1Deep reinforcement learning-based joint task offloading and bandwidth allocation for multi-user mobile edge computing显示文摘The rapid growth of mobile internet services has yielded a variety of computation-intensive applications such as virtual/augmented reality. Mobile Edge Computing (MEC), which enables mobile terminals to offload computation tasks to servers located at the edge of the cellular networks, has been considered as an efficient approach to relieve the heavy computational burdens and realize an efficient computation offloading. Driven by the consequent requirement for proper resource allocations for computation offloading via MEC, in this paper, we propose a Deep-Q Network (DQN) based task offloading and resource allocation algorithm for the MEC. Specifically, we consider a MEC system in which every mobile terminal has multiple tasks offloaded to the edge server and design a joint task offloading decision and bandwidth allocation optimization to minimize the overall offloading cost in terms of energy cost, computation cost, and delay cost. Although the proposed optimization problem is a mixed integer nonlinear programming in nature, we exploit an emerging DQN technique to solve it. 3Characteristics of the microbial communities in the integrated vertical-flow constructed wetlands显示文摘Microorganisms play an important role in removing pollutants from constructed wetlands.We investigated the microbial characteristics in a novel integrated vertical-flow constructed wetland(IVCW) ,which has been in operation in Wuhan,China since 1998.We used phospholipid fatty acid(PLFA) and amoA gene to analyze the structure and diversity of the microbial community within the IVCW.PLFA results suggested that the amount of bacterial PLFA was significantly higher than that of fungal PLFA,but the total microbial biomass represented by PLFA index was low in the system.Microbial spatial distribution showed significantly higher bacterial(both G+and G-) and fungal biomass in the surface than in the subsurface layers.The ratios of monounsaturated to branched PLFA demonstrated that an anaerobic layer sandwiched by two aerobic layers existed in the IVCW,consistent with the redox potential results.Analysis of the amoA revealed the presence of Nitrosomonas-like sequences in the surface substrate of the downflow chamber and apparent diversities of ammonia-oxidizing bacteria in the system.These results suggest that microorganisms,despite their relatively low biomass,have inhabited the IVCW,and the results will offer some valuable information on microbe to system designers and managers.ZHOU Qiaohong,HE Feng,Zhang Liping,WANG Yanfen,WU Zhenbin State Key Laboratory of Freshwater Ecology and Biotechnology,Institute of Hydrobiology,Chinese Academy of Sciences,Wuhan 430072,China Department of Biology and Chemistry,City University of Hong Kong 2009Journal of Environmental Sciences2009,21,9:14

5Characterization of the essential role of bone morphogenetic protein 9 (BMP9) in osteogenic differentiation of mesenchymal stem cells (MSCs) through RNA interference显示文摘Mesenchymal stem cells(MSCs)are multipotent stem cells and capable of differentiating into multiple cell types including osteoblastic,chondrogenic and adipogenic lineages.We previously identified BMP9 as one of the most potent BMPs that induce osteoblastic differentiation of MSCs although exact molecular mechanism through which BMP9 regulates osteogenic differentiation remains to be fully understood.Here,we seek to develop a recombinant adenovirus system to optimally silence mouse BMP9 and then characterize the important role of BMP9 in osteogenic differentiation of MSCs.Using two different siRNA bioinformatic prediction programs,we design five siRNAs targeting mouse BMP9(or simB9),which are expressed under the control of the converging H1 and U6 promoters in recombinant adenovirus vectors.We demonstrate that two of the five siRNAs,simB9-4 and simB9-7,exhibit the highest efficiency on silencing exogenous mouse BMP9 in MSCs.Furthermore,simB9-4 and simB9-7 act synergistically in inhibiting BMP9-induced expression of osteogenic markers,matrix mineralization and ectopic bone formation from MSCs.Thus,our findings demonstrate the important role of BMP9 in osteogenic differentiation of MSCs.The characterized simB9 siRNAs may be used as an important tool to investigate the molecular mechanism behind BMP9 osteogenic signaling.Our results also indicate that recombinant adenovirus-mediated expression of siRNAs is efficient and sustained,and thus may be used as an effective delivery vehicle of siRNA therapeutics.Shujuan Yan Ruyi Zhang Ke Wu Jing Cui Shifeng Huang Xiaojuan Ji Liping An Chengfu Yuan Cheng Gong Linghuan Zhang Wei Liu Yixiao Feng Bo Zhang Zhengyu Dai Yi Shen Xi Wang Wenping Luo Bo Liu Rex C.Haydon Michael J.Lee Russell R.Reid Jennifer Moriatis Wolf Qiong Shi Hue H.Luu Tong-Chuan He Yaguang Weng 2018Genes & Diseases2018,5,2:8
