| 1 | Combination of ribavirin and reduning protects mice against severe pneumonia induced by H1N1 influenza a virus显示文摘OBJECTIVE:To investigate the effects of ribavirin administration combined with Reduning in a mouse model of influenza A(H1N1)-induced severe pneumonia.METHODS:Influenza A/Beijing/501/2009(H1N1)-infected C57BL/6 mice were randomly divided into four experimental groups treated with either a mock injection of phosphate-buffered saline(PBS),ribavirin(66.6 mg/kg daily)or Reduning(86.6 mg/kg daily),or a combination of both,for 7 days.Mice were monitored for clinical signs and survival,and body weight was measured daily for 14 days.Virus titer,lung wet-to-dry ratios,pathology and cytokines including interleukin(IL)-6,IL-10,and interferon(IFN)-γwere assayed on different days.RESULTS:In the untreated group injected with phosphate buffer saline,all the mice died of the infection.The survival rate of mice treated with Reduning was only 10%,whereas 100%of the ribavirin-and the combination-treated mice survived.Low lung viral loads indicated that ribavirin significantly inhibited virus replication,whereas Reduning did not.Lung wet-to-dry ratios demonstrated that both ribavirin and Reduning,administered together or separately,reduced acute lung edema compared with results in the untreated group.Pathology analyses also showed that treatment with a combination of both drugs relieved pathological lesions,whereas the single drug treatment did not.Levels of IL-6,IL-10 and IFN-γin mice treated with ribavirin or the combination of both ribavirin and Reduning were all significantly lower than in the untreated group,especially in the combination-treated group.In addition,Reduning administration significantly decreased both IL-6 and IL-10production but had no effect on IFN-γ.CONCLUSION:Due to the synergistic effect of antiviral and antiinflammation,the combination of ribavirin and Reduning could be an effective treatment for severe H1N1 which was considered to be significant to delayed antiviral and drug resistant. | Ma Yuexia Zhang Wei Zhao Zhongpeng Li Min Liu Jian Wang Yuguang | 2016 | Journal of Traditional Chinese Medicine2016,36,2: | 13 |
| 2 | Experimental studies on the P-T stability conditions and influencing factors of gas hydrate in different systems显示文摘The P-T stability conditions of gas hydrate in different systems (i.e., solution, silica sand, and marine sediment) were studied using multi-step decomposition method with our experimental equipment. The effects of different ions with various concentrations and sediment grains on the P-T stability conditions of gas hydrate were investigated. The results show that different ions have different influences on the phase equilibrium of gas hydrate. However, the influence of ions is in a similar trend: the larger the concentration, the bigger the P-T curve shifts to the left. For the silica sand, the influence of pore capillarity of coarse particles (> 460 μm) can be negligible. The P-T curve measured in coarse silica is in agreement with that in pure water. However, the influence of pore capillarity of fine particles (< 35 μm) is significant. The maximum reduction value of temperature is 1.5 K for methane hydrate under stable state. The sediment from the South China Sea significantly affects the P-T stability conditions of methane hydrate, with an average reduction value of 1.9 K within the experimental conditions. This is mainly the result of both the pore water salinity and the pore capillarity of sediment. | 3 | Expression of human cytomegalovirus components in the brain tissues of patients with Rasmussen's encephalitis显示文摘Rasmussen's encephalitis(RE) is a rare and severe progressive epileptic syndrome with unknown etiology. Infection by viruses, including human cytomegalovirus(HCMV), has been speculated to be a potential trigger for RE. However, no viral antigens have been detected in the brains of patients with RE; thus, a possible clinical linkage between viral infections and RE has not been firmly established. In this study, we evaluated the expression of HCMV pp65 antigen in brain sections from 26 patients with RE and 20 non-RE patients by immunohistochemistry and in situ hybridization, and assessed the associations between HCMV infection and clinical parameters.Elevated expression of HCMV pp65 protein and DNA was observed in 88.5%(23/26) and 69.2%(18/26) of RE cases, respectively. In the non-RE group, HCMV pp65 antigen was detected only in two cases(10%), both of which were negative for DNA staining. Additionally, the intensity of HCMV pp65 staining was correlated with a shorter duration of the prodromal stage, younger age of seizure onset, and more severe unilateral cortical atrophy. Elevated expression of HCMV pp65 was observed in RE brain tissue and was correlated with the clinical features of RE disease. In summary, our results suggested that HCMV infection may be involved in the occurrence and progression of RE disease. | 5 | Effect of SiC particle addition on microstructure of Mg_2Si/Al composite显示文摘In the present study, by adding SiC particles into Al-Si-Mg melt, Mg2Si and SiC particles hybrid reinforced Al matrix composites were fabricated through the Mg2Si in situ synthesis in melt combined with the SiC ex situ stir casting. The as-cast microstructure containing primary Mg2Si and SiC particles that distribute homogenously in Al matrix was successfully achieved. The effects of SiC particle addition on the microstructure of Mg2Si/Al composites were investigated by using scanning electron microscopy(SEM) and XRD. The results show that, with increasing the fraction of the SiC particles from 5wt.