2Direct technetium-99m labeling of anti-hepatoma monoclonal antibody fragment:a radioimmunoconjugate for hepatocellular carcinoma imaging显示文摘AIM To directly radiolabel an anti-hepatomamAb fragment HAb18 F(ab’)2 with 99mTc bystannous-reduced method,and assess thestability,biodistribution and radioimmun-oimaging(RⅡ).METHODS Immunoreactive fraction wasdetermined according to Lindmo’s method.Ellman’s reagent was used to determine thenumber of thiols in the reduced F(ab’)2.Labelingefficiency and homogeneity were measured bypaper chromatography,sodium dodecylsulphatepolyacrylamide gel electrophoresis(SDS-PAGE)and autoradiography.Challenge assay involvedthe incubation of aliquots of labeled antibody inethylenediaminetetraacetate( EDTA )and L-cysteine(L-cys)solutions with different molarratio at 37℃ for 1h,respectively.Investigationsin vivo utilized nude mice bearing humanhepatocellular carcinoma(HHCC)xenograftswith gamma camera imaging and tissuebiodistribution studies at regular intervals.RESULTS The labeling procedure was finishedwithin 1.5 h compared with the'pretinning'method which would take at least 21h.In vitrostudies demonstrated that the radiolabeled mAbfragment was homogeneous and retained itsimmunoreactivity.Challenge studies indicatedthat 99mTc-labeled HAb18 F(ab’)2 in EDTA is morestable than in L-cys.Imaging and biodistribution showed a significant tumor uptake at 24 h post-injection of 99mTc-labeled HAb18 F(ab’)2.Theblood,kidney,liver and tumor uptakes at 24hwere 0.56±0.09,56.45±11.36,1.43±0.27 and6.57±3.01(%ID/g),respectively.CONCLUSION 99mTc-HAb18 F(ab’)2 conjugateprepared by this direct method appears to be aneffective way to detect hepatoma in nude micemodel.Hui Jie Bian Zhi Nan Chen Jing Lan Deng 2000World Journal of Gastroenterology2000,6,3:13

4Catalysis of Manganese(II) on Chromium(VI) Reduction by Citrate显示文摘The catalysis of manganese(II) (Mn2+) on chromium(VI) (Cr6+) reduction by citrate was studied through batch experiments with the concentration of citrate greatly in excess of Cr6+ at 25℃and in pH ranges of 4.0 to 5.0. Results showed that at pH 4.5 within 22 h direct reduction of Cr6+ by citrate was not observed, but for the same time when Mn2+ (50 to 200μmol L-1) was added, nearly all Cr6+ was reduced, with the higher initial Mn2+ concentration having faster Cr6+ reduction. In the initial stage of the reaction, the Cr6+ reduction could be described with a pseudo-first-order kinetics equation. In the later stage of the reaction, plots of lnc(Cr6+) versus t, where c(Cr6+) is the Cr6+ concentration in the reaction and t is the reaction time, deviated from the initial linear trend. The deviations suggested that the pseudo-first-order kinetics did not apply to the whole experimental period and that some reaction intermediates could have greatly accelerated Cr6+ reduction by citrate. The catalysis of the intermediates increased with the reaction time and gradually reached stability. Then, the plot of lnc(Cr6+) versus t in the presence of Mn2+ was linear again, with the rate constant increasing by 102 times compared with the absence of Mn2+. Complexation between Mn2+ and citrate was likely a prerequisite for the catalysis of Mn2+ on the reaction. 5Drift and deposition of pesticide applied by UAV on pineapple plants under different meteorological conditions显示文摘Spray drift has always been a focus research area in the field of unmanned aerial vehicle(UAV)application.Under the fixed premises of UAV operating parameters,such as height,speed and spraying liquid,the droplet drift is mainly affected by meteorological conditions.In this research,the spray drift and deposition tests were conducted using a QuanFeng120 UAV in a pineapple field under various different meteorological conditions.The experimental results showed that with the changes of UAV operating height and wind speed,the start position of the in-swath deposition area changed 4 m in the extreme situation.The percentage of the total spray drift was from 15.42%to 55.76%.The position of cumulative spray drift that accounted for 90%of the total spray drift was from 3.70 m to 46.50 m relative to the flight line.According to the downwind spray drift curve,the nonlinear equations of the same type under the four operating conditions of the UAV were fitted.The spray drift and the deposition of UAV application were significantly affected by different meteorological conditions and UAV operating heights.The results could provide a theoretical basis for UAV spraying in pineapple plants and support for spray drift control and prediction.Juan Wang Yubin Lan Huihui Zhang Yali Zhang Sheng Wen Weixiang Yao Jiajian Deng 2018International Journal of Agricultural and Biological Engineering2018,11,6:10
