1Field Relationships,Geochemistry,Zircon Ages and Evolution of a Late Archaean to Palaeoproterozoic Lower Crustal Section in the Hengshan Terrain of Northern China显示文摘The Hengshan complex forms part of the central zone of the North China Craton and consists predominantly of ductilely-deformed late Archaean to Palaeoproterozoic high-grade, partly migmatitic, granitoid orthogneisses, intruded by mafic dykes of gabbroic composition. Many highly strained rocks were previously misinterpreted as supracrustal sequences and represent mylonitized granitoids and sheared dykes. Our single zircon dating documents magmatic granitoid emplacement ages between 2.52 Ga and 2.48 Ga, with rare occurrences of 2.7 Ga gneisses, possibly reflecting an older basement. A few granitic gneisses have emplacement ages between 2.35 and 2.1 Ga and show the same structural features as the older rocks, indicating that the main deformation occurred after ~2.1 Ga. Intrusion of gabbroic dykes occurred at~1920 Ma, and all Hengshan rocks underwent granulite-facies metamorphism at 1.88-1.85 Ga, followed by retrogression, shearing and uplift.We interpret the Hengshan and adjacent Fuping granitoid gneisses as the lower, plutonic, part of a late Archaean to early Palaeoproterozoic Japan-type magmatic arc, with the upper, volcanic part represented by the nearby Wutai complex. Components of this arc may have evolved at a continental margin as indicated by the 2.7 Ga zircons. Major deformation and HP North China Craton collided to form the Trans-North China orogen. Shear zones in the Hengshan are interpreted as major lower crustal discontinuities post-dating the peak of HP metamorphism, and we suggest that they formed during orogenic collapse and uplift of the Hengshan complex in the late Palaeoproterozoic (<1.85 Ga).A. 6Improvement on Mixograph test through water addition and parameter conversions显示文摘To improve Mixograph testing effect, Farinograph measurements were adopted as a quality standard and changes in water absorption and parameter conversion in Mixograph test were explored. Comparative study showed that increasing water absorption to about 73% and converting original parameters to compound parameters in Mixograph tests significantly increased their predictive power for flour quality. These efforts also enabled the adoption of fixed water addition level in Mixograph test and simplified the test procedure significantly. With the success in parameter conversions, Mixograph test results were successfully described by Farinograph parameters, which allow breeders to compare and exchange test results easily. 9LRP6 in mesenchymal stem cells is required for bone formation during bone growth and bone remodeling显示文摘Lipoprotein receptor-related protein 6(LRP6) plays a critical role in skeletal development and homeostasis in adults. However, the role of LRP6 in mesenchymal stem cells(MSCs), skeletal stem cells that give rise to osteoblastic lineage, is unknown. In this study, we generated mice lacking LRP6 expression specifically in nestin1MSCs by crossing nestin-Cre mice with LRP6floxmice and investigated the functional changes of bone marrow MSCs and skeletal alterations. Mice with LRP6 deletion in nestin1cells demonstrated reductions in body weight and body length at 1 and 3 months of age. Bone architecture measured by microCT(mCT) showed a significant reduction in bone mass in both trabecular and cortical bone of homozygous and heterozygous LRP6mutant mice. A dramatic reduction in the numbers of osteoblasts but much less significant reduction in the numbers of osteoclasts was observed in the mutant mice. Osterix1osteoprogenitors and osteocalcin1osteoblasts significantly reduced at the secondary spongiosa area, but only moderately decreased at the primary spongiosa area in mutant mice. Bone marrow MSCs from the mutant mice showed decreased colony forming, cell viability and cell proliferation. 