Reactive capillary hemangiomas: a novel dermatologic toxicity following anti-PD-1 treatment with SHR-1210显示文摘Objective: SHR-1210 is a new and promising anti-PD-1 agent for solid tumors. During the phase I study of SHR-1210, we encountered a novel but prevalent immune-related dermatologic toxicity: reactive capillary hemangiomas(RCHs). Thus we tried to summarize the features of RCHs and estimate their relationship with tumor response. Methods: This prospective observational study systematically enrolled 98 patients with advanced solid tumors from April 27th,2016 to June 8th, 2017 in the context of the phase I clinical study of SHR-1210. This report focused on the skin toxicities. Patients underwent entire skin inspection every two weeks while taking medication. The clinical course of RCHs was recorded and their association with tumor response was estimated. The data cut-off date was November 15th, 2017.Results: After a median follow-up of 242(range, 29–567) days, RCHs were observed in 85.7%(84/98) of patients on cutaneous/mucosal surfaces; 84.5%(71/84) of the RCHs were evaluated as grade 1 adverse events. No grade 3 or 4 RCHs were observed. The time of onset of RCHs was dose dependent and shortest in the 400 mg-dose cohort(P < 0.001). Spontaneous and complete regression of RCHs was observed both during and after treatment. The objective response rate of tumors for patients with RCHs was 28.9%(24/83). However, no responders were observed among the patients without RCHs.Conclusions: RCHs were prevalent but manageable during treatment with SHR-1210. It might add to the expanding literature regarding immune-related dermatologic adverse events.Xuelian Chen Lanying Ma Xi Wang Hongnan Mo Dawei Wu Bo Lan Dong Qu Hongtu Zhang Jing Huang Binghe Xu 2019Cancer Biology & Medicine2019,16,1:53
Peripheral T-lymphocyte subpopulations in different clinical stages of chronic HBV infection correlate with HBV load显示文摘AIM: To characterize the peripheral T-cell subpopulation profiles and their correlation with hepatitis B virus (HBV) replication in different clinical stages of chronic HBV infection.METHODS: A total of 422 patients with chronic HBV infection were enrolled in this study. The patients were divided into three stages: immune-tolerant stage, immune active stage, and immune-inactive carrier stage. Composition of peripheral T-cell subpopulations was determined by flow cytometry. HBV markers were detected by enzyme-linked immunosorbent assay. SerumHBV DNA load was assessed by quantitative real-time polymerase chain reaction.RESULTS: CD8+ T-cells were significantly higher in patients at the immune-tolerant stage than in patients at the immune-active and -inactive carrier stages (36.87 ± 7.58 vs 34.37 ± 9.07, 36.87 ± 7.58 vs 28.09 ± 5.64, P < 0.001). The peripheral blood in patients at the immune-tolerant and immune active stages contained more CD8+ T-cells than CD4+ T-cells (36.87 ± 7.58 vs 30.23 ± 6.35, 34.37 ± 9.07 vs 30.92 ± 7.40, P < 0.01), whereas the peripheral blood in patients at the immune-inactive carrier stage and in normal controls contained less CD8+ T-cells than CD4+ T-cells (28.09 ± 5.64 vs 36.85 ± 6.06, 24.02 ± 4.35 vs 38.94 ± 3.39, P < 0.01). ANOVA linear trend test showed that CD8+ T-cells were signif icantly increased in patients with a high viral load (39.41 ± 7.36, 33.83 ± 7.50, 31.81 ± 5.95 and 26.89 ± 5.71, P < 0.001), while CD4+ T-cells were signif icantly increased in patients with a low HBV DNA load (37.45 ± 6.14, 33.33 ± 5.61, 31.58 ± 6.99 and 27.56 ± 5.49, P < 0.001). Multiple regression analysis displayed that log copies of HBV DNA still maintained its highly signif icant coefficients for T-cell subpopulations, and was the strongest predictors for variations in CD3+, CD4+ and CD8+ cells and CD4+/CD8+ ratio after adjustment for age at HBV-infection, maternal HBV-infection status, presence of hepatitis B e antigen and HBV mutation.CONCLUSION: Differences in peripheral T-cell subpopulation profi les can be found in different clinical stages of chronic HBV infection. T-cell impairment is signifi cantly associated with HBV load.Jing You Lin Zhuang Yi-Feng Zhang Hong-Ying Chen Hutcha Sriplung Alan Geater Virasakdi Chongsuvivatwong Teerha Piratvisuth Edward McNeil Lan Yu Bao-Zhang Tang .lun-Hua Huang 2009World Journal of Gastroenterology2009,15,27:39
