2A review of topology optimization for additive manufacturing:Status and challenges显示文摘Topology optimization was developed as an advanced structural design methodology to generate innovative lightweight and high-performance configurations that are difficult to obtain with conventional ideas.Additive manufacturing is an advanced manufacturing technique building asdesigned structures via layer-by-layer joining material,providing an alternative pattern for complex components.The integration of topology optimization and additive manufacturing can make the most of their advantages and potentials,and has wide application prospects in modern manufacturing.This article reviews the main content and applications of the research on the integration of topology optimization and additive manufacturing in recent years,including multi-scale or hierarchical structural optimization design and topology optimization considering additive manufacturing constraints.Meanwhile,some challenges of structural design approaches for additive manufacturing are discussed,such as the performance characterization and scale effects of additively manufactured lattice structures,the anisotropy and fatigue performance of additively manufactured material,and additively manufactured functionally graded material issues,etc.It is shown that in the research of topology optimization for additive manufacturing,the integration of material,structure,process and performance is important to pursue high-performance,multi-functional and lightweight production.This article provides a reference for further related research and aerospace applications.Jihong ZHU Han ZHOU Chuang WANG Lu ZHOU Shangqin YUAN Weihong ZHANG 2021Chinese Journal of Aeronautics2021,34,1:33
3How would global-mean temperature change in the 21st century?显示文摘The time series of HadCRUT3 global-mean surface air temperature(GSAT) anomaly,Pacific decadal oscillation(PDO) index,and the equatorial Pacific sea surface temperature(SST) were utilized,and their long-term trends and multiple time-scale periodic oscillations were explored in this study.A long-term trend with a warming rate about 0.44°C /century during 1850-2008,two cool floors occurred respectively around 1910 and during 1950-1970,and three warm flats happened in the 1870s,1940s and since the year 1998 were found in the GSAT.In this duration,the variability of GSAT can be well reconstructed by the quasi-21-year,the quasi-65-year,and century-scale oscillations.The recent decadal warm flat is caused by their positive phase overlapping from these three oscillations.The maximum rising temperature reached 0.26°C was simulated in 2004 by the three oscillations.The quasi-21-year and the quasi-65-year oscillations were possibly caused by solar radiation and internal variability of the ocean-atmosphere system.Therefore,an outlook of GSAT for the 21st century was made based on the long-term trend and these three oscillations.It was expected that a cool floor and a warm flat of the GSAT would appear in the 2030s and 2060s,respectively.However,the highest warming range is predicted about 0.6°C,it is less than the threshold 2°C and IPCC projection.QIAN WeiHong LU Bo ZHU CongWen 2010Chinese Science Bulletin2010,55,19:20
4Multi-scale design and optimization for solid-lattice hybrid structures and their application to aerospace vehicle components显示文摘By integrating topology optimization and lattice-based optimization,a novel multi-scale design method is proposed to create solid-lattice hybrid structures and thus to improve the mechanical performance as well as reduce the structural weight.To achieve this purpose,a two-step procedure is developed to design and optimize the innovative structures.Initially,the classical topology optimization is utilized to find the optimal material layout and primary load carrying paths.Afterwards,the solid-lattice hybrid structures are reconstructed using the finite element mesh based modeling method.And lattice-based optimization is performed to obtain the optimal crosssection area of the lattice structures.Finally,two typical aerospace structures are optimized to demonstrate the effectiveness of the proposed optimization framework.The numerical results are quite encouraging since the solid-lattice hybrid structures obtained by the presented approach show remarkably improved performance when compared with traditional designs.Chuang WANG Jihong ZHU Manqiao WU Jie HOU Han ZHOU Lu MENG Chenyang LI Weihong ZHANG 2021Chinese Journal of Aeronautics2021,34,5:14
