1Clinically relevant factors associated with quantitative optical coherence tomography angiography metrics in deep capillary plexus in patients with diabetes显示文摘Background:To test clinically relevant factors associated with quantitative artifact-free deep capillary plexus(DCP)metrics in patients with diabetes mellitus(DM).Methods:563 eligible eyes(221 with no diabetic retinopathy[DR],135 with mild DR,130 with moderate DR,and 77 with severe DR)from 334 subjects underwent optical coherence tomography-angiography(OCT-A)with a swept-source OCT(Triton DRI-OCT,Topcon,Inc.,Tokyo,Japan).Strict criteria were applied to exclude from analysis those DCP images with artifacts and of poor quality,including projection artifacts,motion artifacts,blurriness,signal loss,B-scan segmentation error,or low-quality score.A customized MATLAB program was then used to quantify DCP morphology from the artifact-free DCP images by calculating three metrics:foveal avascular zone(FAZ),vessel density(VD),and fractal dimension(FD).Results:166(29.5%)eyes were excluded after quality control,leaving in the analysis 397 eyes(170 with no DR,101 with mild DR,90 with moderate DR,36 with severe DR)from 250 subjects.In the multiple regression models,larger FAZ area was associated with more severe DR(β=0.687;p=0.037),shorter axial length(AL)(β=−0.171;p=0.003),thinner subfoveal choroid thickness(β=−0.122;p=0.031),and lower body mass index(BMI)(β=−0.090;p=0.047).Lower VD was associated with more severe DR(β=−0.842;p=0.001),shorter AL(β=0.107;p=0.039),and poorer visual acuity(VA)(β=−0.133;p=0.021).Lower FD was associated with more severe DR(β=−0.891;p<0.001)and with older age(β=−0.142;p=0.004).Conclusions:Quantitative artifact-free DCP metrics are associated with VA,DR severity,AL,subfoveal choroidal thickness,age,and BMI in diabetic patients.The effects of ocular and systemic factors should be considered for meaningful interpretations of DCP changes in DM patients.Fang Yao Tang Erica O.Chan Zihan Sun Raymond Wong Jerry Lok Simon Szeto Jason C.Chan Alexander Lam Clement C.Tham Danny S.Ng Carol Y.Cheung 2020Eye and Vision2020,7,1:7
2Evolutionary Optimization: Pitfalls and Booby Traps显示文摘Evolutionary computation (EC), a collective name for a range of metaheuristic black-box optimization algorithms, is one of the fastest-growing areas in computer science. Many manuals and 'how-to's on the use of different EC methods as well as a variety of free or commercial software libraries are widely available nowadays. However, when one of these methods is applied to a real-world task, there can be many pitfalls and booby traps lurking - certain aspects of the optimization problem that may lead to unsatisfactory results even if the algorithm appears to be correctly implemented and executed. These include the convergence issues, ruggedness, deceptiveness, and neutrality in the fitness landscape, epistasis, non-separability, noise leading to the need for robustness, as well as dimensionality and scalability issues, among others. In this article, we systematically discuss these related hindrances and present some possible remedies. 3Whole-genome sequencing of monozygotic twins discordant for schizophrenia indicates multiple genetic risk factors for schizophrenia显示文摘Schizophrenia is a common disorder with a high heritability, but its genetic architecture is still elusive.We implemented whole-genome sequencing(WGS) analysis of 8 families with monozygotic(MZ) twin pairs discordant for schizophrenia to assess potential association of de novo mutations(DNMs) or inherited variants with susceptibility to schizophrenia. Eight non-synonymous DNMs(including one splicing site) were identified and shared by twins, which were either located in previously reported schizophrenia risk genes(p.V24689 I mutation in TTN, p.S2506 T mutation in GCN1L1, IVS3+1G > T in DOCK1) or had a benign to damaging effect according to in silico prediction analysis. By searching the inherited rare damaging or loss-of-function(LOF) variants and common susceptible alleles from three classes of schizophrenia candidate genes, we were able to distill genetic alterations in several schizophrenia risk genes, including GAD1, PLXNA2, RELN and FEZ1. Four inherited copy number variations(CNVs; including a large deletion at 16p13.11) implicated for schizophrenia were identified in four families, respectively. Most of families carried both missense DNMs and inherited risk variants, which might suggest that DNMs, inherited rare damaging variants and common risk alleles together conferred to schizophrenia susceptibility. 13MR-seq:measuring a single cell's transcriptome repeatedly by RNA-seq显示文摘We described a novel single-cell RNA-seq technique called MR-seq(measure a single-cell transcriptome repeatedly),which permits statistically assessing the technical variation and identifying the differentially expressed genes between just two single cells by measuring each single cell twice. We demonstrated that MR-seq gave sensitivity and reproducibility similar to the standard single-cell RNA-seq and increased the positive predicate value. Application of MR-seq to early mouse embryos identified hundreds of candidate intra-embryonic heterogeneous genes among mouse 2-,4-and 8-cell stage embryos. 