| 2 | Potentiating CD8^(+) T cell antitumor activity by inhibiting PCSK9 to promote LDLR-mediated TCR recycling and signaling显示文摘Metabolic regulation has been proven to play a critical role in T cell antitumor immunity.However,cholesterol metabolism as a key component of this regulation remains largely unexplored.Herein,we found that the low-density lipoprotein receptor(LDLR),which has been previously identified as a transporter for cholesterol,plays a pivotal role in regulating CD8+T cell antitumor activity.Besides the involvement of cholesterol uptake which is mediated by LDLR in T cell priming and clonal expansion,we also found a non-canonical function of LDLR in CD8+T cells:LDLR interacts with the T-cell receptor(TCR)complex and regulates TCR recycling and signaling,thus facilitating the effector function of cytotoxic T-lymphocytes(CTLs).Furthermore,we found that the tumor microenvironment(TME)downregulates CD8+T cell LDLR level and TCR signaling via tumor cell-derived proprotein convertase subtilisin/kexin type 9(PCSK9)which binds to LDLR and prevents the recycling of LDLR and TCR to the plasma membrane thus inhibits the effector function of CTLs.Moreover,genetic deletion or pharmacological inhibition of PCSK9 in tumor cells can enhance the antitumor activity of CD8+T cells by alleviating the suppressive effect on CD8+T cells and consequently inhibit tumor progression.While previously established as a hypercholesterolemia target,this study highlights PCSK9/LDLR as a potential target for cancer immunotherapy as well. | Juanjuan Yuan Ting Cai Xiaojun Zheng Yangzi Ren Jingwen Qi Xiaofei Lu Huihui Chen Huizhen Lin Zijie Chen Mengnan Liu Shangwen He Qijun Chen Siyang Feng Yingjun Wu Zhenhai Zhang Yanqing Ding Wei Yang | 2021 | Protein & Cell2021,12,4: | 12 |
| 3 | The effects of autophagy on vascular endothelial cells induced by airborne PM2.5显示文摘The purpose of this study was to examine the direct toxicity of PM2.5 collected from Beijing on human umbilical vein endothelial cells(HUVEC). A Cell Counting Kit 8(CCK8) assay demonstrated that PM2.5 exposure decreased the proliferation of HUVECs in a dosedependent manner. We also found that PM2.5 exposure induced autophagy in HUVECs, as evidenced by:(1) an increased number of double-membrane vesicles;(2) enhanced conversion and punctuation of the microtubule-associated protein light chain 3(LC3); and(3) decreased levels of the selective autophagy substrate p62 in a time-dependent manner.Furthermore, promoting autophagy in PM2.5-exposed HUVECs with rapamycin increased the cell survival rate, whereas inhibiting autophagy via 3-methyladenine significantly decreased cell survival. These results demonstrate that PM2.5 exposure can induce cytotoxicity and autophagy in HUVECs and that autophagy play a protective role against PM2.5-induced cytotoxicity. | 15 | Different meridian tropism in three Chinese medicines: Tinglizi(Semen Lepidii Apetali), Yiyiren(Semen Coicis), Cheqianzi(Semen Plantaginis)显示文摘OBJECTIVE: To study the meridian tropism(MT) for three Chinese medicines: Tinglizi(Semen Lepidii Apetali, TLZ), Yiyiren(Semen Coicis, YYR), Cheqianzi(Semen Plantaginis, CQZ) in vivo experiments.METHODS: The extracts of TLZ,YYR, CQZ were orally administered to rats at a dose 234, 700 and 350 mg/kg, respectively. The urinary excretion rate, loss in weight of rats and the electrolyte levels in the plasma were measured, in order to assess the mechanisms of the three traditional Chinese diuretic medicines.RESULTS: TLZ, YYR and CQZ exhibited obvious diuretic effect and reduced the weight of saline-loaded animals. | 16 | Reduced Smoothened level rescues Aβ-induced memory deficits and neuronal inflammation in animal models of Alzheimer's disease显示文摘Emerging evidence suggests that neuro-inflammation begins early and drives the pathogenesis of Alzheimer's disease(AD), and anti-inflammatory therapies are under clinical development. However,several anti-inflammatory compounds failed to improve memory in clinical trials, indicating that reducing inflammation alone might not be enough. On the other hand, neuro-inflammation is implicated in a number of mental disorders which share the same therapeutic targets. Based on these observations,we screened a batch of genes related with mental disorder and neuro-inflammation in a classical olfactory conditioning in an amyloid beta(Aβ) overexpression fly model. A Smoothened(SMO) mutant was identified as a genetic modifier of Aβ toxicity in 3-min memory and downregulation of SMO rescued Aβ induced 3-min and 1-h memory deficiency. Also, Aβ activated innate inflammatory response in fly by increasing the expression of antimicrobial peptides, which were alleviated by downregulating SMO.Furthermore, pharmaceutical administration of a SMO antagonist LDE rescued Aβ-induced upregulation of SMO in astrocytes of mouse hippocampus, improved memory in Morris water maze(MWM), and reduced expression of astrocyte secreting pro-inflammatory factors IL-1 b, TNFa and the microglia marker IBA-1 in an APP/PS1 transgenic mouse model. | 20 | A Privacy-Preserving Mechanism Based on Local Differential Privacy in Edge Computing显示文摘With the development of Internet of Things(IoT),the delay caused by network transmission has led to low data processing efficiency.At the same time,the limited computing power and available energy consumption of IoT terminal devices are also the important bottlenecks that would restrict the application of blockchain,but edge computing could solve this problem.The emergence of edge computing can effectively reduce the delay of data transmission and improve data processing capacity.However,user data in edge computing is usually stored and processed in some honest-but-curious authorized entities,which leads to the leakage of users’privacy information.In order to solve these problems,this paper proposes a location data collection method that satisfies the local differential privacy to protect users’privacy.In this paper,a Voronoi diagram constructed by the Delaunay method is used to divide the road network space and determine the Voronoi grid region where the edge nodes are located.A random disturbance mechanism that satisfies the local differential privacy is utilized to disturb the original location data in each Voronoi grid.In addition,the effectiveness of the proposed privacy-preserving mechanism is verified through comparison experiments.Compared with the existing privacy-preserving methods,the proposed privacy-preserving mechanism can not only better meet users’privacy needs,but also have higher data availability. | Mengnan Bi Yingjie Wang Zhipeng Cai Xiangrong Tong | 2020 | China Communications2020,17,9: | 3 |
