1Transgene directionally integrated into C-genome of Brassica napus显示文摘Integration of a transgene into a C-genome chromosome plays an important role in reducing ecological risk of transgenic Brassica napus To obtain C-genome transgenic B. napus, herbi- cide-resistant bar gene was firstly transferred into B. oleracea var. alboglabra mediated by Agrobacterium tumefaciens strain LBA4404. Then using the trans- genic B. oleracea as paternal plants and 8 non- transgenic varieties of B. rapa as maternal plants, C- genome transgenic B. napus with bar gene was arti- ficially resynthesized by means of ovary culture and chromosome doubling. Among 67 lines of the resyn- thesized B. napus, 31 were positive, and 36 were negative according to PCR test for bar gene. At least 2 plants from each line were kept for PPT spray con- firmation. The result was in consistence with the PCR test. 2Comparative sorption and desorption behaviors of PFHx S and PFOS on sequentially extracted humic substances显示文摘The sorption and desorption behaviors of two perfluoroalkane sulfonates(PFSAs),including perfluorohexane sulfonate(PFHx S)and perfluorooctane sulfonate(PFOS)on two humic acids(HAs)and humin(HM),which were extracted from a peat soil,were investigated.The sorption kinetics and isotherms showed that the sorption of PFOS on the humic substances(HSs)was much higher than PFHx S.For the same PFSA compound,the sorption on HSs followed the order of HM>HA2>HA1.These suggest that hydrophobic interaction plays a key role in the sorption of PFSAs on HSs.The sorption capacities of PFSAs on HSs were significantly related to their aliphaticity,but negatively correlated to aromatic carbons,indicating the importance of aliphatic groups in the sorption of PFSAs.Compared to PFOS,PFHx S displayed distinct desorption hysteresis,probably due to irreversible pore deformation after sorption of PFHx S.The sorption of the two PFSAs on HSs decreased with an increase in p H in the solution.This is ascribed to the electrostatic interaction and hydrogen bonding at lower p H.Hydrophobic interaction might also be stronger at lower p H due to the aggregation of HSs.Lixia Zhao Yifeng Zhang Shuhong Fang Lingyan Zhu Zhengtao Liu 2014Journal of Environmental Sciences2014,26,12:3
3A method of elevated temperatures coupled with magnetic stirring to predict real time release from long acting progesterone PLGA microspheres显示文摘The object of the study was to develop a quick and reproducible accelerated in vitro release method to predict and deduce the function of the real time(37 °C) release for long acting PLGA microspheres. The method could be described in several steps. First, the release of the microspheres were studied using the sample and separate method at 37 °C with normal orbital shaking and elevated temperatures with magnetic stirring to further accelerate the release. Second, the most similar profile at elevated temperatures with the real time release was chosen with the help of the n value in the fitted Korsmeyer-Peppas Function. Third,the Weibull function and conversion ratio were used to deduce the function of real time release according to the chosen profile at elevated temperatures. The key point in this study was to provide a quick and precise method to predict the real time release for long acting progesterone PLGA microspheres. So the elevated temperatures coupled with magnetic stirring were used to accelerate the release further, and when there have many similar release profiles with the real time release at elevated temperatures, releasing time at elevated temperatures and the R2 of the final deduced function will be used to help choosing the most similar release profile with the real time release. 4Analysis of F_1 hybrid and BC_1 monosomic alien addition line plants from Brassica oleracea × Sinapis alba by GISH显示文摘Sterile and semi-fertile F_1 plants were obtained by intergeneric sexual hybridization between paternal Brassica oleracea var. alboglabra (genome CC, 2n=18) and maternal Sinapis alba (genome SS, 2n=24), BC_1 plants were obtained by backcrossing between paternal B. oleracea and ma-ternal semi-fertile F_1 plants. Genomic in situ hybridization (GISH) combined with dual-colour fluorescence in situ hybridization (dcFISH) showed that sterile F_1 plants contained 21 chromosomes consist-ing of one B. oleracea chromosome set and one S. alba chromosome set, belonging to expected hybrids, and semi-fertile F_1 plants contained 30 chromosomes consisting of two B. oleracea chromosome sets and one S. alba chromosome set. It is obvious that the semi-fertile F_1 plants belong to unexpected hybrids. 1―3 trivalents were detected at meiotic metaphase I of semi-fertile F_1 pollen mother cells (PMCs). Different separation ratios of S chromosomes were detected at anaphase I. A monosomic alien addition line (MAAL) was identified by GISH-dcFISH from BC_1 plants; it contained 19 chromosomes consisting of 18 C chromosomes and 1 S chromosome. At meiotic metaphase I, 9 divalents from B. oleracea and one univalent from S. alba could be detected. Sometimes, one putative C-S trivalent could also be detected. 