| 3 | Isolation and Identification of Cancer Stem-Like Cells from Murine Melanoma Cell Lines显示文摘In current study,cancer stem-like cells in the murine melanoma B16F10 cells were investigated.CD phenotypes of the B16F10 cells were analyzed by flow cytometry,and the specific CD phenotype cells from the B16F10 cells were isolated by MACS.Then we used colony formation assay in soft agar media,the cell growth assay in serum-free culture media as well as the tumorigenicity investigation of the specific CD phenotype cells in C57BL/6 mice, respectively,to identify cancer stem-like cells in the B16F10 cells.The results showed that the B16F10 cells could form spherical clones in serum-free culture media,and the rate of clonegenesis of CD133 + ,CD44 + and CD44 + CD133 + cells was higher than that of CD133 - ,CD44 - and CD44 + CD133 - cells in soft agar media,respectively. The reconstruction of aqueous outflow drainage (RAOD) has recently been proposed to aid in restoring aqueous outflow drainage in primary open-angle glaucoma. However, the mechanism of RAOD remains to be fully understood. Based on literature review and research studies, the potential mechanisms of RAOD are the following: (i) Circumferential dilation of the Schlemm’s canal (SC) and surrounding collector channels. | 6 | Function identification of miR482b, a negative regulator during tomato resistance to Phytophthora infestans显示文摘Tomato is an important horticultural and economic crop cultivated worldwide.As Phytophthora infestans becomes a huge threat to tomato production,it is necessary to study the resistance mechanisms of tomato against P.infestans.Our previous research has found that miR482 might be involved in tomato–P.infestans interaction.In this study,miR482b precursor was cloned from Solanum pimpinellifolium“L3708”and miR482b was shown to decrease in abundance in tomato following P.infestans infection.Compared to wild-type tomato plants,tomato plants that overexpressed miR482b displayed more serious disease symptoms after P.infestans infection,with more necrotic cells,longer lesion diameters,and increased P.infestans abundance.Meanwhile,silencing of miR482b was performed by short tandem target mimic(STTM),resulting in enhancement of tomato resistance to P.infestans.Using miRNA and degradome data sets,NBS–LRR disease-resistance genes targeted by miR482b were validated.Negative correlation between the expression of miR482b and its target genes was found in all miR482b-overexpressing and-silencing tomato plants.Our results provide insight into tomato miR482b involved in the response to P.infestans infection,and demonstrate that miR482b–NBS–LRR is an important component in the network of tomato–P.infestans interaction. | Ning Jiang Jun Meng Jun Cui Guangxin Sun Yushi Luan | 2018 | Horticulture Research2018,5,1: | 13 |
| 7 | Tomato lncRNA23468 functions as a competing endogenous RNA to modulate NBS-LRR genes by decoying miR482b in the tomato-Phytophthora infestans interaction显示文摘Our previous studies indicated that tomato miR482b could negatively regulate the resistance of tomato to Phytophthora infestans and the expression of miR482b was decreased after inoculation with P.infestans.However,the mechanism by which the accumulation of miR482b is suppressed remains unclear.In this study,we wrote a program to identify 89 long noncoding RNA(lncRNA)-originated endogenous target mimics(eTMs)for 46 miRNAs from our RNA-Seq data.Three tomato lncRNAs,lncRNA23468,lncRNA01308 and lncRNA13262,contained conserved eTM sites for miR482b.When lncRNA23468 was overexpressed in tomato,miR482b expression was significantly decreased,and the expression of the target genes,NBS-LRRs,was significantly increased,resulting in enhanced resistance to P.infestans.Silencing lncRNA23468 in tomato led to the increased accumulation of miR482b and decreased accumulation of NBS-LRRs,as well as reduced resistance to P.infestans.In addition,the accumulation of both miR482b and NBS-LRRs was not significantly changed in tomato plants that overexpressed lncRNA23468 with a mutated eTM site.Based on the VIGS system,a target gene of miR482b,Solyc02g036270.2,was silenced.The disease symptoms of the VIGS-Solyc02g036270.2 tomato plants were in accordance with those of tomato plants in which lncRNA23468 was silenced after inoculation with P.infestans.More severe disease symptoms were found in the modified plants than in the control plants.Our results demonstrate that lncRNAs functioning as eTMs may modulate the effects of miRNAs in tomato and provide insight into how the lncRNA23468-miR482b-NBS-LRR module regulates tomato resistance to P.infestans. | Ning Jiang Jun Cui Yunsheng Shi Guanglei Yang Xiaoxu Zhou Xinxin Hou Jun Meng Yushi Luan | 2019 | Horticulture Research2019,6,1: | 12 |

