1Expression of vascular endothelial growth factor and its receptors KDR and Fit-1 in gastric cancer cells显示文摘AIM: The expression of vascular endothelial growth factor (VEGF) and its receptors KDR and Flt-1 by gastric carcinoma tissues and different gastric carcinoma cell lines was detected to elucidate the molecular mechanism of this growth factor in promoting tumor growth.METHODS: The expression of VEGF, Flt-1 and KDR was determined by reverse transcription-polymerase chain reaction (RT-PCR) in gastric cancer cell lines RF-1, RF-48,AGS-1, NCI-N87, NCI-SNU-1, NCI-SNU-5, NCI-SNU-16 and KATO-Ⅲ. The expression of Flt-1 and KDR in paraffinembedded specimens of gastric cancer was determined by immunohistochemistry. The 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay was used to assess the role of VEGF in tumor cell proliferation.RESULTS: All 8 gastric cancer cell lines analyzed expressed VEGF121 and VEGF16s and six of them expressed both Flt-1 and KDR, while cell line NCI-SNU-5 expressed Flt-1 only and cell line KATOⅢ expressed neither Flt-1 nor KDR. The gastric carcinoma tissues expressed Flt-1 and KDR widely,with the positive rate of expression of Flt-1 and KDR being 84.6 % and 70 % respectively. The exogenous VEGF stimulated the growth of KDR-positive cell lines NCI-N87 and AGS-1 in a dose-dependent manner but exhibited no effect on the growth of KDR-negative cell line NCI-N87.CONCLUSION: VEGF and its receptors KDR and Flt-1 were expressed widely in gastric carcinoma cells and the VEGF stimulated KDR-positive tumor cell growth directly. 2Small sphincterotomy combined with endoscopic papillary large balloon dilation vs sphincterotomy alone for removal of common bile duct stones显示文摘AIM:To evaluate the efficacy and safety of endoscopic papillary large diameter balloon dilation(EPLBD)following limited endoscopic sphincterotomy(EST)and EST alone for removal of large common bile duct(CBD)stones.METHODS:We retrospectively compared EST+EPLBD(group A,n=64)with EST alone(group B,n=89)for the treatment of large or multiple bile duct stones.The success rate of stone clearance,procedure-related complications and incidents,frequency of mechanical lithotripsy use,and recurrent stones were recorded.RESULTS:There was no statistically significant difference between the two groups regarding periampullary diverticula(35.9%vs 34.8%,P>0.05),pre-cut sphincterotomy(6.3%vs 6.7%,P>0.05),size(12.1±2.0mm vs 12.9±2.6 mm,P>0.05)and number(2.2±1.9vs 2.4±2.1,P>0.05)of stones or the diameters of CBD(15.1±3.3 mm vs 15.4±3.6 mm,P>0.05).The rates of overall stone removal and stone removal in the first session were not significantly different between the two groups[62/64(96.9%)vs 84/89(94.4%),P>0.05;and 58/64(90.6%)vs 79/89(88.8%),P>0.05,respectively].The rates of post-endoscopic retrograde cholangiopancreatography pancreatitis and hyperamylasemia were not significantly different between the two groups[3/64(4.7%)vs 4/89(4.5%),P>0.05;7/64(10.9%)vs 9/89(10.1%),P>0.05,respectively].There were no cases of perforation,acute cholangitis,or cholecystitis in the two groups.The rate of bleeding and the recurrence of CBD stones were significantly lower in group A than in group B[1/64(1.6%)vs 5/89(5.6%),P<0.05;1/64(1.6%)vs 6/89(6.7%),P<0.05,respectively].CONCLUSION:EST+EPLBD is an effective and safe endoscopic approach for removing large or multiple CBD stones.Shi-Bin Guo Hua Meng Zhi-Jun Duan Chun-Yan Li 2014World Journal of Gastroenterology2014,20,47:43
3Differential expression of serum miR-126,miR-141 and miR-21 as novel biomarkers for early detection of liver metastasis in colorectal cancer显示文摘Objective:MicroRNAs(miRNAs) have potential as diagnostic biomarkers in cancer.Evaluation of the association between miRNA expression patterns and early detection of liver metastasis in colorectal cancer(CRC) has not been reported.Methods:We investigated the expression of metastasis-associated miRs-31,335,206,141,126,200b,200c,21,Let7a,Let7b and Let7c in localized,liver-metastatic and other organ-metastatic CRC(OM-CRC).Expressions of target miRNAs in serum were evaluated in 116 consecutive localized CRC(L-CRC),72 synchronous liver-metastatic CRC(SLM-CRC) and 36 other OM-CRC by quantitative real-time PCR.Results:Seven of 11 tested miRNAs could be detected from serum.Four miRNAs,miR-126,Let-7a,miR-141 and miR-21 were identified as metastasis-associated miRNAs.Compared with L-CRC,significant upregulated