5Photoelectrocatalytic degradation of high COD dipterex pesticide by using TiO_2 /Ni photo electrode显示文摘A TiO_2 thin film electrode deposited on porous nickel net (TiO_2 /Ni) was prepared by the sol-gel method,and the surface morphology,crystal structure features and the grain size were characterized by Field emission scan electron microscopy (FESEM) and X-ray diffraction (XRD).The photoelectrocatalytic system was set up using a UV high-pressure mercury lamp as the light source,TiO_2 coated on foamed nickel as photo anode,Pt sheet as counter electrode and the pesticide dipterex in synthetic wastewater.Various factors that influence the photoelectrocatalytic decomposition of dipterex pesticide have been studied,such as degradation time,the type of electrolyte,current density,original pH value and different degradation methods.The prepared catalysts were employed to photoelectrocatalytically degrade the pesticide dipterex under UV irradiation,comparing the results with photocatalytic degradation and electrochemical oxidation.The results indicated that under the optimal conditions of 0.02 mol/L NaCl as the supporting electrolyte,current density=2.5 mA/cm 2,pH 6.0 and dipterex pesticide 40 mg/L,and reaction time 2 hr,dipterex chemical oxygen demand (COD) removal rate and organophosphorous conversion of up to 82.6% and 83.5% were achieved,respectively.The method of photoelectrocatalytic degradation is more efficient than photocatalysis and electrochemical oxidation.The possible roles of the electrolytes on the reactions and probable mechanisms were also discussed.Tao Fang Chao Yang Lixia Liao 2012Journal of Environmental Sciences2012,24,6:2

7Oxidative coupling of cinnamic acid derivatives and their radical-scavenging activities显示文摘4-Hydroxycinnamic acid derivatives,including ferulic acid (FA) (1),sinapic acid (SA) (2) and caffeic acid (CA) (3),are widely distributed in the plant kingdom,and undergo oxidative cross-coupling leading to the corresponding dehydrodimers,trimers and even higher oligomers in plants. In order to evaluate the antioxidative ability of these 4-hydroxycinnamic acid derivatives and their oligomers,we synthesized 8-8'-bis-lactone-dimers (8-8'-DiFA (4),8-8'-DiSA (5) and 8-8'-DiCA (6)),as well as a new trimer (7),by the reaction of the corresponding monomers (1-3) with Ag2O in methanol,and assayed their free radical-scavenging activity by the reaction with 2,2-diphenyl-1-picrylhydrazyl radical (DPPH·). 8Dynamic Variations in Brain Glycogen are Involved in Modulating Isoflurane Anesthesia in Mice显示文摘General anesthesia severely affects the metabolites in the brain.Glycogen,principally stored in astrocytes and providing the short-term delivery of substrates to neurons,has been implicated as an affected molecule.However,whether glycogen plays a pivotal role in modulating anesthesia-arousal remains unclear.Here,we demonstrated that isoflurane-anesthetized mice exhibited dynamic changes in the glycogen levels in various brain regions.Glycogen synthase(GS)and glycogen phosphorylase(GP),key enzymes of glycogen metabolism,showed increased activity after isoflurane exposure.Upon blocking glycogenolysis with 1,4-dideoxy-1,4-imino-Darabinitol(DAB),a GP antagonist,we found a prolonged time of emergence from anesthesia and an enhancedδfrequency in the EEG(electroencephalogram).In addition,augmented expression of glycogenolysis genes in glycogen phosphorylase,brain(Pygb)knock-in(PygbH11/H11)mice resulted in delayed induction of anesthesia,a shortened emergence time,and a lower ratio of EEG-δ.Our findings revealed a role of brain glycogen in regulating anesthesiaarousal,providing a potential target for modulating anesthesia.Ze Fan Zhihao Zhang Shiyi Zhao Yuanyuan Zhu Dong Guo Bo Yang Lixia Zhuo Jiao Han Rui Wang Zongping Fang Hailong Dong Yan Li Lize Xiong 2020Neuroscience Bulletin2020,36,12:2

10Effect of compound Guizhu capsule on phosphate and tension homology deleted on chromsome ten and murine double mimute 2 gene expression in lung cancer of mice显示文摘OBJECTIVE: To observe the inhibitory effect of the Chinese herbal compound Guizhu capsule(CGZC)on lung cancer and explore the possible mechanism underlying its actions by evaluating its effects on the expression of phosphate and tension homology deleted on chromsome ten(PTEN) and murine double mimute 2(MDM2) in lung cancer model mice.METHODS: A mouse model of transplanted lung cancer was established by subcutaneous inoculation of cancer cells into the axillae of mice. Twenty-four hours later, the mice were weighed and randomly divided into the model control group, cisplatin group(DDP group), and high, moderate and low dosage CGZC groups, with ten mice in each group. The control mice received an equal volume of distilled water. DDP was intraperitoneally injected at 1 mg/kg in the DDP group, once a day for 3days. CGZC diluted with distilled water was admin-istered at 20 g/kg(10 times the clinical adult dosage) in