1Erianin,a novel dibenzyl compound in Dendrobium extract,inhibits lung cancer cell growth and migration via calcium/calmodulin-dependent ferroptosis显示文摘Ferroptosis,a novel form of programmed cell death,is characterized by iron-dependent lipid peroxidation and has been shown to be involved in multiple diseases,including cancer.Stimulating ferroptosis in cancer cells may be a potential strategy for cancer therapy.Therefore,ferroptosis-inducing drugs are attracting more attention for cancer treatment.Here,we showed that erianin,a natural product isolated from Dendrobium chrysotoxum Lindl,exerted its anticancer activity by inducing cell death and inhibiting cell migration in lung cancer cells.Subsequently,we demonstrated for the first time that erianin induced ferroptotic cell death in lung cancer cells,which was accompanied by ROS accumulation,lipid peroxidation,and GSH depletion.The ferroptosis inhibitors Fer-1 and Lip-1 but not Z-VAD-FMK,CQ,or necrostatin-1 rescued erianin-induced cell death,indicating that ferroptosis contributed to erianin-induced cell death.Furthermore,we demonstrated that Ca^(2+)/CaM signaling was a critical mediator of erianin-induced ferroptosis and that blockade of this signaling significantly rescued cell death induced by erianin treatment by suppressing ferroptosis.Taken together,our data suggest that the natural product erianin exerts its anticancer effects by inducing Ca^(2+)/CaMdependent ferroptosis and inhibiting cell migration,and erianin will hopefully serve as a prospective compound for lung cancer treatment.Peng Chen Qibiao Wu Jiao Feng Lili Yan Yitian Sun Shuiping Liu Yu Xiang Mingming Zhang Ting Pan Xiaying Chen Ting Duan Lijuan Zhai Bingtao Zhai Wengang Wang Ruonan Zhang Bi Chen Xuemeng Han Yicong Li Liuxi Chen Ying Liu Xingxing Huang Ting Jin Wenzheng Zhang Hong Luo Xiaohui Chen Yongqiang Li Qiujie Li Guohua Li Qin Zhang Lvjia Zhuo Zuyi Yang Huifen Tang Tian Xie Xiaoping Ouyang Xinbing Sui 2020Signal Transduction and Targeted Therapy2020,5,1:42
2Preparation, characterization, pharmacokinetics and anticancer effects of PEGylated β-elemene liposomes显示文摘Objective:This study aimed to develop a new polyethylene glycol(PEG)ylatedβ-elemene liposome(PEG-Lipo-β-E)and evaluate its characterization,pharmacokinetics,antitumor effects and safety in vitro and in vivo.Methods:The liposomes were prepared by ethanol injection and high-pressure micro-jet homogenization.Characterization of the liposomes was conducted,and drug content,entrapment efficiency(EE),in vitro release and stability were studied by ultra-fast liquid chromatography(UFLC)and a liquid surface method.Blood was drawn from rats to establish the pharmacokinetic parameters.The anticancer effect was evaluated in a KU-19-19 bladder cancer xenograft model.Histological analyses were performed to evaluate safety.Results:The PEG-Lipo-β-E showed good stability and was characterized as 83.31±0.181 nm in size,0.279±0.004 in polydispersity index(PDI),-21.4±1.06 mV in zeta potential,6.65±0.02 in pH,5.024±0.107 mg/mL inβ-elemene(β-E)content,and 95.53±1.712%in average EE.The Fourier transform infrared spectroscopy(FTIR)and differential scanning calorimetry(DSC)indicated the formation of PEG-Lipo-β-E.Compared to elemene injection,PEG-Lipo-β-E demonstrated a 1.75-fold decrease in clearance,a 1.62-fold increase in half-life,and a 1.76-fold increase in area under the concentration-time curves(AUCs)from 0 hour to 1.5 hours(P<0.05).PEG-Lipo-β-E also showed an enhanced anticancer effect in vivo.Histological analyses showed that there was no evidence of toxicity to the heart,kidney,liver,lung or spleen.Conclusions:The present study demonstrates PEG-Lipo-β-E as a new formulation with ease of preparation,high EE,good stability,improved bioavailability and antitumor effects.Bingtao Zhai Qibiao Wu Wengang Wang Mingming Zhang Xuemeng Han Qiujie Li Peng Chen Xiaying Chen Xingxing Huang Guohua Li Qin Zhang Ruonan Zhang Yu Xiang Shuiping Liu Ting Duan Jianshu Lou Tian Xie Xinbing Sui 2020Cancer Biology & Medicine2020,17,1:6

