2Groundwater hydrochemistry and isotope geochemistry in the Turpan Basin, northwestern China显示文摘The Turpan Basin is located in the arid zone of northwestern China and is a typical closed inland basin surrounded by high mountains. It is one of the most arid regions in the world and, as a result, the groundwater in this area is very important for both domestic and agricultural uses. In the present study, the relationships of major elements(K+, Na+, Ca2+, Mg2+, HCO3-, SO42- and Cl-) and environmental isotopes(δ18O, δ2H and T) in groundwater were analyzed to investigate the evolution of the regional hydrochemistry within the Turpan Basin. The hydrochemistry results demonstrate that groundwater with high total dissolved solids(TDS) concentration is dominated by sodium chloride(Na-Cl) and sodium sulfate(Na-SO4) type water, whereas that with low TDS concentration(typically from near mountain areas) is dominated by calcium bicarbonate(Ca-HCO3) type water. The evolution of groundwater hydrochemistry within the Turpan Basin is a result of calcium carbonate precipitation, evaporation concentration, cation exchange and dissolution of evaporites(i.e. halite, mirabilite and gypsum). Furthermore, evaporite dissolution associated with irrigation practice plays a key role in the groundwater salinization, especially in the central part of the basin. Environmental isotopes reveal that the groundwater is recharged by precipitation in the mountain areas and fast vertical infiltration of irrigation return flow. In the southern sub-basin the shallow groundwater and the deep groundwater is separated at a depth of about 40 m, with substantial differences in terms of hydrochemical and isotopic characteristics. 4Transcriptional analysis and histochemistry reveal that hypersensitive cell death and H2O2 have crucial roles in the resistance of tea plant (Camellia sinensis (L.) O. Kuntze) to anthracnose显示文摘Anthracnose causes severe losses of tea production in China.Although genes and biological processes involved in anthracnose resistance have been reported in other plants,the molecular response to anthracnose in tea plant is unknown.We used the susceptible tea cultivar Longjing 43 and the resistant cultivar Zhongcha 108 as materials and compared transcriptome changes in the leaves of both cultivars following Colletotrichum fructicola inoculation.In all,9015 and 8624 genes were differentially expressed between the resistant and susceptible cultivars and their controls(0 h),respectively.In both cultivars,the differentially expressed genes(DEGs)were enriched in 215 pathways,including responses to sugar metabolism,phytohormones,reactive oxygen species(ROS),biotic stimuli and signalling,transmembrane transporter activity,protease activity and signalling receptor activity,but DEG expression levels were higher in Zhongcha 108 than in Longjing 43.Moreover,functional enrichment analysis of the DEGs showed that hydrogen peroxide(H2O2)metabolism,cell death,secondary metabolism,and carbohydrate metabolism are involved in the defence of Zhongcha 108,and 88 key genes were identified.Protein–protein interaction(PPI)network demonstrated that putative mitogen-activated protein kinase(MAPK)cascades are activated by resistance(R)genes and mediate downstream defence responses.Histochemical analysis subsequently validated the strong hypersensitive response(HR)and H2O2 accumulation that occurred around the hyphal infection sites in Zhongcha 108.Overall,our results indicate that the HR and H2O2 are critical mechanisms in tea plant defence against anthracnose and may be activated by R genes via MAPK cascades.Yuchun Wang Xinyuan Hao Qinhua Lu Lu Wang Wenjun Qian Nana Li Changqing Ding Xinchao Wang Yajun Yang 2018Horticulture Research2018,5,1:11
