| 1 | A two-step lineage reprogramming strategy to generate functionally competent human hepatocytes from fibroblasts显示文摘Terminally differentiated cells can be generated by lineage reprogramming,which is,however,hindered by incomplete conversion with residual initial cell identity and partial functionality. Here, we demonstrate a new reprogramming strategy by mimicking the natural regeneration route, which permits generating expandable hepatic progenitor cells and functionally competent human hepatocytes. Fibroblasts were first induced into human hepatic progenitor-like cells (hHPLCs), which could robustly expand in vitro and efficiently engraft in vivo. Moreover, hHPLCs could be efficiently induced into mature human hepatocytes (hiHeps) in vitro, whose molecular identity highly resembles primary human hepatocytes (PHHs). Most importantly, hiHeps could be generated in large quantity and were functionally competent to replace PHHs for drug-metabolism estimation, toxicity prediction and hepatitis B virus infection modeling. Our results highlight the advantages of the progenitor stage for successful lineage reprogramming. | 2 | GRAIN LENGTH AND AWN 1 negatively regulates grain size in rice显示文摘Grain size is an important factor determining yield in rice. Here, we identified a recessive mutant gene, grain length and awn 1 (gla1), which caused a significant increase in grain length and weight, and was associated with long awns. The gla1 mutation was mapped to a single-nucleotide polymorphism in a gene encoding a cytoplasmically-localized mitogen-activated protein kinase (MAPK) phosphatase. Overexpression of GLA1 caused a decrease in grain length, and the GLA1 protein interacted with OsMAPK6. | 3 | Less and shrunken pollen 1(LSP1) encodes a member of the ABC transporter family required for pollen wall development in rice(Oryza sativa L.)显示文摘Pollen fertility is an agronomic trait that strongly influences rice yield. Recent studies have revealed that the development of the pollen wall is required for pollen fertility and is regulated by several genes. However, the mechanisms underlying pollen and pollen wall development in rice remain largely unknown. In the present study, a point mutation in a gene on chromosome 1 was identified that resulted in the production of less and shrunken pollen(LSP) and led to defects in pollen wall formation. This gene was named LSP1 and was found to encode a member of the adenosine triphosphate-binding cassette(ABC)transporter G subfamily, OsABCG3. Two other loss-of-function mutants of LSP1/OsABCG3,generated using CRISPR/Cas9 technology, showed the same male sterile phenotype. The LSP1/OsABCG3 gene showed a spatio-temporal expression pattern in the developing anthers, and is an ortholog of the Arabidopsis genes At ABCG1 and At ABCG16, which play an important role in pollen wall development. Mutation of LSP1/OsABCG3 affected the expression of several genes involved in pollen and pollen wall formation. | 11 | Influence of Sn substitution for Co in RCo_2 (R= Gd,Tb,Dy) alloys on the structure and magnetocaloric effect显示文摘The phases and the magnetocaloric effect in the alloys R(Co1-xSnx)2 with x=0, 0.025, 0.050, 0.075, and 0.100 were investigated by X-ray diffraction analysis and magnetization measurement. The substitution of Sn in RCo2 is limited. The cubic MgCu2-type structure for the alloys of RCo2 was confirmed by X-ray powder diffraction and the remaining alloys mainly consisted of the RCo2 phase, along with some RCo3 and R5Sn3 impurity phases. The impurity phases increase with the increase of Sn content. The Tc of the alloys is not very sensitive to the Sn substitution for Dy(Co1-xSnx)2 and Tb(Co1-xSnx)2, whereas in Gd(Co1-xSnx)2, the Curie temperatures significantly increase. | 14 | Emerging Carbonyl Polymers as Sustainable Electrode Materials for Lithium-Free Metal-Ion Batteries显示文摘Lithium-ion batteries using inorganic electrode materials have been long demonstrated as the most promising power supplies for portable electronics,electric vehicles,and smart grids.However,the increasing cost and descending availability of lithium resources in combination with the limited electrochemical performance and eco-sustainability have created serious concerns with the competitiveness of lithium-ion batteries.There is a pressing need for the discovery of new redox chemistries between the alternative host materials and charge carriers.Organic nonlithium batteries using organic electrodes have recently attracted considerable interests as the future substitutes for energy storage systems,because of their combined merits(e.g.,natural abundance,rich chemistry of organics,rapid kinetics,and multielectron redox)of Li-free batteries and organic electrodes.Herein,an overview on the state-of-the-art developments of emerging carbonyl polymers for nonlithium metal-ion batteries is comprehensively presented with a primary focus on polyquinones and polyimides from the perspective of chain engineering.Six distinct categories,including monovalent(Na^(+),K^(+)) and multivalent(Mg^(2+),Zn^(2+),Ca^(2+),Al^(3+)) metal-ions batteries are individually outlined.Advantages of polymer electrode materials and characteristics of charge storage mechanisms are highlighted.Some key performance parameters such as specific capacity,rate capability,and cycle stability are carefully discussed.Moreover,aqueous nonlithium batteries based on carbonyl polymers are specially scrutinized due to the less reactivity of Li-free metals when exposed in aqueous electrolytes and ambient atmosphere.Current challenges and future prospects of developing polymer-based batteries are proposed finally.This review provides a fundamental guidance for the future advancement of next-generation sustainable batteries beyond lithium-ion batteries. | Qing Zhang Yu Dou Qiming He Shuyi Deng Qihua Huang Shaozhuan Huang Yingkui Yang | 2022 | Energy & Environmental Materials2022,5,4: | 0 |

Backbone parents play important roles in elite hybrid rice breeding systems. In this study, we performed pedigree-based analysis of the elite backbone parent rice variety, namely, Shuhui 527(SH527, Oryza sativa), to exploit key genome regions during breeding. Twenty-four cultivars(including SH527, its six progenitors and 17 derived cultivars) were collected and analyzed with high-density single nucleotide polymorphism(SNP) array. Scanning all these cultivars with genome-wide SNP data indicated the unique contributions of progenitors to the SH527 genome and identified the key genomic regions of SH527 conserved within all its derivatives. These findings were further supported by known rice yield-related genes or unknown QTLs identified by genome-wide association study.