4An ATF24 peptide-functionalized β-elemene-nanostructured lipid carrier combined with cisplatin for bladder cancer treatment显示文摘Objective:In this study,we aimed to develop an amino-terminal fragment(ATF)peptide-targeted liposome carryingβ-elemene(ATF24-PEG-Lipo-β-E)for targeted delivery into urokinase plasminogen activator receptor-overexpressing bladder cancer cells combined with cisplatin(DDP)for bladder cancer treatment.Methods:The liposomes were prepared by ethanol injection and high-pressure microjet homogenization.The liposomes were characterized,and the drug content,entrapment efficiency,andin vitro release were studied.The targeting efficiency was investigated using confocal microscopy,ultra-fast liquid chromatography,and an orthotopic bladder cancer model.The effects of ATF24-PEG-Lipo-β-E combined with DDP on cell viability and proliferation were evaluated by a Cell Counting Kit-8(CCK-8)assay,a colony formation assay,and cell apoptosis and cell cycle analyses.The anticancer effects were evaluated in a KU-19-19 bladder cancer xenograft model.Results:ATF24-PEG-Lipo-β-E had small and uniform sizes(~79 nm),high drug loading capacity(~5.24 mg/mL),high entrapment efficiency(98.37±0.95%),and exhibited sustained drug release behavior.ATF24-PEG-Lipo-β-E had better targeting efficiency and higher cytotoxicity than polyethylene glycol(PEG)ylatedβ-elemene liposomes(PEG-Lipo-β-E).DDP,combined with ATF24-PEG-Lipo-β-E,exerted a synergistic effect on cellular apoptosis and cell arrest at the G2/M phase,and these effects were dependent on the caspase-dependent pathway and Cdc25C/Cdc2/cyclin B1 pathways.Furthermore,thein vivo antitumor activity showed that the targeted liposomes effectively inhibited the growth of tumors,using the combined strategy.Conclusions:The present study provided an effective strategy for the targeted delivery ofβ-elemene(β-E)to bladder cancer,and a combined strategy for bladder cancer treatment.Bingtao Zhai Peng Chen Wengang Wang Shuiping Liu Jiao Feng Ting Duan Yu Xiang Ruonan Zhang Mingming Zhang Xuemeng Han Xiaying Chen Qiujie Li Guohua Li Ying Liu Xingxing Huang Wenzheng Zhang Ting Pan Lili Yan Ting Jin Tian Xie Xinbing Sui 2020Cancer Biology & Medicine2020,17,3:5
5Bushen Qiangjin capsule(补肾强筋胶囊) inhibits the Wnt/β-catenin pathway to ameliorate papain-induced knee osteoarthritis in rats显示文摘OBJECTIVE:To evaluate the molecular mechanism underlying the beneficial effect of Bushen Qiangjin capsule(补肾强筋胶囊,BSQJ),a Traditional Chinese Medicine,on knee osteoarthritis(KOA).METHODS:In the present study,32 female Sprague-Dawley rats were randomly divided into four groups:control,KOA,high-dose BSQJ(H-BSQJ),and low-dose BSQJ(L-BSQJ).After successfully establishing the KOA model by intra-articular injection of papain,H-BSQJ and L-BSQJ groups were intragastrically administered 0.243 and 0.122 g/kg BSQJ,respectively,daily for 6 weeks.At the end of the experiment,knee articular cartilage tissues of rats were collected for evaluation by hematoxylin and eosin staining,Safranin O-Fast Green staining,and terminal deoxynucleotidyl transferase-mediated d UTP nick-end labeling assay.Serum interleukin-1βand tumor necrosis factor-αlevels of rats were detected with an enzyme-linked immunosorbent assay method.Gene expression of Wnt-4,β-catenin,Frizzled-2,glycogen synthase kinase-3β(GSK-3β),cysteinyl aspartate-specific proteinases 3 and 9(caspases 3 and 9),collagen typeⅡalpha 1(Col2 a1),and matrix metalloproteinases 1 and 13(MMP-1 and MMP-3)of rat knee articular cartilage was quantified by reverse transcription-quantitative polymerase chain reaction analysis.Wnt-4,β-catenin,Frizzled-2,GSK-3β,cleaved caspase-3,and cleaved caspase-9 protein expression in rat knee articular cartilage was determined by western blot analysis.RESULTS:BSQJ obviously reduced pathological damage and matrix degradation of articular cartilage in KOA rats.Compared with the KOA group,H-BSQJ rats exhibited downregulated m RNA and protein expression of Wnt-4,β-catenin,Frizzled-2,and caspase-3,as well as upregulated m RNA and protein expression of GSK-3β.In addition,H-BSQJ significantly increased m RNA expression of Col2 a1 and decreased m RNA expression of MMP-1 and MMP-13.CONCLUSION:BSQJ exerted a beneficial effect on KOA by a mechanism involving downregulation of the Wnt/β-catenin pathway,which inhibited both cartilage extracellular matrix degradation and chondrocyte apoptosis to ameliorate KOA in rats.YAO Nan CHEN Guocai LU Yanyan XU Xuemeng ZHAO Chuanxi HUANG Xuejun LIU Wengang PENG Sha WU Huai 2021Journal of Traditional Chinese Medicine2021,41,6:5

9Multiple omics datasets reveal significant physical and physiological dormancy in alfalfa hard seeds identified by multispectral imaging analysis显示文摘Physical dormancy(PY) commonly present in the seeds of higher plants is believed to be responsible for the germination failure by impermeable seed coat in hard seeds of legume species, instead of physiological dormancy(PD). In this study, a non-destructive approach involving multispectral imaging was used to successfully identify hard seeds from non-hard seeds in Medicago sativa, with accuracy as high as96.8%–99.0%. We further adopted multiple-omics strategies to investigate the differences of physiology,metabolomics, methylomics, and transcriptomics in alfalfa hard seeds, with non-hard seeds as