Collagen-binding basic fibroblast growth factor improves functional remodeling of scarred endometrium in uterine infertile women: a pilot study显示文摘Intrauterine adhesion(IUA) is a common cause of uterine infertility and one of the most severe clinical features is endometrial fibrosis namely endometrial scarring for which there are few cures currently. Blocked angiogenesis is the main pathological change in the scarred endometrium. The fibroblast growth factor 2(b FGF), a member of FGF family, is usually applied to promote healing of refractory ulcer and contributes to angiogenesis of tissues. In this study, the sustained-release system of b FGF100 μg was administrated around scarred endometrium guiding by ultrasound every 4 weeks in 18 patients(2–4 times). Results showed that after treatment, the menstrual blood volume, endometrial thickness and the scarred endometrial area were improved.Histological study showed blood vessel density increased obviously. Three patients(3/18) achieved pregnancy over 20 gestational weeks. Therefore, administrating the b FGF surrounding scarred endometrium may provide a new therapeutic approach for the patients with endometrial fibrosis. | Peipei Jiang Xiaoqiu Tang Huiyan Wang Chenyan Dai Jing Su Hui Zhu Minmin Song Jingyu Liu Ziqing Nan Tong Ru Yaling Li Jingmei Wang Jun Yang Bing Chen Jianwu Dai Yali Hu | 2019 | Science China(Life Sciences)2019,62,12: | 16

A dimeric structure of PD-L1: functional units or evolutionary relics?显示文摘PD-L1 is a member of the B7 protein family, most of whose members so far were identified as dimers in a solution and crystalline state, either complexed or uncomplexed with their ligand(s).The binding of PD-L1 with its receptor PD-1(CD279)delivers an inhibitory signal regulating the T cell function.Simultaneously with the Garboczi group,we successfully solved another structure of human PD-L1(hPD-L1).Our protein crystallized in the space group of C222_(1) with two hPD-L1 molecules per asymmetric unit. After comparison of reported B7 structures,we have found some intrinsic factors involved in the interaction of these two molecules.Based on these results,we tend to believe this uncomplexed hPD-L1 structure demonstrated its potential dimeric state in solution, althougt it could just be an evolutionary relic, too weak to be detected under present technology,or still a functional unit deserved our attentions. | Yong Chen Peipei Liu Feng Gao Hao Cheng Jianxun Qi George F.Gao | 2010 | Protein & Cell2010,1,2: | 10

A new magnetic tapping mode (MAC mode), which is more suitable for living samples, and a novel data collecting system named TREC, are also introduced. | CHEN PeiPei DONG HongTao CHEN Long SUN QuanMei HAN Dong | 2009 | Chinese Science Bulletin2009,54,14: | 8 |
Functional analysis of alternative splicing of the FLOWERING LOCUS T orthologous gene in Chrysanthemum morifolium显示文摘As the junction of floral development pathways, the FLOWERING LOCUS T(FT)protein called‘florigen’plays an important role in the process of plant flowering through signal integration. We isolated four transcripts encoding different isoforms of a FT orthologous gene CmFTL1,from Chrysanthemum morifolium cultivar‘Jimba’.Sequence alignments suggested that the four transcripts are related to the intron 1.Expression analysis showed that four alternative splicing(AS)forms of CmFTL1 varied depending on the developmental stage of the flower. The functional complement experiment using an Arabidopsis mutant ft-10 revealed that the archetypal and AS forms of CmFTL1 had the function of complementing late flower phenotype in different levels. In addition, transgenic confirmation at transcript level showed CmFTL1 and CmFTL1ast coexist in the same tissue type at the same developmental stage, indicating a post-transcriptional modification of CmFTL1 in Arabidopsis. Moreover, ectopic expression of different AS forms in chrysanthemum resulted in the development of multiple altered phenotypes, varying degrees of early flowering. We found that an alternative splicing form(CmFTL1-astE134)without the exon 2 lacked the ability causing the earlier flower phenotype. The evidence in this study indicates that complex alternative processing of CmFTL1 transcripts in C.morifolium may be associated with flowering regulation and hold some potential for biotechnical engineering to create early-flowering phenotypes in ornamental cultivars. | Yachao Mao Jing Sun Peipei Cao Rong Zhang Qike Fu Sumei Chen Fadi Chen Jiafu Jiang | 2016 | Horticulture Research2016,3,1: | 7
