2Generation of B Cell-Deficient Pigs by Highly Efficient CRISPR/Cas9-Mediated Gene Targeting显示文摘Generating B cell-deficient mutant is the first step to produce human antibody repertoires in large animal models. In this study, we applied the clustered regularly interspaced short palindromic repeat(CRISPR)/CRISPR-associated(Cas) system to target the JHregion of the pig Ig M heavy chain gene which is crucial for B cell development and differentiation. Transfection of Ig M-targeting Cas9 plasmid in primary porcine fetal fibroblasts(PFFs) enabled inducing gene knock out(KO) in up to 53.3% of colonies analyzed, a quarter of which harbored biallelic modification, which was much higher than that of the traditional homologous recombination(HR). With the aid of somatic cell nuclear transfer(SCNT) technology, three piglets with the biallelic Ig M heavy chain gene mutation were produced. The piglets showed no antibody-producing B cells which indicated that the biallelic mutation of the Ig M heavy chain gene effectively knocked out the function of the Ig M and resulted in a B cell-deficient phenotype. 3MiR-181b suppresses proliferation of and reduces chemoresistance to temozolomide in U87 glioma stem cells显示文摘MicroRNAs regulate self renewal and differentiation of cancer stem cells.There,we sought to identify the expression of miR-181b in glioma stem cells and investigate the biological effect of miR-181b on glioma stem cells in this study.MiR-181b expression was measured by real-time PCR in glioma stem cells isolated from U87 cells by FACS sorting.After miR-181b was overexpressed in U87 glioma stem cells by miR-181b lentiviral expression vector and/or treatment of temozolomide,secondary neurosphere assay,soft agar colony assay and MTT assay were performed.Compared with U87 cells,the expression of miR-181b was significantly decreased in U87 glioma stem cells.Overexpression of miR-181b decreased neurosphere formation by U87 glioma stem cells in vitro and suppressed colony formation in soft agar,and the cell growth inhibition rates increased in a time-dependent manner in U87 glioma stem cells infected with miR-181b lentivirus.Furthermore,miR-181b had a synergistic effect on temozolomide-induced inhibition of secondary neurosphere and soft agar colony,and on cell growth inhibition rates.MiR-181b functions as a tumor suppressor that suppresses proliferation and reduces chemoresistance to temozolomide in glioma stem cells.Ping Li Xiaoming Lu Yingyi Wang Lihua Sun Chunfa Qian Wei Yan Ning Liu Yongping You Zhen Fu 2010The Journal of Biomedical Research2010,24,6:14
4Reactive oxygen species-based nanomaterials for the treatment of myocardial ischemia reperfusion injuries显示文摘Interventional coronary reperfusion strategies are widely adopted to treat acute myocardial infarction,but morbidity and mortality of acute myocardial infarction are still high.Reperfusion injuries are inevitable due to the generation of reactive oxygen species(ROS)and apoptosis of cardiac muscle cells.However,many antioxidant and anti-inflammatory drugs are largely limited by pharmacokinetics and route of administration,such as short half-life,low stability,low bioavailability,and side effects for treatment myocardial ischemia reperfusion injury.Therefore,it is necessary to develop effective drugs and technologies to address this issue.Fortunately,nanotherapies have demonstrated great opportunities for treating myocardial ischemia reperfusion injury.Compared with traditional drugs,nanodrugs can effectively increase the therapeutic effect and reduces side effects by improving pharmacokinetic and pharmacodynamic properties due to nanodrugs’size,shape,and material characteristics.In this review,the biology of ROS and molecular mechanisms of myocardial ischemia reperfusion injury are discussed.Furthermore,we summarized the applications of ROS-based nanoparticles,highlighting the latest achievements of nanotechnology researches for the treatment of myocardial ischemia reperfusion injury.Tianjiao Zhao Wei Wu Lihua Sui Qiong Huang Yayun Nan Jianhua Liu Kelong Ai 2022Bioactive Materials2022,7,1:11