control.The hard seeds showed dramatically increased antioxidants and 125 metabolites of significant differences in non-targeted metabolomics analysis, which are enriched in the biosynthesis pathways of flavonoids, lipids and hormones, especially with significantly higher ABA, a hormone known to induce dormancy. In our transcriptomics results, the enrichment pathway of “response to abscisic acid” of differential expressed genes(DEG) supported the key role of ABA in metabolomics results. The methylome analysis identified 54,899, 46,216 and 54,452 differential methylation regions for contexts of CpG, CHG and CHH, and 344 DEGs might be regulated by hypermethylation and hypomethylation of promoter and exon regions, including four ABA-and JA-responsive genes. Among 8% hard seeds in seed lots,24.5% still did not germinate after scarifying seed coat, and were named as non-PY hard seeds.Compared to hard seeds, significantly higher contents of ABA/IAA and ABA/JA were identified in nonPY hard seeds, which indicated the potential presence of PD. 10A new progressive open-set recognition method with adaptive probability threshold显示文摘In the traditional pattern classification method,it usually assumes that the object to be classified must lie in one of given(known)classes of the training data set.However,the training data set may not contain the class of some objects in practice,and this is considered as an Open-Set Recognition(OSR)problem.In this paper,we propose a new progressive open-set recognition method with adaptive probability threshold.Both the labeled training data and the test data(objects to be classified)are put into a common data set,and the k-Nearest Neighbors(k-NNs)of each object are sought in this common set.Then,we can determine the probability of object lying in the given classes.If the majority of k-NNs of the object are from labeled training data,this object quite likely belongs to one of the given classes,and the density of the object and its neighbors is taken into account here.However,when most of k-NNs are from the unlabeled test data set,the class of object is considered very uncertain because the class of test data is unknown,and this object cannot be classified in this step.Once the objects belonging to known classes with high probability are all found,we re-calculate the probability of the other uncertain objects belonging to known classes based on the labeled training data and the objects marked with the estimated probability.Such iteration will stop when the probabilities of all the objects belonging to known classes are not changed.Then,a modified Otsu’s method is employed to adaptively seek the probability threshold for the final classification.If the probability of object belonging to known classes is smaller than this threshold,it will be assigned to the ignorant(unknown)class that is not included in training data set.The other objects will be committed to a specific class.The effectiveness of the proposed method has been validated using some experiments.Zhunga LIU Xuemeng HUI Yimin FU 2023Chinese Journal of Aeronautics2023,36,11:0

12Zearalenone induces apoptosis and autophagy by regulating endoplasmic reticulum stress signalling in porcine trophectoderm cells显示文摘Zearalenone(ZEA),a mycotoxin produced mainly by fungi belonging to Fusarium species in foods and feeds,causes a serious hazard to humans and animals.Numerous studies have revealed that ingesting ZEA can disrupt the reproductive function and impair the reproductive process in animals.This experiment was to investigate the toxicological effect and the mechanism of ZEA exposure on reproduction in pigs during early stages of pregnancy.In the present study,we treated with 0 to 80μmol/L ZEA for 12 or 24 h in trophoblast ectoderm(pTr)cells.The results showed that ZEA had significantly decreased cell proliferation(P<0.05),which was accompanied by DNA damage-related cell cycle arrest at G2/M phase,activation of the apoptosis and endoplasmic reticulum(ER)stress,as well as impairment of barrier function(P<0.05).Western blot analysis and transmission electron microscopy(TEM)showed that exposure to ZEA can activation of autophagy in pTr cells.Importantly,pretreatment with chloroquine(CQ)or 3-methyladenine(3-MA)led to increased apoptosis in pTr cells.Interestingly,pTr cells pretreated with 4-phenylbutyric acid(4-PBA),an inhibitor of ER stress,resulted in reduced cell death in pTr cells,indicating a critical role for ER stress in the activation of autophagy.In conclusion,these results reveal that ZEA-triggered ER stress is critical for the cell fate decision of pTr cells during early porcine embryonic development.Application of small molecules with ability of blocking ER stress might be therapeutic option to reduce the deleterious effect of ZEA in pregnant animals.Jun Bai Jun Li Ning Liu Hai Jia Xuemeng Si Yusong Zhou Zhian Zhai Ying Yang Fazheng Ren Zhenlong Wu 2023Animal Nutrition2023,,1:0