6Numerical simulation of land subsidence and verification of its character for an iron mine using sublevel caving显示文摘Combined with a digital bored photography system and in-situ statistics concerning the joints and fissures of both ore-body and surrounding rock,a 2D discrete model was constructed using UDEC.The stress field and displacement field changes of different sublevel stoping systems were also studied.Changes in the overlying rock strata settlement pattern has been analyzed and validated by in-situ monitoring data.The results show that:in the caving process,there exists an obvious delay and jump for the overlying rock strata displacement over time,and a stable arch can be formed in the process of caving,which leads to hidden goafs.Disturbed by the mining activity,a stress increase occurred in both the hanging wall and the foot wall,demonstrating a hump-shaped distribution pattern.From the comparison between simulation results and in-situ monitoring results,land subsidence shows a slow-development,suddenfailure,slow-development cycle pattern,which leads eventually to a stable state.This pattern validates the existence of balanced arch and hidden goafs.Cao Shuai Song Weidong Deng Dan Lei Yuankun Lan Jianqiang 2016International Journal of Mining Science and Technology2016,26,2:9

8Combined effects of p53 and MDM2 polymorphisms on susceptibility and surgical prognosis in hepatitis B virus-related hepatocellular carcinoma显示文摘The p53 signaling pathway works as a potent barrier to tumor progression.Two single nucleotide polymor-phisms(SNPs)in the gene loci of p53 pathway,p53 codon 72 Arg72Pro and MDM2 SNP309(T>G),have been shown to cause perturbation of p53 function,but the effect of the two SNPs on the risk of hepatocellular carcinoma(HCC)remains inconsistent.This study in-vestigated the influence of combined p53 Arg72Pro and MDM2 SNP309 on the risk of developing HCC in pa-tients with chronic hepatitis B virus infection,and evaluated the significance of the two combined SNPs on patient prognosis.In total,350 HCC patients,230 non-HCC patients,and 96 healthy controls were geno-typed for the p53 Arg72Pro and MDM2 SNP309.The combined p53 Pro/Pro and MDM2 G/G genotype was significantly associated with HCC risk(P=0.047).Mul-tivariate analysis indicated that combined p53 Pro/Pro and MDM2 G/G genotype was an independent factor affecting recurrence and survival(P<0.05).Patients with combined p53 Pro/Pro and MDM2 G/G genotypes had a poorer prognosis than other genotypes,P<0.01 for both disease-free survival(DFS)and overall survival(OS).DFS and OS rates also differed significantly be-tween Barcelona Clinic Liver Cancer(BCLC)stage A patients with combined p53 Pro/Pro and MDM2 G/G and other genotypes(P<0.05).Thus,the combined p53 Pro/Pro and MDM2 G/G genotype is associated with increased risk of developing HCC and is an independ-ent adverse prognostic indicator in early stage HCC.Yun Yang Tian Xia Ning Li Jin Zhang Yuan Yang Wenming Cong Qiang Deng Ke Lan Weiping Zhou 2013Protein & Cell2013,4,1:8

10Catalysis of Dissolved and Adsorbed Iron in Soil Suspension for Chromium(VI) Reduction by Sulfide显示文摘The kinetics of Cr(VI) reduction by sulfide in soil suspensions with various pHs, soil compositions, and Fe(II) concentrations was examined using batch anaerobic experimental systems at constant temperature. The results showed that the reaction rate of Cr(VI) reduction was in the order of red soil < yellow-brown soil < chernozem and was proportional to the concentration of HCl-extractable iron in the soils. Dissolved and adsorbed iron in soil suspensions played an important role in accelerating Cr(VI) reduction. The reaction involved in the Cr(VI) reduction by Fe(II) to produce Fe(III), which was reduced to Fe(II) again by sulfide, could represent the catalytic pathway until about 70% of the initially present Cr(VI) was reduced. The catalysis occurred because the one-step reduction of Cr(VI) by sulfide was slower than the two-step process consisting of rapid Cr(VI) reduction by Fe(II) followed by Fe(III) reduction by sulfide. In essence, Fe(II)/Fe(III) species shuttle electrons from sulfide to Cr(VI), facilitating the reaction. The effect of iron, however, could be completely blocked by adding a strong Fe(II)-complexing ligand, 1,10-phenanthroline, to the soil suspensions. In all the experiments, initial sulfide concentration was much higher than initial Cr(VI) concentration. The plots of lnc[Cr(VI)] versus reaction time were linear up to approximately 70% of Cr(VI) reduction, suggesting a first-order reaction kinetics with respect to Cr(VI). Elemental sulfur, the product of sulfide oxidation, was found to accelerate Cr(VI) reduction at a later stage of the reaction, resulting in deviation from linearity for the lnc[Cr(VI)] versus time plots.LAN Ye-Qing YANG Jun-Xiang B. 12Preinduced intestinal HSP70 improves visceral hypersensitivity