2Effect of proteolytic starter culture isolated from Chinese Dong fermented pork(Nanx Wudl)on microbiological,biochemical and organoleptic attributes in dry fermented sausages显示文摘The effect of a proteolytic starter culture isolated from Nanx Wudl,on microbiological,biochemical and organoleptic attributes of dry fermented sausages was investigated during processing.Based on preliminary screening,the combination of Staphylococcus xylosus SX16 and Lactobacillus plantarum CMRC6,showing excellent proteolytic activity in vitro,was selected as the multi-strain starter(starter LS).For comparison,the single-strain starter culture of L.plantarum CMRC6(starter LB)and non-inoculated control were also tested.During fermentation,lactic acid bacteria and staphylococci dominated the microbiota and suppressed the Enterobacteriaceae growth in LS-inoculated sausages.The addition of LS starter accelerated acidification and proteolysis during ripening,improving the contents of total free amino acids and several essential free amino acids(Phe,Ile and Leu).Volatile compounds analysis revealed that LS-fermented sausage showed the highest abundance of 3-methyl-1-butanol,probably due to the inoculated S.xylosus.The inoculation of LS starter improved the sensory properties of sausages,especially the flavor attribute.Therefore,S.xylosus SX16 and L.plantarum CMRC6 are promising candidates for inclusion as multi-strain starters in the manufacture of gourmet fermented dry sausage.Xi Chen Ruifang Mi Biao Qi Suyue Xiong Jiapeng Li Chao Qu Xiaoling Qiao Wenhua Chen Shouwei Wang 2021Food Science and Human Wellness2021,10,1:9
3Chemical dehydration coupling multi-effect evaporation to treat waste sulfuric acid in titanium dioxide production process显示文摘In order to concentrate the diluted sulfuric acid from the titanium dioxide(TiO2)production of sulphate process,a new concentration process was proposed by coupling chemical dehydration and multi-effect evaporation.The ferrous sulfate monohydrate(FeSO4·H2O),as the dehydrant,was added to the diluted sulfuric acid to form ferrous sulfate heptahydrate(FeSO4·7H2O)according to the H2SO4-FeSO4-H2O phase diagrams,which partially removes the water.This process was named as Chemical Dehydration Process.The residual water was further removed by two-effect evaporation and finally 70 wt%sulfuric acid was obtained.The FeSO4·H2O can be regenerated through drying and dehydration of FeSO4·7H2O.The results show that FeSO4·H2O is the most suitable dehydrant,the optimal reaction time of chemical dehydration process is 30 min,and low temperature is favorable for the dehydration reaction.45.17%of the entire removed water can be removed by chemical dehydration from the diluted sulfuric acid.This chemical dehydration process is also energy efficient with 24.76%saving compared with the direct evaporation process.Furthermore,51.21%of the FeSO4 dissolved originally in the diluted sulfuric acid are precipitated out during the chemical dehydration,which greatly reduces the solid precipitation and effectively alleviates the scaling in the subsequent multi-effect evaporation process.Hongyin Pang Ruifang Lu Tao Zhang Li Lü Yanxiao Chen Shengwei Tang 2020Chinese Journal of Chemical Engineering2020,28,4:7
4Differential expression of immune-related genes between healthy volunteers and type 2 diabetic patients with spleen-deficiency pattern显示文摘OBJECTIVE: To investigate the clinical differentiation of spleen-deficiency pattern(SDP), a group of symptoms and signs defined in terms of Traditional Chinese Medicine for its clinical practice.METHODS: Peripheral venous blood(> 3 m L) was collected from each of six type 2 diabetes mellitus(T2DM)-SDP patients and six healthy volunteers. After the isolation of peripheral white blood cells(PWBCs), total RNA was extracted, and quality control was performed on all RNA samples. Microarray experiments were conducted using the Agilent human whole genome gene chip, and genes demonstrating differential expression were screened. Bioinformatics analysis was conducted on these genes using several online databases.RESULTS: We screened a total of 175 differentially expressed genes(DEGs), of which 111(63%) were down-regulated and 64(37%) were up-regulated in T2DM-SDP patients compared with healthy controls. Among the 175 