expression was observed for miR-141 and miR-21 in SLM-CRC and OM-CRC,down-regulated expression was observed for miR-126 in SLM-CRC and OM-CRC,and up-regulated expression of Let-7a in OM-CRC.The receiver operating characteristic(ROC) curve showed serum miR-126 had a cut-off [log 10 relative quantity(log 10 RQ)=–0.2005] with 77.78% sensitivity and 68.97% specificity with an area under curve(AUC) of 0.7564,miR-141 had a cut-off(log 10 RQ=–0.2285) with 86.11% sensitivity and 76.11% specificity with an AUC of 0.8279,and miR-21 had a cut-off(log 10 RQ=–0.1310) with 73.61% sensitivity and 66.38% specificity with an AUC of 0.7479.Conclusions:We identified liver metastasis-associated miRNAs,suggesting serum miR-126,miR-141 and miR-21 may be novel biomarkers for clinical diagnosis of early stage liver-metastatic CRC.Jie Yin Zhigang Bai Jianning Song Yun Yang Jin Wang Wei Han Jun Zhang Hua Meng Xuemei Ma Yao Yang Tingting Wang Weirong Li Zhongtao Zhang 2014Chinese Journal of Cancer Research2014,26,1:22
4含亚稳残余奥氏体HSLA TR IP钢的摩擦磨损性能研究显示文摘在SST-ST型销-盘摩擦磨损试验机上对特殊工艺热处理、成分为0.22C-1.50S i-1.65M n的新型相变诱发塑性(HSLA S i-M n系TR IP)钢的摩擦磨损性能进行研究,采用X射线衍射技术分析了摩擦过程中钢中残余奥氏体的变形诱发转变行为.结果表明,在本试验条件下,与相同成分并在两相区双相化处理的双相钢相比,TR IP钢具有较低摩擦系数和较好耐磨性.由于HSLA S i-M n系TR IP钢中亚稳残余奥氏体在摩擦应力作用下的变形诱发相变作用,使得其磨损区的大量残余奥氏体向马氏体转变,一方面松弛了磨损表面的内应力,另一方面转变的马氏体使基体强化,从而使其具有较好摩擦磨损特性.韦习成 李健 MENG Hua 2006摩擦学学报2006,26,1:16

7Paleogene Tectonic Evolution Controls on Sequence Stratigraphic Patterns in the Central Part of Deepwater Area of Qiongdongnan Basin, Northern South China Sea显示文摘In active rift basins, tectonism is extremely important for sequence stratigraphic patterns, affecting both the sequence architecture and internal makeup. Sequence stratigraphic framework of a Paleogene rift succession in Qiongdongnan Basin, northern South China Sea, was built using seismic profiles, complemented by well logs and cores. One first-order and three second-order sequences were identified on the basis of basin-scale unconformities, and seven third-order sequences are defined by unconformities along the basin margins and correlative conformities within the central basin. Through unconformity analysis and backstripping procedure, the Paleogene synrift tectonic evolution of deepwater area of Qiongdongnan Basin was proved to be episodic, which can be divided into rifting stage-I, rifting stage-II and rifting stage-III. Episodic rifting resulted in the formation of various types of structural slope break belts, which controlled different architectures and internal makeup of sequences. 8Molecular Epidemiological Characteristics of Streptococcus pyogenes Strains Involved in an Outbreak of Scarlet Fever in China,2011显示文摘Objective To investigate molecular characterization of streptococcus pyogenes isolates involved in an outbreak of scarlet fever in China in 2011.Methods Seventy-four Streptococcal pyogenes involved in an outbreak of scarlet fever were isolated from pediatric patients in the areas with high incidence in China from May to August of 2011.Emm genotyping,pulsed-field gel electrophoresis (PFGE),superantigen (SAg) genes and antimicrobial susceptibility profiling were analyzed for these isolates.Results A total of 4 different emm types were identified.Emm12 was the most prevalent type which contained four predominating PFGE patterns corresponding to four different virulence and superantigen profiles.Emm12 (79.7%) and emml (14.9%) accounted for approximately 94%of all the isolates.The speA gene was all negative in emm12 isolates and positive in emml isolates.All strains were resistant to erythromycin,and 89.4%of them were resistant to erythromycin,tracycline,and clindamycin simultaneously.Conclusion Several highly diversified clones with a high macrolide resistance rate comprise a predominant proportion of circulating strains,though no new emm type was found in this outbreak.The data provide a baseline for further surveillance of scarlet fever,which may contribute to the explanation of the outbreak and development of a GAS vaccine in China.YOU Yuan Hai SONG Yan Yan YAN Xiao Mei WANG Hai Bin ZHANG Meng Han TAO Xiao Xia Li Lei Lei ZHANG Yu Xin JIANG Xi Hong ZHANG Bing Hua ZHOU Hao XIAO Di JIN Lian Mei FENG Zi Jian LUO Feng Ji ZHANG Jian Zhong 2013Biomedical and Environmental Sciences2013,26,11:15

