1miR-137 inhibits melanoma cell proliferation through downregulation of GLO1显示文摘Late-stage melanoma is refractory to current therapies. MicroRNAs (miRNAs) can modulate many physiological and pathological processes of melanoma. Studies have demonstrated that miR-137 acts as a tumor suppressor by inhibiting the proliferation of melanoma cells through targeting multiple mRNAs. The glyoxalase system member glyoxalase 1 (GLO1) is the principal scavenging enzyme of methylglyoxal (MG), a toxic byproduct of glycolysis. Using 35S in vivo/vitro labelling analysis for dynamic proteomics (SiLAD), we found that miR-137 downregulated the expression of GLO1 in melanoma cells.Bioinformatics analysis predicted that GLO1 is a direct target of miR-137. This was validated by dual luciferase reporter assay.Quantitative RT-PCR (qRT-PCR) and western blot analysis indicated that miR-137 could decrease endogenous GLO1 expression.Furthermore, siRNA targeting of GLO1 mimicked inhibition of melanoma cell proliferation caused by miR-137 overexpression.Re-expression of GLO1 was able to restore miR-137-mediated suppression of melanoma cell proliferation. 2Effects of antisense CENP-B on the proliferation of HeLa (Tet-off) cells显示文摘In order to study the change of the expression of centromere protein B (CENP-B) caused by antisense transfection, proto-eukaryotically expressed fused protein GST-CENP-B (65 ku) was injected into mouse, and a peculiar anti-CENP-B serum MaCenpB was collected. A strain of transfected HeLa Tet-off cell HaCb, which contains antisense CENP-B expressing vector pBI-EGFP-as-CenpB, was prepared. Northern blot and Western blot were used to analyze the repression of internal CENP-B in transfected cells. According to the growth curve, the proliferation of HeLa (Tet-off) is repressed by antisense CENP-B, and the multiplication time is prolonged for 32.81 h. The analysis of flow cytometry revealed that, compared with HeLa (Tet-off), the G1 cell population of HaCb is increased (AG1=9%) while S fraction is decreased (AS=11%), but the G2/M phase is nearly unchanged (AG2/M=3%). In the meanwhile, the mi-totic index of HaCb declines greatly compared with that of HeLa (Tet-off). 3Synthesis of novel β-amino ketones containing a p-aminobenzoic acid moiety and evaluation of their antidiabetic activities显示文摘The synthesis of two series of β-amino ketones containing a p-aminobenzoic acid moiety (TM-1 and TM-2) using a modified protocol of the Mannich reaction is reported. The molecular structures of a total of tweenty three new target compounds were characterized by 1H NMR, 13C NMR, ESI-MS and HR-MS. Subsequently, their antidiabetic activities were screened in vitro. The α-glucodase inhibition (α-GI) activity of compound 1e reached a remarkable level of 66.50%. The peroxisome proliferatoractivated receptor (PPAR) relative activation activities of six compounds are above 80%, and in particular 2i displays an unprecedentedly high PPAR of 130.91%. 4Structural basis of the ultrasensitive calcium indicator GCaMP6显示文摘GCaMP is one of the most widely used calcium indicators in neuronal imaging and calcium cell biology.The newly developed GCaMP6 shows superior brightness and ultrasensitivity to calcium concentration change.In this study,we determined crystal structures of Ca2+-bound GCaMP6 monomer and dimer and presented detailed structural analyses in comparison with its parent version GCaMP5G.Our analyses reveal the structural basis for the outperformance of this newly developed Ca2+indicator.Three substitution mutations and the resulting changes of local structure and interaction explain the ultrasensitivity and increased fluorescence intensity common to all three versions of GCaMP6.Each particular substitution in the three GCaMP6 is also structurally consistent with their differential sensitivity and intensity,maximizing the potential of using GCaMP6 in solving diverse problems in neuronal research and calcium signaling.Our studies shall also be beneficial to further structure-guided optimization of GCaMP and facilitate the design of novel calcium indicators.DING JingJin LUO Andrew F. 11Non-destructive Testing Method for Crack Based on Diamond Nitrogen-vacancy Color Center显示文摘Magnetic field measurement plays an extremely important role in material science,electronic en-gineering,power system and even industrial fields.In particular,magnetic field measurement provides a safe and reliable tool for industrial non-destructive testing.The sensitivity of