and abnormal intestinal motility in PI-IBS mouse model显示文摘Objective:To investigate the impact of the preinduced intestinal heat shock protein 70(HSP70)on the visceral hypersensitivity and abnormal intestinal motility in a post-infectious irritable bowel syndrome(PI-IBS) mouse model.Methods:Eighty-four female C57BL/6 mice were randomly assigned to four groups:control group(n=21) and induction+PI-IBS group(n=21),PI-IBS group(n=21) and induction group(n=21).The mice in PI-IBS group were infected in vivo with trichinella spiralis by oral administration.The visceral hypersensitivity and intestinal motility were evaluated respectively with abdominal withdrawal reflex and colon transportation test.The intestinal HSP70 protein and mRNA level was measured by Western blot and realtime PCR.Meanwhile,the intestinal proinflammatory cytokines IL-10 and TNF-α level was detected by ELISA.Results:Compared with their counterparts in PI-IBS group,the animals in the Induction+PI-IBS group show significantly increased intestinal level of HSP70 and obviously ameliorative clinical tigurcs.including abdominal withdrawal reflex score,intestine transportation time and Bristol scores(P<0.05).Meanwhile,the intestinal post-inflammatory cytokines remarkably changed,including increased IL-10 level and decreased TNF-αlevel(P<0.05).Conclusions:Intestinal IISP70 may play a potential protective role through improving the imbalance between the intestinal post-inflammatory and anti-inflammatory cytokines in PI-IBS.Cheng Lan Xiao-Ning Sun Xu-Chun Zhou Bo Yang Bai-Li Huang Tao-Zhi Deng Zhou-Tao He Xiang-Yang Han 2016Asian Pacific Journal of Tropical Medicine2016,9,3:7

14Anxiety symptoms in patients with Mayer-Rokitansky-Küster-Hauser syndrome:a cross-sectional study显示文摘Background:As a congenital malformation that results in infertility and an inability to have vaginal intercourse,Mayer-Rokitansky-Küster-Hauser(MRKH)syndrome places a considerable psychological burden on patients,which results in anxiety symptoms.However,only single case studies or a few small to medium-sized cross-sectional studies were identified to focus on anxiety symptoms in MRKH patients.Thus,the aim of this study was to explore the status of anxiety symptoms and the related factors in patients with MRKH syndrome.Methods:This cross-sectional study involving 141 patients with MRKH syndrome and 178 healthy women was conducted from January 2018 to December 2018.All participants were required to complete a demographic questionnaire and the Generalized Anxiety Disorder 7-item scale(GAD-7),Patient Health Questionnaire-9,Eysenck Personality Questionnaire-Revised,Short Scale for Chinese,and Chinese Version of the Female Sexual Function Index.The main outcome was the anxiety symptoms measured by the GAD-7.Main outcome was compared between the MRKH syndrome group and the healthy control group.Then,we explored the related factors by comparing patients with and without anxiety symptoms.Results:Of the respondents,24.1%experienced moderate to severe anxiety symptoms.Patients with MRKH syndrome manifested more severe anxiety symptoms than healthy women.Negative self-evaluation of femininity(odds ratio[OR]2.706,95%confidence interval[CI]1.010-7.247),neurotic personality traits(OR 1.100,95%CI 1.029-1.175),and coexisting depressive symptoms(OR 4.422,95%CI 1.498-13.049)were more prevalent in anxious patients.Conclusion:The findings stress the importance of anxiety symptom screening in MRKH patients and identify patients at risk of anxiety symptoms,providing a possible basis for future intervention.Shuang Song Na Chen Yan-Ping Duan Jia Kang Shan Deng Hong-Xin Pan Lan Zhu 2020Chinese Medical Journal2020,,4:6

18Seroprevalence of Toxoplasma gondii infection among patients with hand, foot and mouth disease in Henan, China: a hospital-based study显示文摘Background:The prevalence of infection with Toxoplasma gondii(T.gondii)in humans has been increasing in China due to the growing number of cats in the country.Hand,foot and mouth disease(HFMD)is a serious public health issue in China and still one of the leading causes of child mortality.However,little is known about the epidemiology of T.gondii infection among HFMD patients.Methods:A case–control study of 281 HFMD patients from the First People’s Hospital of Pingdingshan in Pingdingshan city,Henan province,central China,and 222 controls from Pingdingshan city was conducted.Anti-T.gondii antibodies were serologically detected using the enzyme-linked immunosorbent assay.Results:We found that the overall anti-T.gondii immunoglobulin G(IgG)antibody prevalence among HFMD patients was 12.46%,which was significantly higher than that in clinically healthy children(1.80%).The highest T.gondii seroprevalence was detected in critical cases(22.58%),followed by severe cases(11.50%),and the lowest was detected in mild cases(8.33%).Conclusion:The present study is the first survey of T.gondii seroprevalence among HFMD patients in China;12.46%were defined as seropositive.It is imperative that improved integrated measures are taken to prevent and control T.gondii infection among HFMD patients.Shuai Wang Chunwei Lan Luwen Zhang Haizhu Zhang Zhijun Yao Dong Wang Jingbo Ma Jiarong Deng Shiguo Liu 2015Infectious Diseases of Poverty2015,4,1:5