Immunity-and-matrix-regulatory cells derived from human embryonic stem cells safely and effectively treat mouse lung injury and fibrosis显示文摘Lung injury and fibrosis represent the most significant outcomes of severe and acute lung disorders, including COVID-19.However,there are still no effective drugs to treat lung injury and fibrosis. In this study,we report the generation of clinical-grade human embryonic stem cells(hESCs)-derived immunity-and matrix-regulatory cells(IMRCs)produced under good manufacturing practice requirements, that can treat lung injury and fibrosis in vivo.We generate IMRCs by sequentially differentiating hESCs with serum-free reagents.IMRCs possess a unique gene expression profile distinct from that of umbilical cord mesenchymal stem cells(UCMSCs),such as higher expression levels of proliferative, immunomodulatory and anti-fibrotic genes. Moreover, intravenous delivery of IMRCs inhibits both pulmonary inflammation and fibrosis in mouse models of lung injury, and significantly improves the survival rate of the recipient mice in a dose-dependent manner, likely through paracrine regulatory mechanisms.IMRCs are superior to both primary UCMSCs and the FDA-approved drug pirfenidone, with an excellent efficacy and safety profile in mice and monkeys. In light of public health crises involving pneumonia, acute lung injury and acute respiratory distress syndrome, our findings suggest that IMRCs are ready for clinical trials on lung disorders. | Jun Wu Dingyun Song Zhongwen Li Baojie Guo Yani Xiao Wenjing Liu Lingmin Liang Chunjing Feng Tingting Gao Yanxia Chen Ying Li Zai Wang Jianyan Wen Shengnan Yang Peipei Liu Lei Wang Yukai Wang Liang Peng Glyn Nigel Stacey Zheng Hu Guihai Feng Wei Li Yan Huo Ronghua Jin Ng Shyh-Chang Qi Zhou Liu Wang Baoyang Hu Huaping Dai Jie Hao | 2020 | Cell Research2020,30,9: | 7
Orbital angular momentum mode logical operation using optical diffractive neural network显示文摘Optical logical operations demonstrate the key role of optical digital computing, which can perform general-purpose calculations and possess fast processing speed, low crosstalk, and high throughput. The logic states usually refer to linear momentums that are distinguished by intensity distributions, which blur the discrimination boundary and limit its sustainable applications. Here,we introduce orbital angular momentum(OAM)mode logical operations performed by optical diffractive neural networks(ODNNs).Using the OAM mode as a logic state not only can improve the parallel processing ability but also enhance the logic distinction and robustness of logical gates owing to the mode infinity and orthogonality.ODNN combining scalar diffraction theory and deep learning technology is designed to independently manipulate the mode and spatial position of multiple OAM modes, which allows for complex multilight modulation functions to respond to logic inputs. We show that few-layer ODNNs successfully implement the logical operations of AND,OR,NOT,NAND,and NOR in simulations. The logic units of XNOR and XOR are obtained by cascading the basic logical gates of AND,OR,and NOT,which can further constitute logical half-adder gates. Our demonstrations may provide a new avenue for optical logical operations and are expected to promote the practical application of optical digital computing. | PEIPEI WANG WENJIE XIONG ZEBIN HUANG YANLIANG HE ZHIQIANG XIE JUNMIN LIU HUAPENG YE YING LI DIANYUAN FAN SHUQING CHEN | 2021 | Photonics Research2021,9,10: | 5
