1Glycolysis reprogramming in cancer-associated fibroblasts promotes the growth of oral cancer through the lncRNA H19/miR-675-5p/PFKFB3 signaling pathway显示文摘As an important component of the tumor microenvironment,cancer-associated fibroblasts(CAFs)secrete energy metabolites to supply energy for tumor progression.Abnormal regulation of long noncoding RNAs(lncRNAs)is thought to contribute to glucose metabolism,but the role of lncRNAs in glycolysis in oral CAFs has not been systematically examined.In the present study,by using RNA sequencing and bioinformatics analysis,we analyzed the lncRNA/mRNA profiles of normal fibroblasts(NFs)derived from normal tissues and CAFs derived from patients with oral squamous cell carcinoma(OSCC).LncRNA H19 was identified as a key lncRNA in oral CAFs and was synchronously upregulated in both oral cancer cell lines and CAFs.Using small interfering RNA(siRNA)strategies,we determined that lncRNA H19 knockdown affected proliferation,migration,and glycolysis in oral CAFs.We found that knockdown of lncRNA H19 by siRNA suppressed the MAPK signaling pathway,6-phosphofructo-2-kinase/fructose-2,6-biphosphatase 3(PFKFB3)and miR-675-5p.Furthermore,the lncRNA H19/miR-675-5p/PFKFB3 axis was involved in promoting the glycolysis pathway in oral CAFs,as demonstrated by a luciferase reporter system assay and treatment with a miRNA-specific inhibitor.Our study presents a new way to understand glucose metabolism in oral CAFs,theoretically providing a novel biomarker for OSCC molecular diagnosis and a new target for antitumor therapy.Jin Yang Xueke Shi Miao Yang Jingjing Luo Qinghong Gao Xiangjian Wang Yang Wu Yuan Tian Fanglong Wu Hongmei Zhou 2021International Journal of Oral Science2021,13,1:8
2Coprinus comatus-derived nitrogen-containing biocarbon electrocatalyst with the addition of self-generating graphene-like support for superior oxygen reduction reaction显示文摘The development of nitrogen-rich biomassderived carbon catalysts provides an attractive perspective to substitute for Pt-based electrocatalysts for oxygen reduction reaction(ORR). We here report a facile strategy for synthesis of a nitrogen-doped biocarbon/graphene-like composite electrocatalyst by pyrolyzing a solid-state mixture of coprinus comatus biomass and melamine under nitrogen protection. The graphtic carbon nitride formed by polycondensation of melamine at 600 °C acts as a selfsacrificing template to generate the nitrogen-doped graphene-like sheet, which can function as an inserting agent and self-generating support. The composite catalyst exhibits the most promising catalytic activity towards the fourelectron ORR with a half-wave potential of around 0.83 V(vs. RHE), and more excellent stability and tolerance to methanol/ethanol compared to the commercial Pt/C catalyst. It is interestingly found that both a higher content of nitrogen and a larger ratio of graphitic-nitrogen species,which may derive from self-addition of graphene-like support into the catalyst, can effectively improve the electrocatalytic activity. The planar N group may be the nitrogen functionality that is most responsible for maintaining the ORR activity in alkaline medium. 3Three-dimensional silk fibroin scaffolds enhance the bone formation and angiogenic differentiation of human amniotic mesenchymal stem cells:a biocompatibility analysis显示文摘Silk fibroin(SF)is a fibrous protein with unique mechanical properties,adjustable biodegradation,and the potential to drive differentiation of mesenchymal stem cells(MSCs)along the osteogenic lineage,making SF a promising scaffold material for bone tissue engineering.In this study,hAMSCs were isolated by enzyme digestion and identified by multiple-lineage differentiation.SF scaffold was fabricated by freeze-drying,and the adhesion and proliferation abilities of hAMSCs on scaffolds were determined.Osteoblast differentiation and angiogenesis of hAMSCs on scaffolds were further evaluated,and histological staining of calvarial defects was performed to examine the cocultured scaffolds.We found that hAMSCs expressed the basic surface markers of MSCs.Collagen type I(COLI)expression was observed on scaffolds cocultured with hAMSCs.The scaffolds potentiated the proliferation of hAMSCs and increased the expression of COL-I in hAMSCs.The scaffolds also enhanced the alkaline phosphatase activity and bone mineralization,and upregulated the expressions of osteogeniorelated factors in vitro.The scaffolds also enhanced the an gioge nic differe ntiati on of hAMSCs.The cocultured scaffolds in creased bone formati on in treating critical calvarial defects in mice.This study first demonstrated that the application of 3D SF scaffolds co-cultured with hAMSCs greatly enhaneed osteogenic differentiation and angiogenesis of hAMSCs in vitro and in vivo.Thus,3D SF scaffolds cocultured with hAMSCs may be a better alter native for bone tissue engin eering.Yuwan Li Ziming Liu Yaping Tang Qinghong Fan Wei Feng Changqi Luo Guangming Dai Zhen Ge Jun Zhang Gang Zou Yi Liu Ning Hu Wei Huang 2020Acta Biochimica et Biophysica Sinica2020,52,6:2