the high-dose group, 10 g/kg(5 times the clinical adult dosage) in the moderate-dose group and 5 g/kg(2.5 times the clinical adult dosage) in the low-dose group once a day for 10 days. On the11 th day, the mice were weighed and killed. The tumor tissues were weighed and the tumor inhibition rate was calculated. PTEN and MDM2 protein expression were detected by immunohistochemical analysis of tumor tissues.RESULTS: The CGZC high-and moderate-dose groups showed a significant inhibitory effect on tumor growth(P < 0.05); there was no significant difference between the high dose group and the DDP group(P > 0.05). The CGZC high-dose group also showed enhanced expression of PTEN protein(P <0.01) and decreased expression of MDM2 protein(P < 0.01) in lung cancer cells of the mice. There was no significant difference between the high dose group and the DDP group.CONCLUSION: CGZC has a significant inhibitory effect on transplanted lung cancer in mice. 12Estimation of chlorophyll content in Brassica napus based on unmanned aerial vehicle images显示文摘The chlorophyll content has a direct effect on photosynthesis of crops.In order to explore a quick and convenient method for estimating the chlorophyll content of Brassica napus and facilitate efficient crop monitoring,we measured the actual value of chlorophyll with a SPAD-502 chlorophyll detector,and collected aerial images of B.napus with an unmanned aerial vehicle(UAV)carrying a RGB camera in this study.The total number of 270samples collected images were divided into regions according to the planting conditions of different B.napus varieties in the field.Then,according to the empirical formula,there were 36 colors’characteristic parameters calculated and combined.To estimate the chlorophyll content of rape,189 samples were included in the modeling set,while the other 81 samples were enrolled in the validation set for testing the accuracy of this model.After the combination of R(red),G(green)and B(blue)color channels,the results showed that the color characteristics B/(R+G),b,B/G,(G-B)/(G+B),g-b were highly connected with the measured value of chlorophyll SPAD,and the correlation coefficient between the combination based on B/(R+G)and SPAD value was 0.747.With R2=0.805,RMSE=3.343,and RE=6.84%,the regression model created using random forest had superior outcomes,according to the model comparison.This study offers a new method for quickly estimating the amount of chlorophyll in rapeseed and a workable reference for crop monitoring using the UAV platform.Yayi Huang Qiming Ma Xiaoming Wu Hao Li Kun Xu Gaoxiang Ji Fang Qian Lixia Li Qian Huang Ying Long Xiaojun Zhang Biyun Chen Changhua Liu 2022Oil Crop Science2022,7,3:1

14Highly efficient multiplex base editing:One-shot deactivation of eight genes in Shewanella oneidensis MR-1显示文摘Obtaining electroactive microbes capable of efficient extracellular electron transfer is a large undertaking for the scalability of bio-electrochemical systems.Inevitably,researchers need to pursue the co-modification of multiple genes rather than expecting that modification of a single gene would make a significant contribution to improving extracellular electron transfer rates.Base editing has enabled highly-efficient gene deactivation in model electroactive microbe Shewanella oneidensis MR-1.Since multiplexed application of base editing is still limited by its low throughput procedure,we thus here develop a rapid and efficient multiplex base editing system in S.oneidensis.Four approaches to express multiple gRNAs were assessed firstly,and transcription of each gRNA cassette into a monocistronic unit was validated as a more favorable option than transcription of multiple gRNAs into a polycistronic cluster.Then,a smart scheme was designed to deliver one-pot assembly of multiple gRNAs.3,5,and 8 genes were deactivated using this system with editing efficiency of 83.3%,100%and 12.5%,respectively.To offer some nonrepetitive components as alternatives genetic parts of sgRNA cassette,different promoters,handles,and terminators were screened.This multiplex base editing tool was finally adopted to simultaneously deactivate eight genes that were identified as significantly downregulated targets in transcriptome analysis of riboflavin-overproducing strain and control strain.The maximum power density of the multiplex engineered strain HRF(8BE)in microbial fuel cells was 1108.1 mW/m2,which was 21.67 times higher than that of the wild-type strain.This highly efficient multiplexed base editing tool elevates our ability of genome manipulation and combinatorial engineering in Shewanella,and may provide valuable insights in fundamental and applied research of extracellular electron transfer.Yaru Chen Meijie Cheng Yan Li Lin Wang Lixia Fang Yingxiu Cao Hao Song 2023Synthetic and Systems Biotechnology2023,8,1:1
