| 1 | Gentiopicroside targets PAQR3 to activate the PI3K/AKT signaling pathway and ameliorate disordered glucose and lipid metabolism显示文摘The obstruction of post-insulin receptor signaling is the main mechanism of insulin-resistant diabetes.Progestin and adipoQ receptor 3(PAQR3),a key regulator of inflammation and metabolism,can negatively regulate the PI3 K/AKT signaling pathway.Here,we report that gentiopicroside(GPS),the main bioactive secoiridoid glycoside of Gentiana manshurica Kitagawa,decreased lipid synthesis and increased glucose utilization in palmitic acid(PA) treated HepG2 cells.Additionally,GPS improved glycolipid metabolism in streptozotocin(STZ) treated high-fat diet(HFD)-induced diabetic mice.Our findings revealed that GPS promoted the activation of the PI3 K/AKT axis by facilitating DNA-binding protein 2(DDB2)-mediated PAQR3 ubiquitinated degradation.Moreover,results of surface plasmon resonance(SPR),microscale thermophoresis(MST) and thermal shift assay(TSA) indicated that GPS directly binds to PAQR3.Results of molecular docking and cellular thermal shift assay(CETSA) revealed that GPS directly bound to the amino acids of the PAQR3 NH2-terminus including Leu40,Asp42,Glu69,Tyr125 and Ser129,and spatially inhibited the interaction between PAQR3 and the PI3 K catalytic subunit(P110α) to restore the PI3 K/AKT signaling pathway.In summary,our study identified GPS,which inhibits PAQR3 expression and directly targets PAQR3 to restore insulin signaling pathway,as a potential drug candidate for the treatment of diabetes. | Haiming Xiao Xiaohong Sun Zeyuan Lin Yan Yang Meng Zhang Zhanchi Xu Peiqing Liu Zhongqiu Liu Heqing Huang | 2022 | Acta Pharmaceutica Sinica B2022,12,6: | 7 |

| 4 | Local electronic structure and magnetic properties of (Ga,Cr)N显示文摘The local electronic structure and mag- netic properties of diluted magnetic semiconductor (Ga,Cr)N have been studied by using discrete varia- tional method (DVM) based on density functional theory. The magnetic moments per Cr atom vary significantly with Cr concentration, and the trend of variation is in agreement with that of the experiment. The coupling between Cr atoms in the system with two Cr atoms considered is found to be ferromagnetic, and the magnetic moment per Cr atom is similar to the case in which only one Cr atom is considered in the same doping concentration. | 14 | Impact of living donor liver with steatosis and idiopathic portal inflammation on clinical outcomes in pediatric liver transplantation:Beijing experience显示文摘Background:To evaluate the impact of steatosis and/or idiopathic portal inflammation(IPI)in living donor livers on recipients’clinical outcomes.Methods:We assessed 305 qualified donor liver samples from June 2013 to December 2018.Donors and recipients’clinical characteristics,including follow-up data were retrieved.The graft and overall survival with/without steatosis or portal inflammation were compared by Kaplan-Meier analysis.Results:For living donors,the medium age of was 31.2(28,35.8)years old;liver histopathology showed macrovesicular steatosis:0-5%264/305(86.6%)and 5-30%41/305(13.4%),IPI:no 220/305(72.1%)and mild 85/305(27.9%).For recipients,the medium age was 1.0(0.6,1.5)years old;the median pediatric-end-stage-liver-disease score was 16(5.0,26.0)and medium follow-up time was 32.8(24.8,52.0)months.Biliary atresia(69.5%)was the main indication for liver transplantation(LT).Conclusions:The presence of steatosis and portal inflammation of the donor liver did not impact the clinical outcomes including transaminase or bilirubin normalization,short-/long-term complications and recipients’survival.However,recipients with high pediatric-end-stage-liver-disease score(>16)receiving donor liver with portal inflammation,but not steatosis,had trend negative effect on recipients’survival.In conclusion,donor livers with mild steatosis and portal inflammation were qualified for pediatric living donor LT.However,donor liver with mild portal inflammation would better not be allocated to recipients with high pediatric-end-stage-liver-disease score.This study provided new evidence in pediatric living donor liver allocation. | Xinyan Zhao Yafei He Jimin Liu Qian Zhang Liwei Liu Wei Qu Ying Liu Zhigui Zeng Haiming Zhang Jidong Jia Liying Sun Lin Wei Zhijun Zhu | 2022 | Hepatobiliary Surgery and Nutrition2022,11,3: | 1 |

| 18 | Assessment of the Capability of ENSEMBLES Hindcasts in Predicting Spring Climate in China显示文摘Using the hindcasts provided by the Ensemble-Based Predictions of Climate Changes and Their Impacts(ENSEMBLES) project for the period of 1980–2005, the forecast capability of spring climate in China is assessed mainly from the aspects of precipitation, 2-m air temperature, and atmospheric circulations. The ENSEMBELS can reproduce the climatology and dominant empirical orthogonal function(EOF) modes of precipitation and 2-m air temperature, with some differences arising from different initialization months. The multi-model ensemble(MME) forecast of interannual variability is of good performance in some regions such as eastern China with February initialization.The spatial patterns of the MME interannual and inter-member spreads for precipitation and 2-m air temperature are consistent with those of the observed interannual spread, indicating that internal dynamic processes have major impacts on the interannual anomaly of spring climate in China. We have identified two coupled modes between intermember anomalies of the 850-hPa vorticity in spring and sea surface temperature(SST) both in spring and at a lead of 2 months, of which the first mode shows a significant impact on the spring climate in China, with an anomalous anticyclone located over Northwest Pacific and positive precipitation and southwesterly anomalies in eastern China.Our results also suggest that the SST at a lead of two months may be a predictor for the spring climate in eastern China. Results showed that 15-day EUK1001 treatment via intraperitoneal injection promoted neural cell proliferation in the dentate gyrus, although cell differentiation did not change. The majority of bromodeoxyuridine-positive cells co-expressed the immature neuronal marker doublecortin. In addition, the level of neurogenesis in the subventricular zone was not altered. Brain-derived neurotrophic factor mRNA expression was up-regulated following EUK1001 treatment, but no change was observed in expression of camp-responsive element binding protein 1, paired box gene 6, vascular endothelial growth factor alpha, neurogenic differentiation factor 1, and wingless-related mouse mammary tumor virus integration site 3A mRNA.
