8Induction of Endoplasmic Reticulum Stress by Cadmium and Its Regulation on Nrf2 Signaling Pathway in Kidneys of Rats显示文摘Objective This study was conducted to investigate the regulation of endoplasmic reticulum stress on Nrf2 signaling pathway in the kidneys of rats. Methods Rats were divided into twelve groups of six animals each. Some groups were pre-administered with bacitracin or tauroursodeoxycholic acid(TUDCA), and all of them were treated with 5-20 μmol/kg cadmium(Cd) for 48 h. The oxidative stress levels were analyzed using kits. The mRNA and protein expression levels of endoplasmic reticulum stress-related factors and Nrf2 signaling pathway-related factors were determined using RT-PCR and western blot. Results Cd exposure resulted in oxidative stress in the kidneys of rats and upregulated the expression of endoplasmic reticulum stress(ERS)-related factors and Nrf2 signaling pathway-related factors, especially at doses of 10 and 20 μmol/kg Cd, and the expression changes were particularly obvious. Moreover, after pretreatment with bacitracin, Cd upregulated the expression of ERS-related factors to a certain extent and, at higher doses, increased the mRNA expression of Nrf2. After pretreatment with TUDCA, Cd reduced the level of ERS to a certain extent; however, at these doses, there were no significant changes in the expression of Nrf2. Conclusion Cadmium can result in ERS and oxidative stress in the kidneys of rats, activate Nrf2, and upregulate the transcriptional expression of phase II detoxification enzymes under these experimental conditions. 9Novel ionic liquids as reaction medium for atom transfer radical polymerization of methyl methacrylate显示文摘Atom transfer radical polymerization (ATRP) of methyl methacrylate (MMA) employing ethyl 2-bromoisobutyrate (EBiB)/ CuBr as the initiating system was investigated at 50℃ in the absence of any additional ligand in the three room temperature ionic liquids (RTIL_s), 1-methyl-imidazolium acetate ([mim][CH_3COO]), 1-methylimidazolium propionate ([mim][CH_3CH_2COO]) and 1-methylimidazolium butyrate ([mim][CH_3CH_2CH_2COO]), respectively. All the polymerization in the three RTILs proceeded in a well-controlled manner. 10Magnetic and microwave absorption properties of Ni_(1-x)Zn_xFe_2O_4 nanocrystalline synthesized by sol-gel method显示文摘Ni1-xZnxFe2O4(0≤x≤1,in steps of 0.1) nanocrystallines were synthesized by sol-gel route.The doping effects of zinc on structural,magnetic and microwave absorption properties were investigated in detail.X-ray diffraction(XRD) results show that all the samples are single-phase spinel structure.The magnetic and microwave absorption properties are strongly dependent on the zinc content,which can be understood in terms of the cations redistribution in spinel tetrahedral and octahedral sites with the increase of zinc content.The magnetic measurement shows the antiferromagnetic nature of the samples for x=0.9 and x=1.0.The saturation magnetization reaches the maximum of 3.35μB/f.u.at x=0.5.The optimal reflection loss(RL) of-29.6 dB is found at 6.5 GHz for an absorber thickness of 5 mm.The RL values exceeding 10 dB are obtained for the absorber in the range of 3.9-8.9 GHz.These Ni1-xZnxFe2O4 nanocrystallines may be attractive candidates for electromagnetic wave absorption materials.ZHANG Min LIU QiangChun ZI ZhenFa DAI YuQiang ZHU XueBin SUN YuPing DAI JianMing 2013Science China(Technological Sciences)2013,56,1:4

The lower phase contained 20 mmol/L of carboxymethly-β-cyclodextrin as chiral selector. The enantiomers were separated in 1.2 h and identified by chiral HPLC. 14Mitochondrial Oxidative Stress Enhances Vasoconstriction by Altering Calcium Homeostasis in Cerebrovascular Smooth Muscle Cells under Simulated Microgravity显示文摘Objective Exposure to microgravity results in postflight cardiovascular deconditioning in astronauts.Vascular oxidative stress injury and mitochondrial dysfunction have been reported during this process.To elucidate the mechanism for this condition,we investigated whether mitochondrial oxidative stress regulates calcium homeostasis and vasoconstriction in hindlimb unweighted(HU)rat cerebral arteries.Methods Three-week HU was used to simulate microgravity in rats.The contractile responses to vasoconstrictors,mitochondrial fission/fusion,Ca^(2+) distribution,inositol 1,4,5-trisphosphate receptor(IP3 R)abundance,and the activities of voltage-gated K+channels(KV)and Ca^(2+)-activated K+channels(BKCa)were examined in rat cerebral vascular smooth muscle cells(VSMCs).Results An increase of cytoplasmic Ca^(2+) and a decrease of mitochondrial/sarcoplasmic reticulum(SR)Ca^(2+) were observed in HU rat cerebral VSMCs.The abundance of fusion proteins(mitofusin 1/2[MFN1/2])and fission proteins(dynamin-related protein 1[DRP1]and fission-mitochondrial 1[FIS1])was significantly downregulated and upregulated,respectively in HU rat cerebral VSMCs.The cerebrovascular contractile responses to vasoconstrictors were enhanced in HU rats compared to control rats,and IP3 R protein/mRNA levels were significantly upregulated.The current densities and open probabilities of KV and BKCa decreased and increased,respectively.Treatment with the mitochondrial-targeted antioxidant mitoTEMPO attenuated mitochondrial fission by upregulating MFN1/2 and downregulating DRP1/FIS1.It also decreased IP3 R expression levels and restored the activities of the KV and BKCa channels.MitoTEMPO restored the Ca^(2+) distribution in VSMCs and attenuated the enhanced vasoconstriction in HU rat cerebral arteries.Conclusion The present results suggest that mitochondrial oxidative stress enhances cerebral vasoconstriction by regulating calcium homeostasis during simulated microgravity.LIU Zi Fan WANG Hai Ming JIANG Min WANG Lin LIN Le Jian ZHAO Yun Zhang SHAO Jun Jie ZHOU Jing Jing XIE Man Jiang LI Xin ZHANG Ran 2021Biomedical and Environmental Sciences2021,34,3:3
15Gene Expression Profiles Comparison between 2009 Pandemic and Seasonal H1N1 Influenza Viruses in A549 Cells显示文摘Objective To perform gene expression profiles comparison so that to identify and understand the potential differences in pathogenesis between the pandemic and seasonal A (H1N1) influenza viruses. Methods A549 cells were infected with A/California/07/09 (H1N1) and A/GuangdongBaoan/51/08 (H1N1) respectively at the same MOI of 2 and collected at 2, 4, 8, and 24 h post infection (p.i.). Gene expression profiles of A549 cells were obtained using the 22 K Human Genome Oligo Array, and differentially expressed genes were analyzed at selected time points. Results Microarrays results indicated that both of the viruses suppressed host immune response related pathways including cytokine production while pandemic H1N1 virus displayed weaker suppression of host immune response than seasonal H1N1 virus. Observation on similar anti-apoptotic events such as activation of apoptosis inhibitor and down-regulation of key genes of apoptosis pathways in both infections showed that activities of promoting apoptosis were different in later stage of infection. Conclusion The immuno-suppression and anti-apoptosis events of pandemic H1N1 virus were similar to those seen by seasonal H1N1 virus.
