1Anlotinib has good efficacy and low toxicity: a phase Ⅱ study of anlotinib in pre-treated HER-2 negative metastatic breast cancer显示文摘Objective:Anlotinib is a novel tyrosine kinase inhibitor blocking angiogenesis.This study was performed to assess the efficacy and safety of anlotinib in patients with metastatic breast cancer.Methods:Patients with HER2-negative breast cancer,who were pre-treated with anthracycline or taxanes in a neoadjuvant,adjuvant,or metastatic setting,and had treatment failure after at least one prior chemotherapy regimen in the metastatic setting were enrolled.Anlotinib was administered at 12 mg daily for 14 days in a 21-day cycle until disease progression or unacceptable toxicity occurred.Simultaneously,5–10 m L of venous blood was collected to perform circulating tumor DNA(ct DNA)testing every 2 treatment cycles.The primary endpoint was the objective response rate(ORR).Secondary endpoints included the disease control rate(DCR),progression-free survival(PFS),overall survival,safety,and biomarkers.Results:Twenty-six eligible patients were enrolled,with a median age of 56(30–75)years.The median follow-up time was 10.5 months.The ORR was 15.4%,the DCR was 80.8%,and the median PFS was 5.22 months(95%confidence interval 2.86–6.24).Fourteen(53.8%)patients survived for more than 10 months.The changes in the detectable ct DNA variant allele frequency were consistent with the tumor response.The most common treatment-related adverse events were hypertension(57.7%),thyroidstimulating hormone elevation(34.6%),and hand-foot syndrome(23.1%).Conclusion:Anlotinib showed objective efficacy with tolerable toxicity in heavily pre-treated,metastatic HER2-negative breast cancer.The dynamic changes in the ct DNA variant allele fraction may be predictive of the tumor response.Nanlin Hu Yiran Si Jian Yue Tingting Sun Xue Wang Zhuqing Jia Songlin Gao Qiao Li Yang Shao Jiayu Wang Yang Luo Fei Ma Binghe Xu Peng Yuan 2021Cancer Biology & Medicine2021,18,3:11
2Biomass-derived porous carbon highly efficient for removal of Pb(Ⅱ) and Cd(Ⅱ)显示文摘The utilization of abundant and renewable biomass to fabricate advanced functional materials is considered a promising route for environmental applications.Herein,Lignin-based porous carbon with layered graphene-like structure(LPC)is successfully synthesized and applied to efficiently remove Pb(Ⅱ)and Cd(Ⅱ).The as-synthesized LPC materials are systematically characterized and these results show that LPC has a porous graphene-like structure,facilitating the diffusion and immobilization of heavy metal ions.The influence of different reaction parameters(solution pH,initial concentration of metal ions,contact time and adsorbent amount)on the adsorption performance is investigated in details.The results demonstrate that LPC can achieve superior adsorption capacities of 250.5 mg·g^-1 for Pb(Ⅱ)and 126.4 mg·g^-1 for Cd(Ⅱ),which are far superior to the previously reported adsorbents.Pseudo-second order kinetics model and Freundlich isotherm model describe the adsorption process well.Furthermore,the exhausted LPC can be regenerated easily and exhibits the removal efficiency of 96%and 92%for Pb(Ⅱ)and Cd(Ⅱ)after five continuous runs,respectively.This study shows a sustainable strategy for the design of porous carbon material from na?ve biomass and highlights the great potential in wastewater treatment.Anqi Wang Zhikeng Zheng Ruiqi Li Di Hu Yiran Lu Huixia Luo Kai Yan 2019Green Energy & Environment2019,4,4:10
3Metastatic status of sentinel lymph nodes in breast cancer determined with photoacoustic microscopy via dual-targeting nanoparticles显示文摘Detection of sentinel lymph nodes(SLNs)is critical to guide the treatment of breast cancer.However,distinguishing metastatic SLNs from normal and inflamed lymph nodes(LNs)during surgical resection remains a challenge.Here,we report a CD44 and scavenger receptor class B1 dual-targeting hyaluronic acid nanoparticle(5K-HA-HPPS)loaded with the near-infra-red fluorescent dye DiR-BOA for SLN imaging in breast cancer.The small sized(~40 nm)self-assembled 5K-HA-HPPSs accumulated rapidly in the SLNs after intradermal injection.Compared with normal popliteal LNs(N-LN),there were ~3.2-fold and ~2.4-fold increases in fluorescence intensity in tumour metastatic SLNs(T-MLN)and inflamed LNs(Inf-LN),respectively,6 h after nanoparticle inoculation.More importantly,photoacoustic microscopy(PAM)of 5KHA-HPPS showed a significantly distinct distribution in T-MLN compared with N-LN and Inf-LN.Signals were mainly distributed at the centre of T-MLN but at the periphery of N-LN and Inf-LN.The ratio of PA intensity(R)at the centre of the LNs compared with that at the periphery was 5.93±0.75 for T-MLNs of the 5K-HA-HPPS group,which was much higher than that for the Inf-LNs(R=0.2±0.07)and N-LNs(R=0.45±0.09).These results suggest that 5K-HA-HPPS injection combined with PAM provides a powerful tool for distinguishing metastatic SLNs from pLNs and inflamed LNs,thus guiding the removal of SLNs during breast cancer surgery.Yanfeng Dai Xiang Yu Jianshuang Wei Fanxin Zeng Yiran Li Xiaoquan Yang Qingming Luo Zhihong Zhang 2020Light(Science & Applications)2020,9,1:6

