| 1 | OSBPL2-disrupted pigs recapitulate dual features of human hearing loss and hypercholesterolaemia显示文摘Oxysterol binding protein like 2(OSBPL2), an important regulator in cellular lipid metabolism and transport, was identified as a novel deafness-causal gene in our previous work. To resemble the phenotypic features of OSBPL2 mutation in animal models and elucidate the potential genotypephenotype associations, the OSBPL2-disrupted Bama miniature(BM) pig model was constructed using CRISPR/Cas9-mediated gene editing, somatic cell nuclear transfer(SCNT) and embryo transplantation approaches, and then subjected to phenotypic characterization of auditory function and serum lipid profiles. The OSBPL2-disrupted pigs displayed progressive hearing loss(HL) with degeneration/apoptosis of cochlea hair cells(HCs) and morphological abnormalities in HC stereocilia, as well as hypercholesterolaemia. High-fat diet(HFD) feeding aggravated the development of HL and led to more severe hypercholesterolaemia. | 2 | GLUT1-mediated effective anti-miRNA21 pompon for cancer therapy显示文摘Oncogenic microRNAs are essential components in regulating the gene expression of cancer cells. Especially miR21, which is a major player involved of tumor initiation, progression, invasion and metastasis in several cancers. The delivery of anti-miR21 sequences has significant potential for cancer treatment. Nevertheless, since anti-miR21 sequences are extremely unstable and they need to obtain certain concentration to function, it is intensely difficult to build an effective delivery system for them.The purpose of this work is to construct a self-assembled glutathione(GSH)-responsive system with tumor accumulation capacity for effective anti-miR21 delivery and cancer therapy. A novel drug delivery nanosphere carrying millions of anti-miR21 sequences was developed through the rolling circle transcription(RCT) method. GSH-responsive cationic polymer polyethyleneimine(pOEI) was synthesized to protect the nanosphere from degradation by Dicer or other RNase in normal cells and optimize the pompon-like nanoparticle to suitable size. Dehydroascorbic acid(DHA), a targeting molecule, which is a substrate of glucose transporter 1(GLUT 1) and highly expressed on malignant tumor cells, was connected to pOEI through PEG, and then the polymer was used for contracting a RNA nanospheres into nanopompons. | 4 | Application of remote sensing for investigating mining geological hazards显示文摘To investigate geological mining hazards using digital techniques such as highresolution remote sensing,a semi-automatically geological mining hazards extraction method is proposed based on the case of the Shijiaying coal mine,located in Fangshan District,Beijing,China.In the method,the vegetation is first removed using the normalized difference vegetation index(NDVI)on the GeoEye-1 data.Then,geological mining hazards interpretation features are determined after color enhancement using principal component analysis(PCA)transformation.Bitmaps mainly covered by geological mining hazards are isolated by masking operation in the environment for visualizing images software.Next,each bitmap is classified into a two-valued imagery using support vector machine algorithm.In the two-valued imagery,1 denotes the geological mining hazards,while 0 denotes none.Afterwards,the two-valued imagery is converted into a vector graph by corresponding functions in the ArcGIS software and no geological mining hazards regions in the vector graph are deleted manually.Finally,the correlation between factors(such as mining activity,lithology,geological structure,and slope)and geological mining hazards is analyzed using a logistic regression and a hazardous-area forecasting model is built.The results of field verification show that the accuracy of the geological mining hazards extraction method is 98.1%and the results of the hazardous-area forecasting indicate that the logistic regression is an effective model in assessing geological hazard risks and that mining activity is the main contributing factor to the hazards,while geological structure,slope,lithology,roughness of the surface,and aspect are the secondary. | Qinjun Wang Huadong Guo Yu Chen Qizhong Lin Hui Li | 2013 | International Journal of Digital Earth2013,6,5: | 1 |