19Pseudomonas aeruginosa:pathogenesis,virulence factors,antibiotic resistance,interaction with host,technology advances and emerging therapeutics显示文摘Pseudomonas aeruginosa(P.aeruginosa)is a Gram-negative opportunistic pathogen that infects patients with cystic fibrosis,burn wounds,immunodeficiency,chronic obstructive pulmonary disorder(COPD),cancer,and severe infection requiring ventilation,such as COVID-19.P.aeruginosa is also a widely-used model bacterium for all biological areas.In addition to continued,intense efforts in understanding bacterial pathogenesis of P.aeruginosa including virulence factors(LPS,quorum sensing,two-component systems,6 type secretion systems,outer membrane vesicles(OMVs),CRISPR-Cas and their regulation),rapid progress has been made in further studying host-pathogen interaction,particularly host immune networks involving autophagy,inflammasome,noncoding RNAs,cGAS,etc.Furthermore,numerous technologic advances,such as bioinformatics,metabolomics,scRNA-seq,nanoparticles,drug screening,and phage therapy,have been used to improve our understanding of P.aeruginosa pathogenesis and host defense.Nevertheless,much remains to be uncovered about interactions between P.aeruginosa and host immune responses,including mechanisms of drug resistance by known or unannotated bacterial virulence factors as well as mammalian cell signaling pathways.The widespread use of antibiotics and the slow development of effective antimicrobials present daunting challenges and necessitate new theoretical and practical platforms to screen and develop mechanism-tested novel drugs to treat intractable infections,especially those caused by multi-drug resistance strains.Benefited from has advancing in research tools and technology,dissecting this pathogen’s feature has entered into molecular and mechanistic details as well as dynamic and holistic views.Herein,we comprehensively review the progress and discuss the current status of P.aeruginosa biophysical traits,behaviors,virulence factors,invasive regulators,and host defense patterns against its infection,which point out new directions for future investigation and add to the design of novel and/or alternative therapeutics to combat this clinically significant pathogen.Shugang Qin Wen Xiao Chuanmin Zhou Qinqin Pu Xin Deng Lefu Lan Haihua Liang Xiangrong Song Min Wu 2022Signal Transduction and Targeted Therapy2022,7,7:5
20Grade classification of neuroepithelial tumors using high-resolution magic-angle spinning proton nuclear magnetic resonance spectroscopy and pattern recognition显示文摘Clinical data have shown that survival rates vary considerably among brain tumor patients,according to the type and grade of the tumor.Metabolite profiles of intact tumor tissues measured with high-resolution magic-angle spinning proton nuclear magnetic resonance spectroscopy (HRMAS 1H NMRS) can provide important information on tumor biology and metabolism.These metabolic fingerprints can then be used for tumor classification and grading,with great potential value for tumor diagnosis.We studied the metabolic characteristics of 30 neuroepithelial tumor biopsies,including two astrocytomas (grade I),12 astrocytomas (grade II),eight anaplastic astrocytomas (grade III),three glioblastomas (grade IV) and five medulloblastomas (grade IV) from 30 patients using HRMAS 1H NMRS.The results were correlated with pathological features using multivariate data analysis,including principal component analysis (PCA).There were significant differences in the levels of N-acetyl-aspartate (NAA),creatine,myo-inositol,glycine and lactate between tumors of different grades (P<0.05).There were also significant differences in the ratios of NAA/creatine,lactate/creatine,myo-inositol/creatine,glycine/creatine,scyllo-inositol/creatine and alanine/creatine (P<0.05).A soft independent modeling of class analogy model produced a predictive accuracy of 87% for high-grade (grade III-IV) brain tumors with a sensitivity of 87% and a specificity of 93%.HRMAS 1H NMR spectroscopy in conjunction with pattern recognition thus provides a potentially useful tool for the rapid and accurate classification of human brain tumor grades.CHEN WenXue LOU HaiYan ZHANG HongPing NIE Xiu LAN WenXian YANG YongXia XIANG Yun QI JianPin LEI Hao TANG HuiRu CHEN FenEr DENG Feng 2011Science China(Life Sciences)2011,54,7:5