Phase II clinical trial using camrelizumab combined with apatinib and chemotherapy as the first-line treatment of advanced esophageal squamous cell carcinoma显示文摘Background:Effective therapeutic options are limited for patients with advanced esophageal squamous cell carcinoma(ESCC).The incorporation of an immune checkpoint inhibitor and a molecular anti-angiogenic agent into the commonly adopted chemotherapy may produce synergistic effects. Therefore,we aimed to investigate the efficacy and safety of camrelizumab plus apatinib combined with chemotherapy as the first-line treatment of advanced ESCC.Methods:In this single-arm prospective phase II trial, patients with unresectable locally advanced or recurrent/metastatic ESCC received camrelizumab 200 mg,liposomal paclitaxel 150 mg/m2,and nedaplatin 50 mg/m2 on day 1,and apatinib 250 mg on days 1-14.The treatments were repeated every 14 days for up to 9 cycles, followed by maintenance therapy with camrelizumab and apatinib. The primary endpoint was objective response rate(ORR)according to the Response Evaluation Criteria in Solid Tumors(version 1.1).Secondary endpoints included disease control rate(DCR),progression-free survival(PFS),overall survival(OS),and safety. Results:We enrolled 30 patients between August 7,2018 and February 23,2019.The median follow-up was 24.98 months(95%confidence interval[CI]:23.05-26.16 months). The centrally assessed ORR was 80.0%(95%CI:61.4%-92.3%),with a median duration of response of 9.77 months(range:1.54 to 24.82+months).The DCR reached 96.7%(95%CI:82.8%-99.9%).The median PFS was 6.85 months(95%CI:4.46-14.20 months),and the median OS was 19.43 months(95%CI:9.93 months–not reached). The most common grade 3-4 treatmentrelated adverse events(AEs)were leukopenia(83.3%),neutropenia(60.0%),and increased aspartate aminotransferase level(26.7%).Treatment-related serious AEs included febrile neutropenia, leukopenia, and anorexia in one patient(3.3%),and single cases of increased blood bilirubin level(3.3%)and toxic epidermal necrolysis(3.3%).No treatment-related deaths occurred. Conclusions:Camrelizumab plus apatinib combined with liposomal paclitaxel and nedaplatin as first-line treatment demonstrated feasible anti-tumor activity and manageable safety in patients with advanced ESCC.Randomized trials to evaluate this new combination strategy are warranted.Trial registration:This trial was registered on July 27,2018,at ClinicalTrials.gov(identifier:NCT03603756).Bo Zhang Ling Qi Xi Wang Jianping Xu Yun Liu Lan Mu Xingyuan Wang Lidan Bai Jing Huang 2020Cancer Communications2020,40,12:38
Helicobacter pylori infection generated gastric cancer through p53-Rb tumor-suppressor system mutation and telomerase reactivation显示文摘AIM: To investigate the relationship between Helicobacterpylori (H. pylori) infection and the expressions of the p53,Rb, c-myc, bcl-2 and hTERT mRNA in a series of diseasesfrom chronic gastritis (CG), intestinal metaplasia type Ⅰ or Ⅱ(IMⅠ-Ⅱ), intestinal metaplasia type Ⅲ (IMⅢ), mild or modestdysplasia (DysⅠ-Ⅱ), severe dysplasia (DysⅢ) to gastric cancer(GC) and to elucidate the mechanism of gastriccarcinogenesis relating to H.pyloriinfection.METHODS: 272 cases between 1998 and 2001 wereavailable for the study including 42 cases of CG, 46 cases ofIMⅠ-Ⅱ, 25 cases of IMⅢ, 48 cases of DysⅠ-Ⅱ, 27 cases ofDysⅢ, 84 cases of GC.