| 9 | Effect of transcutaneous electrical acupoint stimulation combined with palonosetron on chemotherapy-induced nausea and vomiting: a single.blind,randomized, controlled trial显示文摘Background: Chemotherapy?induced nausea and vomiting adversely affects the quality of life of patients who receive chemotherapy via intravenous infusion or transcatheter arterial chemoembolization(TACE). This study aimed to investigate the clinical effects of transcutaneous electrical acupoint stimulation(TEAS) on nausea and vomiting after TACE.Methods: A total of 142 patients who received TACE with cisplatin for primary or metastatic liver cancer were assigned to the active?acupuncture(n = 72) or placebo?acupuncture(n Hegu(LI4), Neiguan(P6), an= 70) groups using a covariate?adaptive randomization at a ratio of 1:1. The acupointsd Zusanli(ST36) were stimulated twice daily for 6 days. The effects of TEAS on nausea and vomiting were assessed by using occurrence rate and severity of these symptoms. Anorexia scale and M. D. Anderson Symptom Inventory(MDASI) scores were secondary endpoints and were used to assess the effect of TEAS on patient appetite and quality of life. The safety of the treatments was also monitored.Results: Between the two groups, the differences in occurrence rates and severities of nausea and vomiting after TACE were not significant(all P > 0.05). From the second day after TACE, anorexia scores were significantly lower in the active?acupuncture group than in the placebo?acupuncture group and continued to decrease over time with treat?ment(all P values less than 0.01). On days 0, 1, and 2, the mean MDASI scores for the active?acupuncture group were slightly lower than those for the placebo?acupuncture group, but the differences were not statistically significant(all P > 0.05). | 13 | Effect of microRNA-155 on angiogenesis after cerebral infarction of rats through AT1R/VEGFR2 pathway显示文摘Objective:To explore the function and mechanism of microRNA-155 to regulate the angiogenesis after the cerebral infarction of rats through the angiotensin Ⅱ receptor 1(AT1R)/vascular endothelial growth factor(VEGF) signaling pathway.Methods:Female SD rats were chosen for the construction of cerebral infarction model of rats using the modified right middle cerebral artery occlusion.The real-time PCR(RT-PCR) method was employed to detect the expression of microRNA-155 in each group at different time points after the cerebral infarction(1 h,I d,3 d and 7 d).SD rats were randomly divided into four groups(n=20 rats):sham operation group(Sham group),MACO group,MACO+microRNA-155 mimic group,and MACO+microRNA-155 inhibitor group.Sham group was given the free graft,while MACO+microRNA-155 mimic group and MACO+microRNA-155 inhibitor group were treated with microRNA-155 mimic and microRNA-155 inhibitor respectively.The Zea Longa5-point scale was used to score the neurologic impairment of rats in each group;2,3,5-triphenyl tetrazolium chloride staining to evaluate the volume of cerebral infarction of rats in each group;the immunohistochemistry to detect the expression of CD31;Western blot and RT-PCR to detect the expression of AT1 R and VEGF receptor 2(VEGFR2).Results:The expression of microRNA-155 was increased in the cerebral ischemia tissue after the cerebral infarction.It was significantly increased at 1 d of ischemia and maintained at the high level for a long time.Rats in the Sham group had no symptom of neurologic impairment while rats in the MACO group had the obvious neurologic impairment.After being treated with microRNA-155 inhibitor,the neural function of MACO rats had been improved,with the decreased area of cerebral infarction.But after being treated with microRNA-155 mimic,the neural function was further worsened,with the increased area of cerebral infarction.Results of immunohistoehemical assay indicated that microRNA-155 inhibitor could up-regulate the expression of CD31,while microRNA-155 mimic could down-regulate the expression of CD31.The RT-PCR found that,after being treated with microRNA-155 inhibitor,MACO rats had the increased expression of ATIR and VEGFR2 messenger RNA(mRNA);but after being treated with microRNA-155 mimic,the expression of ATIR and VEGFR2 mRNA was decreased.Results of Western blot showed that,after being treated with microRNA-155 inhibitor,MACO rats had the increased expression of ATIR and VEGFR2 mRNA;but after being treated with microRNA-155 mimic,the expression of ATIR and VEGFR2 mRNA was decreased.Conclusions:The inhibition of microRNA-155 can improve the neurologic impairment of rats with the cerebral infarction,reduce the volume of cerebral infarction and effectively promote the angiogenesis in the region of ischemia,which may be mediated through AT1R/VEGFR2 pathway. | Ying-Chun Meng Zhen-Ying Ding Hai-Quan Wang Li-Ping Ning Chao Wang | 2015 | Asian Pacific Journal of Tropical Medicine2015,8,10: | 10 |