% to 10wt.%, the morphologies of the primary Mg2Si particulates in the prepared samples remain polygonal, but the size of the primary phase decreases slightly. However, when the SiC particle addition reaches 15wt.%, the morphologies of the primary Mg2Si particulates change partially from polygonal to quadrangular with a decrease in size from 50 μm to 30 μm. The size of primary Al dendrites decreases with increasing fraction of the SiC particles from 0wt.% to 15wt.%. The morphology of the eutectic Mg2Si phase changes from a fi ber-form to a short fi ber-form and/or a dot-like shape with increasing fraction of the SiC particles. | 6 | Progress in small-molecule luminescent materials for organic light-emitting diodes显示文摘Organic light-emitting diodes(OLEDs) have been extensively studied since the first efficient device based on small molecular luminescent materials was reported by Tang. Organic electroluminescent material, one of the centerpieces of OLEDs, has been the focus of studies by many material scientists. To obtain high luminosity and to keep material costs low, a few remarkable design concepts have been developed. Aggregation-induced emission(AIE) materials were invented to overcome the common fluorescence-quenching problem, and cross-dipole stacking of fluorescent molecules was shown to be an effective method to get high solid-state luminescence. To exceed the limit of internal quantum efficiency of conventional fluorescent materials, phosphorescent materials were successfully applied in highly efficient electroluminescent devices. Most recently, delayed fluorescent materials via reverse-intersystem crossing(RISC) from triplet to singlet and the 'hot exciton' materials based on hybridized local and charge-transfer(HLCT) states were developed to be a new generation of low-cost luminescent materials as efficient as phosphorescent materials. In terms of the device-fabrication process, solution-processible small molecular luminescent materials possess the advantages of high purity(vs. polymers) and low procession cost(vs. vacuum deposition), which are garnering them increasing attention. | 7 | Influence of foraminifera on formation and occurrence characteristics of natural gas hydrates in fine-grained sediments from Shenhu area, South China Sea显示文摘Marine gas hydrates accumulate primarily in coarse-grained, high-permeability layers; however, highly saturated natural gas hydrates have been discovered in the fine-grained sediments of Shenhu area, South China Sea(SCS). This may be explained by key factors, such as the great abundance of foraminifera shells. In this paper, by analyzing the SCS foraminifera structure and performing hydrate formation experiments in the foraminifera shells, the contribution of foraminifera to hydrate accumulation in the SCS was investigated from a microscopic point of view. Simulations of hydrate formation were carried out in both pure SCS foraminifera shells and the host sediments. Pore structures in typical foraminifera were studied by use of micro-focus X-ray computed tomography(CT) and scanning electron microscopy(SEM). Hydrate growth and occurrence characteristics in the foraminifera shells were observed in-situ. The results showed that the presence of foraminifera significantly enhanced the effective porosity of the SCS sediments. Moreover, while the hydrates grew preferentially in the chambers of the coarse-grained foraminifera by adhering to the inner walls of the foraminifera shells, no apparent hydrate accumulation was observed in the fine-grained or argillaceous matrix. | 11 | The diversity of denitrifying bacteria in the alpine meadow soil of Sanjiangyuan natural reserve in Tibet Plateau显示文摘This is the first time to describe the diversity of denitrifying bacteria in Sanjiangyuan natural reserve in Tibet Plateau by investigating the molecular diversity and phylogenetic analysis of nirK and nosZ genes using PCR-RFLP and sequencing. Four soil samples were collected from alpine meadow communities from over an altitude of 4600 m which had different physicochemical properties by principal component analysis (PCA). For the genes fragment of nirK and nosZ, the diverse PCR products were characterized by cloning, restriction fragment length polymorphism (RFLP) analysis and sequenced. A total of 253 nirK clones and 283 nosZ clones were received in four samples, and 78 operational taxonomic units (OTUs) of nirK and 120 OTUs of nosZ by the restriction enzymes MspI and RsaI digested. The analysis of environmental factors showed that altitude and C/N ratio in soil may be the key factors to the denitrifying bacteria community. 