-H. pyloriinfection and the expressionsof p53, Rb, c-myc, bcl-2 were detected by means ofstreptavidin-peroxidase (SP) immunohistochemical method.HTERT mRNA was detected byin situ hybridization(ISH).RESULTS: The expressions of p53, Rb, c-myc, hTERT mRNAand bcl-2 were higher in the GC than in CG, IN, Dys. Theexpression of c-myc was higher in IMⅢ with-H.pyloriinfection(10/16) than that without infection (1/9) and the positive ratein DysⅠ-Ⅱ and DysⅢ with-H.pyloriinfection was 18/30 and 13/17, respectively, higher than that without infection (4/18 and3/10, respectively). In our experiment mutated p53 had noassociation with H.pyloriinfection, theexpression of Rb wasassociated with-H. pyloriinfection in GC, but the p53-Rb tumor-suppressor system abnormal in DysⅠ-Ⅱ cases, DysⅢⅡ and GCcases with H. pyloriinfection was 21/30, 15/17 and 48/48respecively, higher than non-infection groups (4/18, 3/10, 28/36). Furthermore the level of hTERT mRNA in GC with H. pyloriinfection (47/48) was higher than that without infection (30/36), however the relationship between bcl-2 and H. pyloriwasonly in IMⅢ. C-myc had a close association with hTERT mRNAin DysⅢ and GC (P=0.0 253,0.0 305 respectively).CONCLUSION: In the gastric carcinogenesis, H. pylorimightcause the severe imbalance of proliferation and apoptosisin the precancerous lesions (IMⅢ and GysⅢ) first, leadingto p53-Rb tumor-suppressor system mutation and telomerasereactivation, and finally causes gastric cancer.Jing Lan Yong-Yan Xiong Yi-Xian Lin Bi-Cheng Wang Ling-Ling Gong Hui-Sen Xu Guang-Song Guo Department of Pathology,Zhongnan Hospital,Wuhan University,Wuhan city 430071,Hubei Province,China 2003World Journal of Gastroenterology2003,9,1:39
Effect of viral load on T-lymphocyte failure in patients with chronic hepatitis B显示文摘AIM: To investigate peripheral T-lymphocyte sub-population profile and its correlation with hepatitis B virus (HBV) replication in patients with chronic hepatitis B (CHB).METHODS: Distribution of T-lymphocyte subpopulations in peripheral blood was measured by flow cytometry in 206 CHB patients. HBV markers were detected with ELISA. Serum HBV DNA load was assessed with quantitative real-time polymerase chain reaction (PCR). The relationship between HBV replication and variation in peripheral T-cell subsets was analyzed.RESULTS: CHB patients had significantly decreased CD3+ and CD4+ cells and CD4+/CD8+ ratio, and increased CD8+ cells compared with uninfected controls (55.44 ± 12.39 vs 71.07 ± 4.76, 30.92 ± 7.48 vs 38.94 ± 3.39, 1.01 ± 0.49 vs 1.67 ± 0.33, and 34.39 ± 9.22 vs 24.02 ± 4.35; P < 0.001, respectively). Univariate analysis showed a similar pattern of these parameters was significantly associated with high viral load, presence of serum hepatitis B e antigen (HBeAg) expression, liver disease severity, history of maternal HBV infection, and young age at HBV infection, all with P < 0.01. There was a significant linear relationship between viral load and these parameters of T-lymphocyte subpopulations (linear trend test P < 0.001). There was a negative correlation between the levels of CD3+ and CD4+ cells and CD4+/CD8+ ratio and serum level of viral load in CHB patients (r = -0.68, -0.65 and -0.75, all P < 0.0001), and a positive correlation between CD8+ cells and viral load (r = 0.70, P < 0.0001). There was a significant decreasing trend in CD3+ and CD4+ cells and CD4+/CD8+ ratio with increasing severity of hepatocyte damage and decreasing age at HBV infection (linear trend test P < 0.01). In multiple regression (after adjustment for age at HBV infection, maternal HBV infection status and hepatocyte damage severity) log copies of HBV DNA maintained a highly significant predictive coefficient on T-lymphocyte subpopulations, and was the strongest predictor of variation in CD3+, CD4+, CD8+ cells and CD4+/CD8+ ratio. However, the effect of HBeAg was not significant.CONCLUSION: T-lymphocyte failure was signifi-cantly associated with viral replication level. The substantial linear dose-response relationship and strong independent predictive effect of viral load on T-lymphocyte subpopulations suggests the possibility of a causal relationship between them, and indicates the importance of viral load in the pathogenesis of T cell hyporesponsiveness in these patients.Jing You Hutcha Sriplung Alan Geater Virasakdi Chongsuvivatwong Lin Zhuang Hong-Ying Chen Lan Yu Bao-Zhang Tang Jun-Hua Huang 2008World Journal of Gastroenterology2008,14,7:30