11VdPKS1 is required for melanin formation and virulence in a cotton wilt pathogen Verticillium dahliae显示文摘Verticillium dahliae is a soil-borne phytopathogenic fungus that causes vascular wilt disease in a broad range of hosts.This pathogen survives for many years in soil in the form of melanized microsclerotia.To investigate the melanin synthesis in V.dahliae,we identified a polyketide synthase gene in V.dahliae,namely VdPKS1.PKS1 is known to involve in the dihydroxynaphthalene melanin synthesis pathway in many fungi.We found that VdPKS1 was required for melanin formation but not for microsclerotial production in V dahliae.The VdPKS1 gene-disruption mutant (vdpksl) formed melanin-deficient albino microsclerotia,which did not affect the fungal colonization in host tissues but significantly reduced the disease severity.Gene transcription analysis in the wild-type and the vdpksl strains suggested that VdPKS1 gene-disruption influenced the expression of a series of genes involved in ethylene biosynthesis,microsclerotial formation and pathogenesis.Our results suggest that the VdPKS1-mediated melanin synthesis is important for virulence and developmental traits of V.dahliae.Ting Zhang Bosen Zhang Chenlei Hua Pei Meng Sheng Wang Zhirong Chen Yejuan Du Feng Gao Jiafeng Huang 2017Science China(Life Sciences)2017,60,8:14
12Comparison Between a Tetraploid Turnip and Its Diploid Progenitor(Brassica rapa L.):The Adaptation to Salinity Stress显示文摘Polyploidy is pursued in plant breeding programs due mainly to its ability to yield larger vegetative or reproductive organs. In controlled growth chamber experiments, a tetraploid turnip (cv. Aijiaohuang, 4n) and its diploid progenitor (cv. Aijiaohuang, 2n) were evaluated for their tolerance to salinity stress via investigations on a group of physiological parameters. The results indicate that the tetraploid turnip exhibit better adaptation to a high concentration salt medium (200 mmol L-1), as evidenced by a less-affected germination rate and a healthier morphological appearance at the seedling stage. Furthermore, an extension of salinity stress up to a certain period of time at the 5-7-leaf stage shows differences between the tetraploid turnip and its diploid progenitor. The former had a higher K+/Na+ ratio in the roots, higher glutathione concentration and antioxidant activities in the leaves, and smaller reductions in photosynthetic capacity in terms of leaf chlorophyll content. Studies on the differences between an autopolyploid and its respective relative, from which the autopolyploid originated, in terms of their tolerance to salinity and/or other abiotic stresses, have remained rather limited. 14OsBRXL4 Regulates Shoot Gravitropism and Rice Tiller Angle through Affecting LAZY1 Nuclear Localization显示文摘Rice tiller angle is a key agronomic trait that contributes to ideal plant architecture and grain production.LAZY1 (LA1) was previously shown to control tiller angle via affecting shoot gravitropism,but the underlying molecular mechanism remains largely unknown.In this study,we identified an LA1-interacting protein named Brevis Radix Like 4 (OsBRXL4).We showed that the interaction between OsBRXL4 and LA1 occurs at the plasma membrane and that their interaction determines nuclear localization of LA1.We found that nuclear localization of LA1 is essential for its function,which is different from AtLA1,its Arabidopsis ortho.log.Overexpression of OsBRXL4 leads to a prostrate growth phenotype,whereas OsBRXLs RNAi plants,in which the expression levels of OsBRXLI,OsBRXL4,and OsBRXL5 were decreased,display a compact phenotype.Further genetic analysis also supported that OsBRXL4 controls rice tiller angle by affecting nuclear localization of LA1.Consistently,we demonstrated that OsBRXL4 regulates the shoot gravitropism through affecting polar auxin transport as did LA1.Taken together,our study not only identifies OsBRXL4 as a regulatory component of rice tiller angle but also provides new insights into genetic regulation of rice plant architecture.Zhen Li Yan Liang Yundong Yuan Lei Wang Xiangbing Meng Guosheng Xiong Jie Zhou Yueyue Cai Ningpei Han Lekai Hua Guifu Liu Jiayang Li Yonghong Wang 2019Molecular Plant2019,12,8:12