genes, 158 had biological function annotations: 46(29%) were directly related to an individual's immune regulation or response, 25(16%) were associated with substance and energy metabolism of PWBCs which could also indirectly influence immunity, and the remaining87(55%) were involved in a variety of PWBC biological processes that might eventually influence the immune function. Kyoto Encyclopedia of Genes and Genomes pathway enrichment analysis revealed that the DEGs were predominantly enriched in seven immune-related pathways. Hierarchical cluster analysis identified gene expression patterns that were distinguishable between the two study groups.CONCLUSION: Our results suggest that T2DM-SDP patients experience significant hypoimmunity and/or immune dysfunctions, and possess a specific gene expression profile. 5The heterogeneity of islet autoantibodies and the progression of islet failure in type 1 diabetic patients显示文摘Type 1 diabetes mellitus is heterogeneous in many facets. The patients suffered from type 1 diabetes present several levels of islet function as well as variable number and type of islet-specific autoantibodies. This study was to investigate prevalence and heterogeneity of the islet autoantibodies and clinical phenotypes of type 1 diabetes mellitus; and also discussed the process of islet failure and its risk factors in Chinese type 1 diabetic patients. A total of 1,291 type 1 diabetic patients were enrolled in this study. Demographic information was collected. Laboratory tests including mixed-meal tolerance test, human leukocyte antigen alleles, hemoglobin A1 c, lipids, thyroid function and islet autoantibodies were conducted. The frequency of islet-specific autoantibody in newly diagnosed T1 DM patients(duration shorter than half year) was 73% in East China. According to binary logistic regressions, autoantibody positivity, longer duration and lower Body Mass Index were the risk factors of islet failure. As the disease developed, autoantibodies against glutamic acid decarboxylase declined as well as the other two autoantibodies against zinc transporter 8 and islet antigen 2. The decrease of autoantibodies was positively correlated with aggressive beta cell destruction. Autoantibodies can facilitate the identification of classic T1 DM from other subtypes and predict the progression of islet failure. 6A Self-certified Cross-Cluster Asymmetric Group Key Agreement for Wireless Sensor Networks显示文摘Wireless sensor networks have some obvious characteristics, such as communication range is limited, computing power is limited and energy is limited. Group key agreement in this environment requires a cross-cluster, computation and communication overhead are lightweight and highly safe group key agreement protocol. Aiming at these demands, the paper proposes a Self-certified cross-cluster Asymmetric group key agreement(SC-AGKA). To establish a lightweight and efficient group communication channel among sensor nodes. According to the cluster head as the bridge node to realize the sensor nodes in different cluster have the same group key information, and negotiate a pair of asymmetric group keys to realize the cross cluster secure communication. The group communication adopts asymmetric encryption mechanism. It realizes the group security communication mechanism of message sender unconstraint. The asymmetric group key agreement has the key self-certified, which does not need additional rounds to verify the correctness of group key. 7Modeling of void closure in diffusion bonding process based on dynamic conditions显示文摘The dynamic conditions for plastic deformation mechanism,surface source mechanism,interface source mechanism and creep mechanism in the diffusion bonding process are proposed.Based on these dynamic conditions,a model for void closure in the diffusion bonding process is derived.The effects of diffusion bonding parameters on the bonding mechanisms are analyzed.For the diffusion bonding process of TC4 alloy,at a low diffusion bonding temperature or pressure,or for a short time,the interface source mechanism plays a main role.But,the creep mechanism is the dominant mechanism and operates until the diffusion bonding is fulfilled.As the diffusion bonding time increases,the surface source and interface source mechanism enhance and then stop at a certain stage of diffusion bonding,while the creep mechanism enhances continuously.As the diffusion bonding temperature or pressure increases,the interface source mechanism weakens and the other mechanisms enhance.The maximum and average errors between the calculated and the experimental results in the diffusion bonding for TC4 alloy are 12.86% and 5.79%,respectively.MA RuiFang LI MiaoQuan LI Hong YU WeiXin 2012Science China(Technological Sciences)2012,55,9:5