5Prioritizing natural-selection signals from the deep-sequencing genomic data suggests multi-variant adaptation in Tibetan highlanders显示文摘Human genetic adaptation to high altitudes(>2500 m)has been extensively studied over the last few years,but few functional adaptive genetic variants have been identified,largely owing to the lack of deep-genome sequencing data available to previous studies.Here,w e build a list of putative adaptive variants,including 63 missense,7 loss-of-function;1,298 evolutionarily conserved variants and 509 expression quantitative traits loci.Notably,the top signal of selection is located in TMEM 247,a transmembrane protein-coding gene.The Tibetan version of TMEM 247 harbors one high-frequency(76.3%)missense variant,rsl 16983452(c.248C>T;p.Ala83Val);with the T allele derived from archaic ancestry and carried by>94%of Tibetans but absent or in low frequencies(<3%)in non-Tibetan populations.The rsl 16983452-T is strongly and positively correlated with altitude and significantly associated with reduced hemoglobin concentration(p=5.78×10^-5),red blood cell count(p=5.72×10^-7)and hematocrit(p=2.57×10^-6).In particular,TMEM247-rs 116983452 shows greater effect size and better predicts the phenotypic outcome than any E P A S1 variants in association with adaptive traits in Tibetans.Modeling the interaction between TM£M247-rsl 16983452 and EPAS1 variants indicates weak but statistically significant epistatic effects.Our results support that multiple variants may jointly deliver the fitness of the Tibetans on the plateau,where a complex model is needed to elucidate the adaptive evolution mechanism.Lian Deng Chao Zhang Kai Yuan Yang Gao Yuwen Pan Xueling Ge Yaoxi He Yuan Yuan Yan Lu Xiaoxi Zhang Hao Chen Haiyi Lou Xiaoji Wang Dongsheng Lu Jiaojiao Liu Lei Tian Qidi Feng Asifullah Khan Yajun Yang Zi-Bing Jin Jian Yang Fan Lu Jia Qu Longli Kang Bing Su Shuhua Xu 2019National Science Review2019,6,6:9
6Genome-wide variants of Eurasian facial shape differentiation and a prospective model of DNA based face prediction显示文摘It is a long-standing question as to which genes define the characteristic facial features among different ethnic groups. In this study, we use Uyghurs, an ancient admixed population to query the genetic bases why Europeans and Han Chinese look different. Facial traits were analyzed based on high-dense 3D facial images; numerous biometric spaces were examined for divergent facial features between European and Han Chinese, ranging from inter-landmark distances to dense shape geometrics. Genome-wide association studies(GWAS) were conducted on a discovery panel of Uyghurs. Six significant loci were identified, four of which, rs1868752, rs118078182, rs60159418 at or near UBASH3B, COL23A1, PCDH7 and rs17868256 were replicated in independent cohorts of Uyghurs or Southern Han Chinese. 7A draft sequence of the rice (Oryza sativa ssp. indica) genome显示文摘The sequence of the rice genome holds fundamental information for its biology, including physiology, genetics, development, and evolution, as well as information on many beneficial phenotypes of economic significance. Using a 'whole genome shotgun' approach, we have pro-duced a draft rice genome sequence of Oryza sativa ssp. in-dica, the major crop rice subspecies in China and many other regions of Asia. The draft genome sequence is constructed from over 4.3 million successful sequencing traces with an accumulative total length of 2214.9 Mb. The initial assembly of the non-redundant sequences reached 409.76 Mb in length, based on 3.30 million successful sequencing traces with a total length of 1797.4 Mb from an indica variant cultivar 93-11, giving an estimated coverage of 95.29% of the rice genome with an average base accuracy of higher than 99%. 8Response to climate change of different tree species and NDVI variation since 1923 in the middle arid region of Ningxia,China显示文摘The normalized difference vegetation index(NDVI)is used extensively to describe vegetation cover and ecological environment change.The purpose of this study was to contrast the response of different tree species growing in the same habitat to climate change and retrieve past NDVI using tree-ring width data from tree cores collected from the transitional zone of Pinus tabulaeformis and Picea crassifolia in the Luoshan Mountains in the middle arid region of Ningxia.Correlation analysis indicated that radial growth of P.tabulaeformis is more sensitive to precipitation and temperature change than that of P.crassifolia.Natural factors such as water availability and heat at this elevation are more suited to the growth of P.crassifolia,and are more advantageous to its renewal and succession.P.crassifolia is probably the better of the two species for protecting the forest ecosystem and conserving water in the Luoshan desertification area.Ring width of P.crassifolia correlates significantly with average NDVI for April–May(r=0.641,p<0.01),and both of them are influenced positively by precipitation in April–May.The reconstructed NDVI for 1923–2007 shows the relatively low vegetation cover occurred in the 1920s–1930s,the1960s–1970s,and the early 21st century.The reconstructed NDVI better reflected the drought climate in the study area.YaJun Wang RuiJie Lu YuZhen Ma HongWei Meng ShangYu Gao 2014Research in Cold and Arid Regions2014,6,1:6