14IN VIVO NEAR-INFRAREDFLUORESCENCEIMAGING OF HUMANCOLONADENOCARCINOMABY SPECIFIC IMMUNOTARGETINGOFA TUMOR-ASSOCIATEDMUCIN显示文摘Background and Aims: Accurate endoscopic detection of premalignant lesions and earlycancers in the colon is essential for cure, since prognosis is closely related to lesion size andstage. Although it has great clinical potential, autofluorescence endoscopy has limited tumorto-normal tissue image contrast for detecting small preneoplastic lesions. We have developed amolecularly specific, near-infrared fluorescent monoclonal antibody (CC49) bioconjugate whichtargets tumor-associated glycoprotein 72 (TAG72), as a contrast agent to improve fluorescencebased endoscopy of colon cancer. Methods: The fluorescent anti-TAG72 conjugate was evaluated in vitro and in vivo in athymic nude mice bearing human colon adenocarcinoma (LS174T)subcutaneous tumors. Autofluorescence, a fluorescent but irrelevant antibody and the free fluorescent dye served as controls. Fluorescent agents were injected intravenously, and in vivowhole body fluorescence imaging was performed at various time points to determine pharmacokinetics, followed by ex vivo tissue analysis by confocal fluorescence microscopy and histology Results: Fluorescence microscopy and histology confirmed specific LS174T cell membrane targeting of labeled CC49 in vitro and ex vivo. In vivo fluorescence imaging demonstrated significant tumor-to-normal tissue contrast enhancement with labeled-CC49 at three hours postinjection, with maximum contrast after 48 h. Accumulation of tumor fluorescence demonstratedthat modification of CC49 antibodies did not alter their specific tumor-localizing properties, andwas antibody-dependent since controls did not produce detectable tumor fluorescence. 17The Hong Kong consensus recommendations on the diagnosis and management of pancreatic cystic lesions显示文摘Background:The finding of pancreatic cystic lesions(PCL)on incidental imaging is becoming increasingly common.International studies report a prevalence of 2.2-44.7%depending on the population,imaging modality and indication for imaging,and the prevalence increases with age.Patients with PCL are at risk of developing pancreatic cancer,a disease with a poor prognosis.This publication summarizes recommendations for the diagnosis and management of PCL and post-operative pancreatic exocrine insufficiency(PEI)from a group of local specialists.Methods:Clinical evidence was consolidated from narrative reviews and consensus statements formulated during two online meetings in March 2022.The expert panel included gastroenterologists,hepatobiliary surgeons,oncologists,radiologists,and endocrinologists.Results:Patients with PCL require careful investigation and follow-up due to the risk of malignant transformation of these lesions.They should undergo clinical investigation and pancreas-specific imaging to classify lesions and understand the risk profile of the patient.Where indicated,patients should undergo pancreatectomy to excise PCL.Following pancreatectomy,patients are at risk of PEI,leading to gastrointestinal dysfunction and malnutrition.Therefore,such patients should be monitored for symptoms of PEI,and promptly treated with pancreatic enzyme replacement therapy(PERT).Patients with poor response to PERT may require increases in dose,addition of a proton pump inhibitor,and/or further investigation,including tests for pancreatic function.Patients are also at risk of new-onset diabetes mellitus after pancreatectomy;they should be screened and treated with insulin if indicated.Conclusions:These statements are an accurate summary of our approach to the diagnosis and management of patients with PCL and will be of assistance to clinicians treating these patients in a similar clinical landscape.Tan-To Cheung Yuk Tong Lee Raymond Shing-Yan Tang Wong Hoi She Kai Chi Cheng Chin Cheung Cheung Keith Wan Hang Chiu Kenneth Siu Ho Chok Wing Sun Chow Tak Wing Lai Wai-Kay Seto Thomas Yau 2023Hepatobiliary Surgery and Nutrition2023,12,5:0
18Low-temperature caproate production,microbial diversity,and metabolic pathway in xylose anaerobic fermentation显示文摘Mixed culture fermentation(MCF)is challenged by the unqualified activity of enriched bacteria and unwanted methane dissolution under low temperatures.In this work,caproate production from xylose was investigated by MCF at a low temperature(20°C).The results showed that a 9 d long hydraulic retention time(HRT)in a continuously stirred tank reactor was necessary for caproate production(~0.3 g/L,equal to 0.6 g COD/L)from xylose(10 g/L).The caproate concentration in the batch mode was further increased to 1.6 g/L.However,changing the substrate to ethanol did not promote caproate production,resulting in~1.0 g/L after 45 d of operation.Four genera,Bifidobacterium,Caproiciproducens,Actinomyces,and Clostridium_sensu_stricto_12,were identified as the enriched caproate-producing bacteria.The enzymes in the fatty acid biosynthesis(FAB)pathway for caproate production were identified via metagenomic analysis.The enzymes for the conversion of(C_(n+2))-2,3-Dehydroxyacyl-CoA to(C_(n+2))-Acyl-CoA(i.e.,EC 1.3.1.8 and EC 1.3.1.38)in the reverseβ-oxidation(RBO)pathway were not identified.These results could extend the understanding of low-temperature caproate production.Qingting Wang Kun Dai Jie Tang Sidi Hong Sijie Zheng Ting Sun Raymond Jianxiong Zeng Fang Zhang 2023Frontiers of Environmental Science & Engineering2023,17,3:0
