1A two-step lineage reprogramming strategy to generate functionally competent human hepatocytes from fibroblasts显示文摘Terminally differentiated cells can be generated by lineage reprogramming,which is,however,hindered by incomplete conversion with residual initial cell identity and partial functionality. Here, we demonstrate a new reprogramming strategy by mimicking the natural regeneration route, which permits generating expandable hepatic progenitor cells and functionally competent human hepatocytes. Fibroblasts were first induced into human hepatic progenitor-like cells (hHPLCs), which could robustly expand in vitro and efficiently engraft in vivo. Moreover, hHPLCs could be efficiently induced into mature human hepatocytes (hiHeps) in vitro, whose molecular identity highly resembles primary human hepatocytes (PHHs). Most importantly, hiHeps could be generated in large quantity and were functionally competent to replace PHHs for drug-metabolism estimation, toxicity prediction and hepatitis B virus infection modeling. Our results highlight the advantages of the progenitor stage for successful lineage reprogramming. 3Chloroplast genomic diversity in Bulbophyllum section Macrocaulia(Orchidaceae,Epidendroideae,Malaxideae):Insights into species divergence and adaptive evolution显示文摘Bulbophyllum is the largest genus in Orchidaceae with a pan tropical distribution.Due to highly significant diversifications,it is considered to be one of the most taxonomically and phylogenetically complex taxa.The diversification pattern and evolutionary adaptation of chloroplast genomes are poorly understood in this species-rich genus,and suitable molecular markers are necessary for species determination and phylogenetic analysis.A natural Asian section Macrocaulia was selected to estimate the interspecific divergence of chloroplast genomes in this study.Here,we sequenced the complete chloroplast genome of four Bulbophyllum species,including three species from section Macrocaulia.The four chloroplast genomes had a typical quadripartite structure with a genome size ranged from 156,182 to 158,524 bp.The chloroplast genomes included 113 unique genes encoding 79 proteins,30 tRNAs and 4 rRNAs.Comparison of the four chloroplast genomes showed that the three species from section Macrocaulia had similar structure and gene contents,and shared a number of indels,which mainly contribute to its monophyly.In addition,interspecific divergence level was also great.Several exclusive indels and polymorphism SSR loci might be used for taxonomical identification and determining interspecific polymorphisms.A total of 20 intergenic regions and three coding genes of the most variable hotspot regions were proposed as candidate effective molecular markers for future phylogenetic relationships at different taxonomical levels and species divergence in Bulbophyllum.All of chloroplast genes in four Bulbophyllum species were under purifying selection,while 13 sites within six genes exhibited sitespecific selection.A whole chloroplast genome phylogenetic analysis based on Maximum Likelihood,Bayesian and Parsimony methods all supported the monophyly of section Macrocaulia and the genus of Bulbophyllum.Our findings provide valuable molecular markers to use in accurately identifying species,clarifying taxonomy,and resolving the phylogeny and evolution of the genus Bulbophyllum.The molecular markers developed in this study will also contribute to further research of conservation of Bulbophyllum species.Hanqing Tang Lu Tang Shicheng Shao Yulan Peng Lu Li Yan Luo 2021Plant Diversity2021,43,5:3
4Strain effect on the orientation-dependent harmonic spectrum of monolayer aluminum nitride显示文摘In this study, we theoretically investigate the strain effect on the orientation-dependent high-order harmonic generation(HHG) of monolayer aluminum nitride(Al N) by solving the multiband semiconductor Bloch equations in strong laser fields. Our simulations denote that the efficiency of the orientation-dependent HHG is considerably enhanced when a 15% biaxial tensile strain is applied to Al N, which is attributed to the downshifting energy level of the conduction band. Furthermore, the odd-even feature in the orientation-dependent high harmonic spectra owing to the strain is considerably different when compared with that in the case without strain. The enhanced quantum interference between different energy bands in strained Al N around the Γ-M direction is responsible for the observed odd-even distributions of the orientation-dependent HHG. 8Degradation of antipyrine in the Fenton-like process with a La-doped heterogeneous catalyst显示文摘The aim of this study was to synthesize a novel lanthanum(La)doped catalyst and to investigate