5Tectonic Evolution of the Tianhuan Depression and the Western Margin of the Late Triassic Ordos显示文摘The Ordos Basin is one of the most important oil and gas basins in China. Based on surface outcrop, key exploratory wells and seismic reflection data and by using the technology of 'prototype basin recovery', seismic profile 'layer flattening' and 'restoration of balanced section',and other methods, the sedimentary boundary, structure and the evolution history of the Tianhuan depression on the western margin of the Ordos Basin are reestablished. The following results have been obtained. (1) The west boundary of the Late Triassic Ordos Basin was far beyond the scope of the current basin. The basin is connected with the Late Triassic Hexi Corridor Basin, and its western margin did not have tectonic characteristics of a foreland basin. (2) The Tianhuan depression was first formed in the Late Jurassic. At the late stage it was impacted by the late Yanshanian and Himalayan tectonic movement and the depression axis gradually moved eastwards to the present location with a cumulative migration distance of ~30 km. 6Evolution of Surface Sensible Heat over the Tibetan Plateau Under the Recent Global Warming Hiatus显示文摘Based on regular surface meteorological observations and NCEP/DOE reanalysis data, this study investigates the evolution of surface sensible heat(SH) over the central and eastern Tibetan Plateau(CE-TP) under the recent global warming hiatus. The results reveal that the SH over the CE-TP presents a recovery since the slowdown of the global warming. The restored surface wind speed together with increased difference in ground-air temperature contribute to the recovery in SH.During the global warming hiatus, the persistent weakening wind speed is alleviated due to the variation of the meridional temperature gradient. Meanwhile, the ground surface temperature and the difference in ground-air temperature show a significant increasing trend in that period caused by the increased total cloud amount, especially at night. At nighttime, the increased total cloud cover reduces the surface effective radiation via a strengthening of atmospheric counter radiation and subsequently brings about a clear upward trend in ground surface temperature and the difference in ground-air temperature.Cloud–radiation feedback plays a significant role in the evolution of the surface temperature and even SH during the global warming hiatus. 7Serum containing Tongqiaohuoxue decoction suppresses glutamate-induced PC12 cell injury显示文摘Glutamate application is an established method of inducing PC12 cell injury. PC12 cells were cultured with serum containing Tongqiaohuoxue decoction consisting of moschus, Carthamus tinctorius, Rhizoma chuanxiong, Semen pruni persicae, and Radix Paeoniae Rubra. After 24 hours of co-cultivation, glutamate (12.5 mM) was added to the culture medium. We found that serum containing Tongqiaohuoxue decoction prevented the increase in reactive oxygen species, and the decreases in superoxide dismutase and Na+-K+-ATPase activity, induced by glutamate. It also reduced the concentration of malondialdehyde, enhanced the mitochondrial transmembrane potential, inhibited the elevation of cellular calcium, and decreased phosphorylation of calmodulin-dependent protein kinase II. 8Water-in-salt electrolyte ion-matched N/O codoped porous carbons for high-performance supercapacitors显示文摘Pore size and distribution in carbon-based materials are regarded to be a key factor to affect the electrochemical capacitive performances of the resultant electrodes.In this study,nitrogen and oxygen codoped porous carbons(NOPCs) are fabricated based on a simple Schiff-base reaction between m-phenylenediamine and terephthalaldehyde.The NOPCs have tunable morphologies,high surface areas,abundant heteroatom doping.More importantly,the carbons show a dominant micropores of 0.5-0.8 nm,comparable to the ionic sizes of LiTFSI(Li^+0.069 nm;TFSI-0.79 nm) water-in-salt electrolyte with a high potential window of 2.2 V.Consequently,the fabricated symmetric supercapacitor gives a high energy output of 30.5 Wh/kg at 1 kW/kg,and high stability after successive 10,000 cycles with ^96.8% retention.This study provides promising potential to develop high-energy supercapacitors.Jingjing Yan Dazhang Zhu Yaokang Lv Wei Xiong Mingxian Liu Lihua Gan 2020Chinese Chemical Letters2020,31,2:6