9Synchrotron radiation-based Fourier-transform infrared spectromicroscopy for characterization of the protein/peptide distribution in single microspheres显示文摘The present study establishes a visualization method for the measurement of the distribution and localization of protein/peptide constituents within a single poly-lactide-co-glycolide(PLGA) microsphere using synchrotron radiation–based Fourier-transform infrared spectromicroscopy(SR-FTIR).The representative infrared wavenumbers specific for protein/peptide(Exenatide) and excipient(PLGA) were identified and chemical maps at the single microsphere level were generated by measuring and plotting the intensity of these specific bands.For quantitative analysis of the distribution within microspheres,Matlab software was used to transform the map file into a 3D matrix and the matrix values specific for the drug and excipient were extracted.Comparison of the normalized SR-FTIR maps of PLGA and Exenatide indicated that PLGA was uniformly distributed,while Exenatide was relatively non-uniformly distributed in the microspheres.In conclusion,SR-FTIR is a rapid,nondestructive and sensitive detection technology to provide the distribution of chemical constituents and functional groups in microparticles and microspheres.Manli Wang Xiaolong Lu Xianzhen Yin Yajun Tong Weiwei Peng Li Wu Haiyan Li Yan Yang Jingkai Gu Tiqiao Xiao Min Chen Jiwen Zhang 2015Acta Pharmaceutica Sinica B2015,5,3:5
10Research on modeling of heat source for electron beam welding fusion-solidification zone显示文摘In this paper, the common heat source model of point and linear heat source in the numerical simulation of electron beam welding (EBW) were summarized and introduced. The combined point-linear heat source model was brought forward and to simulate the welding temperature fields of EBW and predicting the weld shape. The model parameters were put forward and regulated in the combined model, which included the ratio of point heat source to linear heat source Qpr and the distribution of linear heat source Lr. Based on the combined model, the welding temperature fields of EBW were investigated. 12The design and synthesis of dextran-doxorubicin prodrug-based pH-sensitive drug delivery system for improving chemotherapy efficacy显示文摘Tumor cells show acidic conditions compared with normal cells,which further inspires scientist to build nanocarrier responsive to tumor microenvironment(TME)for enhancing tumor therapeutic efficacy.Here,we report a pH-sensitive and biocompatible polyprodrug based on dextran-doxorubicin(DOX)prodrug(DOXDT)for enhanced chemotherapy.Highdensity DOX component was covalently decorated on the nanocarrier and the drug molecules could be effectively released in the acidic tumor tissue/cells,improving chemotherapy efficacy.Specifically,a dextran-based copolymer was preliminarily prepared by one-step atom transfer radical polymerization(ATRP);then DOX was conjugated on the copolymer component via pH-responsive hydrazone bond.The structure of DOXDT can be well-controlled.The resulting DOXDT was able to further self-assemble into nanoscale micelles with a hydration diameter of about 32.4 nm,which presented excellent micellar stability.Compared to lipid-based drug delivery system,the DOXDT prodrug showed higher drug load capacity up to 23.6%.In addition,excellent stability and smaller size of the nanocarrier contributed to better tissue permeability and tumor suppressive effects in vivo.Hence,this amphipathic DOXDT prodrug is promising in the development of translational DOX formulations,which would be widely applied in cancer therapy.Xiaoli Zhang Tian Zhang Xianbin Ma Yajun Wang Yi Lu Die Jia Xiaohua Huang Jiucun Chen Zhigang Xu Feiqiu Wena 2020Asian Journal of Pharmaceutical Sciences2020,15,5:3