8Identification of Differentially-expressed Genes in Intestinal Gastric Cancer by Microarray Analysis显示文摘Gastric cancer(GC) is one of the most frequent malignant tumors. In order to systematically characterize the cellular and molecular mechanisms of intestinal GC development,in this study, we used 22 K oligonucleotide microarrays and bioinformatics analysis to evaluate the gene expression profiles of GC in 45 tissue samples, including 20 intestinal GC tissue samples,20 normal appearing tissues(NATs) adjacent to tumors and 5 noncancerous gastric mucosa tissue samples. These profiles allowed us to explore the transcriptional characteristics of GC and determine the change patterns in gene expression that may be of clinical significance. 1519 and 1255 differentially-expressed genes(DEGs) were identified in intestinal GC tissues and NATs, respectively,as determined by Bayesian analysis(P < 0.001). 10Predominant yeasts in Chinese Dong fermented pork(Nanx Wudl)and their aroma-producing properties in fermented sausage condition显示文摘Yeasts are one of the predominant microbial groups in fermented meats.In this study,yeast communities of Chinese Dong fermented pork(Nanx Wudl)were investigated and the technological properties of 73 yeast isolates were evaluated.Through culture-dependent and high-throughput sequencing methods,the main yeast species identified included Pichia membranifaciens,Kazachstania bulderi,Millerozyma farinosa,Candida zeylanoides,Kazachstania exigua,Candida parapsilosis and Saccharomyces cerevisiae.Among these yeasts,P.membranifaciens,M.farinosa,K.exigua and K.bulderi were detected in fermented meats for the first time.A total of 73 yeast isolates was investigated for their lipolytic and proteolytic activities.All yeast species showed lipolytic activity,while proteolytic activity against meat protein was only detected in S.cerevisiae.Assay of aroma-producing potential was performed in a model simulating fermented sausage condition.Inoculation of yeast strains increased volatiles production,especially esters and alcohols.The highest ester production was observed in S.cerevisiae Y70 strain,followed by K.exigua Y12 and K.bulderi Y19.C.zeylanoides Y10 and S.cerevisiae Y70 were the highest producers of benzeneethanol and 3-methyl-1-butanol.S.cerevisiae Y70 with its highest production of branched alcohols and esters could be a promising candidate as aroma enhancer in the manufacture of fermented sausages.Ruifang Mi Xi Chen Suyue Xiong Biao Qi Jiapeng Li Xiaoling Qiao Wenhua Chen Chao Qu Shouwei Wang 2021Food Science and Human Wellness2021,10,2:3
11Efficacy of combined inhibition of mTOR and ERK/MAPK pathways in treating a tuberous sclerosis complex cell model显示文摘Tuberous sclerosis complex (TSC) is an autosomal dominant tumor syndrome which afflicts multiple organs and for which there is no cure, such that TSC patients may develop severe mental retardation and succumb to renal or respiratory failure. TSC derives from inactivating mutations of either the TSC1 or TSC2 tumor suppressor gene, and the resulting inactivation of the TSC1/TSC2 protein complex causes hyperactivation of the mammalian target of rapamycin (mTOR), leading to uncontrolled cell growth and proliferation. Recent clinical trials of targeted suppression of mTOR have yielded only modest success in TSC patients. It was proposed that abrogation of a newly identified mTOR-mediated negative feedback regulation on extracellular signal-regulated kinase/mitogen-activated protein kinase (ERK/MAPK) signaling pathway and on the well-documented RTK-PI3K-AKT signaling cascade could limit the efficacy of mTOR inhibitors in the treatment of TSC patients. Therefore, we speculate that dual inhibition of mTOR and ERK/MAPK pathways may overcome the disadvantage of single agent therapies and boost the efficacy of mTOR targeted therapies for TSC patients. Investigation of this hypothesis in a TSC cell model revealed that mTOR suppression with an mTOR inhibitor, rapamycin (sirolimus), led to up-regulation of ERK/MAPK signaling in mouse Tsc2 knockout cells and that this augmented signaling was attenuated by concurrent administration of a MEK1/2 inhibitor, PD98059. 