| 14 | Macular integrity assessment to determine the association between macular microstructure and functional parameters in diabetic macular edema显示文摘● AIM: To respectively evaluate macular morphological features and functional parameters by using spectraldomain optical coherence tomography(SD-OCT) and macular integrity assessment(MAIA) in patients with diabetic macular edema(DME).● METHODS: This prospective, non-controlled, open study included 61 eyes of 38 consecutive patients with DME. All patients underwent best-corrected visual acuity(BCVA) measurement, MAIA microperimetry, and SDOCT. DME morphology, including central retinal thickness(CRT) and central retinal volume(CRV); integrity of the external limiting membrane(ELM) and photoreceptor inner segment/outer segment(IS/OS) junction; and the deposition of hard macular exudates were assessed within a 1000-μm central subfield area. MAIA microperimetry parameters evaluated were average threshold(AT)-retinal sensitivity, macular integrity index(MI), fixation points within a circle of radius 1°(P1) and 2°(P2), and bivariate contour ellipse area considering 63% and 95% of the fixation points(A63 and A95, respectively). ● RESULTS: MI was significantly higher in eyes with disrupted ELM or IS/OS, compared with eyes with intact ELM and IS/OS. Values of BCVA(log MAR), total AT, AT within 1000-μm diameter, P2, A63, A95, and CRT were significantly worse in eyes with disrupted IS/OS, compared with eyes with intact IS/OS. The values of BCVA(log MAR), AT within 1000-μm diameter, and CRT were significantly worse in eyes with disrupted ELM, compared with eyes with intact ELM. These parameters were not significantly different between eyes with or without hard macular exudate deposition. CRV was not significantly different in the presence or absence of the integrity of ELM, IS/OS, or deposition of hard macular exudates. At the center, nasal and temporal sectors of the fovea, significant negative correlations were observed between retinal thickness and AT of the corresponding area. At the inferior and superior sectors of the fovea, no correlations were observed between retinal thickness and AT of the corresponding area. In the intact IS/OS group, significant negative correlations were observed between CRT and central AT. There was no correlation between retinal sensitivity and thickness when the IS/OS layer was disrupted. Multiple linear regression analyses revealed that IS/OS integrity was an independent factor affecting MI.● CONCLUSION: Functional(BCVA and visual field) and morphological parameters(retinal thickness) were significantly associated with an intact IS/OS. Local photoreceptor integrity was a strong predictor of local visual function throughout the retina. | 15 | Glycerol aerobic oxidation to glyceric acid over Pt/hydrotalcite catalysts at room temperature显示文摘Glycerol(GLY) aerobic oxidation in an aqueous solution is one of the most prospective pathways in biomass transformation, where the supported catalysts based on noble metals(mainly Au, Pd, Pt) are most commonly employed. Herein, Pt nanoparticles supported on rehydrated MgxAl1-hydrotalcite(denoted as re-MgxAl1-LDH-Pt) were prepared via impregnation-reduction method followed by an in situ rehydration process, which showed high activity and selectivity towards GLY oxidation to produce glyceric acid(GLYA) at room temperature. The metal-support interfacial structure and catalyst basicity were modulated by changing the Mg/Al molar ratio of the hydrotalcite precursor, and the optimal performance was achieved on re-Mg6Al1-LDH-Pt with a GLY conversion of 87.6% and a GLYA yield of 58.6%, which exceeded the traditional activated carbon and oxide supports. A combinative study on structural characterizations(XANES, CO-FTIR spectra, and benzoic acid titration) proves that a higher Mg/Al molar ratio promotes the formation of positively charged Ptd+species at metal-support interface, which accelerates bond cleavage of a-C–H and improves catalytic activity. Moreover, a higher Mg/Al molar ratio provides a stronger basicity of support that contributes to the oxidation of terminal-hydroxyl and thus enhances the selectivity of GLYA. | 16 | Long-term postoperative outcomes of bilateral lateral rectus recession vs unilateral recession-resection for intermittent exotropia显示文摘AIM: To discuss the long-term postoperative results of bilateral lateral rectus recession(BLR) and unilateral lateral rectus recession-medial rectus resection(RR) in therapy of intermittent exotropia.METHODS: We retrospectively analyzed 213 cases of intermittent exotropia who underwent surgery between2008 and 2010. The patients were grouped into BLR group and RR group. Motor outcomes were divided into three groups on the basis of the angle of deviation after surgery: overcorrection(esotropia/phoria >5△), orthophoria(esotropia/phoria ≤5△to exotropia/phoria ≤10△), and undercorrection/recurrence(exotropia/phoria >10△).Titmus test was used to evaluate stereoacuity, the stereoacuity <800s of arc meaned the patients had stereopsis. Surgical outcome including motor criteria and sensory status were compared at