15Evaluation of yield,stability and adaptability of national winter rapeseed regional trials in the upper Yangtze River region in 2017-2018显示文摘Rapeseed variety needs to be tested by regional trial in multiple sites for many years before being applied in market in China.Performants of rapeseed were affected by the interaction of sites and varieties.Evaluation of regional trials is very important for guiding rapeseed breeding.GGE(genotype main effects and genotypeenvironment interaction)biplot was used to evaluate yield,stability,adaptability,representativeness and discrimination of national winter rapeseed trials in the upper reaches of Yangtze River in 2017-2018.Results showed that the main effects of genotype(G),environment(E)and genotypeenvironment interaction(GE)were significant(P<0.01)for yield.Among them,E accounted for 46.95%total variation on average,G and GE accounted for 19.34%and 33.71%respectively.Eight varieties were found with high-yield,excellent stability and adaptability,including‘Yiyou 29’,‘Xiwang 920’,‘Liyouza 108’,‘Nanyou 546’,‘Dadi 195’,‘Jiayou 1’,‘Huayouza 28’and‘Yuhua 2’.Test sites included Nanchong,Mianyang,Wanzhou,Shuangliu and Chengdu in Sichuan Province and Zunyi together with Guiyang in Guizhou Province were selected for their excellent representativeness and discrimination.These results would provide theoretical basis for rapeseed breeding.Ze’en Yu Lixia Luo Fang Zhang Meiyan Hong Xiangxiang Zhang Ruixing Guo 2020Oil Crop Science2020,5,3:1

18Target slinging of droplets with a flexible cantilever显示文摘Control of the directional bounce of droplets impacting solid surfaces is crucial for many agricultural and industrial applications.However,for the universal impact process of raindrops on plant leaves,little is known about how the highly coupled and complicated fluid–structure interaction controls the postimpact motion of droplets and endows the leaves with tenacious vitality.Here,we report a leaf-like superhydrophobic cantilever to flexibly bounce droplets with well-defined directionality and controllability.Through theoretical modeling and three-dimensional fluid–solid coupling simulations,we find that the flexible cantilever significantly relieves the impacting forces of raindrops to reduce droplet fragmentation and enhance water repellency.The results further uncover the scaling relations of the droplet bouncing direction with respect to Weber number and cantilever stiffness.By this technique,the seemed disorganized postimpact movements of droplets are programmable and predictable,achieving the goal of where to point and where to hit automatically.This work advances the understanding of natural droplet impact phenomena,opens a new avenue for delicately controlling liquid motion in space with soft materials,and inspires a plethora of applications like soft robots to transport materials and energies,monitor plant growth as well as predict pathogen transmission in plants.Wei Fang Shun Wang Hu Duan Shahid Ali Tahir Kaixuan Zhang Lixia Wang Xi-Qiao Feng Meirong Song 2023Droplet2023,2,3:0
19Maternal organic selenium supplementation during gestation enhances muscle fiber area and muscle fiber maturation of offspring in porcine model显示文摘Background:Organic selenium supplementation during gestation improves the antioxidant status and reproduc-tive performance of sows and increases the antioxidative capacity of the intestines of their offspring.This study was conducted to investigate the effect of maternal basel diet(control)supplemented with an organic Se,2-hydroxy-4-methylselenobutanoic acid(HMSeBA),or inorganic sodium selenite(Na_(2)SeO_(3))during gestation on the antioxidant status and development of muscle in newborn and weaned piglets.Newborn piglets before colostrum intake and weaned piglets were selected for longissimus dorsi(LD)muscle collection and analysis.Results:The results showed that maternal HMSeBA supplementation increased the muscle area and content of Se in the LD muscle of newborn piglets,improved gene expression of selenoproteins,and decreased oxidative status in the LD muscle of both newborn and weaned piglets compared with the control.The expression of muscle development-related genes of newborn piglets in the HMSeBA group was lower than in the control group,whereas the expression of MRF4 in weaned piglets was higher in the HMSeBA group than in the control and Na_(2)SeO_(3)groups.In addition,HMSeBA supplementation decreased the mRNA expressions of myosin heavy chains(MyHC)IIx and MyHC IIb and the percentage of MyHC IIb;increased the expression of PGC-1αin the LD muscle of newborn piglets;increased the gene expression of MyHC IIa;and decreased the protein expression of slow MyHC and the activity of malate dehydrogenase in the LD muscle of weaned piglets compared with the control group.Conclusions:Maternal HMSeBA supplementation during gestation can improve the antioxidative capacity of the muscle of their offspring and promote the maturity of muscle fibres in weaned offspring.Yan Lin Hui Yan Lei Cao Daolin Mou Dajiang Ding Binting Qin Lianqiang Che Zhengfeng Fang Shengyu Xu Yong Zhuo Jian Li Jianping Wang Chao Huang Yuanfeng Zou Lixia Li De Wu Bin Feng 2023Journal of Animal Science and Biotechnology2023,14,2:0