Medusoid Fossils from the Sinian Xingmincun Formation of Southern Liaoning Province显示文摘Abundant medusoid soft-bodied metazoan fossils have been found in the Sinian Xingmincun Formation(with an isotopic age of 650 Ma) at Qipanmo, Qidingshan and Yangtun of the Dalian-Jinzhou area, southern Liaoning Province; in addition, a number of macroscopic algal fossils have been discovered for the first time in the Getun Formation overlying the Xingmincun Formation. Medusoid fossils include 3 genera and 6 species(all of which are new). Judging from their biological characters, these medusoid fossils are considered to have an intimate affinity with the Ediacara fauna in the terminal Precambrian of Australia, as their characters are close to each other. The authors suggest that these medusoids and the Edicara fauna might be the products of the same period. The discovery of the medusoid fauna in the Xingmincun Formation marks an important event in the evolution of the terminal Precambrian organisms in China, and it is of major practical significance in the study of the Sinian System of China and the correlation of the Sinian strata of northern and southern China.Hong Zuomin, Huang Zhenfu, Yang Xinde, Lan Jing, Xian Bingcai, Yang Yajun, Regional Geological Survey Party, Bureau of Geology and Mineral Resources of Liaoning Province, Jinzhou, Liaoning and Liu Xiaoliang Shenyang Institute of Geology and Resources, Chinese Academy of Geological Sciences, Shenyang, Liaoning Fang Xinzhong 1989Acta Geologica Sinica(English Edition)1989,63,1:20
Effects of Short-Term Forest Bathing on Human Health in a Broad-Leaved Evergreen Forest in Zhejiang Province,China显示文摘Objective To investigate the effects of short-term forest bathing on human health. Methods Twenty healthy male university students participated as subjects and were randomly divided into two groups of 10.One group was sent on a two-night trip to a broad-leaved evergreen forest, and the other was sent to a city area.Serum cytokine levels reflecting inflammatory and stress response, indicators reflecting oxidative stress, the distribution of leukocyte subsets, and plasma endothelin-1 (ET-1) concentrations were measured before and after the experiment to evaluate the positive health effects of forest environments.A profile of mood states (POMS) evaluation was used to assess changes in mood states. Results No significant differences in the baseline values of the indicators were observed between the two groups before the experiment.Subjects exposed to the forest environment showed reduced oxidative stress and pro-inflammatory level,as evidenced by decreased malondialdehyde,interleukin-6,and tumor necrosis factor α levels compared with the urban group.Serum cortisol levels were also lower than in the urban group.Notably, the concentration of plasma ET-1 was much lower in subjects exposed to the forest environment. The POMS evaluation showed that after exposure to the forest environment, subjects had lower scores in the negative subscales, and the score for vigor was increased. Conclusion Forest bathing is beneficial to human health, perhaps through preventive effects related to several pathological factors.MAO Gen Xiang LAN Xiao Guang CAO Yong Bao CHEN Zhuo Mei HE Zhi Huat LV Yuan Dong WANG Ya Zhen HU Xi Lian WANG Guo Fu YAN Jing 2012Biomedical and Environmental Sciences2012,25,3:20
Repurposing vitamin D for treatment of human malignancies via targeting tumor microenvironment显示文摘Tumor cells along with a small proportion of cancer stem cells exist in a stromal microenvironment consisting of vasculature, cancer-associated fibroblasts, immune cells and extracellular components.Recent epidemiological and clinical studies strongly support that vitamin D supplementation is associated with reduced cancer risk and favorable prognosis. Experimental results suggest that vitamin D not only suppresses cancer cells, but also regulates tumor microenvironment to facilitate tumor repression. In this review, we have outlined the current knowledge on epidemiological studies and clinical trials of vitamin D. Notably, wesummarized and discussed the anticancer action of vitamin D in cancer cells, cancer stem cells and stroma cells in tumor microenvironment, providing a better understanding of the role of vitamin D in cancer. We presently re-propose vitamin D to be a novel and economical anticancer agent.Xu Wu Wei Hu Lan Lu Yueshui Zhao Yejiang Zhou Zhangang Xiao Lin Zhang Hanyu Zhang Xiaobing Li Wanping Li Shengpeng Wang Chi Hin Cho Jing Shen Mingxing Li 2019Acta Pharmaceutica Sinica B2019,9,2:20

