| 1 | Prevalence and evolution of drug resistance HIV-1 variants in Henan, China显示文摘To understand the prevalence and evolution of drug resistant HIV strains in Henan China after the implementation of free antiretroviral therapy for AIDS patients. 45 drug nave AIDS patients, 118 AIDS patients who received three months antiretroviral therapy and 124 AIDS patients who received six months antiretroviral treatment were recruited in the southern part of Henan province. Information on general condition, antiretroviral medicines, adherence and clinical syndromes were collected by face to face interview. Meanwhile, 14ml EDTA anticoagulant blood was drawn. CD4/CD8 T cell count, viral load and genotypic drug resistance were tested. The rates of clinical improvement were 55.1% and 50.8% respectively three months and six months after antiretroviral therapy. The mean CD4 cell count after antiretroviral therapy was significantly higher than in drug nave patients. The prevalence rate of drug resistant HIV strains were 13. 9%, 45.4% and 62.7% in drug nave patients, three month treatment patients and six month treatment patients, respectively. The number of resistance mutation codons and the frequency of mutations increased significantly with continued antiretroviral therapy. The mutation sites were primarily at the 103, 106 and 215 codons in the three-month treatment group and they increased to 15 codon mutations in the six-month treatment group. From this result, the evolution of drug resistant strains was inferred to begin with the high level NNRTI resistant strain, and then develop low level resistant strains to NRTIs. The HIV strains with high level resistance to NVP and low level resistance to AZT and DDI were highly prevalent because of the AZT+DDI+NVP combination therapy. These HIV strains were also cross resistant to DLV, EFV, DDC and D4T. Poor adherence to therapy was believed to be the main reason for the emergence and prevalence of drug resistant HIV strains. The prevalence of drug resistant HIV strains was increased with the continu- ation of antiretroviral therapy in the southern part of Henan province. | 2 | Microwave Exposure Impairs Synaptic Plasticity in the Rat Hippocampus and PC12 Cells through Over-activation of the NMDA Receptor Signaling Pathway显示文摘Objective The aim of this study is to investigate whether microwave exposure would affect the N-methyl-D-aspartate receptor(NMDAR) signaling pathway to establish whether this plays a role in synaptic plasticity impairment. Methods 48 male Wistar rats were exposed to 30 m W/cm2 microwave for 10 min every other day for three times. Hippocampal structure was observed through H&E staining and transmission electron microscope. PC12 cells were exposed to 30 m W/cm2 microwave for 5 min and the synapse morphology was visualized with scanning electron microscope and atomic force microscope. The release of amino acid neurotransmitters and calcium influx were detected. The expressions of several key NMDAR signaling molecules were evaluated. Results Microwave exposure caused injury in rat hippocampal structure and PC12 cells, especially the structure and quantity of synapses. The ratio of glutamic acid and gamma-aminobutyric acid neurotransmitters was increased and the intracellular calcium level was elevated in PC12 cells. A significant change in NMDAR subunits(NR1, NR2 A, and NR2B) and related signaling molecules(Ca2+/calmodulin-dependent kinase II gamma and phosphorylated c AMP-response element binding protein) were examined. Conclusion 30 m W/cm2 microwave exposure resulted in alterations of synaptic structure, amino acid neurotransmitter release and calcium influx. | 3 | Influence of FasL gene expression on hepatic metastasis of colorectal carcinoma显示文摘BACKGROUND: FasL expression was reported to be asso- ciated with hepatic metastasis of colorectal cancer. The aim of this study was to study FasL gene expression in colorectal carcinoma and its influences on biological behavior and he- patic metastasis of colorectal carcinoma. METHODS: FasL gene expressions were examined with re- verse transcriptase-polymerase chain reaction (RT-PCR) in the primary focus of colorectal carcinoma, adjacent can- cerous mucosae, and metastasized liver focus from colorec- tal cancer. HR-8348 cells of human rectal cancer cell line were transfected with FasL cDNA. Cell growth suppression rate and response to 5-FU and carboplatin were observed and analyzed with the MTT method. RESULTS: FasL gene expression was detected in the prima- ry focus of colorectal cancer ( n = 58), adjacent cancerous mucosae ( n = 58), and metastasized hepatic tumor tissues (n =28). The positive rate of FasL expression was 24% (14/ 58), 8% (5/58), and 100% (58/58) in the primary focus, adjacent cancerous mucosae and metastasized hepatic tumor tissues respectively. FasL expression rate in the me- tastasized hepatic tumor tissues was higher than that in the primary focus (χ2 = 43.49, P<0. 