| 2 | Effects of Wen Dan Tang on insomnia-related anxiety and levels of the brain-gut peptide Ghrelin显示文摘Ghrelin, a brain-gut peptide that induces anxiety and other abnormal emotions, contributes to the effects of insomnia on emotional behavior. In contrast, the traditional Chinese Medicine remedy Wen Dan Tang reduces insomnia-related anxiety, which may perhaps correspond to changes in the brain-gut axis. This suggests a possible relationship between Wen Dan Tang's pharmacological mechanism and the brain-gut axis. Based on this hypothesis, a sleep-deprived rat model was induced and Wen Dan Tang was administered using oral gavage during model establishment. Wen Dan Tang significantly reduced insomnia-related anxiety and prevented Ghrelin level decreases following sleep deprivation, especially in the hypothalamus. | 4 | Effects of Xiangsha Liujunzi decoction drug serum on gastric antrum smooth muscle cells from rats with functional dyspepsia by regulating gastrointestinal hormones显示文摘Objective:To observe the effect and mechanism of Xiangsha Liujunzi decoction(XSLJZD)drug serum on gastric antrum smooth muscle cells(SMCs)in rats with functional dyspepsia(FD).Methods:Gastric antrum SMCs from rats with FD were isolated,cultured,and then divided into six groups as follows:control,model,domperidone,low-dose XSLJZD(LXSLJZD),medium-dose XSLJZD(MXSLJZD),and high-dose XSLJZD(HXSLJZD).Each group was administered the corresponding drug serum for intervention.Drug serum intervention conditions and proliferative activity of SMCs were tested by cholecystokinin octapeptide.Ghrelin,gastrin,somatostatin,and substance P(SP)levels were measured by ELISA.Somatostatin and SP mRNA expression was measured by real-time PCR.Results:A concentration of 10%drug serum for 24 h was decided to be the best intervention condition for later study.The mean optical density value in the model group was lower than that in the control group(P紏.001).Optical density values in the domperidone and HXSLJZD groups were higher than those in the model group(P?.025,P?.032,respectively).Gastrin,SP,and ghrelin levels in the model group were lower(P?.007,P?.037,P?.005,respectively),but somatostatin levels were higher,compared with those in the control group(P?.031).Gastrin,SP,and ghrelin levels in the domperidone,MXSLJZD,and HXSLJZD groups were higher than those in the model group(all P<.05).Somatostatin levels in the four drug-treated groups were lower than those in the model group(P?.002,P?.007,P?.001,P?.009,respectively).SP mRNA levels in the model group were lower than those in the control,domperidone,MXSLJZD,and HXSLJZD groups(P?.037 P?.016,P?.025,P?.002,respectively).Somatostatin mRNA levels in the model group were higher than those in the control and MXSLJZD groups(P紏.042,P紏.035).Conclusions:XSLJZD and domperidone drug serum effectively promote proliferative activity of gastric antrum SMCs in an FD model.The mechanism of this activity may be regulated by gastrointestinal hormones. | Ning Dai Jinghong Hu Yan Liu Dongyu Ge Ruijuan Dong Fengzhi Wu Jiaojiao Yu Mindan Chen Deborah CMa Feng Li | 2020 | Journal of Traditional Chinese Medical Sciences2020,7,1: | 6 |
| 5 | Development of the triazole-fused pyrimidine derivatives as highly potent and reversible inhibitors of histone lysine specific demethylase1(LSD1/KDM1A)显示文摘Histone lysine specific demethylase 1(LSD1) has been recognized as an important modulator in post-translational process in epigenetics. Dysregulation of LSD1 has been implicated in the development of various cancers. Herein, we report the discovery of the hit compound 8 a(IC50=3.93 μmol/L) and further medicinal chemistry efforts, leading to the generation of compound 15 u(IC50=49 nmol/L, and Ki= 16 nmol/L), which inhibited LSD1 reversibly and competitively with H3 K4 me2, and was selective to LSD1 over MAO-A/B. Docking studies were performed to rationalize the potency ofcompound 15 u. Compound 15 u also showed strong antiproliferative activity against four leukemia cell lines(OCL-AML3, K562, THP-1 and U937) as well as the lymphoma cell line Raji with the IC50 values of 1.79, 1.30, 0.45, 1.22 and 1.40 μmol/L, respectively. In THP-1 cell line, 15 u significantly inhibited colony formation and caused remarkable morphological changes. | 7 | Effects of tanshinone Ⅱ sodium sulfonate plus cinepazide maleate on the hemorrheologic indexes and blood lipids in patients with acute cerebral infarction显示文摘BACKGROUND:The severity of cerebral infarction is associated with the increase of blood viscosity caused by