheet formation enhances the therapeutic effects of human umbilical cord mesenchymal stem cells on myocardial infarction as a bioactive material显示文摘Stem cell-based therapy has been used to treat ischaemic heart diseases for two decades.However,optimal cell types and transplantation methods remain unclear.This study evaluated the therapeutic effects of human umbilical cord mesenchymal stem cell(hUCMSC)sheet on myocardial infarction(MI).Methods:hUCMSCs expressing luciferase were generated by lentiviral transduction for in vivo bio-luminescent imaging tracking of cells.We applied a temperature-responsive cell culture surface-based method to form the hUCMSC sheet.Cell retention was evaluated using an in vivo bio-luminescent imaging tracking system.Unbiased transcriptional profiling of infarcted hearts and further immunohistochemical assessment of monocyte and macrophage subtypes were used to determine the mechanisms underlying the therapeutic effects of the hUCMSC sheet.Echocardiography and pathological analyses of heart sections were performed to evaluate cardiac function,angiogenesis and left ventricular remodelling.Results:When transplanted to the infarcted mouse hearts,hUCMSC sheet significantly improved the retention and survival compared with cell suspension.At the early stage of MI,hUCMSC sheet modulated inflammation by decreasing Mcp1-positive monocytes and CD68-positive macrophages and increasing Cx3cr1-positive non-classical macrophages,preserving the cardiomyocytes from acute injury.Moreover,the extracellular matrix produced by hUCMSC sheet then served as bioactive scaffold for the host cells to graft and generate new epicardial tissue,providing mechanical support and routes for revascularsation.These effects of hUCMSC sheet treatment significantly improved the cardiac function at days 7 and 28 post-MI.Conclusions:hUCMSC sheet formation dramatically improved the biological functions of hUCMSCs,mitigating adverse post-MI remodelling by modulating the inflammatory response and providing bioactive scaffold upon transplantation into the heart.Translational perspective:Due to its excellent availability as well as superior local cellular retention and survival,allogenic transplantation of hUCMSC sheets can more effectively acquire the biological functions of hUCMSCs,such as modulating inflammation and enhancing angiogenesis.Moreover,the hUCMSC sheet method allows the transfer of an intact extracellular matrix without introducing exogenous or synthetic biomaterial,further improving its clinical applicability. | Rui Guo Feng Wan Masatoshi Morimatsu Qing Xu Tian Feng Hang Yang Yichen Gong Shuhong Ma Yun Chang Siyao Zhang Youxu Jiang Heqing Wang Dehua Chang Hongjia Zhang Yunpeng Ling Feng Lan | 2021 | Bioactive Materials2021,6,9: | 6 |

| 5 | Trimetazidine enhances myocardial angiogenesis in pressure overload-induced cardiac hypertrophy mice through directly activating Akt and promoting the binding of HSF1 to VEGF-A promoter显示文摘Latest clinical research shows that trimetazidine therapy during the perioperative period relieves endothelial dysfunction in patients with unstable angina induced by percutaneous coronary intervention.In this study we investigated the effects of TMZ on myocardial angiogenesis in pressure overload-induced cardiac hypertrophy mice.Cardiac hypertrophy was induced in mice by transverse aortic constriction(TAC)surgery.TAC mice were administered trimetazidine(2.8 mg/100μL,i.g.)for 28 consecutive days.We showed that trimetazidine administration significantly increased blood vessel density in the left ventricular myocardium and abrogated cardiac dysfunction in TAC mice.Co-administration of a specific HSF1 inhibitor KRIBB11(1.25 mg/100μL,i.h.)abrogated the angiogenesis-promoting effects of trimetazidine in TAC mice.Using luciferase reporter and electrophoretic mobility shift assays we demonstrated that the transcription factor HSF1 bound to the promoter region of VEGF-A,and the transcriptional activity of HSF1 was enhanced upon trimetazidine treatment.In molecular docking analysis we found that trimetazidine directly bound to Akt via a hydrogen bond with Asp292 and a pi–pi bond with Trp80.In norepinephrine-treated HUVECs,we showed that trimetazidine significantly increased the phosphorylation of Akt and the synergistic nuclear translocation of Akt and HSF1,as well as the binding of Akt and HSF1 in the nucleus.These results suggest that trimetazidine enhances myocardial angiogenesis through a direct interaction with Akt and promotion of nuclear translocation of HSF1,and that trimetazidine may be used for the treatment of myocardial angiogenic disorders in hypertensive patients. | Hong-yang Shu Yi-zhong Peng Wei-jian Hang Min Zhang Lan Shen Dao-wen Wang Ning Zhou | 2022 | Acta Pharmacologica Sinica2022,43,10: | 2 |