5Periodic oscillations in millennial global-mean temperature and their causes显示文摘Time series of solar radiation and north Pacific sea surface temperature(SST) index were used to analyze their causality relationship with various periodic oscillations in reconstructed millennial global-mean temperature series.The three long-term periods of the Medieval Warm Period(MWP),Little Ice Age(LIA) and recent Global Warming Period(GWP) were distinct in the temperature series.21-year,65-year,115-year and 200-year oscillations were derived from the temperature series after removing three long-term climatic temperatures.The phases of temperature oscillations significantly lagged behind oceanic SST and solar radiation variability.The recent decadal warm period was caused by the quasi-21-year temperature oscillation.At this century-cross period,the four oscillations reached their peaks simultaneously,which did not occur during the last millennium.Based on the long-term trend during the GWP and the four periodic oscillations,global-mean temperature is expected to drop to a new cool period in the 2030s and then a rising trend would be towards to a new warm period in the 2060s.QIAN WeiHong LU Bo 2010Chinese Science Bulletin2010,55,35:12
6Improvement of pristinamycin I(PI)production in Streptomyces pristinaespiralis by metabolic engineering approaches显示文摘Pristinamycin,produced by Streptomyces pristinaespiralis,which is a streptogramin-like antibiotic consisting of two chemically unrelated components:pristinamycin I(PI)and pristinamycin II(PII),shows potent activity against many antibiotic-resistant pathogens.However,so far pristinamycin production titers are still quite low,particularly those of PI.In this study,we constructed a PI single component producing strain by deleting the PII biosynthetic genes(snaE1 and snaE2).Then,two metabolic engineering approaches,including deletion of the repressor gene papR3 and chromosomal integration of an extra copy of the PI biosynthetic gene cluster(BGC),were employed to improve PI production.The final engineered strain DPIIDpapR3/PI produced a maximum PI level of 132 mg/L,with an approximately 2.4-fold higher than that of the parental strain S.pristinaespiralis HCCB10218.Considering that the PI biosynthetic genes are clustered in two main regions in the 210 kb“supercluster”containing the PI and PII biosynthetic genes as well as a cryptic polyketide BGC,these two regions were cloned separately and then were successfully assembled into the PI BGC by the transformation-associated recombination(TAR)system.Collectively,the metabolic engineering approaches employed is very efficient for strain improvement in order to enhance PI titer.Jiali Meng Rongrong Feng Guosong Zheng Mei Ge Yvonne Mast Wolfgang Wohlleben Jufang Gao Weihong Jiang Yinhua Lu 2017Synthetic and Systems Biotechnology2017,2,2:7
7Metabolic engineering of Streptomyces coelicolor for enhanced prodigiosins (RED) production显示文摘Bacterial prodigiosins are red-colored secondary metabolites with multiple activities,such as anticancer,antimalarial and immunosuppressive,which hold great potential for medical applications.In this study,dramatically enhanced prodigiosins(RED) production in Streptomyces coelicolor was achieved by combinatorial metabolic engineering,including inactivation of the repressor gene ohkA,deletion of the actinorhodin(ACT) and calcium-dependent antibiotic(CDA) biosynthetic gene clusters(BGCs) and multi-copy chromosomal integration of the RED BGC.The results showed that ohkA deletion led to a 1-fold increase of RED production over the wild-type strain M145.Then,the ACT and CDA BGCs were deleted successively based on the AohkA mutant(SBJ101).To achieve multi-copy RED BGC integration,artificial ΦC31 attB site(s) were inserted simultaneously at the position where the ACT and CDA BGCs were deleted.The resulting strains SBJ102(with a single deletion of the ACT BGC and insertion of one artificial attB site) and SBJ103(with the deletion of both BGCs and insertion of two artificial attB sites) produced 1.9-and 6-fold higher RED titers than M145,respectively.Finally,the entire RED BGC was introduced into mutants from SBJ101 to SBJ103,generating three mutants(from SBJ104 to SBJ106) with chromosomal integration of one to three copies of the RED BGC.The highest RED yield was from SBJ106,which produced a maximum level of 96.8 mg g^(-1) cell dry weight,showing a 12-fold increase relative to M145.Collectively,the metabolic engineering strategies employed in this study are very efficient for the construction of high prodigiosin-producing strains.Panpan Liu Hong Zhu Guosong Zheng Weihong Jiang Yinhua Lu 2017Science China(Life Sciences)2017,60,9:7