antipyrine removal in wastewater using the Fenton-like process with the catalyst.The La-doped Co-Cu-Fe catalyst was synthesized using the modified hydrothermal method.Results showed that the Ladoped catalyst had higher specific surface area and lower particle size than the catalyst without La doping(i.e.,the control)(267 vs.163 m2/g and 14 vs.32 nm,respectively).Under the conditions of catalyst dosage 0.5 g/L,H2O2 concentration 1.70 g/L,and NaHCO3 0.1g/L,the antipyrine removal within 60 min using the Fenton-like process with the La-doped catalyst was much higher than that with the control(95%vs.54%).The hydroxyl radical concentration with the La-doped catalyst within 60 min was two times higher than that with the control(49.2 vs.22.1 gg/L).The high catalytic activity of La-doped catalyst was mainly attributed to its high specific surface area based on the X-ray photoelectron spectroscopy result.Our La-doped catalyst should have great potential to remove antipyrine in wastewater using the heterogeneous Fenton-like process.Shicheng Wei Cuiping Zeng Yaobin Lu Guangli Liu Haiping Luo Renduo Zhang 2019Frontiers of Environmental Science & Engineering2019,13,5:0

10Genetic Diversities of 23 Y‑Chromosome Short Tandem Repeat Loci in a Han Population in the Beijing Region显示文摘We investigated the polymorphisms of 23 Y‑short tandem repeat(STR)loci in a Han population in the Beijing region.Blood samples were collected from 255 unrelated Han males.DNA templates were amplified using the PowerPlex®Y23 system,and the amplification products were detected with a 3130 genetic analyzer.A total of 254 haplotypes were detected from the 255 unrelated Han males in the Beijing region.The gene diversity of these 23 Y‑STR loci was 0.3952–0.9721.The haplotype diversity was 0.99996 and discrimination capacity(DC)was more than 99.6%.The 23 Y‑STR loci used in this study are highly polymorphic in Han individuals in the Beijing region and are therefore suitable for paternal kinship identification.Studying allelic deletions such as DYS448 and DYS549 are important for examining Y‑STR polymorphisms and forensic testing.Shicheng Hao Xuan Zhang Yan Liu Di Lu 2018Journal of Forensic Science and Medicine2018,4,2:0
11Coordinatively unsaturated cobalt single-atom nanozymes for visual pesticides detection by smartphone-based platform显示文摘By adjusting the coordination environment of single-atom catalysts,the enzyme-like activity can be finely tuned for highly sensitive biosensing.Herein,we demonstrated that coordinatively unsaturated cobalt-nitrogen sites doped within porous carbon(SA-CoN_(3))could serve as highly efficient oxidase mimic.Compared with the typical planar four-coordination structure(SA-CoN_(4)),the as-obtained single-atom Co nanozymes anchored by three nitrogen atoms are found to display much higher oxidase-like catalytic efficiency.Combined theoretical and experimental analysis revealed that the coordinatively unsaturated Co sites could facilitate adsorption and activation of O_(2) molecule and thus improve their oxidase-like activity.Based on the enhanced oxidase-like activity of SA-CoN_(3),a paper/smartphone sensor for organophosphorus pesticides(OPs)was successfully constructed and used to quantify glyphosate in environmental and food samples with a low detection limit of 0.66μM.This work not only highlights the important role of coordination unsaturation of SA nanozymes for promoting oxidase-like activity,but also provides an easy and cost-effective way to conduct effective quantification of OPs in the field.Fangning Liu Zhe Li Hengya Wei Peng Xu Ge Kang Shicheng Zhu Tingting Wang Ruxue He Chuanxia Chen Yizhong Lu 2024Nano Research2024,17,4:0

13Gridded reconstruction of the population in the traditional cultivated region of China from 1776 to 1953显示文摘Using modern census and environmental factor data,this study first identified the environmental factors that significantly affect the population distribution through Geodetector analysis and then constructed a population spatial distribution model based on the random forest regression algorithm.Finally,with this model and historical population data that were examined and corrected by historians,gridded population distributions with a spatial resolution of 10 km by 10 km in the traditional cultivated region of China(TCRC,hereafter)were reconstructed for six time slices from 1776 to 1953.Using the reconstruction dataset,the spatiotemporal characteristics of the population distribution were depicted.The results showed that(1)the environmental factors that significantly affected the population density differences among counties in the TCRC mainly consisted of elevation,slope,relief amplitude,distances to the nearest prefectural and provincial capitals,distance to the nearest river and the climatology moisture index.