1Genome-wide analysis of heat shock transcription factor families in rice and Arabidopsis显示文摘The heat shock transcription factors (HSFs) are the major heat shock factors regulating the heat stress response. They participate in regulating the expression of heat shock proteins (HSPs), which are critical in the protection against stress damage and many other important biological processes. Study of the HSF gene family is important for understanding the mechanism by which plants respond to stress. The completed genome sequences of rice (Oryza sativa) and Arabidopsis (Arabidopsis thaliana) constitute a valuable resource for comparative genomic analysis, as they are representatives of the two major evolutionary lineages within the angiosperms: the monocotyledons and the dicotyledons. The identification of phylogenetic relationships among HSF proteins in these species is a fundamental step to unravel the functionality of new and yet uncharacterized genes belonging to this family.In this study, the full complement of HSF genes in rice and Arabidopsis has probably been identified through the genome-wide scan. Phylogenetic analyses resulted in the identification of three major clusters of orthologous genes that contain members belonging to both species, which must have been represented in their common ancestor before the taxonomic splitting of the angiosperms. Further analysis of the phylogenetic tree reveals a possible dicot specific gene group. We also identified nine pairs of paralogs, as evidence for studies on the evolution history of rice HSF family and rice genome evolution. Expression data analysis indicates that HSF proteins are widely expressed in plants. 4Overexpression of the maize GRF10, an endogenous truncated growth-regulating factor protein, leads to reduction in leaf size and plant height显示文摘It has long been thought that growth‐regulating factors(GRFs) gene family members act as transcriptional activators to play important roles in multiple plant developmental processes. However, the recent characterization of Arabidopsis GRF7 showed that it functions as a transcriptional repressor of osmotic stress‐responsive genes. This highlights the complex and diverse mechanisms by which different GRF members use to take action. In this study, the maize(Zea mays L.) GRF10 was functionally characterized to improve this concept. The deduced ZmGRF10 protein retains the N‐terminal QLQ and WRC domains, the characteristic regions as protein‐interacting and DNA‐binding domains, respectively. However,it lacks nearly the entire C‐terminal domain, the regions executing transactivation activity. Consistently, ZmGRF10 protein maintains the ability to interact with GRF‐interacting factors(GIFs) proteins, but lacks transactivation activity.Overexpression of ZmGRF10 in maize led to a reduction in leaf size and plant height through decreasing cell proliferation,whereas the yield‐related traits were not affected. Transcriptome analysis revealed that multiple biological pathways were affected by ZmGRF10 overexpression, including a few transcriptional regulatory genes, which have been demonstrated to have important roles in controlling plant growth and development. 12Soil contamination with antibiotics in a typical peri-urban area in eastern China: Seasonal variation, risk assessment, and microbial responses显示文摘The prevalence and persistence of antibiotics in soils has become an emerging environmental issue and an increasing threat to soil security and global public health. The problem is more severe in areas undergoing rapid urbanization; however, the ecological risks of antibiotics,seasonal variability, and associated soil microbial responses in peri-urban soils have not been well-explored. The seasonal soil sampling campaigns were conducted in a typical peri-urban watershed in eastern China to investigate distribution of antibiotics. The results demonstrated higher mean concentrations of most antibiotic compounds in winter than in summer in periurban soils. The seasonal variations of norfloxacin, enrofloxacin, and ciprofloxacin were more significant than those of other antibiotics, due to their higher migration ability and bioavailability.An ecological risk assessment demonstrated that chlortetracycline, ciprofloxacin, doxycycline,and ofloxacin can pose high risks to soil microorganisms. Furthermore, the coexistence of multiple antibiotics obviously poses higher risks than individual compounds. A redundancy analysis demonstrated that tetracyclines mainly showed negative correlations with Firmicutes and Chloroflexi, and quinolones showed obviously negative correlations with Acidobacteria,Gemmatimonadetes, and Nitrospirae, suggesting potential inhibition from antibiotics on biological activities or biodegradation processes. However, the persistence of antibiotics in soil results in a significant decrease in bacterial diversity and a change in dominant species.Our results provide an overview of the seasonal variability of antibiotics and the associated effects on bacterial communities in peri-urban soils. 