8Detection and characterization of diverse alpha-and betacoronaviruses from bats in China显示文摘Bats have been implicated as important reservoir hosts of alpha- and betacoronaviruses. In this study, diverse coronaviruses(CoVs) were detected in 50 of 951(positive rate 5.3%) intestinal specimens of eight bat species collected in four provinces and the Tibet Autonomous Region of China by pan-coronavirus RT-PCR screening. Based on 400-nt RNA-dependent RNA polymerase(Rd RP) sequence analysis, eight belonged to genus Alphacoronavirus and 42 to Betacoronavirus.Among the 50 positive specimens, thirteen gave rise to CoV full-length Rd RP gene amplification for further sequence comparison, of which three divergent sequences(two from a unreported province) were subjected to full genome sequencing. Two complete genomes of betacoronaviruses(JTMC15 and JPDB144) and one nearly-complete genome of alphacoronavirus(JTAC2) were sequenced and their genomic organization predicted. The present study has identified additional numbers of genetically diverse bat-borne coronaviruses with a wide distribution in China. 9Community Characteristics of Wild Fruit Forests Along Elevation Gradients and the Relationships Between the Wild Fruit Forests and Environments in the Keguqin Mountain Region ofⅠii显示文摘In this paper,the quantitative relationship between the wild fruit communities and direct environmental factors is discussed on the basis of detailed data on landscape scale habitats obtained through field vegetation investigation.The results from TWINSPAN and DCCA showed that:1) In the distribution sections of the wild fruit forest in the Keguqin Mountain region,the basic patterns characteristic of the different habitats are due to topographic factors,nutrients and moisture conditions;2) The elevation affected the most basic differentiation of plant communities in the study area,indicating that the elevation condition was the most important factor restricting the distribution of the wild fruit communities in the study area;3) The close relationship between the moisture content in the upper soil layer and the elevation reflected the influence of moisture conditions on both wild fruit and herb-layer communities;4) Nutrient differences not only indicated that the habitat conditions were different in themselves but also showed that the present nutrient conditions of the habitats were seriously affected by human activities.In summary,under complicated mountainous topographic conditions,the habitat conditions for the communities differed very significantly,and the combination of elevation,soil moisture content,total nitrogen,slope aspect,and pH value influenced and controlled the formation of community distribution patterns in the study area.ZHUANG Li TIAN Zhonping CHEN Yaning LI Weihong LI Jiangui LU Shuang 2012Journal of Mountain Science2012,9,1:4
10Synthesis and characterization of novel biocompatible nanocapsules encapsulated lily fragrance显示文摘Fragranced products are wildly used in our daily life. However, their storage, application and quality are severely influenced by their strong volatility. In order to overcome these natural defects of fragrances,fragrance encapsulation approaches have been developed recently. In this study, a mild, simple and lowcost method of fragrance encapsulation was invented. Liposomes composed of soya lecithin/DSPEPEG2000/PLGA were prepared to encapsulate lily fragrance(LF). And these nanosized fragrance products was named LF-NPs. The hydrodynamic diameters of LF-NPs were approximately 100 nm and their encapsulation efficiency was up to 21.9%. Besides, LF-NPs could also prolong the release time of lily fragrance. Finally, the cytotoxicity tests indicated that LF-NPs offered biocompatibility. 