14Deep CLSTM for Predictive Beamforming in Integrated Sensing and Communication-Enabled Vehicular Networks显示文摘Predictive beamforming design is an essential task in realizing high-mobility integrated sensing and communication(ISAC),which highly depends on the accuracy of the channel prediction(CP),i.e.,predicting the angular parameters of users.However,the performance of CP highly depends on the estimated historical channel stated information(CSI)with estimation errors,resulting in the performance degradation for most traditional CP methods.To further improve the prediction accuracy,in this paper,we focus on the ISAC in vehicle networks and propose a convolutional long-short term memory(CLSTM)recurrent neural network(CLRNet)to predict the angle of vehicles for the design of predictive beamforming.In the developed CLRNet,both the convolutional neural network(CNN)module and the LSTM module are adopted to exploit the spatial features and the temporal dependency from the estimated historical angles of vehicles to facilitate the angle prediction.Finally,numerical results demonstrate that the developed CLRNet-based method is robust to the estimation error and can significantly outperform the state-of-the-art benchmarks,achieving an excellent sum-rate performance for ISAC systems.Chang Liu Xuemeng Liu Shuangyang Li Weijie Yuan Derrick Wing Kwan Ng 2022Journal of Communications and Information Networks2022,7,3:0

These compounds, especially phospholipid polycyclic aromatic hydrocarbons, have a wide range of pharmacological applications, including embedded DNA and central nervous system, anti-tumor, anti-virus, anti-parasite, anti-bacterial, and antithrombotic effects. Unfortunately, the insufficient drug sources have limited the development of these compounds. In this study, we isolated salinpostin compounds from a fermentation solution of marine-derived Salinospora sp., which has a common bicyclic enol-phosphotriester core framework, as well as potent and selective antimalarial activities against P. falciparum with EC_(50)=50 nmolL^(-1). The chemical synthesis of these compounds in greater quantities is necessary for their use in bioactivity studies. Thus we explored a short route with high yields and mild reaction conditions, which can generate combinatorial libraries for drug discovery and lead optimization. We developed a new total synthesis method for six cyclic enol-phosphotriester salinipotin compounds and their diastereomers. For the total synthesis of cyclipostin P, we prepared cyclic enol-phosphotriester salinipostin compounds in 10 steps from a readily accessible starting material, 1,3-dihydroxyacetone, and obtained an overall yield of 1.29%. 17Unraveling abnormal buried interface anion defect passivation mechanisms depending on cation-induced steric hindrance for efficient and stable perovskite solar cells显示文摘Although ionic liquids(ILs)have been widely employed to heal the defects in perovskite solar cells(PSCs),the corresponding defect passivation mechanisms are not thoroughly understood up to now.Herein,we first reveal an abnormal buried interface anion defect passivation mechanism depending on cationinduced steric hindrance.The IL molecules containing the same anion([BF4]^(-))and different sizes of imidazolium cations induced by substituent size are used to manipulate buried interface.It was revealed what passivated interfacial defects is mainly anions instead of cations.Theoretical and experimental results demonstrate that the large-sized cations can weaken the ionic bond strength between anions and cations,and facilitate the interaction between anions and SnO2as well as perovskites,which is conducive to interfacial defect passivation and ameliorating interfacial contact.It can be concluded that interfacial chemical interaction strength and defect passivation effect are positively correlated with the size of cations.The discovery breaks conventional thinking that large-sized modification molecules would weaken their chemical interaction with perovskite.Compared with the control device(21.54%),the device based on 1,3-Bis(1-adamantyl)-imidazolium tetrafluoroborate(BAIMBF4)with maximum size cations achieves a significantly enhanced efficiency of 23.61%along with much increased moisture,thermal and light stabilities.Dongmei He Ru Li Baibai Liu Qian Zhou Hua Yang Xuemeng Yu Shaokuan Gong Xihan Chen Baomin Xu Shangfeng Yang Jiangzhao Chen 2023Journal of Energy Chemistry2023,,5:0