01) and adjacent cance- rous mucosae (χ2=57.66, P<0.01). In a group of patients with hepatic metastasis, the FasL expression rate in primary focus was higher than that in patients without hepatic me- tastasis (χ2=3.96, P <0.05). In vitro study positive expres- sion of FasL was shown in transfected HR-8348 cells. When 5-FU or carboplatin was added, there was a significant difference in growth suppression rate between FasL positive and controlled cancer cells (t=9.02, t = 11.93, P<0.01). Under the same concentration of chemotherapeutic agents, the survival rate of FasL positive HR-8348 cells was higher than that of controlled cells. CONCLUSIONS: FasL positive cancer cells are powerfullyresistant to chemotherapeutic agents. | 4 | Prognostic evaluation of postoperative adjuvant therapy for operable cervical cancer:10 years'experience of National Cancer Center in China显示文摘Objective: The aim of this study was to investigate the prognostic factors and to evaluate the impact of adjuvant therapy on clinical outcome for early-stage cervical cancer.Methods: The clinical-pathological data of all 1,335 patients with the International Federation of Gynecology and Obstetrics(FIGO) Ib–IIa cervical cancer treated with primary radical surgery at the Chinese National Cancer Center between May 2007 and Dec 2013 were retrospectively reviewed.The median follow-up was 70 months.Results: Of all the patients,61.6% of the cases received adjuvant therapy,with 5-year disease-free survival(DFS)of 92.1% and 5-year overall survival(OS) of 95.0%.In multivariate analysis,differentiation of G3(P<0.05),lymph node metastasis(LNM,P<0.05) and lymphovascular space invasion(LVSI,P<0.05) were independent predictors for OS,while LNM(P<0.05),deep stroma invasion(DSI,P<0.05) and LVSI(P<0.05) were independent factors for DFS.The samples were stratified by histologic type,and cervical squamous cell carcinoma(SCC) was found to share the same independent factors except for differentiation of OS.As to patients with cervical adenocarcinoma/adenosquamous carcinoma(AC/ASC),differentiation was the independent predictor of OS(P<0.05); and LVSI of DFS(P<0.05).Of 236 patients with high-risk factors,there was no significant difference in survival between concurrent chemoradiotherapy(CCRT,n=195),radiotherapy(RT,n=24),and chemotherapy(CT,n=17).Among the 190 patients with LNM who underwent CCRT,124 cases showed improved DFS after sequential CT(P=0.118),with a recurrence rate decrease of 14%,though the difference was not statistically significant.Patients with single intermediate-risk factors like DSI or LVSI were found to partially benefit from adjuvant therapy,but the difference was not statistically significant.Conclusions: LNM,LVSI,DSI and differentiation were found to be independent prognostic factors for operable cervical cancer.Aggressive postoperative adjuvant therapy based on single risk factors in Chinese National Cancer Center could benefit survival.CCRT+CT outperformed CCRT in high-risk patients.For patients with single non-high-risk factor,the role of adjuvant therapy needs to be further discussed. | Tong Shu Dan Zhao Bin Li Yating Wang Shuanghuan Liu Pingping Li Jing Zuo Ping Ba i RongZhang Lingying Wu | 2017 | Chinese Journal of Cancer Research2017,29,6: | 12 |

| 6 | A double-blind, double-dummy, randomized controlled, multicenter trial of ^(99)Tc-methylene diphosphonate in patients with moderate to severe rheumatoid arthritis显示文摘Background:Clinical observational studies revealed that ^(99)Tc-methylene diphosphonate(^(99)Tc-MDP)could reduce joint pain and swollenness in rheumatoid arthritis(RA)patients.This multicenter,randomized,double-blind,double-dummy study aimed to evaluate the effects of ^(99)Tc-MDP plus methotrexate(MTX)vs.MTX alone or ^(99)Tc-MDP alone on disease activity and structural damage in MTX-naïve Chinese patients with moderate to severe RA.Methods:Eligible patients with moderate to severely active RA were randomized to receive ^(99)Tc-MDP plus MTX(n=59)vs.MTX(n=59)alone or ^(99)Tc-MDP(n=59)alone for 48 weeks from six study sites across four provinces in China.The primary outcomes were the American College of Rheumatology 20%improvement(ACR20)response rates at week 24 and changes in modified total Sharp score at week 48.Results:At week 24,the proportion of participants achieving ACR20 was significantly higher in the MTX+^(99)Tc-MDP combination group(69.5%)than that in the MTX group(50.8%)or ^(99)Tc-MDP group(47.5%)(P=0.03 for MTX+^(99)Tc-MDP vs.MTX,and MTX+^(99)Tc-MDP vs.