9Advertisement Call Variability in the Black-spined Toad Bufo melanostictus (Anura: Bufonidae) during the Breeding Season in Lishui, Zhejiang, China显示文摘Advertisement calls play an important role in influencing female mate choice and are regarded as one of the key characters responsible for reproductive isolation and speciation events in frogs and toads. The black-spined toad, Bufo melanostictus, is one of the most common amphibians in Southeast Asia and is frequently observed in human habitations throughout its distributional range. The advertisement calls of the toad have not been well described. Here, we report the advertisement calls of this species from Lishui, Zhejiang, China during its breeding season. The advertisement calls of B. melanostictus are composed of single notes with a single harmonic frequency emitted sporadically. The call parameters recorded in this study were highly variable across recording times and with different body sizes. Dominant frequency was negatively correlated with body size. Comparing the calls of the B. melanostictus population from Lishui with the populations from five other localities(Bangkok, Thailand; Coorg, India; Burma, Myanmar; Bali, Indonesia; Halimun-Salak National Park, Indonesia), we found that call structure was similar among five different populations with no harmonics, while the Halimun-Salak National Park population contained two clear harmonics. Dominant frequency was similar between Bali(1.56 kHz) and Coorg(1.6 kHz) populations, while it was highly variable among other populations. Besides call duration, call interval and pulse/call rate may also be significantly different among the comparing populations. 11A hybrid inversion method of damped least squares with simulated annealing used for Rayleigh wave dispersion curve inversion显示文摘Surface wave methods are becoming increasingly popular in many geotechnical applications and in earthquake seismology due to their noninvasive characteristics.Inverse surface wave dispersion curves are a crucial step in most surface wave methods.Many inversion methods have been applied to surface wave dispersion curve inversion,including linearized inversion and nonlinearized inversion methods.In this study,a hybrid inversion method of Damped Least Squares(DLS) with Very Fast Simulated Annealing(VFSA) is developed for multi-mode Rayleigh wave dispersion curve inversion.Both synthetic and in situ fi eld data were used to verify the validity of the proposed method.The results show that the proposed method is superior to the conventional VFSA method in aiming at global minimum,especially when parameter searching space is adjacent to real values of the parameters.The advantage of the new method is that it retains both the merits of VFSA for global search and DLS for local search.At high temperatures,the global search dominates the runs,while at a low temperatures,the local search dominates the runs.Thus,at low temperatures,the proposed method can almost directly approach the actual model.Lu Jianqi Li Shanyou Li Wei Tang Lihua 2014Earthquake Engineering and Engineering Vibration2014,13,1:4
12Studies on the structure and catalytic performance of Cu-Zn-Al catalyst prepared by liquid-phase preparation technology under different heat treatment atmosphere显示文摘Cu-Zn-Al slurry catalysts were prepared using a complete liquid-phase preparation technology under different heat treatment atmospheres.The catalysts were characterized using X-ray diffraction,X-ray photoelectron spectroscope,and N2 adsorption-desorption.Their application in the single-step synthesis of dimethyl ether from syngas was also investigated.The results indicate that the type of heat treatment atmosphere has an influence on the Cu species and the Cu0/Cu+ ratio on the catalyst surface.Moreover,the final Cu/Zn ratio on the catalyst surface is mainly dependent on the composition and reaction environment of the catalyst and less on the type of heat treatment atmosphere.The prepared catalysts can suppress sintering of active sites at high temperatures,and the type of heat treatment atmosphere mainly affects the capability of the catalyst for methanol synthesis.The catalysts perform best using N2 as the heat treatment atmosphere.Zhihua Gao Wei Huang Lihua Yin Kechang Xie 2010Journal of Natural Gas Chemistry2010,19,6:3