6A fine acquisition algorithm based on fast three-time FRFT for dynamic and weak GNSS signals显示文摘As high-dynamics and weak-signal are of two primary concerns of navigation using Global Navigation Satellite System(GNSS)signals,an acquisition algorithm based on threetime fractional Fourier transform(FRFT)is presented to simplify the calculation effectively.Firstly,the correlation results similar to linear frequency modulated(LFM)signals are derived on the basis of the high dynamic GNSS signal model.Then,the principle of obtaining the optimum rotation angle is analyzed,which is measured by FRFT projection lengths with two selected rotation angles.Finally,Doppler shift,Doppler rate,and code phase are accurately estimated in a real-time and low signal to noise ratio(SNR)wireless communication system.The theoretical analysis and simulation results show that the fast FRFT algorithm can accurately estimate the high dynamic parameters by converting the traditional two-dimensional search process to only three times FRFT.While the acquisition performance is basically the same,the computational complexity and running time are greatly reduced,which is more conductive to practical application.PAN YI ZHANG Sheng WANG Xiao LIU Manhao LUO Yiran 2023Journal of Systems Engineering and Electronics2023,34,2:1

8Classification of clustered microseismic events in a coal mine using machine learning显示文摘Discrimination of seismicity distributed in different areas is essential for reliable seismic risk assessment in mines.Although machine learning has been widely applied in seismic data processing,feasibility and reliability of applying this technique to classify spatially clustered seismic events in underground mines are yet to be investigated.In this research,two groups of seismic events with a minimum local magnitude(ML) of-3 were observed in an underground coal mine.They were respectively located around a dyke and the longwall face.Additionally,two types of undesired signals were also recorded.Four machine learning methods,i.e.random forest(RF),support vector machine(SVM),deep convolutional neural network(DCNN),and residual neural network(ResNN),were used for classifying these signals.The results obtained based on a primary dataset showed that these seismic events could be classified with at least 91% accuracy.The DCNN using seismogram images as the inputs reached the best performance with more than 94% accuracy.As mining is a dynamic progress which could change the characteristics of seismic signals,the temporal variance in the prediction performance of DCNN was also investigated to assess the reliability of this classifier during mining.A cascaded workflow consisting of database update,model training,signal prediction,and results review was established.By progressively calibrating the DCNN model,it achieved up to 99% prediction accuracy.The results demonstrated that machine learning is a reliable tool for the automatic discrimination of spatially clustered seismicity in underground mining.Yi Duan Yiran Shen Ismet Canbulat Xun Luo Guangyao Si 2021Journal of Rock Mechanics and Geotechnical Engineering2021,13,6:1
9Effects of Grape Seeds and Tea Polyphenols on Serum Enzymatic Indices of Finless Eels ( Monopterus albus )显示文摘[Objectives]This study was conducted to investigate the possibility of adding grape seeds and tea polyphenols to improve the health of finless eels(Monopterus albus).[Methods]Twelve cages were selected in the same water body and divided into 4 groups,each with 3 parallel groups.The control group was fed with common finless eel feed,and the experimental groups were fed with feed supplemented with grape seeds,tea polyphenols,and grape seed-tea polyphenol compound(with a ratio of grape seeds to tea polyphenols at 1∶1),respectively.The feeding amount of each additive was 1%.The experiment was carried out by feeding medicated feed for 3 d and ordinary feed for 3 d,that is,alternately feeding medicated feed and ordinary feed.The experiment lasted for 18 d.