| 2 | Identification of immunoreactive proteins of Brucella melitensis by immunoproteomics显示文摘Infection with Brucella causes brucellosis, a chronic disease in humans, which induces abortion and sterility in livestock. Among the different Brucella species, Brucella melitensis is considered the most virulent and is the predominant species associated with outbreaks in China. To date, no safe human vaccine is available against Brucella infection. The currently used live vaccines against Brucella in livestock induce antibodies that interfere with the diagnosis of field infection in vaccinated animals, which is harmful to eradication programs. However, there is as yet no complete profile of immunogenic proteins of B. melitensis. Towards the development of a safer, equally efficacious, and field infection-distinguishable vaccine, we used immunoproteomics to identify novel candidate immunogenic proteins from B. melitensis M5. Eighty-eight immunoreactive protein spots from B. melitensis M5 were identified by Western blotting and were assigned to sixty-one proteins by mass spectrometry, including many new immunoreactive proteins such as elongation factor G, F0F1 ATP synthase subunit beta, and OMP1. | 8 | Formation mechanism of high-quality reservoirs in intermediate volcanic rocks:A case study of Longfengshan area in Songliao Basin显示文摘The basin margin slope area of Changling fault depression in Songliao Basin is dominated by intermediate-basic volcanic rocks of Huoshiling Formation,and there is still a lack of in-depth understanding of the development pattern and formation mechanism of high-quality reservoirs in intermediate-basic volcanic rocks in this area.Taking the intermediate volcanic rocks in Longfengshan area of Changling fault depression as an example,a comprehensive study was undertaken via core observations,thin section porosity analysis,rock physical property tests,scanning electron microscopy and X-ray diffraction analysis,combined with well log and seismic d ata.As a result,formation mechanism of a high quality reservoir was made clear based on dissolution modification effects of the volcanic rocks,and the development model of the high quality reservoir under t he dual control of lithology and fluid was established.The research shows that the volcanic rocks in this area consist of six lithologies:andesitic hydrothermal breccia,andesite,andesitic volcanic breccia,andesitic tuff,dacitic tuff and sedimentary tuff,which are controlled by structure,eruptive source,eruptive action and palaeotopography.The proximal facies of the volcanic edifice were located on the basin margin,distributed along faults mainly consisting of overflow facies intermixed with eruptive facies,and gradually transformed into thick eruptive facies(middle-source facies)and volcanic sedimentary facies(distal facies).The reservoir space of volcanic rocks was dominated by dissolution pores,which were formed by weathering and dissolution by organic acidic fluids.The thermal debris flow subphase in the middle-source facies of the volcanic edifice had a large amount of easily dissolved volcanic ash,which was close to the source rock in the centre of the basin and was a favourable area for the development of dissolution pores.This study is providing a guide for oil and gas exploration in the intermediate-basic volcanic rocks in the slope of Changling fault depression in Songliao Basin. | XING Jian SHAN Xuanlong REN Xianjun YI Jian LIU Chaoyang NIU Penghui | 2022 | Global Geology2022,25,2: | 1 |
| 9 | N6-methyladenosine modification of CENPF mRNA facilitates gastric cancer metastasis via regulating FAK nuclear export显示文摘Background:N6-methyladenosine(m^(6)A)modification is the most common modification that occurs in eukaryotes.Although substantial effort has been made in the prevention and treatment of gastric cancer(GC)in recent years,the prognosis of GC patients remains unsatisfactory.The regulatory mechanism between m^(6)A modification and GC development needs to be elucidated.In this study,we examined m^(6)A modification and the downstream mechanism in GC.Methods:Dot blotting assays,The Cancer Genome Atlas analysis,and quantitative real‑time PCR(qRT-PCR)were used to measure the m^(6)A levels in GC tissues.Methylated RNA-immunoprecipitation sequencing and RNA sequencingwere performed to identify the targets ofm^(6)Amodification.Western blotting,Transwell,wound healing,and angiogenesis assays were conducted to examine the role of centromere protein F(CENPF)in GC in vitro.Xenograft,immunohistochemistry,and in vivo metastasis experiments were conducted to examine the role of CENPF in GC in vivo.Methylated RNA-immunoprecipitation-qPCR,RNA immunoprecipitation-qPCR and RNA pulldown assays were used to verify the m^(6)A modification sites of CENPF.Gain/loss-of-function and rescue experiments were conducted to determine the relationship between CENPF and the mitogen-activated protein kinase(MAPK)signaling pathway in GC cells.Coimmunoprecipitation,mass spectrometry,qRT-PCR,and immunofluorescence assays were performed to explore the proteins that interact with CENPF and elucidate the regulatory mechanisms between them.Results:CENPF was upregulated in GC and facilitated the metastasis of GC both in vitro and in vivo.Mechanistically,increasedm^(6)A modification of CENPF was mediated by methyltransferase 3,and this modified molecule could be recognized by heterogeneous nuclear ribonucleoprotein A2/B1(HNRNPA2B1),thereby promoting its mRNA stability.In addition,the metastatic phenotype of CENPF was dependent on the MAPK signaling pathway.Furthermore,CENPF could bind to FAK and promote its localization in the cytoplasm.Moreover,we discovered that high expression of CENPF was related to lymphatic invasion and overall survival in GC patients.Conclusions:Our findings revealed that increased m^(6)A modification of CENPF facilitates the metastasis and angiogenesis of GC through the CENPF/FAK/MAPK and epithelial-mesenchymal transition axis.CENPF expression was correlated with the clinical features of GC patients;therefore,CENPF may serve as a prognostic marker of GC. | Penghui Xu Jing Yang Zetian Chen Xing Zhang Yiwen Xia Sen Wang Weizhi Wang Zekuan Xu | 2023 | Cancer Communications2023,43,6: | 0 |