15Effect of acupuncture plus conventional treatment on brain activity in ischemic stroke patients: a regional homogeneity analysis显示文摘OBJECTIVE: To determine differences in cerebral activity evoked by acupuncture and conventional stroke treatment, and identify the treatment targets.METHODS: In total, 21 patients were randomly divided into two groups. Group A(11 patients) received both acupuncture and conventional treatment, while group B(10 patients) received conventional treatment only. Resting-state functional magnetic resonance imaging(f MRI) was performed on each participant before and after treatment. Regional homogeneity analysis was performed to investigate the potential mechanism of acupuncture treatment by comparing differences in cerebral activity between treatments.RESULTS: Group A showed higher Re Ho in the frontal lobe(BA6, BA46), supra-marginal gyrus(BA40),middle temporal gyrus(BA21), cerebellum, and insula. Group B showed higher Re Ho in the frontal lobe(BA6) and parietal lobe(BA3, BA7).CONCLUSION: Acupuncture and conventional treatment triggered relatively different clinical efficacy and brain responses. Acupuncture treatment more significantly improved the symptoms of stroke patients. 16Preparation and evaluation of bis(diallyl alkyl tertiary ammonium salt) polymer as a promising adsorbent for phosphorus removal显示文摘Problems associated with water eutrophication due to high phosphorus concentrations and related environmentally safe solutions have attracted wide attention.A novel bis(diallyl alkyl tertiary ammonium salt)polymer,particularly poly(N1,N1,N6,N6-tetraallylhexane-1,6-diammonium dichloride)(PTAHDADC),was synthesized and characterized by Fourier transform infrared spectroscopy,nuclear magnetic resonance,scanning electron microscopy,mercury intrusion method,and thermogravimetric analysis.The adsorption characteristics in phosphorus were evaluated in dilute solution,and the recycling properties of PTAHDADC were investigated.Results showed that PTAHDADC possessed macropores with a size distribution ranging from 30 to 130μm concentrating at 63μm in diameter and had 46.52%of porosity,excellent thermal stability below 530 K,and insolubility.PTAHDADC could effectively remove phosphorus at p H=7–11 and had a removal efficiency exceeding 98.4%at pH=10–11.The adsorption equilibrium data of PTAHDADC for phosphorus accorded well with the Langmuir and pseudo-second-order kinetic models.Maximum adsorption capacity was 52.82 mg/g at 293 K.PTAHDADC adsorbed phosphorus rapidly and reached equilibrium within 90 min.Calculated activation energy Eawas 15.18 k J/mol.PTAHDADC presented an excellent recyclability with only 8.23%loss of removal efficiency after five adsorption–desorption cycles.The morphology and structure of PTAHDADC slightly changed as evidenced by the pre-and post-adsorption of phosphorus,but the process was accompanied by the partial deprotonation of the(–CH2)3-NH+group of PTAHDADC.The adsorption was a spontaneous exothermic process driven by entropy through physisorption,electrostatic attraction,and ion exchange.Survey results showed that PTAHDADC was a highly efficient and fast-adsorbing phosphorus-removal material prospective in treating wastewater.Zhengchi Yang Lihua Liu Lu Zhao Gang Su Zuoxiao Wei Anping Tang Jianrong Xue 2019Journal of Environmental Sciences2019,31,12:3
17A cabazitaxel liposome for increased solubility, enhanced antitumor effect and reduced systemic toxicity显示文摘The potential side effects of cabazitaxel(CBZ)in the field of cancer treatment have become a great limitation to its further clinical application.Liposomal delivery is a well-established approach to increase the therapeutic index of hydrophobic drugs.In this study,a PEGmodified liposome was developed for efficiently encapsulating CBZ,thus enhancing its specific tumor inhibition effect and reducing the systemic toxicity.It was found that the loading efficiency of CBZ into the liposome could be improved with the increase of lipophilic materials,as it could be over 80%under the weight ratio of 20:1(total lipid:CBZ).The diameter of CBZ loaded liposome(CBZ@Lipo)was^100 nm.And the liposome suspending in aqueous medium was stable at 4°C for at least one month,according to the change of its size distribution.The killing ability of CBZ@Lipo to cancer cells was significantly lower comparing to that of CBZ solution,which could be attributed to the slow release of CBZ from the liposomes.However,CBZ@Lipo could induce an obvious apoptosis of the cancer cells at low concentration.Furthermore,CBZ@Lipo exhibited an expressively enhanced tumor growth inhibition effect comparing to CBZ solution.More importantly,CBZ@Lipo showed an obviously higher biosafety proved by lower hemolysis probability,stable body weight of mice during the whole experiment and no obvious lesion in histology analysis.Our work provided a useful reference of the formulation of CBZ,which had potential for greater clinical application.Xiaoyi Yin Lihua Luo Wei Li Jie Yang Chunqi Zhu Mengshi Jiang Bing qin Xiaoling Yuan Hang Yin Yichao Lu Yongzhong Du Dawei Chen Jian You 2019Asian Journal of Pharmaceutical Sciences2019,14,6:3