11Effects of serum containing natural cerebrolysin on glucose-regulated protein 78 and CCAAT enhancer-binding protein homologous protein expression in neuronal PC12 cells following tunicamycin-induced endoplasmic reticulum stress显示文摘BACKGROUND:Glucose-regulated protein 78(GRP78),a marker of endoplasmic reticulum stress,can prolong cell survival.Alternatively,CCAAT enhancer-binding protein homologous protein(CHOP),a transcription factor specific for endoplasmic reticulum stress,can cause cell cycle arrest and cell apoptosis. OBJECTIVE:To study the protective effects of serum containing natural cerebrolysin on endoplasmic reticulum stress in tunicamycin-induced neuronal PC12 cells,and analyze the influence on GRP78 and CHOP expressions. DESIGN,TIME AND SETTING:A parallel controlled study was performed at the Institute of Integrated Western and Traditional Chinese Medicine,Shenzhen Hospital,Southern Medical University,between March 2006 and August 2008. MATERIALS:Adult Sprague-Dawley rats were perfused with natural Cerebrolysin aqueous extract(0.185 g/kg/d) to produce serum containing natural Cerebrolysin.Physiological saline was used to produce blank serum.PC12 cell line was provided by Shanghai Institute of Cell Biology, Chinese Academy of Science.Tunicamycin was provided by Sigma(St.Louis,USA),and natural Cerebrolysin,containing ginseng,rhizoma gastrodiae,and gingko leaf(1:2:2),by Shengzhen Institute of Integrated Western and Traditional Chinese Medicine. METHODS:PC12 cells were treated with DMEM culture media containing 10%blank serum (normal control group),tunicamycin(1μg/mL;model group),and 5%,10%,and 15%serum containing natural cerebrolysin and tunicamycin(1μg/mL;low-,moderate-,and high-dose serum containing natural cerebrolysin groups),for 2 hours. MAIN OUTCOME MEASURES:PC12 cells were treated with tunicamycin for 48 hours after which apoptosis was measured using the TUNEL method to calculate apoptotic index.GRP78 expression was detected using immunocytochemistry.After 24 hours of treatment with tunicamycin,GRP78 and CHOP mRNA expressions were measured using RT-PCR. RESULTS:The apoptotic index and CHOP mRNA expression were in the model group and three cerebrolysin groups were significantly increased when compared to the normal control group(P<0.05).In contrast,GRP78 mRNA and protein expressions were significantly decreased(P<0.05). CONCLUSION:Serum containing natural cerebrolysin significantly reduced apoptosis in neuronal PC12 cells following tunicamycin-induced endoplasmic reticulum stress.These results may be related to an up-regulation of GRP78 expression and down-regulation of CHOP expression,both of which displayed dose-dependent effects.Zhengzhi Wu Ming Li Andrew C.J. 15Dysfunctional stem and progenitor cells impair fracture healing with age显示文摘Successful fracture healing requires the simultaneous regeneration of both the bone and vasculature;mesenchymal stem cells (MSCs) are directed to replace the bone tissue, while endothelial progenitor cells (EPCs) form the new vasculature that supplies blood to the fracture site. In the elderly, the healing process is slowed, partly due to decreased regenerative function of these stem and progenitor cells. MSCs from older individuals are impaired with regard to cell number, proliferative capacity, ability to migrate, and osteochondrogenic differentiation potential. The proliferation, migration and function of EPCs are also compromised with advanced age. Although the reasons for cellular dysfunction with age are complex and multidimensional, reduced expression of growth factors, accumulation of oxidative damage from reactive oxygen species, and altered signaling of the Sirtuin-1 pathway are contributing factors to aging at the cellular level of both MSCs and EPCs. Because of these geriatric-specific issues, effective treatment for fracture repair may require new therapeutic techniques to restore cellular function. Some suggested directions for potential treatments include cellular therapies, pharmacological agents, treatments targeting age-related molecular mechanisms, and physical therapeutics. 