| 5 | Emergence of superconductivity from fully incoherent normal state in an iron-based superconductor (Ba_(0.6)K_(0.4))Fe_2As_2显示文摘In unconventional superconductors, it is generally believed that understanding the physical properties of the normal state is a pre-requisite for understanding the superconductivity mechanism. In conventional superconductors like niobium or lead, the normal state is a Fermi liquid with a well-defined Fermi surface and well-defined quasipartcles along the Fermi surface. Superconductivity is realized in this case by the Fermi surface instability in the superconducting state and the formation and condensation of the electron pairs(Cooper pairing). The high temperature cuprate superconductors, on the other hand, represent another extreme case that superconductivity can be realized in the underdoped region where there is neither well-defined Fermi surface due to the pseudogap formation nor quasiparticles near the antinodal regions in the normal state. Here we report a novel scenario that superconductivity is realized in a system with well-defined Fermi surface but without quasiparticles along the Fermi surface in the normal state.High resolution laser-based angle-resolved photoemission measurements have been performed on an optimally-doped iron-based superconductor(Ba_(0.6)K_(0.4))Fe_2As_2. We find that, while sharp superconducting coherence peaks emerge in the superconducting state on the hole-like Fermi surface sheets, no quasiparticle peak is present in the normal state. Its electronic behaviours deviate strongly from a Fermi liquid system. The superconducting gap of such a system exhibits an unusual temperature dependence that it is nearly a constant in the superconducting state and abruptly closes at Tc. | 7 | Intravenous route to choroidal neovascularization by macrophage-disguised nanocarriers for mTOR modulation显示文摘Retinal pigment epithelial(RPE) is primarily impaired in age-related macular degeneration(AMD), leading to progressive loss of photoreceptors and sometimes choroidal neovascularization(CNV). mTOR has been proposed as a promising therapeutic target, while the usage of its specific inhibitor,rapamycin, was greatly limited. To mediate the mTOR pathway in the retina by a noninvasive approach, we developed novel biomimetic nanocomplexes where rapamycin-loaded nanoparticles were coated with cell membrane derived from macrophages(termed as MRaNPs). Taking advantage of the macrophage-inherited property, intravenous injection of MRaNPs exhibited significantly enhanced accumulation in the CNV lesions, thereby increasing the local concentration of rapamycin. Consequently, MRaNPs effectively downregulated the mTOR pathway and attenuate angiogenesis in the eye. Particularly, MRaNPs also efficiently activated autophagy in the RPE, which was acknowledged to rescue RPE in response to deleterious stimuli. | 8 | Morphological and physiological plasticity response to low nitrogen stress in black cottonwood(Populus deltoides Marsh.)显示文摘It is important to evaluate nitrogen use efficiency and nitrogen tolerance of trees in order to improve their productivity.In this study,both were evaluated for 338 Populus deltoides genotypes from six provenances.The plants were cultured under normal nitrogen(750μM NH_4 NO_3)and low nitrogen(5μM NH_4 NO_3)conditions for 3 months.Growth,chlorophyll content and glutamine synthetase activity of each genotype were measured.Under low nitrogen,heights,ground diameter,leaf area,leaf and root biomass,and chlorophyll contents were significantly lower than those under normal nitrogen level.Correlation analysis showed that nutrient distribution changed under different nitrogen treatments.There was a negative correlation between leaf traits and root biomass under normal nitrogen level,however,the correlation became positive in low nitrogen treatment.Moreover,with the decrease of nitrogen level,the negative correlation between leaf morphology and chlorophyll levels became weakened.The growth of the genotypes under the two treatments was evaluated by combining principal component analysis with a fuzzy mathematical membership function;the results showed that leaf traits accounted for a large proportion of the variation in the evaluation model.According to the results of comprehensive evaluation of plants under the two treatments,the 338 P.deltoides genotypes could be divided into nine categories,with wide genotypic diversity in nitrogen use efficiency and low nitrogen tolerance.As a result,26 N-efficient genotypes and 24 N-inefficient genotypes were selected.By comparative analysis of their morphological and physiological traits under the two treatments,leaf traits could be significant indicators for nitrogen use efficiency and nitrogen tolerance,which is of considerable significance for breeding poplar varieties with high nitrogen use efficiencies. | Cun Chen Yanguang Chu Qinjun Huang Changjun Ding Weixi Zhang Bo Li Jing Zhang Xiaohua Su | 2022 | Journal of Forestry Research2022,33,1: | 1 |
