| 2 | Identifying changes in the wheat kernel proteome under heat stress using iTRAQ显示文摘Wheat(Triticum aestivum L.) is one of the three major global food crops. Hightemperature stress can affect its yield and quality. Studies of the effect of hightemperature stress on wheat kernel development are important because they can reveal the stability of wheat quality and lead to the genetic improvement of wheat quality traits. In this study, the isobaric tags for relative and absolute quantitation(iTRAQ)method was adopted to analyze changes in the protein expression profile of wheat cultivars under high temperature stress. The protein content of wheat grain increased under heat stress, while the SDS-sedimentation value and starch content decreased.Grain filling was deficient under high temperature stress, which reduced thousandkernel weight but did not affect wheat kernel length. The 207 differentially expressed proteins identified in Gaocheng 8901 under heat stress were associated with energy metabolism, growth and development, and stress response. Gene Ontology enrichment analysis showed that the annotated proteins that were differentially expressed in Gaocheng 8901 under heat stress were involved mainly in stimulus response, abiotic stress response, stress response, and plasma membrane. A set of 78 differentially expressed proteins were assigned to 83 KEGG signaling/metabolic pathways. KEGG pathway enrichment analysis showed that this set of proteins was significantly enriched in members of 51 pathways, and the proteins participated mainly in protein synthesis in the endoplasmic reticulum, starch and sucrose metabolism, and reaction on ribosomes. Five differentially expressed proteins were involved in protein–protein interaction networks that may greatly influence the yield and quality of wheat grain. | 3 | Effects of Huatan Tongluo decoction on vascular endothelial growth factor receptor 2 expression in synovial tissues of rats with collagen-induced arthritis显示文摘OBJECTIVE: To determine the therapeutic effect and potential mechanism of Huatan Tongluo decoction on rats with collagen-induced arthritis.METHODS: Forty specific pathogen-free Wistar rats were selected, and 10 were randomly selected as the control(group 1). The remaining rats were injected intradermally with emulsified type Ⅱ bovine collagen at the tail base and back, followed by a booster 7 d post first immunization. After establishing collagen-induced arthritis(CIA), rats were randomly divided into three groups(n = 10). The rats were treated orally for 30 d as follows: group 1, saline; group 2, model(saline); group 3, tripterygium polyglycoside(TP; 7.81 mg/kg, positive control);group 4, Huatan Tongluo decoction(HTTL; 7.5 g/kg). Body weight, ankle swelling and arthritis index were measured over the course of the study. The rats were sacrificed 30 d after treatment. Morphological changes in the synovium were observed by hematoxylin and eosin staining. Pannus formation and synovial thickness in the left ankle were observed by color Doppler ultrasoundVascular endothelial growth factor(VEGF) and VEGFR2 protein levels were measured by immunohistochemistry.VEGF/VEGFR2 mRNA levels were measured by real-time quantitative polymerase chain reaction.RESULTS: Compared with the model group, a significantly lower arthritis index was observed in the positive control group(P < 0.05) and HTTL group(P < 0.01), after treatment. Both positive control and HTTL reduced intra-articular pannus formation and synovial thickening. | 8 | TaMADS29 interacts with TaNF-YB1 to synergistically regulate early grain development in bread wheat显示文摘Grain development is a crucial determinant of yield and quality in bread wheat(Triticum aestivum L.).However,the regulatory mechanisms underlying wheat grain development remain elusive.Here we report how Ta MADS29 interacts with Ta NF-YB1 to synergistically regulate early grain development in bread wheat.The tamads29 mutants generated by CRISPR/Cas9 exhibited severe grain filling deficiency,coupled with excessive accumulation of reactive oxygen species(ROS)and abnormal programmed cell death that occurred in early developing grains,while overexpression of Ta MADS29 increased grain width and1,000-kernel weight.Further analysis revealed that Ta MADS29 interacted directly with Ta NF-YB1;null mutation in Ta NF-YB1caused grain developmental deficiency similar to tamads29 mutants.The regulatory complex composed of Ta MADS29 and Ta NF-YB1 exercises its possible function that inhibits the excessive accumulation of ROS by regulating the genes involved in chloroplast development and photosynthesis in early developing wheat grains and prevents nucellar projection degradation and endosperm cell death,facilitating transportation of nutrients into the endosperm and wholly filling of developing grains.Collectively,our work not only discloses the molecular mechanism of MADS-box and NF-Y TFs in facilitating bread wheat grain development,but also indicates that caryopsis chloroplast might be a central regulator of grain development rather than merely a photosynthesis organelle.More importantly,our work offers an innovative way to breed high-yield wheat cultivars by controlling the ROS level in developing grains. | Guoyu Liu Runqi Zhang Sen Li Rehmat Ullah Fengping Yang Zihao Wang Weilong Guo Mingshan You Baoyun Li Chaojie Xie Liangsheng Wang Jie Liu Zhongfu Ni Qixin Sun Rongqi Liang | 2023 | Science China(Life Sciences)2023,66,7: | 1 |