4Fabrication of high-capacity cation-exchangers for protein adsorption: Comparison of grafting-from and grafting-to approaches显示文摘In this work, we have synthesized two polymer-grafted cation exchangers: one via the grafting-from approach, in which sulfopropyl methacrylate (SPM) is grafted through atom transfer radical polymerization onto Sepharose FF (the thus resulting exchanger is referred as Sep-g-SPM), and another via the grafting-to approach, in which the polymer of SPM is directly coupled onto Sepharose FF (the thus resulting exchanger is called as Sep-pSPM). Protein adsorption on these two cation exchangers have been also investigated. At the same ligand density, Sep-g-SPM has a larger accessible pore radius and a smaller depth of polymer layer than Sep-pSPM, due to the controllable introduction of polymer chains with the regular distribution of the ligand. Therefore, high-capacity adsorption of lysozyme and γ-globulin could be achieved simultaneously in Sep-g-SPM with an ionic capacity (IC) of 308 mmol·L^-1. However, Sep-pSPM has an irregular chain distribution and different architecture of polymer layer, which lead to more serious repulsive interaction to proteins, and thus Sep-pSPM has a lower adsorption capacity for γ-globulin than Sep-g-SPM with the similar IC. Moreover, the results from protein uptake experiments indicate that the facilitated transport of adsorbed γ-globulin occurs only in Sep-pSPM and depends on the architecture of polymer layers. 7The Effect of Martian Ionospheric Dispersion on SAR Imaging显示文摘When passing through the ionosphere,the high-frequency(HF)pulse signal of the Mars Exploration Radar is affected by the dispersion effect error,which results in signal attenuation and time delay and brings about a phase advance in such a way that the echo cannot be matched and filtered.In this paper,a high-order phase model is built to overcome the above problems and enable echo matching and filtering.Most studies on the dispersion effect approximate the additional phase after the effect,assuming that the ionosphere is a thin-layer structure.In this paper,an effective model for the HF waveband is constructed to analyze the change of signal propagation paths in the ionosphere.The additional phase is expanded in a Taylor series and retained these expansions as high-order terms to calculate the cumulative additional phase along the path.We show the rangeoffset variables of signal frequency,bandwidth,and electron density,simulate the effects of the ionosphere under different conditions,and conclude that the model can effectively estimate Mars without considering the effects of magnetic fields and anomalous solar activity and the effect of the ionosphere on synthetic aperture radar(SAR)echoes.The results obtained using ray tracing calculations are different from those obtained by simplifying assumptions,and we can simulate the Martian ionospheric effects by the former.Bo Wang Xijin Luo Qinghong Sheng Zhijun Yan 2022Space(Science & Technology)2022,,1:0
8A self-powered ultraviolet photodetector with van der Waals Schottky junction based on TiO_(2) nanorod arrays/Au-modulated V2CTx MXene显示文摘A self-powered ultraviolet photodetector(UV PD)with van der Waals(vdW)Schottky junction based on TiO_(2) nanorod arrays/Au-modulated V2 CTx MXene is reported.The Schottky junction enables the device to operate in self-powered mode.The dangling bond-free surface of V2 CTx MXene reduces the charge recombination at the junction interface.Meanwhile,V2 CTx MXene,with the work function(WF)increasing to 5.35 eV,forms a hole transport layer by contacting with Au electrode,which facilitates the carrier extraction.The electron lifetime in the device has prolonged to 8.95μs.As a result,the responsivity and detectivity of the PD have achieved 28 mA/W and 1.2×10^(11) cm Hz1/2/W(340 nm,65 mW/cm2,0 V),respectively.In addition,the presence of the Au electrode prevents the vanadium from coming into contact with oxygen and oxidizing,preserving the properties of the V2 CTx films.After 180 days of exposure to the atmosphere,the device performance remained at a particularly high level,indicating enhanced durability.This work points out an effective approach to modulate the properties of V2 CTx to obtain the high performance and stability of the UV PD.Guangcan Luo Ziling Zhang Yabing Wang Qun Deng Shengtao Pan Tengfei Wang Qinghong Li Kaixiang Liu Pengfei Kong Jing Zhang Shengyun Luo Hong Lin 2023Journal of Materials Science & Technology2023,,25:0