13Sertoli cell and spermatogonial development in pigs显示文摘Background:Spermatogenesis is an intricate developmental process during which undifferentiated spermatogonia,containing spermatogonial stem cells(SSCs),undergo self-renewal and differentiation to generate eventually mature spermatozoa.Spermatogenesis occurs in seminiferous tubules within the testis,and the seminiferous tubules harbor Sertoli and germ cells.Sertoli cells are an essential somatic cell type within the microenvironment that support and steer male germ cell development,whereas spermatogonia are the primitive male germ cells at the onset of spermatogenesis.While the developmental progression of Sertoli cells and spermatogonia has been well established in mice,much less is known in other mammalian species including pigs.Results:To acquire knowledge of Sertoli cell and spermatogonial development in pigs,here we collected as many as nine ages of Duroc porcine testes from the neonate to sexual maturity,i.e.,testes from 7-,30-,50-,70-,90-,110-,130-,150-and 210-day-old boars,and performed histological and immunohistochemical analyses on testis sections.We first examined the development of spermatogenic cells and seminiferous tubules in porcine testes.Then,by immunofluorescence staining for marker proteins(AMH,SOX9,DBA,UCHL1,VASA,KIT,Ki67 and/or PCNA),we delved into the proliferative activity and development of Sertoli cells and of spermatogonial subtypes(pro-,undifferentiated and differentiating spermatogonia).Besides,by immunostaining forβ-catenin and ZO-1,we studied the establishment of the blood-testis barrier in porcine testes.Conclusions:In this longitudinal study,we have systematically investigated the elaborate Sertoli cell and spermatogonial developmental patterns in pigs from the neonate to sexual maturity that have so far remained largely unknown.The findings not only extend the knowledge about spermatogenesis and testicular development in pigs,but also lay the theoretical groundwork for porcine breeding and rearing.Yi Zheng Qiang Gao Tianjiao Li Ruifang Liu Zechao Cheng Ming Guo Jinhong Xiao De Wu Wenxian Zeng 2022Journal of Animal Science and Biotechnology2022,13,4:2

16Single-cell RNA-sequencing reveals the dynamic process and novel markers in porcine spermatogenesis显示文摘Background:Spermatogenesis is the process by which male gametes are formed from spermatogonial stem cells and it is essential for the reliable transmission of genetic information between generations.To date,the dynamic transcriptional changes of defined populations of male germ cells in pigs have not been reported.Results:To characterize the atlas of porcine spermatogenesis,we profiled the transcriptomes of~16,966 testicular cells from a 150-day-old pig testis through single-cell RNA-sequencing(scRNA-seq).The scRNA-seq analysis identified spermatogonia,spermatocytes,spermatids and three somatic cell types in porcine testes.The functional enrichment analysis demonstrated that these cell types played diverse roles in porcine spermatogenesis.The accuracy of the defined porcine germ cell types was further validated by comparing the data from scRNA-seq with those from bulk RNA-seq.Since we delineated four distinct spermatogonial subsets,we further identified CD99 and PODXL2 as novel cell surface markers for undifferentiated and differentiating spermatogonia,respectively.Conclusions:The present study has for the first time analyzed the transcriptome of male germ cells and somatic cells in porcine testes through scRNA-seq.Four subsets of spermatogonia were identified and two novel cell surface markers were discovered,which would be helpful for studies on spermatogonial differentiation in pigs.The datasets offer valuable information on porcine spermatogenesis,and pave the way for identification of key molecular markers involved in development of male germ cells.Lingkai Zhang Fuyuan Li Peipei Lei Ming Guo Ruifang Liu Ling Wang Taiyong Yu Yinghua Lv Tao Zhang Wenxian Zeng Hongzhao Lu Yi Zheng 2022Journal of Animal Science and Biotechnology2022,13,2:2