Splitting and Merging Based Multi-model Fitting for Point Cloud Segmentation显示文摘This paper deals with the massive point cloud segmentation processing technology on the basis of machine vision, which is the second essential factor for the intelligent data processing of three dimensional conformation in digital photogrammetry. In this paper, multi-model fitting method is used to segment the point cloud according to the spatial distribution and spatial geometric structure of point clouds by fitting the point cloud into different geometric primitives models. Because point cloud usually possesses large amount of 3D points, which are uneven distributed over various complex structures, this paper proposes a point cloud segmentation method based on multi-model fitting. Firstly, the pre-segmentation of point cloud is conducted by using the clustering method based on density distribution. And then the follow fitting and segmentation are carried out by using the multi-model fitting method based on split and merging. For the plane and the arc surface, this paper uses different fitting methods, and finally realizing the indoor dense point cloud segmentation. The experimental results show that this method can achieve the automatic segmentation of the point cloud without setting the number of models in advance. Compared with the existing point cloud segmentation methods, this method has obvious advantages in segmentation effect and time cost, and can achieve higher segmentation accuracy. After processed by method proposed in this paper, the point cloud even with large-scale and complex structures can often be segmented into 3D geometric elements with finer and accurate model parameters, which can give rise to an accurate 3D conformation. | Liangpei ZHANG Yun ZHANG Zhenzhong CHEN Peipei XIAO Bin LUO | 2019 | Journal of Geodesy and Geoinformation Science2019,2,2: | 5

Roles of low?density lipoproteinreceptor?related protein 1 in tumors显示文摘Low-density lipoprotein receptor-related protein 1(LRP1,also known as CD91),a multifunctional endocytic and cell signaling receptor,is widely expressed on the surface of multiple cell types such as hepatocytes, fibroblasts, neurons, astrocytes, macrophages, smooth muscle cells, and malignant cells.Emerging in vitro and in vivo evidence demonstrates that LRP1 is critically involved in many processes that drive tumorigenesis and tumor progression. For example,LRP1 not only promotes tumor cell migration and invasion by regulating matrix metalloproteinase(MMP)-2and MMP-9 expression and functions but also inhibits cell apoptosis by regulating the insulin receptor, the serine/threonine protein kinase signaling pathway, and the expression of Caspase-3.LRPI-mediated phosphorylation of the extracellular signal-regulated kinase pathway and c-jun N-terminal kinase are also involved in tumor cell proliferation and invasion. In addition,LRP1 has been shown to be down-regulated by microRNA-205 and methylation of LRP1CpG islands.Furthermore,a novel fusion gene,LRP1-SNRNP25,promotes osteosarcoma cell invasion and migration.Only by understanding the mechanisms of these effects can we develop novel diagnostic and therapeutic strategies for cancers mediated by LRP1. | Peipei Xing Zhichao Liao Zhiwu Ren Jun Zhao Fengju Song Guowen Wang Kexin Chen Jilong Yang | 2016 | Chinese Journal of Cancer2016,35,1: | 4

Identification of Gesture Based on Combination of Raw sEMG and sEMG Envelope Using Supervised Learning and Univariate Feature Selection显示文摘In this paper,we propose a novel study for gesture identification using surface electromyography(sEMG)signal, and the raw sEMG signal and the sEMG envelope signal are collected by the sensor at the same time.An efficient method of gesture identification based on the combination of two signals using supervised learning and univariate feature selection is implemented. In previous research techniques, researchers tend to use the raw sEMG signal and extract several constant features for classification, which inevitably causes a result of ignoring individual differences. Our experiment shows that both the optimal feature set and redundant feature set are not same for different subjects. In order to address this problem,we extract all the common features from two signals,up to 76 features, most of which has been established as the common EMG-based gesture index. In addition, extracting too many features in an application can reduce operational efficiency,so we apply for feature selection to get the optimal feature set and decrease the number of extracting feature. As a result, the combination of two signals is better than using a single signal. The feature selection can be used to select optimal feature set from all features to achieve the best classification performance for each subject. The experimental results demonstrate that the proposed method achieves the performance with the highest accuracy of 95%for identifying up to nine gestures only using two sensors. Finally,we develop a real-time intelligent sEMG-driven bionic hand system by using the proposed method. | Shili Liang Yansheng Wu Jianfei Chen Ling Zhang Peipei Chen Zongqian Chai Chunlei Cao | 2019 | Journal of Bionic Engineering2019,16,4: | 4

