1Adaptive Gaussian sum squared-root cubature Kalman filter with split-merge scheme for state estimation显示文摘The paper deals with state estimation problem of nonlinear non-Gaussian discrete dynamic systems for improvement of accuracy and consistency. An efficient new algorithm called the adaptive Gaussian-sum square-root cubature Kalman filter(AGSSCKF) with a split-merge scheme is proposed. It is developed based on the squared-root extension of newly introduced cubature Kalman filter(SCKF) and is built within a Gaussian-sum framework. Based on the condition that the probability density functions of process noises and initial state are denoted by a Gaussian sum using optimization method, a bank of SCKF are used as the sub-filters to estimate state of system with the corresponding weights respectively, which is adaptively updated. The new algorithm consists of an adaptive splitting and merging procedure according to a proposed split-decision model based on the nonlinearity degree of measurement. 3The efficacy and safety of nimodipine in acute ischemic stroke patients with mild cognitive impairment: a double-blind, randomized,placebo-controlled trial显示文摘Nimodipine might be effective in subcortical vascular dementia(VaD). Its benefit in preventing further cognitive decline in patients with acute ischemic stroke(AIS) and vascular mild cognitive impairment(VaMCI) remains to be established. In this multicenter, double-blind trial, we randomly assigned 654 eligible patients to nimodipine 30 mg three times a day or placebo. The primary outcome was any cognitive decline defined by the changes on the Mini-Mental State Examination(DMMSE à3) or vascular AD assessment scale cognitive subscale(DADAS-cog ! 4) at 6 months. Secondary outcomes included any distribution shift of DADAS-cog, DMMSE or cognitive improvement defined by DADAS-cog à2, or DMMSE ! 0. The primary outcome in the nimodipine group and placebo group were similar for DMMSE à3(4.18% and 7.22%, respectively, P = 0.15) and DADAS-cog ! 4(8.36% and 8.93% respectively,P = 0.88). The distribution shift of DADAS-cog and DMMSE differed significantly between the two groups(P = 0.03 and P = 0.05 respectively). Cognitive improvement occurred in 55.4% in the nimodipine group and 43.6% in the placebo group measured by DADAS-cog à2(Odds Ratio, 1.54; 95% confidence interval[CI] 1.10–2.14, P < 0.01) or 84.0% and 74.6% respectively by DMMSE ! 0(Odds Ratio, 1.79; 95% CI 1.18–2.70, P < 0.01). Nimodipine was associated with better cognitive function in the memory domain. The adverse events rate was similar in two groups. This study is registered with ClinicalTrials.gov,NCT01220622. 5Accelerated evolution of an Lhx2 enhancer shapes mammalian social hierarchies显示文摘Social hierarchies emerged during evolution,and social rank influences behavior and health of individuals.However,the evolutionary mechanisms of social hierarchy are still unknown in amniotes.Here we developed a new method and performed a genome-wide screening for identifying regions with accelerated evolution in the ancestral lineage of placental mammals,where mammalian social hierarchies might have initially evolved.Then functional analyses were conducted for the most accelerated region designated as placental-accelerated sequence 1(PAS1,P=3.15 × 10^-18).Multiple pieces of evidence show that PAS1 is an enhancer of the transcription factor gene Lhx2 involved in brain development.PAS1s isolated from various amniotes showed different c/s-regulatory activity in vitro,and affected the expression of Lhx2 differently in the nervous system of mouse embryos.PAS1 knock-out mice lack social stratification.PAS1 knock-in mouse models demonstrate that PAS1s determine the social dominance and subordinate of adult mice,and that social ranks could even be turned over by mutated PAS1.All homozygous mutant mice had normal huddled sleeping behavior,motor coordination and strength.Therefore,PAS1-Lhx2 modulates social hierarchies and is essential for establishing social stratification in amniotes,and positive Darwinian selection on PAS1 plays pivotal roles in the occurrence of mammalian social hierarchies.Yuting Wang Guangyi Dai Zhili Gu Guopeng Liu Ke Tang Yi-Hsuan Pan Yujie Chen Xin Lin Nan Wu Haoshan Chen Su Feng Shou Qiu Hongduo Sun Qian Li Chuan Xu Yanan Mao Yong Edward Zhang Philipp Khaitovich Yan-Ling Wang Qunxiu Liu Jing-Dong Jackie Han Zhen Shao Gang Wei Chun Xu Naihe Jing Haipeng Li 2020Cell Research2020,30,5:4