13Reconstructing the annual precipitation variation since 1899 based on tree-ring width in the western Hedong sandy land of Ningxia显示文摘Based on the analysis of the correlation between the tree-ring width of Pinus tabulaeformis and the climate factors in the western Hedong sandy land of Ningxia, a conversion equation between the annual precipitation and the tree-ring width since 1899 was reconstructed. The results of cross verification indicated that the conversion equation is stable and the reconstructed results are reliable. The result of reconstructed annual precipitation showed the remarkable fluctuation of precipitation and dry-to-wet variation before the 1940s. The smaller fluctuation and high frequent changes of precipitation occurred during the period of 1940s-1980s and after the 1980s the change trend of the precipitation became high periodic extent and low frequent. The study found that there were some coincidences with the climate change in Changling Mountains, Helan Mountains and the east of Qilian Mountains. The relatively dry periods in the beginning of 20th century, 1920s to 1930s, the end of the 20th century and 2004 to 2006 in the western Hedong sandy land of Ningxia accelerated the desertification, while the relatively humid period during the periods of the 1910s-1920s, 1930s-1940s and 1990s is favorable to prevent and control the desertification, and to weaken the climate warming and drying. 16Identification and bioinformatics analysis of lactate dehydrogenase genes from Echinococcus granulosus显示文摘Objective:To identify full length cDNA sequence of lactate dehydrogenase(LDH) from adult Echinococcus granulosus(E.granulosus) and to predict the structure and function of its encoding protein using bioinformatics methods.Methods:With the help of NCBI,EMBI, Expasy and other online sites,the open reading frame(ORF),conserved domain,physical and chemical parameters,signal peptide,epitope,topological structures of the protein sequences were predicted and a homology tertiary structure model was created:Vector NT1 software was used for sequence alignment,phylogenetic tree construction and tertiary structure prediction. Results:The target sequence was 1 233 bp length with a 996 bp biggest ORF encoding 331 amino acids protein with typical L-LDH conserved domain.It was confirmed as full length cDNA of LDH from E.granulosus and named as EgLDH(GenBank accession number:HM748917).The predicted molecular weight and isoelectric point of the deduced protein were 3 5516.2Da and 6.32 respectively.Compared with LDHs from Taenia solium,Taenia saginata asiatica,Spirometra erinaceieuropaei.Schistosoma japonicum,Clonorchis sinensis and human,it showed similarity of 86% ,85% ,55% ,58% ,58% and 53% ,respectively.EgLDH contained 3 putative transmembrane regions and 4 major epitopes(54aa-59aa.81aa-87aa,97aa-102aa,307aa-313aa),the latter were significant different from the corresponding regions of human LDH.In addition,some NAD and substrate binding sites located on epitopes 54aa-59aa and 97aa-102aa,respectively.Tertiary structure prediction showed that 3 key catalytic residues 105R,165D and 192H forming a catalytic center near the epitope 97aa-102aa,most NAD and substrate binding sites located around the center.Conclusions:The full length cDNA sequences of EgLDH were identified.It encoded a putative transmembrane protein which might be an ideal target molecule for vaccine and drugs.Gang Lu Yajun Lu Lihua Li Lixian Wu Zhigang Fan Dazhong Shi Hu Wang Xiumin Han 2010Asian Pacific Journal of Tropical Medicine2010,3,10:2
17Control system for cryogenic permanent magnet undulator(CPMU)of high energy photon source(HEPS)显示文摘Background High energy photon source(HEPS)is the fourth-generation light source,which uses a large number of highperformance insertion devices to generate synchrotron radiation.The control system is an important part of the insertion device(ID).Purpose Cryogenic permanent magnet undulator(CPMU)is one kind of IDs that works in liquid nitrogen temperature and ultra-high vacuum environment,and its control system is more difficult and complex than in-air ID.The design of the control system for CPMU will be introduced in detail.Method The sub-systems include high-precision magnetic gap control,safety protection,measurement and compensation of magnetic gap at cryogenic temperature and cryogenic temperature monitoring.Mature,reliable,stable technical schemes are designed to meet the technical specifications of sub-systems.Results The experiment results show that the magnetic gap motion accuracy can be controlled within 0.2–0.3μm