magnetic field measurement deter-mines the highest level of detection.The diamond nitrogen-vacancy(NV)color center is a new type of quan-tum sensor developed in recent years.The external magnetic field will cause Zeeman splitting of the ground state energy level of the diamond NV color center.Optical detection magnetic resonance(ODMR),using a mi-crowave source and a lock-in amplifier to detect the resonant frequency of the NV color center,and finally the change of the resonant frequency can accurately calculate the size of the external magnetic field and the sensi-tivity of the external magnetic field change.In the experiment,a diamond containing a high concentration of NV color centers is coupled with an optical fiber to realize the preparation of a magnetic field scanning probe.Then,the surface cracks of the magnetized iron plate weld are scanned,and the scanning results are drawn into a two-dimensional magnetic force distribution map,according to the magnetic field gradient change of the magnetic force distribution map,the position and size of the crack can be judged very accurately,which pro-vides a very effective diagnostic tool for industrial safety.ZHAO Long WANG Xin ZHAO Bowen WU Guiyuan LUO Dacheng ZHANG Shaochun 2022Instrumentation2022,9,1:0
12Region-adaptive Concept Aggregation for Few-shot Visual Recognition显示文摘Few-shot learning (FSL) aims to learn novel concepts from very limited examples. However, most FSL methods suffer from the issue of lacking robustness in concept learning. Specifically, existing FSL methods usually ignore the diversity of region contents that may contain concept-irrelevant information such as the background, which would introduce bias/noise and degrade the performance of conceptual representation learning. To address the above-mentioned issue, we propose a novel metric-based FSL method termed region-adaptive concept aggregation network or RCA-Net. Specifically, we devise a region-adaptive concept aggregator (RCA) to model the relationships of different regions and capture the conceptual information in different regions, which are then integrated in a weighted average manner to obtain the conceptual representation. Consequently, robust concept learning can be achieved by focusing more on the concept-relevant information and less on the conceptual-irrelevant information. We perform extensive experiments on three popular visual recognition benchmarks to demonstrate the superiority of RCA-Net for robust few-shot learning. 14Doping strategy on improving the overall cathodic performance of double perovskite LnBaCo_(2)O_(5+σ)(Ln=Pr,Gd)as potential SOFC cathode materials显示文摘A series of single-phase double perovskite Pr1-xGdxBaCo_(2)-yFeyO_(5+σ)(x=0,0.5 and 1,0≤y≤1)materials were engineered through A/B site co-doping strategy to improve the mechanical,electrical and electrochemical properties as potential cathode materials for the application of intermediate solid oxide fuel cells(IT-SOFCs).The corresponding thermochemical stability,thermal expansion behavior,electrical conductivity and cathodic polarization resistance of the materials were systematically investigated.It was found that the A-site dual lanthanide doped Pr_(0.5)Gd_(0.5)BaCo_(2)O_(5+σ)(PGBCO)exhibits improved electrical conductivity,reduced thermal expansion,and comparatively low electrochemical polarization resistance versus single lanthanide double perovskite,PrBaCo_(2)O_(5+σ)(PBCO)and GdBaCo_(2)O_(5+σ)(GBCO)materials.Further investigation on the effect of B-site Fe-doping on Pr_(0.5)Gd_(0.5)BaCo_(2)-yFeyO_(5)+σ(PGBCF-y,0≤y≤1)reveals that all the PGBCF-y compositions exhibit excellent chemical stability with Gd-doped ceria(GDC)at operating temperatures not higher than 1100℃.Besides,doping of Fe in B-site can effectively reduce the thermal expansion coefficients(TECs)of the Pr_(0.5)Gd_(0.5)BaCo_(2)O_(5)+σceramics at 30e1000℃.And the electrochemical impedance spectra(EIS)results show that the PGBCF-y|GDC|PGBCF-y symmetric cells have acceptable low area specific polarization resistances.Further examination of the cathodic polarization and characteristic capacitance from the AC impedance spectra by employing the relaxation time distribution(DRT)method demonstrated that charge transfer is the dominating subprocess for the oxygen transport through the materials.Fangze Chen Dacheng Zhou Xueqing Xiong Juntao Pan Donglin Cai Ze Wei Xiyong Chen Yihui Liu Nengneng Luo Jialin Yan Toyohisa Fujita 2023Journal of Materiomics2023,9,5:0