12Changes in fossil-fuel carbon emissions in response to interannual and interdecadal temperature variability显示文摘Relationships on interannual and interdecadal timescales among global mean air temperature,CO2 concentrations and fossil-fuel carbon emissions in four major developed countries (the United States,the United Kingdom,France,and Germany) were analyzed.On an interannual timescale,the United States fossil-fuel carbon emissions tend to increase during cold winters and decrease during warm winters,which is opposite to the situation in summer.On an interdecadal timescale,cold (warm) periods both in the United States and globally agree with high (low) periods of fossil-fuel carbon emissions,with the temperature variability leading by 5–7 years.The leading correlation on the interdecadal timescale and the asymmetry in seasonal correlation on the interannual timescale indicate that temperature variability is a possible cause of changes in fossil-fuel carbon emissions.QIAN WeiHong LU Bo LIANG HaoYuan 2011Chinese Science Bulletin2011,56,3:2
13Overexpression of the diguanylate cyclase CdgD blocks developmental transitions and antibiotic biosynthesis in Streptomyces coelicolor显示文摘Cyclic dimeric GMP(c-di-GMP)has emerged as the nucleotide second messenger regulating both development and antibiotic production in high-GC,Gram-positive streptomycetes.Here,a diguanylate cyclase(DGC),CdgD,encoded by SCO5345 from the model strain Streptomyces coelicolor,was functionally identified and characterized to be involved in c-di-GMP synthesis through genetic and biochemical analysis.cdgD overexpression resulted in significantly reduced production of actinorhodin and undecylprodigiosin,as well as completely blocked sporulation or aerial mycelium formation on two different solid media.In the cdgD-overexpression strain,intracellular c-di-GMP levels were 13-27-fold higher than those in the wild-type strain.In vitro enzymatic assay demonstrated that CdgD acts as a DGC,which could efficiently catalyze the synthesis of c-di-GMP from two GTP molecules.Heterologous overproduction of cdgD in two industrial Streptomyces strains could similarly impair developmental transitions as well as antibiotic biosynthesis.Collectively,our results combined with previously reported data clearly demonstrated that c-di-GMP-mediated signalling pathway plays a central and universal role in the life cycle as well as secondary metabolism in streptomycetes.Xiaocao Liu Guosong Zheng Gang Wang Weihong Jiang Lei Li Yinhua Lu 2019Science China(Life Sciences)2019,62,11:2
14Multiplex genome editing using a dCas9-cytidine deaminase fusion in Streptomyces显示文摘CRISPR/Cas-mediated genome editing has greatly facilitated the study of gene function in Streptomyces. However, it could not be efficiently employed in streptomycetes with low homologous recombination(HR) ability. Here, a deaminase-assisted base editor d Cas9-CDA-ULstr was developed in Streptomyces, which comprises the nuclease-deficient Cas9(dCas9), the cytidine deaminase from Petromyzon marinus(PmCDA1), the uracil DNA glycosylase inhibitor(UGI) and the protein degradation tag(LVA tag). Using d Cas9-CDA-ULstr , we achieved single-, double-and triple-point mutations(cytosine-to-thymine substitutions)at target sites in Streptomyces coelicolor with efficiency up to 100%, 60% and 20%, respectively. This base editor was also demonstrated to be highly efficient for base editing in the industrial strain, Streptomyces rapamycinicus, which produces the immunosuppressive agent rapamycin. Compared with base editors derived from the cytidine deaminase rAPOBEC1, the PmCDA1-assisted base editor dCas9-CDA-ULstr could edit cytosines preceded by guanosines with high efficiency, which is a great advantage for editing Streptomyces genomes(with high GC content). Collectively, the base editor dCas9-CDA-ULstr could be employed for efficient multiplex genome editing in Streptomyces. 15Influence of Coal Nature and Structure on Ash Size Formation Characteristic and Related Pollutant Emissions During CFB Combustion显示文摘The size distribution of coal particles in a Circulating Fluidized Bed (CFB) boiler plays a crucial role in the complicated combustion, heat exchange and pollutant emissions in such a plat. Therefore, it is fundamental to study the different factors having influence on the size distribution of coal particles. Above all, the coal itself and in particular, the coal combination phenomenon is a very influent factor. In the frame of this work, the coal nature (elementary composition) and coal internal structure (mineral components) are studied in detail. At this intermediary stage, experiments on three typical Chinese coals on a l.5 MWt CFBC pilot plant have been made. Some primary fragmentation tests have also been made in a small lab scale fluidized bed reactor. The results from the hot pilot test show i) the variation of coal ash distributions and other CFB performance data due to the cyclone and the coal characteristics and ii) the variation of desulfurization efficiency with limestone. 