16Prohibitin(PHB)interacts with AKT in mitochondria to coordinately modulate sperm motility显示文摘Prohibitin(PHB),an evoluti on arily con served mitochondrial inner membra ne protein,is highly expressed in cells that require strong mitoch on drial function.Recently,we dem on strated that the deleti on of Phb in spermatocytes results in impaired mitochondrial function.In addition,PHB expression in the mitochondrial sheath of human sperm has a significantly negative correlation with mitochondrial reactive oxygen species levels,but a positive one with mitochondrial membrane potential and sperm motility.These results suggest that mitochondrial PHB expression plays a role in sperm motility.However,the mechanism of PHB-mediated regulation of sperm motility remai ns unk nown.Here,we dem on strate for the first time that PHB interacts with protei n kinase B(AKT)and exists in a complex with phospho-PHB(pT258)and phospho-AKT in the mitochondrial sheath of murine sperm,as determined using colocalization and coimmunoprecipitation assays.After blocking AKT activity using wortmannin(a phosphatidylinositol 3-kinase[PI3K]inhibitor),murine sperm have significantly(P<0.05)decreased levels of phospho-PHB(pT258)and the total and progressive motility.Furthermore,significantly(P<0.05)lower levels of phospho-PI3K P85 subunit a+γ(pY199 and pY46)and phospho-AKT(pS473;pT308)are found in sperm from infertile asthenospermic and oligoasthenospermic men compared with no rmospermic subjects,which suggest a reduced activity of the PI3K/AKT pathway in these in fertile subjects.Importantly,these sperm from infertile subjects also have a significantly(P<0.05)lower level of phospho-PHB(pT258).Collectively,our findings suggest that the interaction of PHB with AKT in the mitochondrial sheath is critical for sperm motility,where PHB phosphorylation(pT258)level and PI3K/AKT activity are key regulatory factors.Xiao-Hui Li Ran-Ran Chai Guo-Wu Chen Ling-Fei Zhang Wen-Jing Tan-Tai Hui-Juan Shi Patricia A Martin-DeLeon Wai-Sum O Hong Chen 2020Asian Journal of Andrology2020,22,6:3

18Safety of endovascular therapy for symptomatic intracranial artery stenosis:a national prospective registry显示文摘Introduction The safety outcomes of endovascular therapy for intracranial artery stenosis in a real-world stetting are largely unknown.The Clinical Registration Trial of Intracranial Stenting for Patients with Symptomatic Intracranial Artery Stenosis(CRTICAS)was a prospective,multicentre,real-world registry designed to assess these outcomes and the impact of centre experience.Methods 1140 severe,symptomatic intracranial arterial stenosis(ICAS)patients treated with endovascular therapy were included from 26 centres,further divided into three groups according to the annual centre volume of intracranial angioplasty and stent placement procedures over 2 years:(1)high volume for≥25 cases/year;(2)moderate volume for 10–25 cases/year and(3)low volume for<10 cases/year.Results The rate of 30-day stroke,transient ischaemic attack or death was 9.7%(111),with 5.4%,21.1%and 9.7%in high-volume,moderate-volume and low-volume centres,respectively(p<0.05).Multivariable logistic regression confirmed high-volume centres had a significantly lower primary endpoint compared with moderate-volume centres(OR=0.187,95%CI:0.056 to 0.627;p≤0.0001),while moderate-volume and low-volume centres showed no significant difference(p=0.8456).Conclusion Compared with the preceding randomised controlled trials,this real-world,prospective,multicentre registry shows a lower complication rate of endovascular treatment for symptomatic ICAS.Non-uniform utilisation in endovascular technology,institutional experience and patient selection in different volumes of centres may have an impact on overall safety of this treatment.Yabing Wang Tao Wang Adam Andrew Dmytriw Kun Yang Liqun Jiao Huaizhang Shi Jie Lu Tianxiao Li Yujie Huang Zhenwei Zhao Wei Wu Jieqing Wan Qinjian Sun Bo Hong Yongli Li Liyong Zhang Jianfeng Chu Qiong Cheng Yiling Cai Pengfei Wang Qi Luo Hua Yang Baijing Dong Yang Zhang Jun Zhao Zuoquan Chen Wei Li Xiaoxin Bai Weiwen He Xueli Cai Maimai Ti Osama O Zaidat 2022Stroke & Vascular Neurology2022,7,2:3