postoperative 6, 12,24 mo and at 36 mo examination between groups. RESULTS: At 12, 24 mo after surgery, the motor outcomes had no difference(P >0.05) between groups.However, the motor outcomes at 6, 36 mo were signally different in each group, indicating the success rate in RR group at 6mo was higher than that in BLR group(83.02%vs 82.24%, P <0.05) but the result was contrary at the 3y examination(60.75% vs 43.40%, P <0.05). No statistical significance were found in the sensory outcomes between the groups at mean of 3.7y follow-up.CONCLUSION: The motor outcomes in RR group were better than in BLR group at 6mo after surgery, while the3 y outcomes were better in BLR group. Lam.) varieties显示文摘Simple sequence repeat(SSR) markers have been shown to be a powerful tool for varieties identification in plants.However,SSR fingerprinting of sweetpotato varieties has been a little reported.In this study,a total of 1 294 SSR primer pairs,including 1 215 genomic-SSR and 79 expressed sequence tag(EST)-SSR primer pairs,were screened with sweetpotato varieties Zhengshu 20 and Luoxushu 8 and their 2 F1 individuals randomly sampled,and 273 and 38 of them generated polymorphic bands,respectively.Four genomic-SSR and 3 EST-SSR primer pairs,which showed good polymorphism,were selected to amplify 203 sweetpotato varieties and gave a total of 172 bands,85(49.42%) of which were polymorphic.All of the 203 sweetpotato varieties showed unique fingerprint patterns,indicating the utility of SSR markers in variety identification of this crop.Polymorphism information content(PIC) ranged from 0.5824 to 0.9322 with an average of 0.8176.SSR-based genetic distances varied from 0.0118 to 0.6353 with an average of 0.3100 among these varieties.Thus,these sweetpotato varieties exhibited high levels of genetic similarity and had distinct fingerprint profiles.The SSR fingerprints of the 203 sweetpotato varieties have been successfully constructed.The highly polymorphic SSR primer pairs developed in this study have the potential to be used as core primer pairs for variety identification,genetic diversity assessment and linkage map construction in sweetpotato and other plants. | MENG Yu-sha ZHAO Ning LI Hui ZHAI Hong HE Shao-zhen LIU Qing-chang | 2018 | Journal of Integrative Agriculture2018,17,1: | 7 |
| 18 | Stability improvement under high efficiency—next stage development of perovskite solar cells显示文摘With efficiency of perovskite solar cells(PSCs) overpassing 23%, to realize their commercialization, the biggest challenge now is to boost the stability to the same level as conventional solar cells. Thus, tremendous effort has been directed over the past few years toward improving the stability of these cells. Various methods were used to improve the stability of bulk perovskites,including compositional engineering, interface adjustment, dimensional manipulation, crystal engineering, and grain boundary decoration. Diverse device configurations, carrier transporting layers, and counter electrodes are investigated. To compare the stability of PSCs and clarify the degradation mechanism, diverse characterization methods were developed. Overall stability of PSCs has become one central topic for the development of PSCs. | 19 | Relationship between higher-order aberrations and myopia progression in schoolchildren: a retrospective study显示文摘AIM: To investigate the relationship between higher- order aberration (HOA) and myopic progression in schoolchildren. ·METHODS: Between April 23 2011 and August 29, 2011 in the children's myopia outpatient clinic of the West China Hospital of Sichuan University, 148 eyes of 74 schoolchildren were reviewed. HOAs for a 6 -mm pupil were measured with an aberrometer. Myopic progression rate was defined according to the change in spherical equivalent refraction (SER) divided by the time span (years). Subjects with myopic progression rate of ≥0.50 diopters (D) were classified as the 'fast'group and the subjects with myopic progression rate of <0.50D were classified as the'slow'group. A retrospective study was conducted to compare HOA between the two groups, using root mean square (RMS) values and Zernike coefficients. ·RESULTS: The RMS values of HOA (t =2.316, P =0.02), HOA without Z 4 0 (t =2.224, P =0.03),third-order aberrations (t' =2.62, P =0.01), and coma (t' =2.49, P =0.01) were significantly higher in the fast group than those in the slow group. The individual Zernike coefficients of Z 3 -1 (t = -2.072, P =0.04) and Z 5 1 (Z =-2.627, P =0.01) displayed statistically significant differences between the two groups. Significant correlations were found between the RMS values of HOA (r =0.193, P =0.019), RMS values of HOA without Z 4 0 (r =0.23, P =0.005), RMS values of coma(r =0.235,P =0.004),RMS values of third-order aberrations (r =0.243, P =0.003), and the progression rate. · CONCLUSION: Our results provide evidence of a relationship between HOA and myopic progression.