hyperfibrinogenemia and hyperlipidemia,etc.Thus it has become one of the target for treating cerebral infarction to decrease blood viscosity by integrated Chinese and western medicine.OBJECTIVE:To investigate the influence and clinical therapeutic effects of cinepazide maleate combined with tanshinone Ⅱ A sodium sulfonate on the hemorrheologic indexes and blood lipids of patients with acute cerebral infarction,and compare the results with those of simple cinepazide maleate treatment.DESIGN:A non-randomized case-controlled observation.SETTINGS:Hebei North University;the Second Affiliated Hospitals of Hebei North University;the Third Affiliated Hospitals of Hebei North University.PARTICIPANTS:Eighty-six inpatients with cerebral infarction were selected from the infirmary,the Second and Third Affiliated Hospitals of Hebei North University from September 2004 to October 2006.They were all diagnosed to have acute cerebral infarction by CT or MRI,and accorded with the diagnostic standards for acute cerebral infarction set by the Fourth National Academic Meeting for Cerebrovascular Disease in 1995.Meanwhile,40 teachers and medical staff of voluntary physical examinees were selected as the control group.Informed contents were obtained from all the patients and their relatives.METHODS:The patients were divided into combined treatment group(n=43)and simple treatment group(n=43).In the combined treatment group,the patients were administrated with 160 mg cinepazide maleate injection(Beijing Four-ring Pharmaceutical,Co.,Ltd,No.H200220125;80 mg/2 mL)added in 5% glucose,and 40 mg tanshinone Ⅱ sodium sulfonate(Shanghai No.1 Biochemical & Pharmaceutical Co.,Ltd.,No.H31022558,10 mg/2 mL)added in 250 mL normal saline.In the simple treatment group,the patients were only administrated with cinepazide maleate 320 mg added in 5% glucose or 250 mL normal saline.They were treated for 1 or 2 courses,once a day,and 14 days as a course.The patients were detected before treatment and at 14 and 28 days after treatment respectively.① Determination of hemorrheologic indexes:Whole blood viscosity was determined with LBY-N6B automatic hemorrheologic meter;Plasma viscosity with LBY-F200B automatic plasma viscosity meter;Volume of fibrinogen was determined by the method of 12.5% sodium nitrate depositing biuret reaction.② Determination of blood lipids:The serum levels of total cholesterol(TC),triglyceride(TG),low density lipoprotein cholesterol(LDL-C)and high density lipoprotein cholesterol(HDL-C)were determined.③ Severity of neurological deficit:The total score of neurological deficit score(NDS)ranged from 0 to 45 points,0-15 points was taken as mild,16-30 points as moderate and 31-45 points as severe.④ Evaluation of curative effects:Generally cured:NDS decreased by 91%-100%,and disabled severity of grade 0;Significantly improved:NDS decreased by 46%-90%,and disabled severity of grades 1-3;Improved:NDS decreased by 18%-45%;No change:NDS decreased by less than 18%;Aggravated:NDS increased by more than 18%.Generally cured and significant improved were taken as significant effect.⑤ The adverse events and side effects after medication were observed.MAIN OUTCOME MEASURES:① Results of hemorrheologic indexes and blood lipids;② NDS results in the combined treatment group and simple treatment group;③ Therapeutic effects and adverse events.RESULTS:All the 86 patients with cerebral infarction and 40 healthy controls were involved in the analysis of results.① Results of hemorrheologic indexes and blood lipids:The hemorrheologic indexes and blood lipids before treatment were manifested as abnormalities to different extents in both the combined treatment group and simple treatment group;The hemorrheologic indexes after treatment were obviously improved in both groups.But the hemorrheologic indexes were improved more obviously in the combined treatment group as compared with those in the simple treatment group(P < 0.05);The levels of TC,TG and LDL-Cafter treatment in the combined treatment group were obviously lowered(P < 0.05),whereas those in the simple treatment group were not significantly changed(P > 0.05).② NDS results:The NDS scores at 14 and 28 days after treatment in the combined treatment group(6.23±2.34),(4.27±1.83)points were obviously lower than those in the simple treatment group(8.76±3.41),(6.65±2.49)points,P < 0.05.