| 10 | Phase noise floor suppression of the output carrier from double sideband-carrier suppressed modulation system显示文摘A theoretical model of all the influencing factors on the phase noise floor of the output carrier from the double sideband-carrier suppressed (DSB-CS) modulation system is presented. Based on the established DSB-CS modulation system with different cases, the phase noise of the input carrier and the output doubled frequency carrier is measured and calculated using the theoretical model, respectively. The calculation results basically agree with the measurement results. It is shown that the phase noise floor of the output carrier is primarily influenced by the signal-spontaneous beat noise and the relative intensity noise (RIN) of the laser. | 11 | Stress corrosion cracking behaviors of RE-containing ME21 magnesium alloy processed by equal-channel angular pressing显示文摘A commercial as-cast ME21 magnesium alloy containing rare-earth (RE) element was processed by equalchannel angular pressing to obtain fine-grained micro structure. Stress corrosion cracking (SCC) behaviors of the fine-grained samples were studied by slow-strain-rate testing in air, distilled water and Hanks’solution at the strain rate of 1×10^(-6) s^(-1). All samples show a relatively low SCC sensitivity in distilled water but a great SCC tendency in Hanks’ solution. The microscopic observations of the fracture surfaces and the side surfaces reveal obvious active anodic dissolution and hydrogen embrittlement cracks, which indicate the higher SCC susceptibility in Hanks'solution. | 14 | Sessile droplet freezing and ice adhesion on aluminum with different surface wettability and surface temperature显示文摘This paper focused on the sessile droplet freezing and ice adhesion on aluminum with different wettability(hydrophilic, common hydrophobic, and superhydrophobic surfaces, coded as HIS, CHS, SHS, respectively) over a surface temperature range of ?9°C to ?19°C. It was found that SHS could retard the sessile droplet freezing and lower the ice adhesion probably due to the interfacial air pockets(IAPs) on water/SHS interface. However, as surface temperature decreasing, some IAPs were squeezed out and such freezing retarding and adhesion lowering effect for SHS was reduced greatly. For a surface temperature of ?19°C, ice adhesion on SHS was even greater than that on CHS. To discover the reason for the squeezing out of IAPs, forces applied to the suspended water on IAPs were analyzed and it was found that the stability of IAPs was associated with surface micro-structures and surface temperature. We firstly establish the relationship between denoising model and ill-posed inverse problem via convex half-quadratic regularization, and compare the difference between the estimator variance obtained from the iterative formula and biased Cramer-Rao bound, which proves the theoretic flaw of the existent methods of parameter selection. Then, the analytic expression of the model solution as the function with respect to the regularization parameter is obtained. On this basis, we study the method for selecting the regularization parameter through minimizing mean-square error of estimators and obtain the final analytic expression, which resulted in the direct calculation, high processing speed, and adaptability. Finally, the effect of regularization parameter selection on the resolution of point targets is analyzed. | 19 | Effects of Boscalid on the Antioxidant Enzyme System of Adult Zebrafish (Danio rerio)显示文摘[Objective] This study was conducted to evaluate the ecotoxicity of boscalid to adult zebrafish(Danio rerio). [Method]The activities of antioxidant enzymes superoxide dismutase(SOD), catalase(CAT), peroxidase(POD), glutathione peroxidase(GPx), as well as a non-enzymatic antioxidant malondialdehyde(MDA), in the liver were measured 3, 7, 14 and 21 d post exposure(dpe) to 0.02(1/100 of acute toxicity), 0.036(monitored concentration), 0.08(1/20 of acute toxicity), 0.16(1/10 of acute toxicity) and 0.32 mg/L(1/5 of acute toxicity) boscalid using a semi-static method. [Result] SOD, CAT, POD, GPx and MDA activity in the liver of zebrafish varied with boscalid concentration and exposure time. Boscalid significantly enhanced MDA content at 21 dpe. A significant upregulation of the activity of SOD, CAT, POD and GPx at 7 dpe was observed, suggesting that boscalid resulted in oxidative stress and lipid peroxidation.