Effect of the number of freeze-thaw cycles number on the quality of Pacific white shrimp (Litopenaeus vannamei): An emphasis on moisture migration and microstructure by LF-NMR and SEM显示文摘The objective of this study was to evaluate the effects of the number of freeze-thaw(F-T)cycles on moisture migration, protein degradation, microstructure and quality in Litopenaeus vannamei. The quality of samples with different F-T cycles were determined by low-field nuclear magnetic resonance(LF-NMR),magnetic resonance imaging(MRI)and scanning electron microscopy(SEM),combined with sodium dodecyl sulphate polyacrylamide gel electrophoresis(SDS-PAGE),texture properties analysis(TPA),color difference, polyphenol oxidase(PPO)activity, total volatile basic nitrogen(TVB-N),total viable count(TVC),and sensory evaluation. The results showed that F-T cycles caused a significantly increase in transverse relaxation time in T22 and T23 and a decreased brightness of pseudo-color diagram after 4 F-T cycles, indicating that water mobility increased as immobilized water was shifted to free water. The texture of samples declined as well, especially after 4 F-T cycles. The rapid growth of PPO activity caused a decrease in brightness(L*)and an increase in redness(a∗)after 3 F-T cycles. The increase of TVB-N,TVC,and sensory score explained the changes in shrimp quality which became obvious after 3 F-T cycles and unacceptable after 6 cycles.Meanwhile, fewer than 4 F-T cycles accelerated protein aggregation, while denaturation occurred after 4 cycles. Therefore, repeated F-T cycles could accelerate the changes of protein, microstructure, water distribution, and quality deterioration especially after 3 F-T cycles, with a threshold was 6 F-T cycles.Weiqing Lan Xiaoyu Hu Xiaohong Sun Xi Zhang Jing Xie 2020Aquaculture and Fisheries2020,5,4:15
Calcium-binding ability of soy protein hydrolysates显示文摘This present study investigated the ability of various soy protein hydrolysates(SPHs)in binding calcium. It was demonstrated that the amount of Ca-bound depended greatly on the SPHs obtained using different proteases, which included:neutrase, flavourzyme, protease M and pepsin. The maximum level of Ca-bound(66.9 mg/g)occurred when protease M was used to hydrolyze soy protein.Peptide fragments exhibiting high Ca-binding capacity had molecular weights of either 14.4 or 8-9 kDa.The level of Ca-bound increased linearly with the increment of carboxyl content in SPHs,and further deamidation on SPHs from protease M improved Ca-binding of the hydrolysate.Xiao Lan Bao Mei Song Jing Zhang Yang Chen Shun Tang Guo 2007Chinese Chemical Letters2007,18,9:14
The Genetic Architecture of Flowering Time and Photoperiod Sensitivity in Maize as Revealed by QTL Review and Meta Analysis显示文摘The control of flowering is not only important for reproduction, but also plays a key role in the processes of domestication and adaptation. To reveal the genetic architecture for flowering time and photoperiod sensitivity,a comprehensive evaluation of the relevant literature was performed and followed by meta analysis.A total of 25 synthetic consensus quantitative trait loci(QTL)and four hot-spot genomic regions were identified for photoperiod sensitivity including 11 genes related to photoperiod response or flower morphogenesis and development.Besides,a comparative analysis of the QTL for flowering time and photoperiod sensitivity highlighted the regions containing shared and unique QTL for the two traits.Candidate genes associated with maize flowering were identified through integrated analysis of the homologous genes for flowering time in plants and the consensus QTL regions for photoperiod sensitivity in maize(Zea mays L.).Our results suggest that the combination of literature review,meta-analysis and homologous blast is an efficient approach to identify new candidate genes and create a global view of the genetic architecture for maize photoperiodic flowering.Sequences of candidate genes can be used to develop molecular markers for various models of marker-assisted selection, such as marker-assisted recurrent selection and genomic selection that can contribute significantly to crop environmental adaptation.Jie Xu Yaxi Liu Jian Liu Moju Cao Jing Wang Hai Lan Yunbi Xu Yanli Lu Guangtang Pan Tingzhao Rong 2012Journal of Integrative Plant Biology2012,54,6:14
