1The discovery of annually laminated sediments (varves) from shallow Sugan Lake in inland arid China and their paleoclimatic significance显示文摘Detailed examination of sedimentary cores retrieved from Sugan Lake in the northern Qaidam Basin of northwest China’s Tibetan Plateau reveal that fine laminated beddings form in the sediments where water depth exceeds 3 m. Seasonal surface sediments trapped at the bottom of the lake suggest that sediments deposited during summer and autumn are mainly light colored monohydrocalcites, while those deposited in winter are dark organic matter, indicating that varve layers form under modern limnological conditions. Continuous varve sediments comprising four types have accumulated in the upper 5.5 m of Core SG03I from the center of the lake. All types exhibit clear seasonality indicative of annual deposition. Varve counts correspondence with 210Pb dates on recent sediments in the upper core suggest the continuous varves of the upper 5.5 m of the core formed in the late Holocene (2670 a BP). 3Emerging roles of activating transcription factor (ATF) family members in tumourigenesis and immunity: Implications in cancer immunotherapy显示文摘Activating transcription factors, ATFs, are a group of bZIP transcription factors that act as homodimers or heterodimers with a range of other bZIP factors. In general, ATFs respond to extracellular signals, indicating their important roles in maintaining homeostasis. The ATF family includes ATF1, ATF2, ATF3, ATF4, ATF5, ATF6, and ATF7. Consistent with the diversity of cellular processes reported to be regulated by ATFs, the functions of ATFs are also diverse. ATFs play an important role in cell proliferation, apoptosis, differentiation and inflammation-related pathological processes. The expression and phosphorylation status of ATFs are also related to neurodegenerative diseases and polycystic kidney disease. Various miRNAs target ATFs to regulate cancer proliferation, apoptosis, autophagy, sensitivity and resistance to radiotherapy and chemotherapy. Moreover, ATFs are necessary to maintain cell redox homeostasis. Therefore, deepening our understanding of the regulation and function of ATFs will provide insights into the basic regulatory mechanisms that influence how cells integrate extracellular and intracellular signals into genomic responses through transcription factors. Under pathological conditions, especially in cancer biology and response to treatment, the characterization of ATF dysfunction is important for understanding how to therapeutically utilize ATF2 or other pathways controlled by transcription factors. 8Early T-cell precursor leukemia:a subtype of high risk childhood acute lymphoblastic leukemia显示文摘Acute lymphoblastic leukemia includes T-cell acute lymphoblastic leukemia(T-ALL)and B-cell acute lymphoblastic leukemia(B-ALL).In children,T-ALL usually has a worse prognosis than B-ALL,although childhood T-ALL prognoses have improved remarkably.The varying outcomes among T-ALL cases suggest that an unrecognized biological heterogeneity may contribute to chemo-resistance.Deep exploration of T-lymphocyte development in recent years has found a subgroup of patients with a phenotype that resembles early T-cell precursor,which confers a much poorer prognosis than any other form of T-ALL.This novel subtype of T-ALL was called early T-cell precursor acute lymphoblastic leukemia(ETP-ALL).Flow cytometry data from T-ALL patients enrolled in Shanghai Children’s Medical Center between July 2002 and October 2010 were assessed according to Dr.Campana’s protocol.Among total 89 T-ALL cases,74 cases had enough immunophenotype data available to differentiate between ETP(CD1a^(-),CD8^(-),CD5^(dim),at least one marker of stem cell or myeloid lineage)and non-ETP.From these 74 subjects,12 ETP-ALL cases(16.2%)were identified.The event-free survival(EFS)rate at 66.8 months was 11.1%±10.1% for ETP-ALL and 57.6%±5.6% for non-ETP-ALL(P=0.003).The overall survival rates were 13.3%±11.0% for ETP-ALL and 64.7%±6.3% for non-ETP-ALL(P=0.002).Our findings demonstrate that early T-cell precursor leukemia is a very high-risk subtype of acute lymphoblastic leukemia with poor prognosis.Meilin Ma Xiang Wang Jingyan Tang Huiliang Xue Jing Chen Ci Pan Hua Jiang Shuhong Shen 2012Frontiers of Medicine2012,6,4:2