under the step size of 1μm.The safety protection system has been tested in turn,and the predetermined protection can be achieved.The average value of magnetic gap cold contraction is 1.512 mm measured by optical micrometer,and the compensation is realized by software.The average temperature of the main magnet structure measured by the temperature sensors is 81.0 K,and the temperature gradient is 0.7 K/m.The temperature monitoring is reliable and stable.Conclusion The CPMU control system which is based on EPICS has been successfully applied to the CPMU prototype,and the test results have met the design specifications.Shutao Zhao Jianshe Cao Huihua Lu Xiaoyu Li Shuchen Sun Yajun Sun Xiangzhen Zhang Lei Zhang Yuhui Li Zhiqiang Li Wan Chen Lingling Gong Qing Guo Keyun Gong Yongsheng Huang Yanwei Yang Yufeng Yang 2021Radiation Detection Technology and Methods2021,5,1:2
18Microwave triggered multifunctional nanoplatform for targeted photothermal-chemotherapy in castration-resistant prostate cancer显示文摘Lacking a precise targeting strategy,castration-resistant prostate cancer(CRPC)is still hard to be treat effectively.Exploring treatment options that can accurately target CPRC is an important issue with urgent need.In this study,a novel nanotechnologybased strategy had been developed for the precise target treatment of CRPC.By combining microwaves and photothermal therapy(PTT),this nanoplatform,cmHSP70-PL-AuNC-DOX,targets tumor tissues with outstanding precision and achieves better anti-tumor activity by simultaneously eliciting photothermal and chemotherapeutic effects.From nanotechnology,cmHSP70-modified and thermo-sensitive liposome-coated AuNC-DOX were prepared and used for CRPC-targeted photothermal ablation and chemotherapy.Doxorubicin(DOX)was selected as the chemotherapeutic agent for cytotoxicity.In terms of the curative scheme,prostate tissues were firstly pre-treated with microwaves to induce the expression of heat shock protein 70(HSP70)and its migration to the cell membrane,which was then targeted by HSP70 antibody(cmHSP70)coated on the nanoparticles to achieve accurate drug delivery.The nanoplatform then achieved precise ablation and controlled release of DOX under external near-infrared(NIR)irradiation.Through the implementation,the targeting,cell killing,and safety of this therapeutical strategy had been verified in vivo and in vitro.This work establishes an accurate,controllable,efficient,non-invasive,and safe treatment platform for targeting CRPC,provides a rational design for CRPC’s PTT,and offers new prospects for nanomedicines with great precision.Pengyuan Liu Yajun Wu Xiaogang Xu Xudong Fan Chuan Sun Xiaojie Chen Jindan Xia Shiting Bai Li Qu Huasong Lu Jing Wu Jun Chen Ji-Gang Piao Zhibing Wu 2023Nano Research2023,16,7:2
19Transferrin plays a central role in coagulation balance by interacting with clotting factors显示文摘Coagulation balance is maintained through fine-tuned interactions among clotting factors,whose physiological concentrations vary substantially.In particular,the concentrations of coagulation proteases(pM to nM)are much lower than their natural inactivator antithrombin(AT,〜3μM),suggesting the existence of other coordinators.In the current study,we found that transferrin(norm plasma concentration〜40 μM)interacts with fibrinogen,thrombin,factor Xlla(FXlla),and AT with different affinity to maintain coagulation balance.Normally,transferrin is sequestered by binding with fibrinogen(normal plasma concentration〜10 μM)at a molar ratio of 4:1.In atherosclerosis,abnormally up-regulated transferrin interacts with and potentiates thrombin/FXlla and blocks ATs inactivation effect on coagulation proteases by binding to AT,thus inducing hypercoagulability.In the mouse model,transferrin overexpression aggravated atherosclerosis,whereas transferrin inhibition via shRNA knockdown or treatment with antitransferrin antibody or designed peptides interfering with transferrin-thrombin/FXlla interactions alleviated atherosclerosis.Collectively,these findings identify that transferrin is an important clotting regulator and an adjuster in the maintenance of coagulation balance and modifies the coagulation cascade.Xiaopeng Tang Zhiye Zhang Mingqian Fang Yajun Han Gan Wang Sheng Wang Min Xue Yaxiong Li Li Zhang Jian Wu Biqing Yang James Mwangi Qiumin Lu Xiaoping Du Ren Lai 2020Cell Research2020,30,2:2