Impact of mechanical circulatory support and immunomodulation therapy on outcome of patients with fulminant myocarditis: Chinese registry of fulminant myocarditis显示文摘Dear Editor,Patients presenting with acute myocarditis and sudden hemodynamic instability (termed fulminant myocarditis [FM]) still have a high mortality and need for heart transplantation, up to 28% at 60 days.1,2,3 Recent scientific statements and expert opinion consensus suggests early use of temporary mechanical circulatory supports (t-MCS).3,4 Specifically, Chinese scientific statement proposed an extensive use of t-MCS combined with immunoregulatory therapy (IT),4 although formal trials are lacking. We present a multicenter, retrospective study to compare the outcome of patients who were treated with t-MCS and IT vs. patients who didn’t receive these treatments. We included patients with the diagnosis of FM based on the presence of viral prodromal signs/symptoms followed by acute onset of severe heart failure (HF) without other relevant differential diagnosis or pre-existing cardiac disorders. Patients who received both t-MCS and IT during hospitalization were classified as t-MCS+IT group.Ning Zhou Yuhua Zhao Jiangang Jiang Lan Shen Junming Li Jing Wan Xueping Ma Jing Zhang Enrico Ammirati Dao Wen Wang 2021Signal Transduction and Targeted Therapy2021,6,11:14

Direct technetium-99m labeling of anti-hepatoma monoclonal antibody fragment:a radioimmunoconjugate for hepatocellular carcinoma imaging显示文摘AIM To directly radiolabel an anti-hepatomamAb fragment HAb18 F(ab’)2 with 99mTc bystannous-reduced method, and assess thestability, biodistribution and radioimmun-oimaging(RⅡ).METHODS Immunoreactive fraction wasdetermined according to Lindmo’s method.Ellman’s reagent was used to determine thenumber of thiols in the reduced F(ab’)2.Labelingefficiency and homogeneity were measured bypaper chromatography, sodium dodecylsulphatepolyacrylamide gel electrophoresis(SDS-PAGE)and autoradiography.Challenge assay involvedthe incubation of aliquots of labeled antibody inethylenediaminetetraacetate( EDTA )and L-cysteine(L-cys)solutions with different molarratio at 37℃ for 1h,respectively.Investigationsin vivo utilized nude mice bearing humanhepatocellular carcinoma(HHCC)xenograftswith gamma camera imaging and tissuebiodistribution studies at regular intervals.RESULTS The labeling procedure was finishedwithin 1.5 h compared with the'pretinning'method which would take at least 21h.In vitrostudies demonstrated that the radiolabeled mAbfragment was homogeneous and retained itsimmunoreactivity.Challenge studies indicatedthat 99mTc-labeled HAb18 F(ab’)2 in EDTA is morestable than in L-cys.Imaging and biodistribution showed a significant tumor uptake at 24 h post-injection of 99mTc-labeled HAb18 F(ab’)2.Theblood,kidney,liver and tumor uptakes at 24hwere 0.56±0.09,56.45±11.36,1.43±0.27 and6.57±3.01(%ID/g),respectively.CONCLUSION 99mTc-HAb18 F(ab’)2 conjugateprepared by this direct method appears to be aneffective way to detect hepatoma in nude micemodel.Hui Jie Bian Zhi Nan Chen Jing Lan Deng 2000World Journal of Gastroenterology2000,6,3:13