③ Therapeutic effects and side effects:The total significant effective rates in the combined treatment group and simple treatment group were 93% and 81% respectively.In the combined treatment group,1 case suffered from palpitation,dizziness and agrypnia.In the simple treatment group,1 case suffered from palpitation,dizziness and agrypnia,1 case had itch of skin.All the above symptoms disappeared gradually after the transfusing speed was adjusted to be slower.No drug withdrawal occurred in the patients due to the adverse events.CONCLUSION:Cinepazide maleate combined with tanshinon can obviously improve the abnormalities of hemorrheologic indexes and blood lipids and nerve function in patients with acute cerebral infarction,and its curative effect is faster than that of simple cinepazide maleate treatment. | Chunliang Li | 9 | Human and climatic drivers of land and water use from 1997 to 2019 in Tarim River basin,China显示文摘Climate and human activities change spatial and temporal distribution of water and land use.The Tarim River,the largest inland river in China,faced a long-term exploitation of land and water over a rapid economic development.We analyzed land and water use from 1997 to 2019 in Tarim River Basin by Landsat images,and data on hydrology,climate,population,economy and PM_(2.5)(air particulate matter≤2.5μm).Agricultural land expanded the fastest(4-11%),followed by natural vegetation(15-16%)and water area(4-5%)with population and economic increase.Air quality(PM_(2.5)μg m^(−3))improved in upper(62-27)and middle(48-17)reaches.The water area in lower increase 5%because of ecological water delivery since 2000.Land use in the lower reach was dominated by agriculture,where the downstream runoff consumption increased by 6.8 times.The average annual air temperature and precipitation gradually increased by 0.5℃and 51 mm in source and 0.9℃and 30 mm in main reaches.The average annual water consumption in upper and middle reaches was 4×10^(9)m^(3),accounting for 87%of input runoff in the main reach.Water consumption in middle reach increased by 33 times in 2009-2017.The industry structure was changing from primary to secondary and tertiary industry.To sum up,implementation of water saving strategies and ecological water delivery restored local ecology.Sustainable strategies should be applied facing industrialization.Furthermore,changing the industry structure and restoring the degraded farmlands to grasslands or forests would keep sustainability of Tarim River Basin. | Wenwen Li Fan Huang Fengzhi Shi Xiaorong Wei Kazem Zamanian Xiaoning Zhao | 2021 | International Soil and Water Conservation Research2021,9,4: | 2 |

| 11 | Interventional effect of scorpion venom from Buthus martensii Karsch in Liaoning on somatic pain显示文摘BACKGROUND: Scorpion venom from Buthus martensii Karsch is a kind of protein toxin secreted by scorpion tail. According to records of Pen-ts’ao Kan-mu, the dried scorpion’s body and tail are used as medicine, which mainly treating rheumatism numbness, pain and convulsion. Some researches consider that scorpion toxin is the main pharmacological active component of the dried scorpion, and it has 4 times stronger analgesic effect on somatic pain than morphine. OBJECTIVE: To observe the interventional effect of scorpion venom of Buthus martensii Karsch analgesic active peptide on somatic pain. DESIGN: A randomized and controlled trial. SETTING: Department of Physiology, Shenyang Medical College. MATERIALS: Totally 80 Wistar rats (provided by Animal Center of Shenyang Medical College), male and female in half, aged 3 to 4 months, weighed 250 to 350 g, were used in this trial. They were randomly divided into 4 groups: experimental group, control group, morphine group and naloxone group, with 20 in each group. Analgesia active peptide from buthus martensii karsch venom (provided by Biochemical Department of Shenyang Pharmaceutical University, 1 mL/dosage), morphine (produced by Shanghai First Pharmaceutical Manufacturing Plant), naloxone (opium acceptor inhibitor, American Sigma Corporation), ATAC-350 data-processing machine (Nihon Kohden Corporation), X-Y recording instrument (Nihon Kohden Corporation). METHODS: The experiment was conducted in the Department of Physiology, Shenyang Medical College from July 2003 to July 2005. ① After all the animals were anesthetized, common peroneal nerve is dissected and ligated at middle part. Posterior nucleus group of thalamus (PO) and the tail nuclear were oriented according to G. Paxino rat brain three-dimensional orientation atlas. Glass micro electrode was inserted into PO as guiding electrode, connected with ATAC-350 data-processing machine and X-Y recording instrument, to record the unit evoked potential of PO, taking the evoked potential discharge of the common peroneal nerve of PO as the somatic pain index.