| 1 | A life support-based comprehensive treatment regimen dramatically lowers the in-hospital mortality of patients with fulminant myocarditis: a multiple center study显示文摘Fulminant myocarditis(FM) has unacceptable high mortality. This study aimed to evaluate the therapeutic efficacy of a life support-based comprehensive treatment regimen(LSBCTR), a completely novel treatment regimen, for FM. A total of 169 FM patients recruited from January 2008 to December 2018 were divided into two groups: patients receiving LSBCTR(81 cases),which includes(i) mechanical life support(positive pressure respiration, intra-aortic balloon pump with or without extracorporeal membrane oxygenation),(ii) immunomodulation therapy using sufficient doses of glucocorticoids and immunoglobulins, and(iii) application of neuraminidase inhibitors, and those receiving conventional treatment(88 cases). The endpoints were in-hospital death and heart-transplantation. Of all the population, 44 patients(26.0%) died in hospitals. Inhospital mortality was 3.7%(3/81) for LSBCTR group and 46.6%(41/88) for traditional treatment(P<0.001). Early application of LSBCTR, mechanical life support, neuraminidase inhibitors, and immunomodulation therapy significantly contributed to reduction in in-hospital mortality. This study describes a novel treatment regimen for FM patients that dramatically reduces inhospital mortality. | 2 | The Relationship between Glucose Excursion and Cognitive Function in Aged Type 2 Diabetes Patients显示文摘Objective Evidence suggests that type 2 diabetes (T2DM) is associated with an increased risk of dementia and that glucose variability is an independent risk factor for diabetic complications.This study investigated the relationship between glucose excursion and cognitive function in aged T2DM patients.Methods A total of 248 aged T2DM patients wore a continuous glucose monitoring system (CGMS) for 3 days in order to evaluate glucose excursion,including mean amplitude of glycemic excursions (MAGE) and mean of daily difference (MODD).All subjects were evaluated with a number of accepted cognitive function tests,including themini‐mental status examination (MMSE).The relationship between MAGE and MODD and performance on these cognitive tests was assessed.Results The MAGE and MMSE score were negatively correlated,likewise with the correlation between MODD and MMSE.Liner multivariate regression analysis showed that MAGE and MODD were also negatively related to MMSE independent of age,sex,glycemic control,hypertension,smoking,or coronary heart disease history.Conclusion Glucose excursion is related to cognitive function in aged T2DM patients.Elevated glucose excursion decreased the MMSE score,which reflects general cognitive function.Thus,therapy aimed at controlling glucose excursion may be beneficial for maintaining cognitive function in aged T2DM patients. | ZHONG Yuan ZHANG Xiao Yan MIAO Ya ZHU Jie Hua YAN Hong WANG Bei Yun JIN Jun HU Ting Jun JIA Wei Ping | 2012 | Biomedical and Environmental Sciences2012,25,1: | 34 |

| 6 | Analysis of air quality characteristics of Beijing–Tianjin–Hebei and its surrounding air pollution transport channel cities in China显示文摘Beijing–Tianjin–Hebei(BTH)and its surrounding areas are very important to air pollution control in China.To analyze the characteristics of BTH and its surrounding areas of China,we collected 5,641,440 air quality data from 161 air monitoring stations and 37,123,000 continuous monitoring data from air polluting enterprises in BTH and surrounding cities to establish an indicator system for urban air quality portraits.The results showed that particulate matter with aerodynamic diameters of<2.5μm(PM2.5),particulate matter with aerodynamic diameters of<10μm(PM10)and SO2 improved significantly in 31 cities from2015 to 2018,but ozone deteriorated.Air quality in BTH and the surrounding areas showed obvious seasonal characteristics,among which PM2.5,PM10,SO2,and NO2 showed a'U'type distribution from January to December,while O3 had an'inverted U'distribution.The hourly changes in air quality revealed that peaks of PM2.5,PM10 and NO2 appeared from 8:00 to 10:00,while those for O3 appeared at 15:00–16:00.The exposure characteristics of the 31 cities showed that six districts in Beijing had the highest air quality population exposure,and that exposure levels in Zhengzhou,Puyang,Anyang,Jincheng were higher than the average of the 31 investigated cities.Additionally,multiple linear regression revealed a negative correlation between meteorological factors(especially wind and precipitation)and air quality,while a positive correlation existed between industrial pollution emissions and air quality in most of BTH and its surrounding cities. | Cuicui Xiao Miao Chang Peikun Guo Meifeng Gu Yang Li | 2020 | Journal of Environmental Sciences2020,32,1: | 15 |