18The enhanced X-ray Timing and Polarimetry mission—eXTP显示文摘In this paper we present the enhanced X-ray Timing and Polarimetry mission—eXTP. eXTP is a space science mission designed to study fundamental physics under extreme conditions of density, gravity and magnetism. The mission aims at determining the equation of state of matter at supra-nuclear density, measuring effects of QED, and understanding the dynamics of matter in strong-field gravity. In addition to investigating fundamental physics, eXTP will be a very powerful observatory for astrophysics that will provide observations of unprecedented quality on a variety of galactic and extragalactic objects. 19Antibacterial Mechanisms of Berberine and Reasons for Little Resistance of Bacteria显示文摘Objective To study the antibacterial mechanisms of berberine and try to understand the reasons why bacteria cells difficultly resisted to it.Methods Detecting the minimal inhibitory concentration(MIC) of bacterial cultures incubated under sub-MIC concentration of berberine,Huanglian,and Neomycin for more than 200 generations,in order to analyze the bacteria resistance.Detecting the binding kinetics of berberine to DNA,RNA,and proteins.Observing the changes in bacterial cell surface structure with scanning electron microscopy.Detecting the Ca2 +and K +released from berberine-treated bacterial cells with atomic absorption spectrum.Detection the absorption of methyl-3H-thymine (3 H-dT),3 H-uridine(3 H-U),and 3 H-tyrosine(3 H-Tyr)into berberine-treated bacterial cells.Results MICs of bacterial cultures,growing more than 200 generations in MH medium with 1/2 MIC of berberine(BA200) or Huanglian(HA200),did not increase compared to the control,while remarkably increased in MH medium with 1/2 MIC of Neomycin (NA200).In addition,from the culture NA200 it was easy to isolate resistant mutant strains which could grow in MH medium with more than four times MIC Neomycin,but from the culture BA200 and HA200 it was difficult to isolate berberine or Huanglian mutant strains could grow in MH medium with more than four times MIC berberine or Huanglian.The binding kinetics of berberine to DNA,RNA,and proteins illustrated that berberine could easily and tightly bind to DNA and RNA,and hardly dis-bind from DNA-and RNA-berberine complexes.Berberine could easily bind to protein too,but also easily dis-bind from berberine-protein complex.The bacterial cells treated with berberine sharply decreased the absorption of 3 H-dT,3 H-U,and 3 H-Tyr,as the radioactive precursors of DNA,RNA,and protein biosynthesis.Berberine could damage bacterial cell surface structure,especially for Gram-negative bacteria.Ca2 +and K + released from berberine-treated cells increased significantly compared to the control.Conclusion All of above results indicate that bacterial cells could not easily become resistant mutants to berberine.The mechanisms for the bactericidal effect of berberine include:inhibiting DNA duplication,RNA transcription,and protein biosynthesis;influencing or inhibiting enzyme activities;destructing the bacterial cell surface structure and resulting in Ca2 +and K +released from cells.All of the berberine bactericidal mechanisms are the most essential physiological functions for a live cell,if influenced any one such function,the mutation would be lethal mutation,so that it is difficult to get berberine resistant cells.The results in this paper also prefigure that berberine and its related Chinese medicines would provide a feasible way to control antibiotic resistance problem.JIN Jian-ling,HUA Guo-qiang,MENG Zhen,GAO Pei-ji State Key Laboratory of Microbial Technology,School of Life Sciences,Shandong University,Jinan 250100,China 2011Chinese Herbal Medicines2011,3,1:11
20Evaluation of a New Real-time PCR Assay for Detection of Mycoplasma Pneumoniae in Clinical Specimens显示文摘Objective To establish and evaluate a real-time PCR assay to detect Mycoplasma pneumoniae (M.pneumoniae) in clinical specimens.Methods By analysing the whole p1 gene sequence of 60 M.pneumoniae clinical isolates in Beijing of China,an optimized real-time PCR assay (MpP1) using p1 gene conserved region was designed.The specificity and sensitivity of this assay were evaluated and compared with other two reported assays (RepMp1 and Mp181) using 40 positive and 100 negative clinical specimens.Results The detection limit of the new assay was 8.1 fg (about 1~3CFU) M.pneumoniae DNA.The sensitivity of MpP1,RepMp1,and Mp181 assays appeared to be 100%,100%,and 85%,respectively.Conclusion MpP1 assay is suitable for the detection of M.pneumoniae in Chinese clinical specimens.ZHAO Fei CAO Bin HE Li Hua YIN Yu Dong TAO Xiao Xia SONG Shu Fan MENG Fan Liang ZHANG Jian Zhong 2012Biomedical and Environmental Sciences2012,25,1:11