6Significance of ultrasound evaluation of carotid atherosclerotic plaque for diagnosing ischemic cerebrovascular disease显示文摘BACKGROUND: Carotid artery is the main source for craniocerebral blood supply. Its intimal plaque formation and arterial stenosis degree both are the risk factors for ischemic cerebrovascular disease. Therefore, the close relationship of carotid atherosclerotic plaque and ischemic cerebrovascular disease, and ultrasound evaluation of carotid atherosclerotic plaque have become the hot spot in studying ischemic cerebrovascular disease. OBJECTIVE: This study was to detect the degree of carotid atherosclerosis of ischemic cerebrovascular disease patients by ultrasonography, and to analyze the situation of carotid atherosclerosis and its relationship with clinic. DESIGN: Clinical randomized concurrent control experiment. SETTING: Lintong Convalescent Hospital of Lanzhou Military Area Command of Chinese PLA. PARTICIPANTS: Totally 60 outpatients and inpatients with ischemic cerebrovascular disease, 42 males and 18 females, admitted to Lintong Convalescent Hospital of Lanzhou Military Area Command of Chinese PLA between January 2006 and December 2006 were involved in the patient group. They met the diagnosis criteria of ischemic cerebrovascular disease constituted by the 4th Cerebrovascular Disease Conference in 1996, and were confirmed to suffer from ischemic cerebrovascular disease by skull CT and MRI. Another 20 subjects who received healthy examination concurrently in the same hospital, 12 males and 8 females, were involved in the control group. Informed consents of detected items were obtained from involved subjects. METHODS: The plaque thickness of mid portion, distal end and crotch of common carotid artery (CCA), internal carotid artery (ICA), external carotid artery (ECA) and vertebral artery (VA) of involved subjects, who received health examination was separately detected with color Doppler ultrasonograph (HDI-5000). Then, total integral of plaque was calculated. The intima-media thickness (IMT) was measured with two-dimensional ultrasonography. The inner diameter stenosis degree of subjects who had plaque was measured. Blood flow parameters were recorded, and stenosis degree and plaque area were calculated. Blood flow volume of bilateral carotid artery and VA was separately measured with ultrasound equipment software, and brain blood flow volume was calculated. MAIN OUTCOME MEASURES: Atherosclerotic degree and blood flow volume of patients of two groups. RESULTS: Sixty patients with ischemic cerebrovascular disease and twenty subjects who received health examination participated in the final analysis. ①The IMT thickness, total plaque score, and total plaque area of patient group was significantly superior to that of control group, respectively(t=5.216–10.158,P < 0.05). ② There were significant differences in the stenosis degree of CCA, ICA and VA between patient group and control group (t=6.720–12.816,P < 0.05). ③ The blood flow volume of CCA, ICA, VA and brain of patient group was significantly lower than that of control group, respectively (t=2.872–10.860, P < 0.05). 7Hydrodynamic process of Tibetan Plateau lake revealed by grain size:Case study of Pumayum Co显示文摘Under the background of global warming,some lakes on the Tibetan Plateau(TP) are potentially sensitive to temperature change.With a case study of Pumayum Co,where glacier meltwater is important to supply(we call this a glacier-fed lake hereafter),we analyze the sensitivity of lake sediment grain size to temperature change.This is done by resolving the modern hydrodynamic process,coupled with comparison of paleoclimatic proxies.According to the spatial distributions of parameters,percentage of grain size and the grain size frequency distribution curve,hydrodynamic processes are analyzed.Five clastic sedimentation types are thereby discriminated.In the open lake area,suspended load transport is the main transport agent.Grain Size Trend Analysis(GSTA),a sediment dynamics model,reveals a trend toward eastward transport.This indicates that the largest and glacier-fed river,the Jiaqu River,influences the entire lake(not just the subsurface alluvial fan),and that lake sediment grain size may serve as a temperature indicator.Time series comparison between grain size of a short core from the central lake and meteorological data confirms this temperature indication,which in turn shows reliability of the method discriminating the hydrodynamic process.This case study will improve the ability of paleoclimatic reconstruction using lake sediment in glacier-fed lakes on the Tibetan Plateau.JU JianTing ZHU LiPing FENG JinLiang WANG JunBo WANG Yong XIE ManPing PENG Ping ZHEN XiaoLin LÜXinMiao 2012Chinese Science Bulletin2012,57,19:7