Glycyrrhizic acid attenuates CCl_4-induced hepatocyte apoptosis in rats via a p53-mediated pathway显示文摘AIM: To investigate the effect of glycyrrhizic acid (GA) on carbon tetrachloride (CCl4)-induced hepatocyte apo-ptosis in rats via a p53-dependent mitochondrial path-way. METHODS: Forty-five male Sprague-Dawley rats were randomly and equally divided into three groups, the control group, the CCl4 group, and the GA treatment group. To induce liver fibrosis in this model, rats were given a subcutaneous injection of a 40% solution of CCl4 in olive oil at a dose of 0.3 mL/100 g body weight biweekly for 8 wk, while controls received the same isovolumetric dose of olive oil by hypodermic injection, with an initial double-dose injection. In the GA group, rats were also treated with a 40% solution of CCl4 plus 0.2% GA solution in double distilled water by the intraperitoneal injection of 3 mL per rat three times a week from the first week following previously published methods, with modifications. Controls were given the same isovolumetric dose of double distilled water. Liver function parameters, such as alanine aminotransferase (ALT) and aspartate aminotransferase (AST) were de-termined. Pathologic changes in the liver were detected by hematoxylin and eosin staining. Collagen fibers were evaluated by Sirius red staining. Hepatocyte apoptosis was investigated using the terminal deoxynucleotidyl transferase-mediated deoxyuridine 5-triphosphate nick end labeling (TUNEL) assay and the cleaved caspase-3 immunohistochemistry assay. The expression levels of p53 and apoptosis-related proteins were evaluated by immunohistochemistry or Western blotting analysis. RESULTS: After 8 wk of treatment, GA significantly re-duced serum activity of ALT (from 526.7 ± 57.2 to 342 ± 44.8, P<0.05) and AST (from 640 ± 33.7 to 462.8 ± 30.6, P<0.05), attenuated the changes in liver his-topathology and reduced the staging score (from 3.53 ± 0.74 to 3.00 ± 0.76, P<0.05) in CCl4 -treated rats. GA markedly reduced the positive area of Sirius red and the ratio of the hepatic fibrotic region (from 7.87% ± 0.66% to 3.68% ± 0.32%, P<0.05) compared with the CCl4 group. GA also decreased the expression level of cleaved caspase-3 compared to the CCl4 group. TU-NEL assay indicated that GA significantly diminished the number of TUNEL-positive cells compared with the CCl4 group (P<0.05). GA treatment clearly decreased the level of p53 (P<0.05) detected by immunohis-tochemistry and Western blotting analysis. Compared with the CCl4 group, we also found that GA reduced the Bax/Bcl-2 ratio (P<0.05), the expression of cleaved caspase-3 (P<0.05), cleaved caspase-9 (P<0.05), and inhibited cytochrome C and second mitochondria-derived activator of caspases (Smac) release from mito-chondria to cytoplasm, i.e. , GA reduced the expressionlevel of Smac, which inhibited c-IAP1 activity (P<0.05), ultimately inhibiting the activity of caspase-3, according to Western blotting analysis. As a result, GA suppressed activation of the caspase cascades and prevented he-patocyte apoptosis.Xiao-Ling Guo Bo Liang Xue-Wei Wang Fu-Gang Fan Jing Jin Rui Lan Jing-Hui Yang Xiao-Chun Wang Lei Jin Qin Cao 2013World Journal of Gastroenterology2013,19,24:12

Reproductive management through integration of PGD and MPS-based noninvasive prenatal screening/diagnosis for a family with GJB2-associated hearing impairment显示文摘A couple with a proband child of GJB2(encoding the gap junction protein connexin 26)-associated hearing impairment and a previous pregnancy miscarriage sought for a reproductive solution to bear a healthy child. Our study aimed to develop a customized preconception-to-neonate care trajectory to fulfill this clinical demand by integrating preimplantation genetic diagnosis(PGD), noninvasive prenatal testing(NIPT), and noninvasive prenatal diagnosis(NIPD) into the strategy. Auditory and genetic diagnosis of the proband child was carried out to identify the disease causative mutations. The couple then received in-vitro-fertilization treatment, and eight embryos were obtained for day 5 biopsy. PGD was performed by short-tandem-repeat linkage analysis and Sanger sequencing of GJB2 gene. Transfer of a GJB2 c.235del C heterozygous embryo resulted in a singleton pregnancy. At the 13 th week of gestation, genomic DNA(g DNA) from the trio family and cell-free DNA(cf DNA) from maternal plasma were obtained for assessment of fetal chromosomal