| 8 | Genetic dissection of wheat uppermost-internode diameter and its association with agronomic traits in five recombinant inbred line populations at various field environments显示文摘Uppermost-internode diameter(UID)is a key morphological trait associated with spike development and yield potential in wheat.Our understanding of its genetic basis remains largely unknown.Here,quantitative trait loci(QTLs)for UID with high-density genetic maps were identified in five wheat recombinant inbred line(RIL)populations.In total,25 QTLs for UID were detected in five RIL populations,and they were located on chromosomes 1A,1D(3 QTL),2B(2),2D(3),3B,3D,4A,4B(3),4D,5A(5),5B(2),6B,and 7D.Of them,five major and stable QTLs(QUid.sau-2CN-1D.1,QUid.sau-2SY-1D,QUid.sau-QZ-2D,QUid.sau-SC-3D,and QUid.sau-AS-4 B)were identified from each of the five RIL populations in multiple environments.QUid.sau-2CN-1D.1,QUid.sau-2SY-1D and QUid.sau-SC-3D are novel QTLs.Kompetitive Allele Specific PCR(KASP)markers tightly linked to them were further investigated for developing near-isogenic lines(NILs)carrying the major loci.Furthermore,candidate genes at these intervals harboring major and stable QTLs were predicted,and they were associated with plant development and water transportation in most cases.Comparison of physical locations of the identified QTL on the‘Chinese Spring’reference genome showed that several QTLs including two major ones,QUid.sau-2CN-1D.1 and QUid.sau-2 SY-1 D,are likely allelic confirming their validity and effectiveness.The significant relationships detected between UID and other agronomic traits and a proper UID were discussed.Collectively,our results dissected the underlying genetic basis for UID in wheat and laid a foundation for further fine mapping and map-based cloning of these QTLs. | LIU Hang TANG Hua-ping LUO Wei MU Yang JIANG Qian-tao LIU Ya-xi CHEN Guo-yue WANG Ji-rui ZHENG Zhi QI Peng-fei JIANG Yun-feng CUI Fa SONG Yin-ming YAN Gui-jun WEI Yu-ming LAN Xiu-jin ZHENG You-liang MA Jian | 2021 | Journal of Integrative Agriculture2021,20,11: | 1 |

| 12 | Ecological and behavioural risk factors of scrub typhus in central Vietnam:a case-control study显示文摘Background The risk factors for scrub typhus in Vietnam remain unknown.Scrub typhus caused by Orientia tsutsugamushi often presents as an undifferentiated febrile illness and remains under appreciated due to the limited availability of diagnostic tests.This tropical rickettsial illness is increasingly recognized as an important cause of non-malaria acute undifferentiated fever in Asia.This study aimed to investigate behavioural and ecological related risk factors of scrub typhus to prevent this potentially life-threatening disease in Vietnam.Methods We conducted a clinical hospital-based active surveillance study,and a retrospective residence-enrolment date-age-matched case–control study in Khanh Hoa province,Vietnam,from August 2018 to March 2020.Clinical examinations,polymerase chain reaction and enzyme-linked immunosorbent assay IgM tests were applied to define cases and controls.All enrolled participants filled out a questionnaire including demographic socio-economic status,personal behaviors/protective equipment,habitat connections,land use,and possible exposure to the vector.Multivariable conditional logistic regression was used to define the scrub typhus associated risk factors.Results We identified 44 confirmed cases and matched them with 152 controls.Among cases and controls,the largest age group was the 41–50 years old and males accounted for 61.4%and 42.8%,respectively.There were similarities in demographic characteristics between the two groups,with the exception of occupation.Several factors were significantly associated with acquisition of scrub typhus,including sitting/laying directly on household floor[adjusted OR(aOR)=4.9,95%CI:1.6–15.1,P=0.006],household with poor sanitation/conditions(aOR=7.9,95%CI:1.9–32.9,P=0.005),workplace environment with risk(aOR=3.0,95%CI:1.2–7.6,P=0.020),always observing mice around home(aOR=3.7,95%CI:1.4–9.9,P=0.008),and use of personal protective equipment in the field(aOR=0.4,95%CI:0.1–1.1,P=0.076).Conclusions Ecological and household hygiene-related factors were more associated with scrub typhus infection,than individual-level exposure activities in the hyper-endemic area.These findings support local education and allow people to protect themselves from scrub typhus,especially in areas with limitations in diagnostic capacity. | Hanh Thi Duc Tran Jan Hattendorf Hung Manh Do Thanh Tien Hoang Hang Thi Hai Hoang Hoa Ngoc Lam Mai Kim Huynh Lan Thi Hoang Vu Jakob Zinsstag Daniel Henry Paris Esther Schelling | 2021 | Infectious Diseases of Poverty2021,10,4: | 1 |

| 17 | Effect of Amino-,Methyl- and Epoxy-Silane Coupling as a Molecular Bridge for Formatting a Biomimetic Hydroxyapatite Coating on Titanium by Electrochemical Deposition显示文摘The objective of this study was to determine the role of functional groups of silane coupling on bioactive titanium(Ti) surface by electrochemical deposition, and calcium phosphate(CaP) coating, as well as bone cell adhesion and proliferation. Methyl group(—CH_3), amino group(—NH_2), and epoxy group(—glyph name—C(O)C) were introduced onto the bioactive Ti surface using self-assembled monolayers(SAMs)with different silane coupling agents as molecular bridges. The effect of the surface functional groups on the growth features of the CaP crystals was analyzed(including chemical compositions, element content,minerals morphology and crystal structure etc.). CH_3- terminated SAMs showed a hydrophobic surface and others were hydrophilic by contact angle measurement; NH_2-terminated SAMs showed a positive charge and others were negatively charged using zeta-potential measurement. Scanning electron microscopy results confirmed that flower-like structure coatings consisting of various pinpoint-like crystals were formatted by different functional groups of silane coupling, and the CaP coatings were multicrystalline consisting of hydroxyapatite(HA) and precursors. Ca P coating of CH_3- terminated SAMs exhibited more excellent crystallization property as compared to coatings of —NH_2 and —C(O)C groups. In vitro MC3T3-E1 cells adhesion and proliferation were performed. The results showed that CaP coatings on silane coupling functionalized surfaces supported cell adhesion and proliferation.
