6Whole genome sequencing of 10K patients with acute ischaemic stroke or transient ischaemic attack: design, methods and baseline patient characteristics显示文摘Background and purpose Stroke is the second leading cause of death worldwide and the leading cause of mortality and long-term disability in China,but its underlying risk genes and pathways are far from being comprehensively understood.We here describe the design and methods of whole genome sequencing(WGS)for 10914 patients with acute ischaemic stroke or transient ischaemic attack from the Third China National Stroke Registry(CNSR-III).Methods Baseline clinical characteristics of the included patients in this study were reported.DNA was extracted from white blood cells of participants.Libraries are constructed using qualified DNA,and WGS is conducted on BGISEQ-500 platform.The average depth is intended to be greater than 30×for each subject.Afterwards,Sentieon software is applied to process the sequencing data under the Genome Analysis Toolkit best practice guidance to call genotypes of single nucleotide variants(SNVs)and insertion-deletions.For each included subject,21 fingerprint SNVs are genotyped by MassARRAY assays to verify that DNA sample and sequencing data originate from the same individual.The copy number variations and structural variations are also called for each patient.All of the genetic variants are annotated and predicted by bioinformatics software or by reviewing public databases.Results The average age of the included 10914 patients was 62.2±11.3 years,and 31.4%patients were women.Most of the baseline clinical characteristics of the 10914 and the excluded patients were balanced.Conclusions The WGS data together with abundant clinical and imaging data of CNSR-III could provide opportunity to elucidate the molecular mechanisms and discover novel therapeutic targets for stroke.Si Cheng Zhe Xu Yang Liu Jinxi Lin Yong Jiang Yilong Wang Xia Meng Anxin Wang Xinying Huang Zhimin Wang Guohua Chen Songdi Wu Zhengchang Jia Yongming Chen Xuerong Qiu Jun Wu Binbin Song Weizhong Ji Zhongping An Wenjun Xue Lili Zhao Yu Geng Hongyan Li Hao Li Yongjun Wang 2021Stroke & Vascular Neurology2021,6,2:8
7Optical observations of LIGO source GW 170817 by the Antarctic Survey Telescopes at Dome A,Antarctica显示文摘The LIGO detection of gravitational waves(GW) from merging black holes in 2015 marked the beginning of a new era in observational astronomy. The detection of an electromagnetic signal from a GW source is the critical next step to explore in detail the physics involved. The Antarctic Survey Telescopes(AST3),located at Dome A, Antarctica, is uniquely situated for rapid response time-domain astronomy with its continuous night-time coverage during the austral winter. We report optical observations of the GW source(GW 170817) in the nearby galaxy NGC 4993 using AST3. The data show a rapidly fading transient at around 1 day after the GW trigger, with the i-band magnitude declining from 17:23 ± 0:13 magnitude to 17:72 ± 0:09 magnitude in ~1:8 h. The brightness and time evolution of the optical transient associated with GW 170817 are broadly consistent with the predictions of models involving merging binary neutron stars. 8IGHG1 promotes motility likely through epithelial-mesenchymal transition in ovarian cancer显示文摘Objective: Ovarian cancer(OC) is one of the leading causes of death for female cancer patients. COC166-9 is an OC-specific monoclonal antibody and we have identified immunoglobulin γ-1 heavy chain constant region(IGHG1) as its antigen. We explore the function of IGHG1 in proliferation, apoptosis and motility of OC cells further in this research.Methods: IGHG1 expression in OC specimens was detected through immunohistochemistry. Real-time quantitative polymerase chain reaction(RT-q PCR) or western blotting assay was used to test IGHG1 expression in OC cells. Viability of OC cells was tested by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT)assay. Flow cytometry or western blotting assay was used to detect cell cycle and apoptosis. Cellular motility was analyzed by using transwell assay and the markers of epithelial-mesenchymal transition(EMT) were tested through immunoblots.Results: Although it exerts negligible effect on the viability and apoptosis of OC cells, IGHG1 could promote migration and invasion of malignant cells in vitro. Mechanistically, IGHG1 increases the expression of N-cadherin and Vimentin while decreases E-cadherin expression. Additionally, IGHG1 expression in OC specimens is higher relative to the paired normal counterparts. Further analysis demonstrates that the increased IGHG1 expression correlates positively with the lymph node metastasis of OC.Conclusions: IGHG1 promotes the motility of OC cells likely through executing the EMT program. 9Nano-needle strontium-substituted apatite coating enhances osteoporotic osseointegration through promoting osteogenesis and inhibiting osteoclastogenesis显示文摘Implant loosening remains a major clinical challenge for osteoporotic patients.This is because osteoclastic bone resorption rate is higher than osteoblastic bone formation rate in the case of osteoporosis,which results in poor bone repair.Strontium(Sr)has been widely accepted as an anti-osteoporosis element.In this study,we fabricated a series of apatite and Sr-substituted apatite coatings via electrochemical deposition under different acidic conditions.The results showed that Ca and Sr exhibited different mineralization behaviors.The main mineralization products for Ca were CaHPO4⋅2H2O and Ca3(PO4)2 with the structure changed from porous to spherical as the pH values increased.The main