Inhibiting Hv1 channel in peripheral sensory neurons attenuates chronic inflammatory pain and opioid side effects显示文摘Both opioids and nonsteroidal anti-inflammatory drugs(NSAIDS)produce deleterious side effects and fail to provide sustained relief in patients with chronic inflammatory pain.Peripheral neuroinflammation(PN)is critical for initiation and development of inflammatory pain.A better understanding of molecular mechanisms underlying PN would facilitate the discovery of new analgesic targets and the development of new therapeutics.Emerging evidence suggests that peripheral sensory neurons are not only responders to painful stimuli, but are also actively engaged in inflammation and immunity, whereas the intrinsic regulatory mechanism is poorly understood. Here we report the expression of proton-selective ion channel Hv1 in peripheral sensory neurons in rodents and humans, which was previously shown as selectively expressed in microglia in mammalian central nervous system.Neuronal Hv1 was up-regulated by PN or depolarizing stimulation, which in turn aggravates inflammation and nociception.Inhibiting neuronal Hv1 genetically or by a newly discovered selective inhibitor YHV98-4 reduced intracellular alkalization and ROS production in inflammatory pain, mitigated the imbalance in downstream SHP-1-pAKT signaling, and also diminished pro-inflammatory chemokine release to alleviate nociception and morphine-induced hyperalgesia and tolerance. Thus, our data reveal neuronal Hv1 as a novel target in analgesia strategy and managing opioids-related side effects. | Qiansen Zhang Yimin Ren Yiqing Mo Peipei Guo Ping Liao Yuncheng Luo Jie Mu Zhuo Chen Yang Zhang Ya Li Linghui Yang Daqing Liao Jie Fu Juwen Shen Wei Huang Xuewen Xu Yanyan Guo Lianghe Mei Yunxia Zuo Jin Liu Huaiyu Yang Ruotian Jiang | 2022 | Cell Research2022,32,5: | 3

Genomic characterization of SARS-CoV-2 identified in a reemerging COVID-19 outbreak in Beijing's Xinfadi market in 2020显示文摘After 56 days without coronavirus disease 2019(COVID-19)cases,reemergent cases were reported in Beijing,China on June 11,2020.Here,we report the genetic characteristics of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)sequenced from the clinical specimens of 4 human cases and 2 environmental samples. The nucleotide similarity among six SARS-CoV-2 genomes ranged from 99.98%to 99.99%.Compared with the reference strain of SARS-CoV-2(GenBank No.NC_045512),all six genome sequences shared the same substitutions at nt241(C→T),nt3037(C→T),nt14408(C→T),nt23403(A→G),nt28881(G→A),nt28882(G→A),and nt28883(G→C),which are the characteristic nucleotide substitutions of L-lineage European branch I.This was also proved by themaximum likelihood phylogenetic tree based on the full-length genome of SARS-CoV-2.They also have a unique shared nucleotide substitution,nt6026(C→T),which is the characteristic nucleotide substitution of SARS-CoV-2 in Beijing's Xinfadi outbreak. It is noteworthy that there is an amino acid D614Gmutation caused by nt23403 substitution in all six genomes, which may enhance the virus's infectivity in humans and help it become the leading strain of the virus to spread around the world today. It is necessary to continuously monitor the genetic variation of SARS-CoV-2,focusing on the influence of key mutation sites of SARS-CoV-2 on viral transmission, clinical manifestations, severity, and course of disease. | Yong Zhang Yang Pan Xiang Zhao Weifeng Shi Zhixiao Chen Sheng Zhang Peipei Liu Jinbo Xiao Wenjie Tan Dayan Wang William J.Liu Wenbo Xu Quanyi Wang Guizhen Wu | 2020 | Biosafety and Health2020,2,4: | 3