13p53 upregulated by HIF-la promotes hypoxiainduced G2/M arrest and renal fibrosis in vitro and in vivo显示文摘Hypoxia plays an important role in the genesis and progression of renal fibrosis.The underlying mechanisms, however, have not been sufficiently elucidated. We examined the role of p53 in hypoxia-induced renal fibrosis in cell culture (human and rat renal tubular epithelial cells) and a mouse unilateral ureteral obstruction (UUO) model. Cell cycle of tubular cells was determined by flow cytometry, and the expression of profibrogenic factors was determined by RT-PCR, immunohistochemistry, and western blotting. Chromatin immunoprecipitation and luciferase reporter experiments were performed to explore the effect of HIF-lα on p53 expression. We showed that, in hypoxic tubular cells, p53 upregulation suppressed the expression of CDK1 and cyclins Bl and DI, leading to cell cycle (G2/M) arrest (or delay) and higher expression of TGF-β, CTGF, collagens, and fibronectin. p53 suppression by siRNA or by a specific p53 inhibitor (PIF-α) triggered opposite effects preventing the G2/M arrest and profibrotic changes. In vivo experiments in the UUO model revealed similar antifibrotic results following intraperitoneal administration of PIF-α(2.2 mg/kg). Using gain-of-function, loss-of-function, and luciferase assays, we further identified an HRE3 region on the p53 promoter as the HIF-lα-binding site. The HIF-la-HRE3 binding resulted in a sharp transcriptional activation of p53. Collectively, we show the presence of a hypoxia-activated, p53-responsive profibrogenic pathway in the kidney. 15A Transcriptional Network Promotes Anthocyanin Biosynthesis in Tomato Flesh显示文摘Dietary anthocyanins are important health-promoting antioxidants that make a major contribution to the quality of fruits. It is intriguing that most tomato cultivars do not produce anthocyanins in fruit. However, the purple tomato variety Indigo Rose, which has the dominant Aft locus combined with the recessive atv locus from wild tomato species, exhibits light-dependent anthocyanin accumulation in the fruit skin. Here, we report that Aft encodes a functional anthocyanin activator named SlAN2-like, while atv encodes a nonfunctional version of the anthocyanin repressor SlMYBATV. The expression of SlAN2-like is responsive to light, and the functional SlAN2-like can activate the expression of both anthocyanin biosynthetic genes and their regulatory genes, suggesting that SlAN2-like acts as a master regulator in the activation of anthocyanin biosynthesis. We further showed that cultivated tomatoes contain nonfunctional alleles of SlAN2-like and therefore fail to produce anthocyanins. Consistently, expression of a functional SlAN2-like gene driven by the fruit-specific promoter in a tomato cultivar led to the activation of the entire anthocyanin biosynthesis pathway and high-level accumulation of anthocyanins in both the peel and flesh. 16Geographical characteristics of China’s wetlands derived from remotely sensed data显示文摘In this paper, we report the first wetland mapping of the entire China using Landsat enhanced thematic mapper plus (ETM+) data. These data were obtained from the Global Land Cover Facility at the University of Maryland spanning from 1999 to 2002. A total of 597 scenes of Landsat images were georeferenced and mosaiced. Manual image interpretation of satellite images was aided with elevation data, soil data, land cover/land use data and Google Earth. The minimum mapping unit is 10 pixel × 10 pixel, equivalent to 9 ha. The aim of our first round of mapping was only targeted at the boundary delineation of any type of wetland except those wetlands that are under agricultural use (i.e., paddy fields), which has already been well mapped by others. Our interpretation results indicate that a total of 359478 km2 of wetlands are of non-agricultural use. Among our preliminarily mapped wetland, 339353 km2 are inland wetland, 2786 km2 are non-agricultural artificial wetland, and 17609 km2 are coastal wetland. Because low-tide is rarely captured in satellite images, an under-estimation of coastal wetland is inevitable. We conducted some statistics based on our mapped wetlands and compared them with those previously obtained from a number of sources including a land cover/land use map made with satellite images during the late 1990s and early 2000s, a marshland map developed in approximately the same period, survey data of coastal wetland in early 1980s, and area data for approximately 400 larger patches of marshland in China compiled in 1996. Because some inconsistencies exist in the guidelines of those different wetland surveys, difference in area is expected. Some further comparison indicates that the wetland distributions derived from the preliminary wetland map are reasonable and more objective than other sources. The mapping process also indicated that the method adopted by us was efficient and cost-effective. We also found that in order to ensure comparability of the wetland maps developed at different times, a set of standard guidelines on the wetland categories to be mapped, and the mapping methods to be used must be well conceived, developed and effectively employed. 19Broadband tunable liquid crystal terahertz waveplates driven with porous graphene electrodes显示文摘Versatile devices,especially tunable ones,for terahertz imaging,sensing and high-speed communication,are in high demand.Liquid crystal based components are perfect candidates in the optical range;however,they encounter significant challenges in the terahertz band,particularly the lack of highly transparent electrodes and the drawbacks induced by a thick cell.Here,a strategy to overcome all these challenges is proposed:Few-layer porous graphene is employed as an electrode with a transmittance of more than 98%.A subwavelength metal wire grid is utilized as an integrated high-efficiency electrode and polarizer.The homogeneous alignment of a high-birefringence liquid crystal is implemented on both frail electrodes via a non-contact photo-alignment technique.A tunable terahertz waveplate is thus obtained.Its polarization evolution is directly demonstrated.Furthermore,quarter-wave plates that are electrically controllable over the entire testing range are achieved by stacking two cells.The proposed solution may pave a simple and bright road toward the development of various liquid crystal terahertz apparatuses.Lei Wang Xiao-Wen Lin Wei Hu Guang-Hao Shao Peng Chen Lan-Ju Liang Biao-Bing Jin Pei-Heng Wu Hao Qian Yi-Nong Lu Xiao Liang Zhi-Gang Zheng Yan-Qing Lu 2015Light(Science & Applications)2015,4,1:13