[Results]Six serum enzymatic indices were detected after the breeding.The results showed that compared with the control group,the three groups of samples had a certain protective effect on the liver in terms of liver function indices.In terms of immunity evaluation,the tea polyphenol group and the grape seed-tea polyphenol complex group could increase the immunity of eels to varying degrees.In terms of antioxidant capacity,all three groups of samples could increase the body's antioxidant capacity.The results of this study showed that adding tea polyphenols and grape seeds to the feed simultaneously could effectively protect the liver of fish,and simultaneously enhance their antioxidant and non-specific immune functions.[Conclusions]This study provides a reference for the application of the two Chinese herbal medicines,grape seeds and tea polyphenols,in finless eel farming.Wenhuan ZHU Zhaohui WEI Tianle HUANG Yangzhi LUO Shaomei YU Yiran LIU Shengyuan ZHANG Ping DENG Guizhen DING Hongliang XU Liqiang ZHANG 2022Agricultural Biotechnology2022,11,2:0
10Analysis of the Immune Response by Standardized Whole‑Blood Stimulation with Metabolism Modulation显示文摘The immune system defends the body from infection and plays a vital role in a wide range of health conditions.Metabolism afects a series of physiological processes,including those linked to the function of human immune system.Cellular metabolism modulates immune cell activation and cytokine production.Understanding the relationship between metabolism and immune response has important implications for the development of immune-based therapeutics.However,the deployment of large-scale functional assays to investigate the metabolic regulation of immune response has been limited by the lack of standardized procedures.Here,we present a protocol for the analysis of immune response using standardized whole-blood stimulation with metabolism modulation.Diverse immune stimuli including pattern recognition receptor(PRR)ligands and microbial stimuli were incubated with fresh human whole blood.The metabolic inhibitors were used to modulate metabolic status in the immune cells.The variable immune responses after metabolic interventions were evaluated.We described in detail the main steps involved in the whole-blood stimulation and cytokines quantifcation,namely,collection and treatment of whole blood,preparation of samples and controls,cytokines detection,and stimulation with metabolic interventions.The metabolic inhibitors for anabolic pathways and catabolic pathways exert selective efects on the production of cytokines from immune cells.In addition to a robust and accurate assessment of immune response in cohort studies,the standardized whole-blood stimulation with metabolic regulation might provide new insights for modulating immunity.Jialin Zhao Xuling Han Helian Li Yali Luo Yan Fang Yun Wang Jian Gao Yiran Zhao Jingxuan Han Feng Qian 2024Phenomics2024,4,1:0

12Creating a semi-opened micro-cavity ovary through sacrificial microspheres as an in vitro model for discovering the potential effect of ovarian toxic agents显示文摘The bio-engineered ovary is an essential technology for treating female infertility.Especially the development of relevant in vitro models could be a critical step in a drug study.Herein,we develop a semi-opened culturing system(SOCS)strategy that maintains a 3D structure of follicles during the culture.Based on the SOCS,we further developed micro-cavity ovary(MCO)with mouse follicles by the microsphere-templated technique,where sacrificial gelatin microspheres were mixed with photo-crosslinkable gelatin methacryloyl(GelMA)to engineer a micro-cavity niche for follicle growth.The semi-opened MCO could support the follicle growing to the antral stage,secreting hormones,and ovulating cumulus-oocyte complex out of the MCO without extra manipulation.The MCO-ovulated oocyte exhibits a highly similar transcriptome to the in vivo counterpart(correlation of 0.97)and can be fertilized.Moreover,we found that a high ROS level could affect the cumulus expansion,which may result in anovulation disorder.The damage could be rescued by melatonin,but the end of cumulus expansion was 3h earlier than anticipation,validating that MCO has the potential for investigating ovarian toxic agents in vitro.We provide a novel approach for building an in vitro ovarian model to recapitulate ovarian functions and test chemical toxicity,suggesting it has the potential for clinical research in the future.Min Ye Yiran Shan Bingchuan Lu Hao Luo Binhan Li Yanmei Zhang Zixuan Wang Yuzhi Guo Liliang Ouyang Jin Gu Zhuo Xiong Ting Zhang 2023Bioactive Materials2023,,8:0
