1The RALF1-FERONIA Complex Phosphorylates eIF4E1 to Promote Protein Synthesis and Polar Root Hair Growth显示文摘The molecular links between extracellular signals and the regulation of localized protein synthesis in plant cells are poorly understood.Here,we show that in Arabidopsis thaliana,the extracellular peptide RALF1 and its receptor,the FERONIA receptor kinase,promote root hair(RH)tip growth by modulating protein synthesis.We found that RALF1 promotes FERONIA-mediated phosphorylation of elF4E1,a eukaryotic translation initiation factor that plays a crucial role in the control of mRNA translation rate.Phosphorylated elF4E1 increases mRNA affinity and modulates mRNA translation and,thus,protein synthesis.The mRNAs targeted by the RALF1-FERONIA-elF4E1 module include ROP2 and RSL4,which are important regulators of RH cell polarity and growth.RALF1 and FERONIA are expressed in a polar manner in RHs,which facilitate elF4E1 polar丨ocalization and thus may control local f?OP2 translation.Moreover,we demonstrated that high-level accumulation of RSL4 exerts negative-feedback regulation of RALF1 expression by directly binding the RALF1 gene promoter,determining the final RH size.Our study reveals that the link between RALF1-FERONIA signaling and protein synthesis constitutes a novel component regulating cell expansion in these polar growing cells.Sirui Zhu Jose Manuel Estevez Hongdong Liao Yonghua Zhu Tao Yang Chiyu Li Yichuan Wang Lan Li Xuanming Liu Javier Martinez Pacheco Hongwei Guo Feng Yu 2020Molecular Plant2020,13,5:3
2Nematode-Encoded RALF Peptide Mimics Facilitate Parasitism of Plants through the FERONIA Receptor Kinase显示文摘The molecular mechanism by which plants defend against plant root-knot nematodes(RKNs)is largely unknown.The plant receptor kinase FERONIA and its peptide ligands,rapid alkalinization factors(RALFs),regulate plant immune responses and cell expansion,which are two important factors for successful RKN parasitism.In this study,we found that mutation of FERONIA in Arabidopsis thaliana resulted in plants showing low susceptibility to the RKN Meloidogyne incognita.To identify the underlying mechanisms associated with this phenomenon,we identified 18 novel RALF-likes from multiple species of RKNs and showed that two RALF-likes(i.e.,MiRALF1 and MiRALF3)from M.incognita were expressed in the esophageal gland with high expression during the parasitic stages of nematode development.These nematode RALF-likes also possess the typical activities of plant RALFs and can directly bind to the extracellular domain of FERONIA to modulate specific steps of nematode parasitism-related immune responses and cell expansion.Genetically,both MiRALF1/3 and FERONIA are required for RKN parasitism in Arabidopsis and rice.Collectively,our study suggests that nematode-encoded RALFs facilitate parasitism via plant-encoded FERONIA and provides a novel paradigm for studying host-pathogen interactions.Xin Zhang Huan Peng Sirui Zhu Junjie Xing Xin Li Zhaozhong Zhu Jingyuan Zheng Long Wang Bingqian Wang Jia Chen Zhenhua Ming Ke Yao Jinzhuo Jian Sheng Luan Devin Coleman-Derr Hongdong Liao Yousong Peng Deliang Peng Feng Yu 2020Molecular Plant2020,13,10:3
3Biochar can Increase Chinese Cabbage(Brassica oleracea L.)Yield,Decrease Nitrogen and Phosphorus Leaching Losses in Intensive Vegetable Soil显示文摘There are few evidences on the effect of biochar on vegetable yield,nitrogen(N)and phosphorus(P)leaching losses under intensive vegetable production soil.The current field plot scale study evaluated responses of Chinese cabbage(Brassica oleracea L.)yield,N and P leaching losses using five N treatments of common N application rate according to local farmers’practice(N100%),reducing 20%or 40%N fertilizer(N80%and N60%),and reducing 40%N fertilizer but incorporating 10 or 20 t/ha biochar(N60%+BC10 and N60%+BC20).Results showed that N80%and N60%decreased both the cabbage economic and leaf yields by 6.8%-36.3%and 27.4%-37.7%,respectively.Incorporation of biochar with reduced N fertilizer rates improved the cabbage yield,in particular the N60%+BC20 matched the yield that observed in N100%treatment.Enhanced N and P uptake capacities of cabbage shoot probably contributed the higher vegetable production under both biochar amendment schemes.Biochar application mitigated the NH_(4)^(+)-N and total P leaching losses by 20%-30%and 29%-32%,respectively,compared with their counterpart treatment N60%.Nevertheless,biochar exerted no influence on the NO_(3)^(-)-N leaching.In addition,soil organic matter content was recorded with 7.4%-28.7%higher following 10-20 t/ha biochar application.In conclusion,biochar application can increase economic yield of cabbage via increasing N and P use efficiency,decrease N and P leaching losses,and improve soil quality in an intensive vegetable production system.Haijun Sun Paramsothy Jeyakumar Hongdong Xiao Xuewen Li Jiayou Liu Min Yu Prabal Bir Jung Rana Weiming Shi 2022Phyton-International Journal of Experimental Botany2022,91,1:3