(2)Using the census data of 1934 counties in the TCRC in 2000 and the abovementioned environmental factor data,a random forest regression algorithm-based population spatial distribution model was constructed.Its determination coefficient(R^(2))is 0.81.In 88.4%of the counties(districts),the relative errors of the model predictions were less than 50%.(3)From 1776 to 1953,the total population in the study area showed an uptrend.Prior to 1851,the population increased mainly in the Yangtze River Delta.During this period,the number of grid cells in which the population densities were greater than 500 persons per km^(2) increased from 292 to 683.From 1851 to 1953,the population increased extensively across the study area.In the North China Plain and the Pearl River Delta,the number of grid cells in which the population densities were greater than 500 persons per km^(2) increased from 36 to 88 and from 4 to 35,respectively.The spatial clustering pattern of the population distribution varied temporally.The potential reasons included the shifts in economic development hot spots,traditional beliefs,wars,famine,and immigration policies.(4)Between our reconstructions and the HYDE dataset,there are large differences in the data sources,selected environmental factors and modeling methods.As a consequence,in comparison to our reconstructions,there were fewer populations in the eastern area and more populations in the western area from 1776 to 1851 and more populations in urban areas and fewer populations in rural areas after 1851 in the HYDE dataset.Xuezhen ZHANG Fahao WANG Weidong LU Shicheng LI Jingyun ZHENG 2022Science China Earth Sciences2022,65,2:0
14Activation of lineage competence in hemogenic endothelium precedes the formation of hematopoietic stem cell heterogeneity显示文摘Hematopoietic stem and progenitor cells (HSPCs) are considered as a heterogeneous population, but precisely when, where and how HSPC heterogeneity arises remain largely unclear. Here, using a combination of single-cell multi-omics, lineage tracing and functional assays, we show that embryonic HSPCs originate from heterogeneous hemogenic endothelial cells (HECs) during zebrafish embryogenesis. Integrated single-cell transcriptome and chromatin accessibility analysis demonstrates transcriptional heterogeneity and regulatory programs that prime lymphoid/myeloid fates at the HEC level. Importantly, spi^(2+) HECs give rise to lymphoid/myeloid-primed HSPCs (L/M-HSPCs) and display a stress-responsive function under acute inflammation. Moreover, we uncover that Spi2 is required for the formation of L/M-HSPCs through tightly controlling the endothelial-to-hematopoietic transition program. Finally, single-cell transcriptional comparison of zebrafish and human HECs and human induced pluripotent stem cell-based hematopoietic differentiation results support the evolutionary conservation of L/M-HECs and a conserved role of SPI1 (spi2 homolog in mammals) in humans. 16Electronic properties of monolayer copper selenide with one-dimensional moirépatterns显示文摘Strain engineering is a vital way to manipulate the electronic properties of two-dimensional(2D)materials.As a typical representative of transition metal mono-chalcogenides(TMMs),a honeycomb CuSe monolayer features with one-dimensional(1D)moirépatterns owing to the uniaxial strain along one of three equivalent orientations of Cu(111)substrates.Here,by combining low-temperature scanning tunneling microscopy/spectroscopy(STM/S)experiments and density functional theory(DFT)calculations,we systematically investigate the electronic properties of the strained CuSe monolayer on the Cu(111)substrate.Our results show the semiconducting feature of CuSe monolayer with a band gap of 1.28 eV and the 1D periodical modulation of electronic properties by the 1D moirépatterns.Except for the uniaxially strained CuSe monolayer,we observed domain boundary and line defects in the CuSe monolayer,where the biaxial-strain and strain-free conditions can be investigated respectively.STS measurements for the three different strain regions show that the first peak in conduction band will move downward with the increasing strain.DFT calculations based on the three CuSe atomic models with different strain inside reproduced the peak movement.The present findings not only enrich the fundamental comprehension toward the influence of strain on electronic properties at 2D limit,but also offer the benchmark for the development of 2D semiconductor materials.Gefei Niu Jianchen Lu Jianqun Geng Shicheng Li Hui Zhang Wei Xiong Zilin Ruan Yong Zhang Boyu Fu Lei Gao Jinming Cai 2023Frontiers of physics2023,18,1:0