10The interaction between polyphyllin I and SQLE protein induces hepatotoxicity through SREBP-2/HMGCR/SQLE/LSS pathway显示文摘Polyphyllin I(PPI)and polyphyllin II(PII)are the main active substances in the Paris polyphylla.However,liver toxicity of these compounds has impeded their clinical application and the potential hepatotoxicity mechanisms remain to be elucidated.In this work,we found that PPI and PII exposure could induce significant hepatotoxicity in human liver cell line L-02 and zebrafish in a dose-dependent manner.The results of the proteomic analysis in L-02 cells and transcriptome in zebrafish indicated that the hepatotoxicity of PPI and PII was associated with the cholesterol biosynthetic pathway disorders,which were alleviated by the cholesterol biosynthesis inhibitor lovastatin.Additionally,3-hydroxy-3-methy-lglutaryl CoA reductase(HMGCR)and squalene epoxidase(SQLE),the two rate-limiting enzymes in the cholesterol synthesis,selected as the potential targets,were confirmed by the molecular docking,the overexpression,and knockdown of HMGCR or SQLE with siRNA.Finally,the pull-down and surface plasmon resonance technology revealed that PPI could directly bind with SQLE but not with HMGCR.Collectively,these data demonstrated that PPI-induced hepatotoxicity resulted from the direct binding with SQLE protein and impaired the sterol-regulatory element binding protein 2/HMGCR/SQLE/lanosterol synthase pathways,thus disturbing the cholesterol biosynthesis pathway.The findings of this research can contribute to a better understanding of the key role of SQLE as a potential target in drug-induced hepatotoxicity and provide a therapeutic strategy for the prevention of drug toxic effects with similar structures in the future.Zhiqi Li Qiqi Fan Meilin Chen Ying Dong Farong Li Mingshuang Wang Yulin Gu Simin Guo Xianwen Ye Jiarui Wu Shengyun Dai Ruichao Lin Chongjun Zhao 2023Journal of Pharmaceutical Analysis2023,13,1:2

12Influence of freeze tube deviation on the development of frozen wall during long cross-passage construction显示文摘This paper investigates the influence of the deviation in freeze pipe installation on the development of the frozen wall in long cross passages by numerical simulation with ANSYS software.The study case is from the artificial ground freezing project along the Fuzhou Metro Line 2 between Ziyang Station and Wuliting Station.Two freezepipe configurations,i.e.,one with perfectly aligned pipes without any deviation from design and another with randomly distributed deviation,are included for comparison.The effect of the random deviation in the freeze pipes on frozen wall interconnection time,the thickness of the frozen wall and the development of the temperature field is explored.For the characteristic section of the numerical model at a depth of 25 m,it is found that the frozen wall interconnection time under the random deviation case and no deviation case is 24 days and 18 days,respectively.The difference in the thickness of the thinnest frozen wall segment between the random deviation and no deviation cases is the largest in the early freezing stage(up to 0.75 m),which decreases with time to 0.31 m in the late freezing stage.The effects of random deviation are more significant in the early freezing stage and diminish as the freezing time increases.JunHao Chen JianLin Wang LeXiao Wang Han Li MeiLin Chen 2022Research in Cold and Arid Regions2022,14,4:2
13Flavonoids of Rosa rugosa Thunb.inhibit tumor proliferation and metastasis in human hepatocellular carcinoma HepG2 cells显示文摘Rosa rugosa Thunb.is recognized as both medicine and edible in China.The article investigated the antitumor activity of rose flavonoids.Water-extracted rose flavonoids(RFW)and ethanol-extracted rose flavonoids(RFE)were achieved by extracting with distilled water and 70%ethanol,respectively.The effects of the two extracts on proliferation inhibition,apoptosis inducement and metastasis prevention of human HepG2 hepatocellular carcinoma cell lines were tested,via optical/fluorescence microscopy,MTT detection,Transwell assay,flow cytometry and Western blot,etc.The results indicated that rose flavonoids at low concentration(10-40μg/mL)had a better inhibitory effect on migration and invasion of HepG2 cells in a dose-dependent manner,while rose flavonoids at high concentration(80-160μg/mL)could induce apoptosis of HepG2 cells by up-regulating the expression of pro-apoptotic proteins p53 and Bax,and down-regulating the expression of anti-apoptotic proteins Bcl-2,leading to the functioning of caspase-3 and caspase-9.The effect of RFE at the same concentration was significantly better than that of RFW.Conclusion,this study found that rose flavonoids had a certain inhibitory effect on proliferation and metastasis of human liver cancer cells HepG2,indicating the application of rose flavonoids in preventing and treating of liver cancer.Zhiguo Zhang Meilin Wang Shu Xing Chen Zhang 2022Food Science and Human Wellness2022,11,2:2