② Single square-wave stimulation of intensity 17-19 volt, wave wide 0.2 ms, time delay 20 ms was exerted on common peroneal nerve. The time interval was 5 minutes. X-Y recording instrument was used to draw the graph. 0.002 mg scorpion venom analgesic active peptide was injected into the rats of the experimental group; 0.002 mg normal saline was injected into the rats of the control group through caudate nucleus; 0.002 mg morphine was injected into the rats of morphine group through caudate nucleus; 1.0 mg/kg naloxone was intraperitoneally into the rats of naloxone group, then 0.002 mg scorpion venom was injected into the rats of control group through caudate nucleus.Changes of evoked potential of PO of 3 groups were observed. ③After experiment, 1 μA direct current was given to the guiding electrode and micro electrode , and it lasted for 5 minutes. Pontamine sky blue was used to label the peak of electrode by electrophoresis. Brain tissue was soaked in formalin for 1 week then sliced into 1.0 to 1.5 mm sections. Electrode was orientated according to blue spots. Compared with G . Parino rat brain three-dimensional orientation atlas, we confirmed if the electrode orientation is consistent with PO orientation of G. Parino rat brain three-dimensional orientation atlas. MAIN OUTCOME MEASURES: Changes of the evoked potential of PO in each group. RESULTS: Totally 80 Wistar rats were enrolled in this experiment. Four rats in the experimental group and two in the morphine group died of overdose of anesthesia, and finally 74 rats entered the stage of result analysis. The inhibitory action time of evoked potential of PO has no statistical difference in between experiment group and morphine group (P > 0.05). The whole inhibitory action time, timeof initiate recovery and time of complete recovery of PO of experiment group were longer than those in the morphine group [(45±0.7),(50±9.2),(65±8.1);(35±7.8),(40±8.9),(50±7.6) min,P < 0.05].The change of evoked potential of PO was not obviously in the control group and naloxone group (P > 0.05), and the above-mentioned 4 indexes were nearly 0. CONCLUSION: Scorpion venom possesses obviously inhibitory effect on somatic pain, and its inhibitory effect is stronger than that of the same dosage and concentration of morphine. | 20 | Correlation of gut microbiota and neurotransmitters in a rat model of post-traumatic stress disorder显示文摘Objective:To determine the effect of gut microbiota on a rat model of post-traumatic stress disorder(PTSD)and explore the correlation of gut microbiota with behavior and neurotransmitters.Methods:We established a single prolonged stress(SPS)model to examine the pathogenesis of PTSD on rat behavior,gut microbiota,and neurotransmitter levels.Rats were separated into control and model groups,and neurotransmitter levels were measured using enzyme linked immunosorbent assay.Then,16 S rRNA sequencing was used to compare the gut microbiota between the control and model groups.Results:Compared with those in the control group,freezing time significantly increased,while number of standing upright,crossing frequency,time spent in the central arena,and total distance traveled were significantly reduced in the model group after exposure to SPS(all P<.05).Meanwhile,serotonin,or 5-hydroxytryptamine,levels in the brain in the model group were significantly lower than those the control group(P紏.0332).In addition,changes were observed in the gut microbiota diversity and relative abundances of bacterial phyla,orders,families,and genera in the model group.Especially,changes in Firmicutes,Bacteroidetes,Cyanobacteria,and Proteobacteria levels were most pronounced after SPS exposure.Correlation analysis showed that the strongest positive correlation was found between Bacteroidaceae and 5-HT(P?.0009).Moreover,RF32 abundance was the most negatively related to 5-HT(P?.0009),crossing frequency(P?.0007),and total distance(P?.0003).Conclusion:Our results suggest that SPS model rats showed differences in behavior,neurotransmitter levels,and gut microbiota with control rats.Moreover,Firmicutes,Bacteroidetes,Cyanobacteria,and Proteobacteria were most relevant to the exhibited fear-like and anxiety-like behaviors and significant serotonin content reduction in SPS model rats. | Qin Zhou Tianshi Sun Fengzhi Wu Feng Li Yan Liu Weihong Li Ning Dai Libo Tan Tenghui Li Yuehan Song | 2020 | Journal of Traditional Chinese Medical Sciences2020,7,4: | 1 |