mineralization products for Sr were SrHPO4 and Sr5(PO4)3OH with the structure changed from flake to needle as the pH values increased.The in vitro experiment demonstrated that coatings fabricated at high pH condition with the presence of Sr were favorable to MSCs adhesion,spreading,proliferation,and osteogenic differentiation.In addition,Sr-substituted apatite coatings could evidently inhibit osteoclast differentiation and fusion.Moreover,the in vivo study indicated that nano-needle like Sr-substituted apatite coating could suppress osteoclastic activity,improve new bone formation,and enhance bone-implant integration.This study provided a new theoretical guidance for implant coating design and the fabricated Sr-substituted coating might have potential applications for osteoporotic patients.Zhen Geng Luli Ji Zhaoyang Li Jing Wang Hongyan He Zhenduo Cui Xianjin Yang Changsheng Liu 2021Bioactive Materials2021,6,4:4
10Hybrid inversion method for equivalent electric charge of thunder cloud based on multi-station atmospheric electric field显示文摘This article proposes the hybrid method to inverse the equivalent electric charge of thunder cloud based on the data of multi-station atmospheric electric field. Firstly,the method combines the genetic algorithm( GA) and New ton method through the mosaic hybrid structure. In addition,the thunder cloud equivalent charge is inversed based on the forw ard modeling results by giving the parameters of the thunder cloud charge structure. Then an ideal model is built to examine the performance compared to the nonlinear least squares method. Finally,a typical thunderstorms process in Nanjing is analyzed by Genetic-New ton algorithm with the help of weather radar. The results show the proposed method has the strong global searching capability so that the problem of initial value selection can be solved effectively,as well as gets the better inversion results. 11A mobile polar atmospheric parameter measurement system:Ⅱ. First atmospheric turbulence observation at Antarctic Taishan Station显示文摘This is the second paper of a series devoted to atmospheric optical turbulence C2 n observation using a mobile polar atmospheric parameter measurement system. We present the initial results of C2 n measurement at Antarctic Taishan Station using micro-thermal sensors and a three-dimensional sonic anemometer at height ~2.0 m above the snow surface. The site testing experiments were carried out during the 30 th Chinese National Antarctic Research Expedition(CHINARE). We collected about 1 000 h of data between 30 December 2013 and 10 February 2014. The C2 n curve exhibits clear daily structures, with two peaks around midnight and midday and two troughs around 7:30 and 17:00 local time(UTC+5). The mean C2 n is 2.7×10-15 m-2/3 and the 25 th and 75 th percentiles of the C215 n cumulative distribution are 9.6×10-16 m-2/3 and 6.2×10- m-2/3, respectively. 12Dual-responsive nanovaccine for cytosolic delivery of antigens to boost cellular immune responses and cancer immunotherapy显示文摘Cancer vaccine contributing to the success of the treatment and prevention of tumors has attracted a huge attention as a strategy for tumor immunotherapy in recent years.A major challenge of cancer vaccine is to target cytosols of dendritic cells(DCs)in the lymph nodes(LNs)to enhance efficiency of antigen cross-presentation,which elicits high levels of cytotoxic T-lymphocytes to destruct tumor cells.Here,we address this issue by conjugating ovalbumin(OVA)to PEG-PCL using disulfide bond(-ss-),and the degradable pH-responsive polymer-PEI-PCL as delivery carrier.In addition,the mol ratio of PEG-PCL to PEI-PCL in the mixed micelles was tailored to deliver the OVA to LNs.Subsequently,CpG ODN_(1826),a TLR-9 agonist,was further introduced into a mixed micelle of 30nm or less as a unique tumor vaccine.Importantly,the results demonstrated the mixed micelles with 1:1mol of PCL-PEG and PCL-PEI can effectively migrate to distal LNs where antigen were efficiently captured by DCs,meanwhile,OVA was modified to the surface of mixed micelles via disulfide bonds(-ss-)for promotion efficiency of antigen cross-presentation.More surprisingly,combination of tumor vaccine with anti-PD-1,the therapy of ectopic melanoma(B16-OVA)and lung metastasis melanoma(B16-OVA)is excellent therapeutic effect.Taken together,our works offers a novel strategy for the cytosol delivery of antigens to achieve potent cancer immunotherapy.Yang Sui Ji Li Jiqiang Qu Ting Fang Hongyan Zhang Jian Zhang Zheran Wang Mingyu Xia Yinghui Dai Dongkai Wang 2022Asian Journal of Pharmaceutical Sciences2022,17,4:1

17Three-dimensional biogenic C-doped Bi_2MoO_6/In_2O_3-ZnO Z-scheme heterojunctions derived from a layered precursor显示文摘Novel 3D biogenic C-doped Bi_2 MoO_6/In_2O_3-ZnO Z-scheme heterojunctions were synthesized for the first time, using cotton fiber as template. The as-prepared samples showed excellent adsorption and photodegradation performance toward the hazardous antibiotic doxycycline under simulated sunlight irradiation. The morphology, phase composition and in situ carbon doping could be precisely controlled by adjusting processing parameters. The carbon doping in Bi_2 MoO_6/In_2O_3-ZnO was derived from the cotton template, and the carbon content could be varied in the range 0.9–4.4 wt.% via controlling the heat treatment temperature. The sample with Bi_2 MoO_6/In_2O_3-ZnO molar ratio of 1:2 and carbon content of1.1 wt.% exhibited the highest photocatalytic activity toward doxycycline degradation,which was 3.6 and 4.3 times higher than those of pure Bi_2 MoO_6 and Zn In Al-CLDH(calcined layered double hydroxides), respectively.