9Using ORYZA2000 to model cold rice yield response to climate change in the Heilongjiang province, China显示文摘Rice(Oryza sativa L.) is one of the most important staple crops in China. Increasing atmospheric greenhouse gas concentrations and associated climate change may greatly affect rice production. We assessed the potential impacts of climate change on cold rice production in the Heilongjiang province, one of China's most important rice production regions. Data for a baseline period(1961–1990) and the period 2010–2050 in A2 and B2 scenarios were used as input to drive the rice model ORYZA2000 with and without accounting for the effects of increasing atmospheric CO2 concentration. The results indicate that mean,maximum, and minimum temperature during the rice growing season, in the future period considered, would increase by 1.8 °C under the A2 scenario and by 2.2 °C under the B2 scenario compared with those in the baseline. The rate of change in average maximum and minimum temperatures would increase by 0.6 °C per 10-year period under the A2 scenario and by 0.4 °C per 10-year period under the B2 scenario. Precipitation would increase slightly in the rice growing season over the next 40 years. The rice growing season would be shortened and the yield would increase in most areas in the Heilongjiang province. Without accounting for CO2 effect, the rice growing season in the period 2010–2050 would be shortened by 4.7 and 5.8 days,and rice yields would increase by 11.9% and 7.9%, under the A2 and B2 scenarios, respectively.Areas with simulated rice yield increases greater than 30.0% were in the Xiaoxing'an Mountain region. The simulation indicated a decrease in yield of less than 15% in the southwestern Songnen Plain. The rate of change in simulated rice yield was 5.0% and 2.5% per 10 years under the A2 and B2 scenarios, respectively. When CO2 effect was accounted for, rice yield increased by 44.5% and 31.3% under the A2 and B2 scenarios, respectively. The areas of increasing yield were sharply expanded. The area of decreasing yield in the western region of Songnen Plains disappeared when increasing CO2 concentration was considered. The stability of rice yield would increase from 2010 to 2050. 10The efficacy and safety of nimodipine in acute ischemic stroke patients with mild cognitive impairment: a double-blind, randomized,placebo-controlled trial显示文摘Nimodipine might be effective in subcortical vascular dementia(VaD). Its benefit in preventing further cognitive decline in patients with acute ischemic stroke(AIS) and vascular mild cognitive impairment(VaMCI) remains to be established. In this multicenter, double-blind trial, we randomly assigned 654 eligible patients to nimodipine 30 mg three times a day or placebo. The primary outcome was any cognitive decline defined by the changes on the Mini-Mental State Examination(DMMSE à3) or vascular AD assessment scale cognitive subscale(DADAS-cog ! 4) at 6 months. Secondary outcomes included any distribution shift of DADAS-cog, DMMSE or cognitive improvement defined by DADAS-cog à2, or DMMSE ! 0. The primary outcome in the nimodipine group and placebo group were similar for DMMSE à3(4.18% and 7.22%, respectively, P = 0.15) and DADAS-cog ! 4(8.36% and 8.93% respectively,P = 0.88). The distribution shift of DADAS-cog and DMMSE differed significantly between the two groups(P = 0.03 and P = 0.05 respectively). Cognitive improvement occurred in 55.4% in the nimodipine group and 43.6% in the placebo group measured by DADAS-cog à2(Odds Ratio, 1.54; 95% confidence interval[CI] 1.10–2.14, P < 0.01) or 84.0% and 74.6% respectively by DMMSE ! 0(Odds Ratio, 1.79; 95% CI 1.18–2.70, P < 0.01). Nimodipine was associated with better cognitive function in the memory domain. The adverse events rate was similar in two groups. This study is registered with ClinicalTrials.gov,NCT01220622. 14NEDD9 promotes cancer stemness by recruiting myeloid-derived suppressor cells via CXCL8 in esophageal squamous cell carcinoma显示文摘Objective:Esophageal squamous cell carcinoma(ESCC)has high morbidity and mortality rates worldwide.Cancer stem cells(CSCs)may cause tumor initiation,metastasis,and recurrence and are also responsible for chemotherapy and radiotherapy failures.Myeloid-derived suppressor cells(MDSCs),in contrast,are known to be involved in mediating immunosuppression.Here,we aimed to investigate the mechanisms of interaction of CSCs and MDSCs in the tumor microenvironment.Methods:ESCC tissues and cell lines were evaluated.Neural precursor cell expressed,developmentally downregulated 9(NEDD9)was knocked down and overexpressed by lentiviral transfection.Quantitative PCR,Western blot,immunohistochemistry,cell invasion,flow cytometry,cell sorting,multiplex chemokine profiling,and tumor growth analyses were performed.Results:Microarray analysis revealed 10 upregulated genes in esophageal CSCs.Only NEDD9 was upregulated in CSCs using the sphere-forming method.NEDD9 expression was correlated with tumor invasion(P=0.0218),differentiation(P=0.0153),and poor prognosis(P=0.0373).Additionally,NEDD9 was required to maintain the stem-like phenotype.Screening of chemokine expression in ESCC cells with NEDD9 overexpression and knockdown showed that NEDD9 regulated C-X-C motif chemokine ligand 8(CXCL8)expression via the ERK pathway.CXCL8 mediated the recruitment of MDSCs induced by NEDD9 in vitro and in vivo.MDSCs promoted the stemness of ESCC cells through NEDD9 via the Notch pathway.Conclusions:As a marker of ESCC,NEDD9 maintained the stemness of ESCC cells and regulated CXCL8 through the ERK pathway to recruit MDSCs into the tumor,suggesting NEDD9 as a therapeutic target and novel prognostic marker for ESCC.Dongli Yue Shasha Liu Tengfei Zhang Yong Wang Guohui Qin Xinfeng Chen Huanyu Zhang Dong Wang Lan Huang Feng Wang Liping Wang Song Zhao Yi Zhang 2021Cancer Biology & Medicine2021,18,3:4