| 11 | Regulatory effect of vascular endothelial growth factor on blood spinal cord barrier in presyrinx state of experimental syringomyelia显示文摘BACKGROUND:Vascular endothelial growth factor (VEGF) is able to regulate blood spinal cord barrier function as well as influence neovascularization and cause edema. OBJECTIVE: Through establishment of a rabbit model of syringomyelia,to explore the correlation between VEGF protein and mRNA expressions and function of blood spinal cord barrier and edema degree of spinal cord in presyrinx state. DESIGN,TIME AND SETTING: Randomized controlled animal study was performed in the Tumor Institute of the Fourth Hospital,Hebei Medical University from January to June 2007. MATERIALS: A total of 0.6 mL Kaolin solution (250 g/L,37 ℃) was injected into the cisterna magna of 40 rabbits in the kaolin group to establish syringomyelia models. Goat anti-rabbit VEGF monoclonal antibody was provided by DIACLONE Company,USA; RT-PCR related reagents were provided by Huamei Bioengineering Co.,Ltd.,Beijing. METHODS: Sixty Chinese white rabbits were divided randomly into two groups: Kaolin group (n=40) and control group (n=20). Physiological saline (0.6 mL at 37 ℃) was injected in rabbits of control group. On days 1,3,7,14 and 21 after kaolin injection,cervical cords samples were harvested after sacrifice of animal. MAIN OUTCOME MEASURES: VEGF protein and mRNA expressions were detected by immunohistochemistry and RT-PCR on days 1,3,7,14,and 21 after kaolin injection. A quantitative analysis of blood spinal cord barrier function was performed by Evans blue technique. Water content of the spinal cord was measured by dry-wet weight technique. The correlation between the expression of VEGF protein and mRNA and the function of blood spinal cord barrier in the upper cervical cord of the presyrinx state was analyzed by linear correlation analysis. RESULTS: The water content and Evans blue content increased in the kaolin group on days 1 and 3 postoperatively compared with the control group (F=7.387,61.35,P<0.05,0.01),and reached a peak on day 7 (F=135.94,528.35,P<0.01). They declined slowly to day 21 postoperatively,but both contents were still higher than the control group (F=11.51,58.63,P<0.01). VEGF protein expression increased on day 1,and stronger positive expression was seen on days 3,7 and 14. It decreased on day 21. VEGF protein expression was higher than the control group at each time point (F=137.4-468.5,P<0.01). VEGF mRNA expression showed the same pattern in the cervical cord at different time points. By statistical analysis,the expression of VEGF protein and mRNA had a significantly positive correlation with the structural and functional changes of the blood spinal cord barrier in the presyrinx state (r=0.604-0.979,P<0.05). | 12 | Microscopic pore characteristics of andesite and implication from mineral content:A case study in Huoshiling Formation of Changling fault depression,Songliao Basin显示文摘In order to study the microscopic pore characteristics of andesite reservoir and the effect of mineral content on the andesite pore,this study takes the andesite of the Huoshiling Formation in Longfengshan of Changling fault depression,Songliao Basin as a subject.The andesite reservoir space was discerned through the observation of cores and casting thin sections.Besides,the pore size distribution of andesites and their mineral contents were quantitatively characterized by high-pressure mercury injection,nitrogen adsorption and XRD,respectively.The results show that:(1)There are various types of reservoir space in andesites,including vesicles,amygdule,intergranular pores,matrix dissolution pores and dissolution pores of amygdala,and three types of fractures including dissolution,structural and explosion fractures.(2)The pore size distribution of andesite is complex.The main pore size of andesite is mid-pore(10-20 nm)with some large-pores(>50 nm).Mid-pore and large-pore provide the main specific surface area,which are the main space for gas storage.(3)The andesite reservoir space in the study area is mainly controlled by dissolution,as supported by the relationship between the change of mineral content and porosity evolution.The porosity of andesites decreases with the increase of quartz and chlorite content,but increases with the increase of soluble mineral,e.g.,feldspar content. | NIU Penghui SHAN Xuanlong REN Xianjun YI Jian LIU Chaoyang XING Jian | 2022 | Global Geology2022,25,2: | 0 |
| 13 | Study on Treating Glioblastoma with (125)~I Seed Implantation during Operations显示文摘OBJECTIVE To evaluate the therapeutic efficacy of treating glioblastoma with interstitial irradiation using radioactive seeds. METHODS First,the tumor was removed as completely as possible under an operating microscope.Then 125I seeds which had been selected were implanted in the tumor bed followed by routine radiotherapy after the operation.The time of progression(TTP)and overall survival(OS)of the patients were recorded,and statistical y compared with patients who had received only surgery and radiotherapy for glioblastoma during the same period. RESULTS Nineteen patients,who were diagnosed with glioblastoma for the first time,with a mean age of 51.2 years,were involved in this study. Radioactive seeds(8~20)were implanted into the tumor bed after tumorectomy with a mean surrounding therapeutic dose of 31.6 Gy,fol owed by an external irradiation dose of 42~54 Gy.The average TTP in the group with implanted seeds was 42.1±2.4 w(95%confidence interval,37.2~46.5 w),and 27.1±2.7 w(95%confidence interval,24.3~31.8 w)in the control group.The average OS was 66.3±3.2 w(95%confidence interval,61.9~70.4 w)and 47.7±2.6 w(95%confidence interval,44.4~50.5 w),respectively.Statistical analysis showed there was a significant difference between two groups. CONCLUSION The survival time of patients with glioblastoma can be prolonged through radioactive seed implantation at the time of operation.