17Development of lunar regolith composite and structure via laser-assisted sintering显示文摘Aiming at the exploration and resource utilization activities on the Moon,in situ resource utilization and in situ manufacturing are proposed to minimize the dependence on the ground transportation supplies.In this paper,a laser-assisted additive manufacturing process is developed to fabricate lunar regolith composites with PA12/SiO2 mixing powders.The process parameters and composite material compositions are optimized in an appropriate range through orthogonal experiments to establish the relationship of process–structure–property for lunar regolith composites.The optimal combination of composite material compositions and process parameters are mixing ratio of 50/50 in volume,laser power of 30 W,scanning speed of 3500 mm/s,and scanning hatch space of 0.2 mm.The maximum tensile strength of lunar regolith composites reaches 9.248 MPa,and the maximum depth of surface variation is 120.79μm,which indicates poor powder fusion and sintering quality.Thereafter,the mechanical properties of laser-sintered lunar regolith composites are implemented to the topology optimization design of complex structures.The effectiveness and the feasibility of this laser-assisted process are potentially developed for future lightweight design and manufacturing of the solar panel installed on the lunar rover.Hua ZHAO Lu MENG Shaoying LI Jihong ZHU Shangqin YUAN Weihong ZHANG 2022Frontiers of Mechanical Engineering2022,17,1:1

20Effect of valproic acid on endogenous neural stem cell proliferation in a rat model of spinal cord injury显示文摘BACKGROUND:Valproic acid has been reported to decrease apoptosis,promote neuronal differentiation of brain-derived neural stem cells,and inhibit glial differentiation of brain-derived neural stem cells. OBJECTIVE:To investigate the effects of valproic acid on proliferation of endogenous neural stem cells in a rat model of spinal cord injury. DESIGN,TIME AND SETTING:A randomized,controlled,neuropathological study was performed at Key Laboratory of Trauma,Burrning,and Combined Injury,Research Institute of Surgery,Daping Hospital,the Third Military Medical University of Chinese PLA between November 2005 and February 2007. MATERIALS:A total of 45 adult,Wistar rats were randomly divided into sham surgery(n=5),injury (n=20),and valproic acid(n=20) groups.Valproic acid was provided by Sigma,USA. METHODS:Injury was induced to the T_(10) segment in the injury and valproic acid groups using the metal weight-dropping method.The spinal cord was exposed without contusion in the sham surgery group.Rats in the valproic acid group were intraperitoneally injected with 150 mg/kg valproic acid every 12 hours(twice in total). MAIN OUTCOME MEASURES:Nestin expression(5 mm from injured center) was detected using immunohistochemistry at 1,3 days,1,4,and 8 weeks post-injury. RESULTS:Low expression of nestin was observed in the cytoplasm,but rarely in the white matter of the spinal cord in the sham surgery group.In the injury group,nestin expression was observed in the ependyma and pia mater one day after injury,and expression reached a peak at 1 week(P<0.05). Expression was primarily observed in the ependymal cells,which expanded towards the white and gray matter of the spinal cord.Nestin expression rapidly decreased by 4 weeks post-injury,and had almost completely disappeared by 8 weeks.At 24 hours after spinal cord injury,there was no significant difference in nestin expression between the valproic acid and injury groups.At 1 week, there was a significant increase in the number of nestin-positive cells surrounding the central canal in valproic acid group compared with the injury group(P<0.05).Expression reached a peak by 4 weeks,and it was still present at 8 weeks.