aneuploidy and GJB2 mutations. NIPT and NIPD showed the absence of chromosomal aneuploidy and GJB2-associated disease in the fetus, which was later confirmed by invasive procedures and postnatal genetic/auditory diagnosis. This strategy successfully prevented the transmission of hearing impairment in the newborn, thus providing a valuable experience in reproductive management of similar cases and potentially other monogenic disorders.XIONG WenPing WANG DaYong GAO Yuan GAO Ya WANG HongYang GUAN Jing LAN Lan YAN JunHao ZONG Liang YUAN Yuan DONG Wei HUANG SeXin WU KeLiang WANG YaoShen WANG ZhiLi PENG HongMei LU YanPing XIE LinYi ZHAO Cui WANG Li ZHANG QiuJing GAO Yun LI Na YANG Ju YIN ZiFang HAN Bing WANG Wei CHEN Zi-Jiang WANG QiuJu 2015Science China(Life Sciences)2015,58,9:12
Antibacterial mechanism of Ginkgo biloba leaf extract when applied to Shewanella putrefaciens and Saprophytic staphylococcus显示文摘The antimicrobial mechanism of Ginkgo biloba leaf extracts(GBLE)when applied to predominant spoilage bacteria(Shewanella putrefaciens and Saprophytic staphylococcus)on refrigerated pomfret and minimal inhibitory concentrations(MICs)were measured by the plate counting method.GBLE at MIC and 2MIC were prepared in tryptic soy broth(TSB)medium and equivalent amounts of sterile distilled water were used in place of GBLE as a control group. The impact of GBLE on the growth of bacteria, the permeability of cell membrane, and cell wall were also investigated by growth curve of bacteria, alkaline phosphates activity(AKP),and electrical conductivity.A scanning electron microscope(SEM)was used to study the effects of GBLE on the cellular structure of S.putrefaciens and S.staphylococcus.The results showed that the MICs of GBLE when applied to S.putrefaciens and S.staphylococcus were 100 mg/mL,the inhibitory rates of MIC and 2MIC concentrations of GBLE when applied to S.putrefaciens were 36.11%and 100%,while 27.78%and 62.22%for S.staphylococcus.Meanwhile,GBLE inhibited the growth of S.putrefaciens and S.staphylococcus until the number of cells at 2MIC values decreased to 0 and 4.29 log CFU/mL,respectively,after 24 h.The electrical conductivity of bacteria increased with GBLE treatment, which was followed by an increased leakage of AKP.The SEM revealed that the structure of bacterial cells was destroyed and the bacteria began to be adhere to each other. The inhibition effect of GBLE when applied to S.putrefaciens and S.staphylococcus was related to the damage of cell membrane and cell wall.It was also revealed that GBLE damages the morphology of bacteria and had stronger effects on the cell membrane of S.putrefaciens than that of S.staphylococcus.Nannan Zhang Weiqing Lan Qian Wang Xiaohong Sun Jing Xie 2018Aquaculture and Fisheries2018,3,4:11
Influences of Technological Parameters on Smelting-separation Process for Metallized Pellets of Vanadium-bearing Titanomagnetite Concentrates显示文摘The smelting-separation process for metallized pellets of vanadium-bearing titanomagnetite concentrates was studied. The influences of smelting temperature, smelting time, and the basicity of the metallized pellet on vanadium and iron recovery were investigated. The characteristics of titanium slag were analyzed using X-ray diffraction, energy dispersive spectroscopy, and mineralographic microscopic analysis. The results demonstrate that appropriate increases in smelting temperature and smelting time can improve the vanadium and iron recovery from metallized pellets and are beneficial for the slag-iron separation.Although increasing the basicity of the metallized pellet can considerably improve the vanadium and iron recovery, the TiO_2 grade of titanium slag was decreased.Under the optimal conditions,90.17% of vanadium and 92.98% of iron in the metallized pellet were recovered, and the TiO_2 grade of titanium slag was 55.01%.It was found that anosovite, augite, spinel, glassiness, and metallic iron were the main mineral phases of the titanium slag.En-hui WU Rong ZHU Shao-li YANG Lan MA Jun LI Jing HOU 2016Journal of Iron and Steel Research(International)2016,23,7:11