| 7 | Performance of transient elastography in assessing liver fibrosis in patients with autoimmune hepatitis-primary biliary cholangitis overlap syndrome显示文摘AIM To investigate the performance of transient elastography(TE) for diagnosis of fibrosis in patients with autoimmune hepatitis-primary biliary cholangitis(AIHPBC) overlap syndrome.METHODS A total of 70 patients with biopsy-proven AIH-PBC overlap syndrome were included. Spearman correlation test was used to analyze the correlation of liver stiffness measurement(LSM) and fibrosis stage. Independent samples Student's t-test or one-way analysis of variance was used to compare quantitative variables. Receiver operating characteristics(ROC) curve was used to calculate the optimal cut-off values of LSM for predicting individual fibrosis stages. A comparison on the diagnostic accuracy for severe fibrosis was made between LSM and other serological scores.RESULTS Patients with AIH-PBC overlap syndrome had higher median LSM than healthy controls(11.3 ± 6.4 k Pa vs 4.3 ± 1.4 k Pa, P < 0.01). LSM was significantly correlated with fibrosis stage(r = 0.756, P < 0.01). LSM values increased gradually with an increased fibrosis stage. The areas under the ROC curves of LSM for stages F ≥ 2, F ≥ 3, and F4 were 0.837(95%CI: 0.729-0.914), 0.910(0.817-0.965), and 0.966(0.893-0.995), respectively. The optimal cut-off values of LSM for fibrosis stages F ≥ 2, F ≥ 3, and F4 were 6.55, 10.50, and 14.45 k Pa, respectively. | 10 | Essential roles of exosome and circRNA_101093 on ferroptosis desensitization in lung adenocarcinoma显示文摘Background:Resistance to ferroptosis,a regulated cell death caused by irondependent excessive accumulation of lipid peroxides,has recently been linked to lung adenocarcinoma(LUAD).Intracellular antioxidant systems are required for protection against ferroptosis.The purpose of the present studywas to investigate whether and how extracellular system desensitizes LUAD cells to ferroptosis.Methods:Established human lung fibroblasts MRC-5,WI38,and human LUAD H1650,PC9,H1975,H358,A549,and H1299 cell lines,tumor and matched normal adjacent tissues of LUAD,and plasma from healthy individuals and LUAD patients were used in this study.Immunohistochemistry and immunoblotting were used to analyze protein expression,and quantitative reverse transcription-PCR was used to analyze mRNA expression.Cell viability,cell death,and the lipid reactive oxygen species generationwere measured to evaluate the responses to ferroptosis.Exosomes were observed using transmission electron microscope.The localization of arachidonic acid(AA)was detected using click chemistry labeling followed by confocal microscopy.Interactions between RNAs and proteins were detected using RNA pull-down,RNA immunoprecipitation and photoactivatable ribonucleoside-enhanced crosslinking and immunoprecipitation methods.Proteomic analysis was used to investigate RNA-regulated proteins,and metabolomic analysis was performed to analyze metabolites.Cellderived xenograft,patient-derived xenograft,cell-implanted intrapulmonary LUAD mouse models and plasma/tissue specimens from LUAD patients were used to validate the molecular mechanism.Results:Plasma exosome from LUAD patients specifically reduced lipid peroxidation and desensitized LUAD cells to ferroptosis.A potential explanation is that exosomal circRNA_101093(cir93)maintained an elevation in intracellular cir93 in LUAD to modulate AA,a poly-unsaturated fatty acid critical for ferroptosisassociated increased peroxidation in the plasma membrane.Mechanistically,cir93 interacted with and increased fatty acid-binding protein 3(FABP3),which transported AA and facilitated its reaction with taurine.Thus,global AA was reduced,whereas N-arachidonoyl taurine(NAT,the product of AA and taurine)was induced.Notably,the role of NAT in suppressing AA incorporation into the plasma membrane was also revealed.In pre-clinical in vivo models,reducing exosome improved ferroptosis-based treatment.Conclusion:Exosome and cir93 are essential for desensitizing LUAD cells to ferroptosis,and blocking exosome may be helpful for future LUAD treatment. | Xiao Zhang Yunhua Xu Lifang Ma Keke Yu Yongjie Niu Xin Xu Yi Shi Susu Guo Xiangfei Xue Yikun Wang Shiyu Qiu Jiangtao Cui Hong Wang Xiaoting Tian Yayou Miao Fanyu Meng Yongxia Qiao Yongchun Yu Jiayi Wang | 2022 | Cancer Communications2022,42,4: | 10 |