36 nirK clones and 17 nosZ clones were sequenced, and their levels of nucleotide identity were from 69% to 98% and 57% to 97%, respectively. The phylogenetic tree was constructed using the Clustal W and Mega softwares, and all the sequenced clones could be subdivided into 4 groups. Some of clone sequences were related to the nirK and nosZ genes belonging to three phylogenetic subdivisions (α-, β- and γ subclasses of the Proteobacteria), while most of the clones were closely related to the genes of the uncultured bacteria. | 12 | Detection of EBV and HHV6 in the Brain Tissue of Patients with Rasmussen's Encephalitis显示文摘Rasmussen's encephalitis (RE) is a rare pediatric neurological disorder, and the exact etiology is not clear. Viral infection may be involved in the pathogenesis of RE, but conflicting results have reported. In this study, we evaluated the expression of both Epstein-Barr virus (EBV) and human herpes virus (HHV) 6 antigens in brain sections from 30 patients with RE and16 control individuals by immunohistochemistry. In the RE group, EBV and HHV6 antigens were detected in 56.7% (17/30) and 50% (15/30) of individuals, respectively. In contrast, no detectable EBV and HHV6 antigen expression was found in brain tissues of the control group. The co-expression of EBV and HHV6 was detected in 20.0% (6/30) of individuals. In particular, a 4-year-old boy had a typical clinical course, including a medical history of viral encephalitis,intractable epilepsy, and hemispheric atrophy. The co-expression of EBV and HHV6 was detected in neurons and astrocytes in the brain tissue, accompanied by a high frequency of CD8?T cells. | 14 | Elevated expression of EBV and TLRs in the brain is associated with Rasmussen's encephalitis显示文摘Rasmussen's encephalitis(RE) is a rare pediatric neurological disorder, the etiology of which remains unclear. It has been speculated that the immunopathogenesis of RE involves damage to neurons, which eventually leads to the occurrence of RE. Viral infection may be a critical factor in triggering RE immunopathogenesis. In this study, we analyzed the expression of Epstein-Barr virus(EBV) antigens as well as of Toll-like receptor 3(TLR3), TLR9, and downstream adapter TIRdomain-containing adapter-inducing interferon-β(TRIF) in the brain tissues of 26 patients with RE and 16 control individuals using immunohistochemistry(IHC). In the RE group, EBV antigens were detected in 53% of individuals at various expression levels. In contrast, there was no detectable EBV antigen expression in control brain tissues. Moreover, we found marked increases in the expression of TLR3, TLR9, and TRIF in the brain tissues of RE patients compared with levels in the control group. Furthermore, among RE cases, EBV expression and high TLR3 expression were associated with more severe brain atrophy. | 15 | Mixture prepared with Chinese medicines in terms of releasing exterior cold and clearing interior heat: its efficacy and safety in patients with seasonal influenza-a randomized controlled trial显示文摘OBJECTIVE: To assess the efficacy and safety in 105 patients with seasonal Influenza in Beijing, the mixture prepared with Chinese medicines follows the treatment regimen of releasing exterior cold andclearing interior heat.METHODS: Total 330 patients with seasonal influenza were enrolled and randomly and averagely divided into the Chinese herbal medicine, the western medicine and the Chinese patent medicine group. They were treated with Chinese medicine Oseltamivir Phosphate Capsules and the Scattering Wind and Resolving Toxins Capsules. The main efficacy indicators were the antifebrile onset time and recovery time of body temperature. The efficacy and safety of the mixture was scientifically evaluated. Comparisons of several variables were analyzed.RESULTS: Median antifebrile onset time of the Chinese herbal medicine group was significantly shorter than the western medicine group(P < 0.05) and the Chinese patent medicine group(P < 0.05). The median antifebrile recovery time of the Chinese herbal medicine group was significantly shorter than the Chinese patent medicine group(P <0.05). The groups evaluated by TCM symptom pattern effect, both the Chinese herbal medicine group and Western Medicine group were better than the Chinese patent medicine group(P <0.05). In this paper, the seafloor water sample and sediment sample (saturated with pore water) from Shenhu Area of South China Sea were used to synthesize methane hydrates, and the stability conditions of methane hydrates were investigated by multi-step heating dissociation method. Preliminary experimental results show that the dissociation temperature of methane hydrate both in seafloor water and marine sediment, under any given pressure, is depressed by approximately -1.4 K relative to the pure water system.