17Clay nanosheet-mediated delivery of recombinant plasmids expressing artificial miRNAs via leaf spray to prevent infection by plant DNA viruses显示文摘Whitefly-transmitted begomoviruses are economically important plant pathogens that cause severe problems in many crop plants,such as tomato,papaya,cotton,and tobacco.Tomato yellow leaf curl virus(TYLCV)is a typical monopartite begomovirus that has been extensively studied,but methods that can efficiently control begomoviruses are still scarce.In this study,we combined artificial microRNA(amiRNA)-mediated silencing technology and clay nanosheetmediated delivery by spraying and developed a method for efficiently preventing TYLCV infection in tomato plants.We designed three amiRNAs that target different regions of TYLCV to silence virus-produced transcripts.Three plant expression vectors expressing pre-amiRNAs were constructed,and recombinant plasmid DNAs(pDNAs)were loaded onto nontoxic and degradable layered double hydroxide(LDH)clay nanosheets.LDH nanosheets containing multiple pDNAs were sprayed onto plant leaves.We found that the designed amiRNAs were significantly accumulated in leaves 7 days after spraying,while the pDNAs were sustainably detected for 35 days after the spray,suggesting that the LDH nanosheets released pDNAs in a sustained manner,protected pDNAs from degradation and efficiently delivered pDNAs into plant cells.Importantly,when the LDH nanosheets coated with pDNAs were sprayed onto plants infected by TYLCV,both the disease severity and TYLCV viral concentration in sprayed plants were significantly decreased during the 35 days,while the levels of H_(2)O_(2) were significantly increased in those plants.Taken together,these results indicate that LDH nanosheets loaded with pDNAs expressing amiRNAs can be a sustainable and promising tool for begomovirus control.Qili Liu Yanpeng Li Kedong Xu Dongxiao Li Haiyan Hu Feng Zhou Puwen Song Yongang Yu Qichao Wei Qian Liu Weipeng Wang Ruifang Bu Haili Sun Xiaohui Wang Jianjun Hao Honglian Li Chengwei Li 2020Horticulture Research2020,7,1:1

20Dynamic changes in O-GlcNAcylation regulate osteoclast differentiation and bone loss via nucleoporin 153显示文摘Bone mass is maintained by the balance between osteoclast-induced bone resorption and osteoblast-triggered bone formation.In inflammatory arthritis such as rheumatoid arthritis(RA),however,increased osteoclast differentiation and activity skew this balance resulting in progressive bone loss.O-GlcNAcylation is a posttranslational modification with attachment of a single O-linkedβ-D-N-acetylglucosamine(O-GlcNAc)residue to serine or threonine residues of target proteins.Although O-GlcNAcylation is one of the most common protein modifications,its role in bone homeostasis has not been systematically investigated.We demonstrate that dynamic changes in O-GlcNAcylation are required for osteoclastogenesis.Increased O-GlcNAcylation promotes osteoclast differentiation during the early stages,whereas its downregulation is required for osteoclast maturation.At the molecular level,O-GlcNAcylation affects several pathways including oxidative phosphorylation and cell-cell fusion.TNFαfosters the dynamic regulation of O-GlcNAcylation to promote osteoclastogenesis in inflammatory arthritis.Targeted pharmaceutical or genetic inhibition of O-GlcNAc transferase(OGT)or O-GlcNAcase(OGA)arrests osteoclast differentiation during early stages of differentiation and during later maturation,respectively,and ameliorates bone loss in experimental arthritis.Knockdown of NUP153,an O-GlcNAcylation target,has similar effects as OGT inhibition and inhibits osteoclastogenesis.These findings highlight an important role of O-GlcNAcylation in osteoclastogenesis and may offer the potential to therapeutically interfere with pathologic bone resorption.Yi-Nan Li Chih-Wei Chen Thuong Trinh-Minh Honglin Zhu Alexandru-Emil Matei Andrea-Hermina Györfi Frederic Kuwert Philipp Hubel Xiao Ding Cuong Tran Manh Xiaohan Xu Christoph Liebel Vladyslav Fedorchenko Ruifang Liang Kaiyue Huang Jens Pfannstiel Min-Chuan Huang Neng-Yu Lin Andreas Ramming Georg Schett Jörg H.W.Distler 2022Bone Research2022,10,4:1