However, direct conversion of syngas via the conventional Fischer-Tropsch synthesis produces little aromatics. We presented herein that a bifunctional composite of ZSM-5 in combination with Zn Cr Oxcatalyzes syngas conversion to aromatics. Particularly, ZSM-5 crystals with a sheet-like morphology can enhance significantly the aromatization activity. The lower length ratio of the b/a axes of the crystals, the more aromatics form but without influencing the selectivity of small molecules such as CH4 and C2–C4. The underlying mechanism and targeted intervention remain largely to be determined. Ginsenoside Rb1 was reported to be neuroprotective. We therefore set out to determine whether ginsenoside Rb1 can attenuate isoflurane/surgery-induced cognitive dysfunction via inhibiting neuroinflammation and oxidative stress. Methods Five-months-old C57BL/6J female mice were treated with 1.4% isoflurane plus abdominal surgery for two hours. Sixty mg/kg ginsenoside Rb1 were given intraperitoneally from 7 days before surgery. Cognition of the mice were assessed by Barnes Maze. Levels of postsynaptic density-95 and synaptophysin in mice hippocampus were measured by Western blot. Levels of reactive oxygen species, tumor necrosis factor-α and interleukin-6 in mice hippocampus were measured by ELISA. Results Here we show for the first time that the ginsenoside Rb1 treatment attenuated the isoflurane/surgery-induced cognitive impairment. Moreover, ginsenoside Rb1 attenuated the isoflurane/surgery-induced synapse dysfunction. Finally, ginsenoside Rb1 mitigated the isoflurane/surgery-induced elevation levels of reactive oxygen species, tumor necrosis factor-α and interleukin-6 in the mice hippocampus. Ginsenoside Rg1 is reported to be neuroprotective. We therefore set out to determine whether ginsenoside Rg1 can attenuate isoflurane-induced caspase activation via inhibiting mitochondrial dysfunction. Methods We investigated the effects of ginsenoside Rg1 at concentrations of 12.5, 25, and 50 μmol/L and pretreatment times of 12 h and 24 h on isoflurane-induced caspase-3 activation in H4 na?ve and stably transfected H4 human neuroglioma cells that express full-length human amyloid precursor protein(APP)(H4-APP cells). For mitochondrial dysfunction, we assessed mitochondrial permeability transition pore(m PTP) and adenosine-5'-triphosphate(ATP) levels. We employed Western blot analysis, chemiluminescence, and flowcytometry. Results Here we show that pretreatment with 50 μmol/L ginsenoside Rg1 for 12 h attenuated isoflurane-induced caspase-3 activation and mitochondrial dysfunction in H4-APP cells, while pretreatment with 25 and 50 μmol/L ginsenoside Rg1 for 24 h attenuated isoflurane-induced caspase-3 activation and mitochondrial dysfunction in both H4 na?ve and H4-APP cells. Conclusion These data suggest that ginsenoside Rg1 may ameliorate isoflurane-induced caspase-3 activation by inhibiting mitochondrial dysfunction. | 20 | Chromium-containing traditional Chinese medicine, Tianmai Xiaoke Tablet improves blood glucose through activating insulin-signaling pathway and inhibiting PTP1B and PCK2 in diabetic rats显示文摘OBJECTIVE: Chromium is an essential mineral that is thought to be necessary for normal glucose homeostasis.Numerous studies give evidence that chromium picolinate can modulate blood glucose and insulin resistance.The main ingredient of Tianmai Xiaoke(TMXK) Tablet is chromium picolinate.In China, TMXK Tablet is used to treat type 2 diabetes.This study investigated the effect of TMXK on glucose metabolism in diabetic rats to explore possible underlying molecular mechanisms for its action.METHODS: Diabetes was induced in rats by feeding a high-fat diet and subcutaneously injection with a single dose of streptozotocin(50 mg/kg, tail vein).One week after streptozotocin-injection, model rats were divided into diabetic group, low dose of TMXK group and high dose of TMXK group.Eight normal rats were used as normal control.After 8 weeks of treatment, skeletal muscle was obtained and was analyzed using Roche NimbleGen mRNA array and quantitative polymerase chain reaction(qPCR).Fasting blood glucose, oral glucose tolerance test and homeostasis model assessment of insulin resistance(HOMA-IR) index were also measured.RESULTS: The authors found that the administration of TMXK Tablet can reduce the fasting blood glucose and fasting insulin level and HOMA-IR index.The authors also found that 2 223 genes from skeletal muscle of the high-dose TMXK group had significant changes in expression(1 752 increased, 471 decreased).Based on Kyoto encyclopedia of genes and genomes pathway analysis, the most three significant pathways were 'insulin signaling pathway', 'glycolysis/gluconeogenesis' and 'citrate cycle(TCA)'.qPCR showed that relative levels of forkhead box O3(FoxO3), phosphoenolpyruvate carboxykinase 2(Pck2), and protein tyrosine phosphatase 1B(Ptp1b) were signifi cantly decreased in the high-dose TMXK group, while v-akt murine thymoma viral oncogene homolog 1(Akt1) and insulin receptor substrate 2(Irs2) were increased.CONCLUSION: Our data show that TMXK Tablet reduces fasting glucose level and improves insulin resistance in diabetic rats.The mechanism may be linked to the inactivation of PTP1B and PCK enzymes, or through intracellular pathways, such as the insulin signaling pathway. | Qian Zhang Xin-hua Xiao Ming Li Wen-hui Li Miao Yu Hua-bing Zhang Fan Ping Zhi-xin Wang Jia Zheng | 2014 | Journal of Integrative Medicine2014,12,3: | 8 |