| 9 | EMLA■ Cream coated on endotracheal tube with or without epidural lidocaine reduces isoflurane requirement during general anesthesia显示文摘This study was designed to evaluate the effect of tracheal topical anesthesia using EMLA■Cream(EC)coated on the endotracheal tube(ETT)with or without epidural anesthesia(EA)on isoflurane requirement during general anesthesia(GA)and investigate whether EC coated on the ETT with EA was associated with the additive effect compared with the effect when each anesthetic was administered independently.The prospective randomized,double-blinded,and controlled study included 60 ASA Ⅰ-Ⅱ patients scheduled for upper abdominal surgery requiring GA.Patients were randomly assigned to one of the following groups:group 1 received GA,group 2 received EC+GA,group 3 received GA+EA,and group 4 received EC+GA+EA.Isoflurane was administered at the required concentrations to maintain the mean arterial pressure at a level not exceeding 20% of preoperative values.The percentage mean expired concentration(%MEC)was used in calculating the isoflurane requirement.Emergence agitation,postoperative sore throat,and hoarseness were recorded.Groups 2,3,and 4 exhibited a significant reduction on isoflurane requirement compared with group 1(P<0.05).The isoflurane requirement evaluated by%MEC decreased by 12%,38%,and 50% in groups 2,3,and 4,respectively.The incidence of emergence agitation was significantly lower in groups 2 and 4 than those in groups 1 and 3(P<0.05).Tracheal topical anesthesia using EC coated on ETT with or without EA reduced the isoflurane requirement during GA,indicating that EC combined with EA exhibited an additive effect on the requirement of general anesthetic. | Hai Yu Qinjun Chu Jin Liu Li Chen Ying Wang Yunxia Zuo | 2012 | Frontiers of Medicine2012,6,3: | 1 |

| 18 | Electricity-driven ammonia oxidation and acetate production in microbial electrosynthesis systems显示文摘Microbial electrosynthesis(MES)is an emerging technology for producing chemicals,and coupling MES to anodic waste oxidation can simultaneously increase the competitiveness and allow additional functions to be explored.In this study,MES was used for the simultaneous removal of ammonia from synthetic urine and production of acetate from CO_(2).Using graphite anode,83.2%±5.3%ammonia removal and 28.4%±9.9%total nitrogen removal was achieved,with an energy consumption of 1.32 kWh/g N for total nitrogen removal,0.45 kWh/g N for ammonia nitrogen removal,and 0.044 kWh/g for acetate production.Using boron-doped diamond(BDD)anode,70.9%±12.1%ammonia removal and 51.5%±11.8%total nitrogen removal was obtained,with an energy consumption of 0.84 kWh/g N for total nitrogen removal,0.61 kWh/g N for ammonia nitrogen removal,and 0.043 kWh/g for acetate production.A difference in nitrate accumulation explained the difference of total nitrogen removal efficiencies.Transport of ammonia and acetate across the membrane deteriorated the performance of MES.These results are important for the development of novel elcctricity-driven technologies for chemical production and pollution removal. | Qinjun Liang Yu Gao Zhigang Li Jiayi Cai Na Chu Wen Hao Yong Jiang Raymond Jianxiong Zeng | 2022 | Frontiers of Environmental Science & Engineering2022,16,4: | 0 |
| 19 | Activation of p38/HSP27 pathway counters melatonin-induced inhibitory effect on proliferation of human gastric cancer cells显示文摘Overexpression of heat shock protein 27(HSP27)in gastric cancer is correlated with poor clinical prognosis.Melatonin,an endogenous hormone,shows promise in gastric cancer therapy.However,there is limited study on the biological activity of HSP27 in response to melatonin treatment.In this study,we show an anti-proliferative action of melatonin on human gastric cancer cell lines BGC-823 and MGC-803.Biochemically,the inhibitory effect of melatonin is accompanied by the upregulation of HSP27 phosphorylation level.Transfection of gastric cancer cells with HSP27-specific siRNA effectively reduces HSP27 phosphorylation and potentiated melatonininduced inhibitory effect on cell proliferation.The reduction of cyclin D1 in melatonin-treated cells is also aggravated by HSP27 depletion.Moreover,melatonin stimulation increases p38 phosphorylation.Pretreatment with p38 inhibitor SB203580 not only remarkably suppresses melatonin-induced HSP27 phosphorylation,but also augment the inhibitory effect of melatonin on cyclin D1 expression as well as cell proliferation.Taken together,our study indicates the protective pathway of p38/HSP27 against melatonin-induced inhibitory effect on gastric cancer cell proliferation,suggesting that combined with p38/HSP27 pathway inhibitor,the therapeutic efficacy of melatonin on gastric cancer may be improved. | Chenchen Zhu Haonan Jiang Wenjie Deng Shuo Zhao Kaiquan Li Yuting Wang Qinjun Wei Jun Du | 2019 | The Journal of Biomedical Research2019,33,5: | 0 |
