| 8 | Porcine FcεRI Mediates Porcine Reproductive and Respiratory Syndrome Virus Multiplication and Regulates the Inflammatory Reaction显示文摘Porcine reproductive and respiratory syndrome virus(PRRSV) shows characteristic antibody-dependent enhancement(ADE) of infection and causes porcine systemic inflammation, which is similar to a type I allergic reaction; however, the role of porcine FceεRI in ADE is still unclear. In this study, the expression of different Fc receptors(Fc Rs) on macrophages was investigated in a PRRSV 3D4/21 cell infection model in the presence or absence of PRRSV antibody. The transcription level of Fcc II and FceεRI was significantly up-regulated under PRRSV-antibody complex infection. Internalization and proliferation of PRRSV were promoted by the ADE mechanism when FceεRI was expressed in permissive 3D4/21 cells and the non-permissive cell line HEK 293T. Transcriptome sequencing data showed that the expression levels of AKT,ERK and other signal molecules in the anti-inflammatory pathway were significantly increased, especially in the cells infected with the PRRSV-antibody immune complex. Inflammatory regulatory molecules such as PLA2G6, LOX, TRPM8 and TRPM4 were significantly up-regulated following PRRSV infection but significantly down-regulated in the cells infected with the PRRSV-antibody immune complex. | 9 | Fabrication and application of a graphene polarizer with strong saturable absorption显示文摘By transferring 100 nm gold-coated CVD monolayer graphene onto the well-polished surface of D-shaped fiber, we achieve a graphene in-line polarizer with a high polarization extinction ratio of ~27 d B and low insertion loss of 5 d B at 1550 nm, meanwhile achieving a strong saturable absorption effect of 14%. The manufacture of this graphene in-line polarizer also simplifies the graphene transfer process. To explore the potential applications of the new device, we also demonstrate noise-like pulse generation and supercontinuum spectrum generation. By launching the designed graphene device into a fiber ring laser cavity, 51 nm bandwidth noise-like pulse is obtained. Although several studies have shown that PRRSV can affect the cell cycle of infected cells, it is still unclear how it manipulates the cell cycle to facilitate its proliferation. In this study, we analyzed the mRNA expression profiles of transcription factors in PRRSV-infected 3D4/21 cells by RNA-sequencing. The result shows that the expression of transcription factor DP2(TFDP2) is remarkably upregulated in PRRSV-infected cells. Further studies show that TFDP2 contributes to PRRSV proliferation and the PRRSV nucleocapsid(N) protein induces TFDP2 expression by activating C/EBPb. TFDP2 positively regulates cyclin A expression and triggers a less proportion of cells in the S phase, which contributes to PRRSV proliferation. This study proposes a novel mechanism by which PRRSV utilizes host protein to regulate the cell cycle to favor its infection. | 14 | CD49a^(+)CD49b^(+) NK cells induced by viral infection reflect an activated state of conventional NK cells显示文摘Natural killer(NK) cells are important innate effectors that play a pivotal role in the defense against tumors and infections and participate in regulating adaptive immunity. Recent studies have revealed phenotypic and functional heterogeneity of NK cells.Here, using murine models of acute and chronic lymphocytic choriomeningitis virus infection, we observed that a CD49a^(+)CD49b^(+) NK cell subset emerged in the liver and other tissues, and underwent vigorous expansion following viral infection,before progressively decreasing in cell number. These viral infection-induced CD49a^(+)CD49b^(+) NK cells displayed an activated and mature phenotype. Moreover, compared with liver-resident NK cells and conventional NK(cNK) cells, CD49a^(+)CD49b^(+) NK cells showed increased functional competence, as evidenced by higher amounts of IFN-γ production and stronger cytotoxic capabilities during viral infection. Generation of these CD49a^(+)CD49b^(+) NK cells was shown to be independent of the T-bet transcription factor. Adoptive transfer experiments revealed that c NK cells could convert into CD49a^(+)CD49b^(+) NK cells following viral infection. In this paper, a low-complexity multiple signal representation (MSR) scheme is proposed to control the PAPR problem in downlink OFDM-CDMA systems. The proposed scheme generates multiple candidate signals by a novel user grouping scheme, which is without distortion and can provide more PAPR reduction than the conventional MSR schemes, such as partial transmit sequence (PTS) and selective mapping (SLM). Furthermore, a low-complexity processing structure is developed using a novel joint spreading and inverse fast Fourier transform (S-IFFT) to simplify the generation of multiple candidate signals. | 20 | Growing impacts of low-flow events on vegetation dynamics in hydrologically connected wetlands downstream Yangtze River Basin after the operation of the Three Gorges Dam显示文摘Wetland vegetation is intimately related to floodplain inundations,which can be seriously affected by dam operation.Poyang Lake is the largest floodplain wetland in China and naturally connected with the Yangtze River and the Three Gorges Dam(TGD)upstream.To understand the potential impacts of TGD on Poyang Lake wetlands,we collected remote sensing imagery acquired during dry season from 1987 to 2020 and extracted vegetation coverage data in the Ganjiang Northern-branch Delta(GND)and the Ganjiang Southern-branch Delta(GSD),using the Object-oriented Artificial Neural Network Regression.Principal components analysis,correlation analysis,and the random forest model were used to explore the interactions between vegetation extent in the two deltas and 33 hydrological variables regarding magnitude,duration,timing,and variation.The implementation of the TGD advanced and extended the low-flow periods in Poyang Lake.Vegetation coverage in the GND and GSD increased at the rates of 0.39 and 0.22 km2/year,respectively.The reservoir storage at the end of September accelerated the runoff recession in the GND and the GSD,making low-flow events more influential for vegetation dynamics and shortening the response time of vegetation to the water regime.This study provides an important reference for evaluating the impacts of dam engineering on downstream wetlands. | WANG Dandan LIU Ying ZHENG Lilin LI Dahui | 2023 | Journal of Geographical Sciences2023,33,4: | 1 |