15Optical dating of the Jingshuiwan Paleolithic site of Three Gorges, China显示文摘The Jingshuiwan Paleolithic site occu- pies the second terrace on the right bank of the Yangtze River. The lithic assemblage is characterized by choppers and scrapers made on pebbles and large flakes——the typical lithic industry of South China. Deposits of the site are mainly composed of fluvial sediments. Quartz grains extracted from these fluvial sediments from which the artifacts were un- covered were dated using the optically stimulated luminescence/single-aliquot regenerative-dose tech- nique. The dating results show that ancient human activities at this site took place in the early Upper Pleistocene (ca. 70 ka). The successful age analysis of the Jingshuiwan site is considered as a major break- through in chronological analysis of Paleolithic open site in the Three Gorges region and even in South China. 16Highly antifouling double network hydrogel based on poly(sulfobetaine methacrylate)and sodium alginate with great toughness显示文摘Hydrogels with good antifouling and mechanical properties as well as biocompatibility have great application potential in the field of biomedicine.In this paper,a newly double network(DN)hydrogel was prepared based on zwitterionic material sulfobetaine methacrylate(SBMA)and natural polysaccharide,sodium alginate(SA).The PSBMA network is covalently crosslinked while the SA network is ionically crosslinked by Ca^(2+).The hybrid crosslinked double network structure endows the DN hydrogel with excellent mechanical properties(E=0.19±0.01 MPa,σ=0.73±0.03 MPa),fast self-recovery ability as well as excellent fatigue resistance.Moreover,the results show that the PSBMA/SA-Ca^(2+)DN hydrogel is biocompatible and resists the absorption of non-specific proteins and adhesion of microorganisms,such as cells and algae,exhibiting outstanding antifouling properties.These unique characteristics of PSBMA/SA-Ca^(2+)DN hydrogel make it a promising candidate for biomedical application,such as artificial connective tissues,implantable devices,and underwater equipment.Jing Zhang Sunxiang Qian Lingdong Chen Liqun Chen Liping Zhao Jie Feng 2021Journal of Materials Science & Technology2021,,26:4
17Loss of Wip1 aggravates brain injury after ischaemia/reperfusion by overactivating microglia显示文摘Background and purpose The inflammatory response mediated by microglia/macrophages is closely related to cerebral ischaemia/reperfusion injury.Wild-type p53-induced protein phosphatase 1(Wip1),a serine/threonine phosphatase,is expressed in various tissues.A growing number of reports have suggested that Wip1 is a negative regulator of inflammation in peripheral tissue;however,its role in the central nervous system(CNS)remains unclear.This study aimed to clarify whether Wip1 can inhibit CNS inflammation by regulating microglia/macrophage functions after ischaemic injury.Methods A model of middle cerebral artery occlusion and reperfusion was established in mice.CNS inflammation was simulated by lipopolysaccharide treatment of primary microglia.Laser speckle imaging was used to monitor regional cerebral blood flow.Behavioural outcomes were assessed with a TreadScan gait analysis system.TTC staining was used to evaluate the infarct volume,and western blotting and immunofluorescence staining were applied to detect the phenotypical transformation of microglia.ELISA was performed to detect the levels of inflammatory factors.Results Wip1 expression was increased after ischaemia/reperfusion.Wip1-knockout(KO)mice displayed more severe brain injury than wild-type mice,as indicated by aggravated motor dysfunction,greater brain infarct volumes and higher expression of inflammatory cytokines(interleukin-6 and tumour necrosis factor alpha)in the brain.We also found that Wip1 depletion increased microglial/macrophage activation in both in vitro and in vivo models,which all showed activation of microglia/macrophages.Lentivirus-Ppm1d reversed the injury induced by Wip1-KO.Conclusions Our results suggest that Wip1 may inhibit neuroinflammation by inhibiting microglial/macrophage activation after brain ischaemia/reperfusion injury.Feng Yan Xiang Cheng Ming Zhao Shenghui Gong Ying Han Liping Ding Di Wu Yumin Luo Wei Zuo Lingling Zhu Ming Fan Xunming Ji 2021Stroke & Vascular Neurology2021,6,3:4
