1Laminar Distribution of Neurochemically-Identified Interneurons and Cellular Co-expression of Molecular Markers in Epileptic Human Cortex显示文摘Inhibitory GABAergic interneurons are fundamental elements of cortical circuits and play critical roles in shaping network activity. Dysfunction of interneurons can lead to various brain disorders, including epilepsy,schizophrenia, and anxiety. Based on the electrophysiological properties, cell morphology, and molecular identity,interneurons could be classified into various subgroups. In this study, we investigated the density and laminar distribution of different interneuron types and the coexpression of molecular markers in epileptic human cortex.We found that parvalbumin(PV) and somatostatin(SST)neurons were distributed in all cortical layers except layer I, while tyrosine hydroxylase(TH) and neuropeptide Y(NPY) were abundant in the deep layers and white matter.Cholecystokinin(CCK) neurons showed a high density in layers IV and VI. Neurons with these markers constituted*7.2%(PV), 2.6%(SST), 0.5%(TH), 0.5%(NPY), and4.4%(CCK) of the gray-matter neuron population. Doubleand triple-labeling revealed that NPY neurons were also SST-immunoreactive(97.7%), and TH neurons were more likely to express SST(34.2%) than PV(14.6%). A subpopulation of CCK neurons(28.0%) also expressed PV, but none contained SST. 2Self-assembled PEG-b-PDPA-b-PGEM copolymer nanoparticles as protein antigen delivery vehicles to dendritic cells:preparation,characterization and cellular uptake显示文摘Antigen uptake by dendritic cells(DCs)is a key step for initiating antigen-specific T cell immunity.In the present study,novel synthetic polymeric nanoparticles were prepared as antigen delivery vehicles to improve the antigen uptake by DCs.Well-defined cationic and acid-responsive copolymers,monomethoxy poly(ethylene glycol)-block-poly(2-(diisopropyl amino)ethyl methacrylate)-block-poly(2-(guanidyl)ethyl methacrylate)(mPEG-b-PDPA-b-PGEM,PEDG)were synthesized by reversible addition-fragmentation chain transfer polymerization of 2-(diisopropylamino)ethyl methacrylate and N-(tert-butoxycarbonyl)amino ethyl methacrylate monomers,followed by deprotection of tert-butyl protective groups and guanidinylation of obtained primary amines.1H NMR,13C NMR and GPC results indicated the successful synthesis of well-defined PEDG copolymers.PEDG copolymers could self-assemble into nanoparticles in aqueous solution,which were of cationic surface charges and showed acid-triggered disassembly contributed by PGEM and PDPA moieties,respectively.Significantly,PEDG nanoparticles could effectively condense with negatively charged model antigen ovalbumin(OVA)to form OVA/PEDG nanoparticle formulations with no influence on its secondary and tertiary structures demonstrating by far-UV circular dichroism and UV-vis spectra.In vitro antigen cellular uptake by bone marrow DCs(BMDCs)indicated using PEDG nanoparticles as antigen delivery vehicles could significantly improve the antigen uptake efficiency of OVA compared with free OVA or the commercialized Alum adjuvant.Moreover,as the surface cationic charges of OVA/PEDG nanoparticle formulations reduced,the uptake efficiency decreased correspondingly.Collectively,our work suggests that guanidinylated,cationic and acid-responsive PEDG nanoparticles represent a new kind of promising antigen delivery vehicle to DCs and hold great potential to serve as immunoadjuvants in the development of vaccines.Pan Li Junhui Zhou Pingsheng Huang Chuangnian Zhang Weiwei Wang Chen Lia Deling Kong 2017Regenerative Biomaterials2017,4,1:2

14Infant attraction:why social bridging matters for female leadership in Tibetan macaques显示文摘Leadership is a key issue in the study of collective behavior in social animals.Affiliation-leadership models predict that dyadic partner preferences based on grooming relationships or allianee formation positively affect an in dividua I's decision to follow or support a con spec ific.In the case of many primate species,females without young infants are attracted to mother-infant dyads.However,the effects of mother-infant-female associations on affiliation-leadership models remain less clear.In free-rangi ng Tibeta n macaques Macaca thibetana,we used social network analysis to examine the importance of mother-inf a nt-adult female'social bridging eve nts as a predictor of who leads and who follows during group movement.Social bridging is a common behavior in Tibetan macaques and occurs whe n 2 adults,gen erally females,engage in coordinated in fa nt handling.Using eigenvector centrality coefficients of social bridging as a measure of social affiliation,we found that among lactating females,initiating bridging behavior with another female played a sign讦icant role in leadership success,with the assisting female following the mother during group movement.Among non lactati ng females,this was not the case.Our results in dicate that in fa nt attracti on can be a strong trigger in collective action and directing group movement in Tibetan macaques and provides benefits to mothers who require helpers and social support in order to ensure the safety of their infants.Our study provides new insights into the importance of the third-party effect in rethinking affiliation-leadership models in group-living animals.Xi WANG Dong-Po XIA Lixing SUN Paul A.Garber Randall C.Kyes Lori K.Sheeran Bing-Hua SUN Bo-Wen Lia Jin-Hua LI 2020Current Zoology2020,66,6:1