4Development and application of tumor-targeting magnetic nanoparticles FA-StNP@Fe_2O_3 for hyperthermia显示文摘Modified magnetic starch nanoparticles (FA-StNP@Fe2O3) were synthesized by conjugating folic acid (FA-PEG-NH2) onto the surface of magnetic starch nanoparticles (StNP@Fe2O3) prepared by reverse microemulsion method. The synthesized FA-StNP@Fe2O3 was investigated by transmission electron microscopy and zeta potential analysis. The average size of its well dispersed particles was 250 nm. The iron concentration of 2 mg/g was detected by phenanthroline method. Placing FA-StNP@Fe2O3 nanoparticles in the alternating magnetic field for 30 min resulted in an increase in the suspension temperature from ambient temperature (37℃) to a value between 42℃ and 43℃. Co-cultured nanoparticles and Hela cell line or normal HUEC-12 cell line, and the biological effects at the cellular level were investigated in the alternating magnetic field using MTT assay, Hochest-PI double staining and flow cytometry analysis. Experimental results showed that FA-StNP@Fe2O3 within acertain concentration range has no obvious effect on cell proliferation. When treated in the magnetic field, apoptosis rate on Hela induced by FA-StNP@Fe2O3 was 13.4%. Prussian blue staining analysis confirmed that the nanoparticles modified with folic acid had improved ability in tumor cell-targeting, and therefore, potential applications in biomedical and magnetocaloric areas. 5Functional modules with disease discrimination abilities for various cancers显示文摘Selecting differentially expressed genes(DEGs) is one of the most important tasks in microarray applications for studying multi-factor diseases including cancers.However,the small samples typically used in current microarray studies may only partially reflect the widely altered gene expressions in complex diseases,which would introduce low reproducibility of gene lists selected by statistical methods.Here,by analyzing seven cancer datasets,we showed that,in each cancer,a wide range of functional modules have altered gene expressions and thus have high disease classification abilities.The results also showed that seven modules are shared across diverse cancers,suggesting hints about the common mechanisms of cancers.Therefore,instead of relying on a few individual genes whose selection is hardly reproducible in current microarray experiments,we may use functional modules as functional signatures to study core mechanisms of cancers and build robust diagnostic classifiers.YAO Chen ZHANG Min ZOU JinFeng LI HongDong WANG Dong ZHU Jing GUO Zheng 2011Science China(Life Sciences)2011,54,2:3
6Coordination engineering of cobalt phthalocyanine by functionalized carbon nanotube for efficient and highly stable carbon dioxide reduction at high current density显示文摘Coordination engineering can enhance the activity and stability of the catalyst in heterogeneous catalysis.However,the axial coordination engineering between different groups on the carbon carrier and molecular catalysts in the electrocatalytic carbon dioxide reduction reaction(CO_(2)RR)has been studied rarely.Through coordination engineering strategy,a series of amino(NH_(2)),hydroxyl(OH),and carboxyl(COOH)groups functionalized carbon nanotubes(CNT)immobilized cobalt phthalocyanine(CoPc)catalysts are designed.Compared with no groups,OH groups and COOH groups,NH_(2)groups can effectively change the coordination environment of the central metal Co,thereby significantly increasing the turnover frequency(TOF)(31.4 s^(-1)at-0.6 V vs.RHE,CoPc/NH_(2)-CNT>CoPc/OH-CNT>CoPc/COOH-CN>CoPc/CNT).In the flow cell,the CoPc/NH_(2)-CNT catalyst has high carbon monoxide(CO)selectivity at high current density(~100%at-225 mA·cm^(-2),~96%at-351 mA·cm^(-2)).Importantly,the CoPc/NH_(2)-CNT catalyst can operate stably for 100 h at 225 mA·cm^(-2).Theoretical calculations reveal that CoPc/NH_(2)-CNT catalyst is beneficial to the formation of^(*)COOH and desorption of^(*)CO,thus promoting CO_(2)RR.This work provides an excellent platform for understanding the effect of coordination engineering on electrocatalytic performance and promotes a way to explore efficient and stable catalysts in other applications.Hongdong Li Yue Pan Zuochao Wang Yaodong Yu Juan Xiong Haoyang Du Jianping Lai Lei Wang Shouhua Feng 2022Nano Research2022,15,4:2
