| 1 | Quality analysis of commercial samples of Ziziphi spinosae semen(suanzaoren) by means of chromatographic fingerprinting assisted by principal component analysis显示文摘Due to the scarcity of resources of Ziziphi spinosae semen(ZSS), many inferior goods and even adulterants are generally found in medicine markets. To strengthen the quality control, HPLC fingerprint common pattern established in this paper showed three main bioactive compounds in one chromatogram simultaneously. Principal component analysis based on DAD signals could discriminate adulterants and inferiorities. Principal component analysis indicated that all samples could be mainly regrouped into two main clusters according to the first principal component(PC1, redefined as Vicenin II)and the second principal component(PC2, redefined as zizyphusine). PC1 and PC2 could explain 91.42%of the variance. Content of zizyphusine fluctuated more greatly than that of spinosin, and this result was also confirmed by the HPTLC result. Samples with low content of jujubosides and two common adulterants could not be used equivalently with authenticated ones in clinic, while one reference standard extract could substitute the crude drug in pharmaceutical production. Giving special consideration to the well-known bioactive saponins but with low response by end absorption, a fast and cheap HPTLC methodfor quality control of ZSS was developed and the result obtained was commensurate well with that of HPLC analysis. Samples having similar fingerprints to HPTLC common pattern targeting at saponins could be regarded as authenticated ones. | 2 | A Geochemical Study of an REE-rich Carbonatite Dyke at Bayan Obo,Inner Mongolia,Northern China显示文摘An REE-rich carbonatite dyke was found in Dulahala, close to the Bayan Obo superlarge REE-Nb-Fe mineral deposit in Inner Mongolia, northern China. The REE content in the dyke varies greatly, from 1% up to 20% (wt), which might constitute rich REE ores. Light REEs in the carbonatite are enriched and highly fractionated relative to heavy REEs and there is no Eu anomaly. The REE and trace element distribution patterns of the carbonatite are identical to those of fine-grained dolomite marble which is the host rock of the Bayan Obo REE-Nb-Fe superlarge mineral deposit. | 4 | Immunomodulatory properties of mesenchymal stem cells/dental stem cells and their therapeutic applications显示文摘Mesenchymal stem/stromal cells(MsCs)are widely distributed in the body and play essential roles in tissue regeneration and homeostasis.MsCs can be isolated from discarded tissues,expanded in vitro and used as therapeutics for autoimmune diseases and other chronic disorders.MsCs promote tissue regeneration and homeostasis by primarily acting on immune cells.At least six different types of MsCs have been isolated from postnatal dental tissues and have remarkable immunomodulatory properties.Dental stem cells(DsCs)have been demonstrated to have therapeutic effects on several systemic inflammatory diseases.Conversely,MsCs derived from nondental tissues such as the umbilical cord exhibit great benefits in the management of periodontitis in preclinical studies.Here,we discuss the main therapeutic uses of MSCs/DSCs,their mechanisms,extrinsic inflammatory cues and the intrinsic metabolic circuitries that govern the immunomodulatory functions of MSCs/DSCs.Increased understanding of the mechanisms underpinning the immunomodulatory functions of MSCs/DSCs is expected to aid in the development of more potent and precise MSC/DSC-based therapeutics. | Peishan Li Qianmin Ou Songtao Shi Changshun Shao | 2023 | Cellular & Molecular Immunology2023,20,6: | 4 |

| 6 | CUL4B negatively regulates Toll-like receptor-triggered proinflammatory responses by repressing Pten transcription显示文摘Toll-like receptors (TLRs) play critical roles in innate immunity and inflammation. The molecular mechanisms by which TLR signaling is fine-tuned remain to be completely elucidated. Cullin 4B (CUL4B), which assembles the CUL4B-RING E3 ligase complex (CRL4B), has been shown to regulate diverse developmental and physiological processes by catalyzing monoubiquitination for histone modification or polyubiquitination for proteasomal degradation. Here, we identified the role of CUL4B as an intrinsic negative regulator of the TLR-triggered inflammatory response. Deletion of CUL4B in macrophages increased the production of proinflammatory cytokines and decreased anti-inflammatory cytokine IL-10 production in response to pathogens that activate TLR3, TLR4, or TLR2. Myeloid cell-specific Cul4b knockout mice were more susceptible to septic shock when challenged with lipopolysaccharide, polyinosinic-polycytidylic acid or Salmonella typhimurium infection. We further demonstrated that enhanced TLR-induced inflammatory responses in the absence of CUL4B were mediated by increased GSK3β activity. Suppression of GSK3β activity efficiently blocked the TLR-triggered increase in proinflammatory cytokine production and attenuated TLR-triggered death in Cul4b mutant mice. Mechanistically, CUL4B was found to negatively regulate TLR-triggered signaling by epigenetically repressing the transcription of Pten, thus maintaining the anti-inflammatory PI3K-AKT-GSK3β pathway. The upregulation of PTEN caused by CUL4B deletion led to uncontrolled GSK3β activity and excessive inflammatory immune responses. | 11 | NAD^(+) salvage governs the immunosuppressive capacity of mesenchymal stem cells显示文摘Mesenchymal stem/stromal cells(MSCs)possess robust immunoregulatory functions and are promising therapeutics for inflammatory disorders.This capacity is not innate but is activated or‘licensed’by inflammatory cytokines.The licensing mechanism remains unclear.Here,we examined whether inflammatory cytokines metabolically reprogrammed MSCs to confer this immunoregulatory capacity.In response to stimulation by inflammatory cytokines,MSCs exhibited a dramatic increase in the consumption of glucose,which was accompanied by an enhanced use of nicotinamide adenine dinucleotide(NAD^(+))and increased expression of nicotinamide phosphoribosyltransferase(NAMPT),a central enzyme in the salvage pathway for NAD^(+) production.When NAD^(+) synthesis was blocked by inhibiting or depleting NAMPT,the immunosuppressive function of MSCs induced by inflammatory cytokines was greatly attenuated.Consequently,when NAD^(+) metabolism in MSCs was perturbed,their therapeutic benefit was decreased in mice suffering from inflammatory bowel disease and acute liver injury.Further analysis revealed that NAMPT-driven production of NAD^(+) was critical for the inflammatory cytokine-induced increase in glycolysis in MSCs.Furthermore,the increase in glycolysis led to succinate accumulation in the tricarboxylic acid cycle,which led to hypoxia-inducible factor 1α(HIF-1α)stabilization and subsequently increased the transcription of key glycolytic genes,thereby persistently maintaining glycolytic flux.This study demonstrated that unlike its proinflammatory role in immune cells,NAD^(+) metabolism governs the anti-inflammatory function of MSCs during inflammation. | Jiankai Fang Pengbo Hou Shisong Liu Muqiu Zuo Zhanhong Liu Wangwang Chen Yuyi Han Yanan Li Tingting Wang Chao Feng Peishan Li Changshun Shao Yufang Shi | 2023 | Cellular & Molecular Immunology2023,20,10: | 1 |