6First experimental constraints on WIMP couplings in the effective field theory framework from CDEX显示文摘We present weakly interacting massive particles(WIMPs) search results performed using two approaches of effective field theory from the China Dark Matter Experiment(CDEX), based on the data from both CDEX-1B and CDEX-10 stages. In the nonrelativistic effective field theory approach, both time-integrated and annual modulation analyses were used to set new limits for the coupling of WIMP-nucleon effective operators at 90% confidence level(C.L.) and improve over the current bounds in the low mχregion. In the chiral effective field theory approach, data from CDEX-10 were used to set an upper limit on WIMP-pion coupling at 90% C.L. 7Direct control of store-operated calcium channels by ultrafast laser显示文摘Ca^(2+) channels are essential to cell birth,life,and death.They can be externally activated by optogenetic tools,but this requires robust introduction of exogenous optogenetic genes for expression of photosensitive proteins in biological systems.Here we present femtoSOC,a method for direct control of Ca^(2+)channels solely by ultrafast laser without the need for optogenetic tools or any other exogenous reagents.Specifically,by focusing and scanning wavelength-tuned low-power femtosecond laser pulses on the plasma membrane for multiphoton excitation,we directly induced Ca^(2+) influx in cultured cells.Mechanistic study reveals that photoexcited flavins covalently bind cysteine residues in Orai1 via thioether bonds,which facilitates Orai1 polymerization to form store-operated calcium channels(SOCs)independently of STIM1,a protein generally participating in SOC formation,enabling all-optical activation of Ca^(2+) influx and downstream signaling pathways.Moreover,we used femtoSOC to demonstrate direct neural activation both in brain slices in vitro and in intact brains of living mice in vivo in a spatiotemporal-specific manner,indicating potential utility of femtoSOC.Pan Cheng Xiaoying Tian Wanyi Tang Juan Cheng Jin Bao Haipeng Wang Sisi Zheng Youjun Wang Xunbin Wei Tunan Chen Hua Feng Tian Xue Keisuke Goda Hao He 2021Cell Research2021,31,7:2

18C-176 loaded Ce DNase nanoparticles synergistically inhibit the cGAS-STING pathway for ischemic stroke treatment显示文摘The neuroinflammatory responses following ischemic stroke cause irreversible nerve cell death.Cell free-double strand DNA(dsDNA)segments from ischemic tissue debris are engulfed by microglia and sensed by their cyclic GMP-AMP synthase(cGAS),which triggers robust activation of the innate immune stimulator of interferon genes(STING)pathway and initiate the chronic inflammatory cascade.The decomposition of immunogenic dsDNA and inhibition of the innate immune STING are synergistic immunologic targets for ameliorating neuroinflammation.To combine the anti-inflammatory strategies of STING inhibition and dsDNA elimination,we constructed a DNase-mimetic artificial enzyme loaded with C-176.Nanoparticles are self-assembled by amphiphilic copolymers(P[CL35-b-(OEGMA20.7-co-NTAMA14.3)]),C-176,and Ce^(4+)which is coordinated with nitrilotriacetic acid(NTA)group to form corresponding catalytic structures.Our work developed a new nano-drug that balances the cGAS-STING axis to enhance the therapeutic impact of stroke by combining the DNase-memetic Ce^(4+)enzyme and STING inhibitor synergistically.In conclusion,it is a novel approach to modulating central nervus system(CNS)inflammatory signaling pathways and improving stroke prognosis.Zhixin Zhu Haipeng Lu Lulu Jin Yong Gao Zhefeng Qian Pan Lu Weijun Tong Pik Kwan Lo Zhengwei Mao Haifei Shi 2023Bioactive Materials2023,,11:1