18Reconstruction of tyrosol synthetic pathways in Escherichia coli显示文摘Tyrosol is a pharmacologically active phenolic compound widely used in the medicine and chemical industries.Traditional methods of plant extraction are complicated and chemical synthesis of tyrosol is not commercially viable. In this study, a recombinant Escherichia coli strain was constructed by overexpressing the phenylpyruvate decarboxylase ARO10 from Saccharomyces cerevisiae, which could produce tyrosol from glucose. Furthermore,genes encoding key enzymes from the competing phenylalanine and tyrosine synthesis pathways and the repression protein TyrR were eliminated, and the resulting engineered strain generated 3.57 mmol·L^(-1) tyrosol from glucose. More significantly, codon optimization of ARO10 increased expression and tyrosol titer. Using the novel engineered strain expressing codon-optimized AR10 in shake-flask culture, 8.72 mmol·L^(-1) tyrosol was obtained after 48 h. Optimization of the induction conditions improved tyrosol production to 9.53 mmol·L^(-1)(1316.3 mg·L^(-1)). 19Auxin response factor 6A regulates photosynthesis,sugar accumulation,and fruit development in tomato显示文摘Auxin response factors(ARFs)are involved in auxin-mediated transcriptional regulation in plants.In this study,we performed functional characterization of SlARF6A in tomato.SlARF6A is located in the nucleus and exhibits transcriptional activator activity.Overexpression of SlARF6A increased chlorophyll contents in the fruits and leaves of tomato plants,whereas downregulation of SlARF6A decreased chlorophyll contents compared with those of wild-type(WT)plants.Analysis of chloroplasts using transmission electron microscopy indicated increased sizes of chloroplasts in SlARF6A-overexpressing plants and decreased numbers of chloroplasts in SlARF6A-downregulated plants.Overexpression of SlARF6A increased the photosynthesis rate and accumulation of starch and soluble sugars,whereas knockdown of SlARF6A resulted in opposite phenotypes in tomato leaves and fruits.RNA-sequence analysis showed that regulation of SlARF6A expression altered the expression of genes involved in chlorophyll metabolism,photosynthesis and sugar metabolism.SlARF6A directly bound to the promoters of SlGLK1,CAB,and RbcS genes and positively regulated the expression of these genes.Overexpression of SlARF6A also inhibited fruit ripening and ethylene production,whereas downregulation of SlARF6A increased fruit ripening and ethylene production.SlARF6A directly bound to the SAMS1 promoter and negatively regulated SAMS1 expression.Taken together,these results expand our understanding of ARFs with regard to photosynthesis,sugar accumulation and fruit development and provide a potential target for genetic engineering to improve fruit nutrition in horticulture crops.Yujin Yuan Xin Xu Zehao Gong Yuwei Tang Mengbo Wu Fang Yan Xiaolan Zhang Qian Zhang Fengqing Yang Xiaowei Hu Qichen Yang Yingqing Luo Lihua Mei Wenfa Zhang Cai-Zhong Jiang Wangjin Lu Zhengguo Li Wei Deng 2019Horticulture Research2019,6,1:3
20Metal porphyrin intercalated reduced graphene oxide nanocomposite utilized for electrocatalytic oxygen reduction显示文摘In this paper, we report a simple and facile self-assembly method to successfully fabricate cationic metal porphyrin-MtTMPyP(Mt= Cobalt(Ⅱ), Manganese(Ⅲ), or Iron(Ⅲ); TMPyP = 5, 10, 15, 20-tetrakis(N-methylpyridinium-4-yl) porphyrin) intercalated into the layer of graphene oxide(GO) by the cooperative effects of electrostatic and π-π stacking interaction between positively charged metal porphyrin and negatively charged GO sheets. Followed by reduction with hydrazine vapor, a series of novel 2 D MtTMPyP/rGO_n were fabricated. The as-prepared 2 D hybrids were fully characterized and tested as non-noble metal catalysts for oxygen reduction reaction(ORR) in an alkaline medium.