^(99)Tc-MDP,respectively).The participants in the MTX+^(99)Tc-MDP group and the ^(99)Tc-MDP group had significantly less important radiographic progression than the participants in the MTX group over the 48 weeks(MTX+^(99)Tc-MDP vs.MTX:P=0.03,^(99)Tc-MDP vs.MTX:P=0.03,respectively).There was no significant difference in terms of adverse events(AEs)among the groups.No serious AEs were observed.Conclusions:This study demonstrated that the combination of ^(99)Tc-MDP with MTX inhibited structural damage and improved disease activity in RA patients compared with MTX and ^(99)Tc-MDP monotherapies,without increasing the rate of AEs.Additional clinical studies of ^(99)Tc-MDP therapy in patients with RA are warranted.Trial Registration:Chictr.org,ChiCTR-IPR-14005684;http://gffzz62a03ee25e6f4a77hvq5o5qucvb9n66nw.ffgz.tsg.suse.edu.cn/showproj.aspx?proj=10088. | Qiong Fu Ping Feng Ling-Yun Sun Xiao-Xia Zuo Dong-Bao Zhao Dong-Yi He Hua-Xiang Wu Wei Zhang Wei Zhang Fang Du Chun-De Bao | 2021 | Chinese Medical Journal2021,,12: | 5 |
| 7 | Proteomic analysis of differentially expressed proteins involving in liver metastasis of human colorectal carcinoma显示文摘BACKGROUND:Early diagnosis of liver metastasis of colorectal carcinoma is very important for the appropriate treatment of such patients.However,there has been no effective approach available for clinical application.The present study aimed to investigate the differential expression of proteins in patients with liver metastasis of colorectal carcinomas using proteomic analysis and evaluate its potentiality in clinical diagnosis. METHODS:Fluorescence two-dimensional differential in-gel electrophoresis(2-D DIGE)was used to analyze and compare the protein expression between normal mucosa,the primary focus,and liver metastases.Proteomic analysis was made to identify the differentially expressed proteins.Immunohistological staining was used to confirm the expression of differentially expressed proteins in colorectal carcinomas and areas of liver metastasis. RESULTS:A 1.5-fold difference was found with 46 differentially expressed proteins.In 20 differentially expressed proteins,3 were down-regulated and 17 up-regulated in liver metastases. Proteomic analysis showed that the S-adenosylmethionine transgelin variant was down-regulated in liver metastasis tissues.Zinc finger protein 64 homolog(Zfp64),guanine nucleotide exchange factor 4(GEF4),human arginase,glutathione S-transferases(GSTs)A3,and tumor necrosis factorα(TNF-α)-induced protein 9 were up-regulated in liver metastasis tissues.Immunohistochemical staining confirmed that human arginase expression was higher in liver metastases than in the primary focus. CONCLUSIONS:There was a significant difference in protein expression between the primary focus of colorectal carcinoma and liver metastases.The differentially regulated proteins were closely related to liver metastasis of colorectal carcinoma. | 17 | Rett syndrome-causing mutations compromise MeCP2- mediated liquid—liquid phase separation of chromatin显示文摘Rett syndrome(RTT),a severe postnatal neurodevelopmental disorder,is caused by mutations in the X-linked gene encoding methyl-CpG-binding protein 2(MeCP2).MeCP2 is a chromatin organizer regulating gene expression.RTT-causing mutations have been shown to affect this function.However,the mechanism by which MeCP2 organizes chromatin is unclear.In this study,we found that MeCP2 can induce compaction and liquid-liquid phase separation of nudeosomal arrays in vitro,and DNA methylation further enhances formation of chromatin condensates by MeCP2.Interestingly,RTT-causing mutations compromise MeCP2-mediated chromatin phase separation,while benign variants have little effect on this process.Moreover,MeCP2 competes with linker histone H1 to form mutually exclusive chromatin condensates in vitro and distinct heterochromatin foci in vivo.RTT-causing mutations reduce or even abolish the ability of MeCP2 to compete with histone H1 and to form chromatin condensates.Together,our results identify a novel mechanism by which phase separation underlies MeCP2-mediated heterochromatin formation and reveal the potential link between this process and the pathology of RTT. | Liang Wang Mingli Hu Mei-Qing Zuo Jicheng Zhao Di Wu Li Huang Yongxin Wen Yunfan Li Ping Chen Xinhua Bao Meng-Qiu Dong Guohong Li Pilong Li | 2020 | Cell Research2020,30,5: | 3 |
