2Growth-inhibiting effects of taxol on human liver cancer in vitro and in nude mice显示文摘AIM To investigate the effects of taxol onSMMC-7721 human hepatoma and itsmechanisms.METHODS In vitro cell growth was assessedby trypan blue exclusion method.Experimentalhepatoma model was established by seedingSMMC-7721 cells subcutaneously into Balb/c(nu/nu)nude mice.In vivo tumor growth wasdetermined by measurement of tumor diameterwith Vernier calipers.The syntheses of DNA,RNA and protein were analyzed by incorporationof ~3H-thymidine,~3H-uridine and ~3H-leucinerespectively.Using light and electronmicroscopes to observe the morphologicalchanges of cells including mitosis andapoptosis.RESULTS Taxol was effective against SMMC-7721 human hepatoma cell growth in the rangesof 2.5 nmol/L-10 nmol/L with mitotic arrestand apoptosis in vitro.DNA,RNA and proteinsyntheses in cells were also obviouslysuppressed by in vitro treatment of taxol for72 h.Taxol at 2.5 nmol/L reduced ~3H-thymidineuptake to about 34% of the control value(P<0.05).Increasing the dose of taxol to20 nmol/L resulted in a greater decrease in ~3H-thymidine incorporation to 60% of the controlvalue(P<0.01).At a concentration of 20 nmol/L,the ~3H-uridine and ~3H-leucine uptakeswere reduced to 52%(P<0.05)and 63%(P<0.01),respectively.In vivo,taxolsignificantly inhibited SMMC-7721 tumor growthat 10 mg/kg,i.p.,once daily for 10 d.A morethan 90% decrease in tumor volume wasobserved by day 11(P<0.01)similarly withmitotic arrest and cell apoptosis.CONCLUSION Taxol has a marked anticanceractivity in SMMC-7721 human hepatoma both invitro and in nude mice.Its mechanisms might beassociated with mitotic arrest,subsequently,apoptosis of the hepatoma cells.No obvioustoxicity was observed with in vivoadministration of taxoi.Jin Hui Yuan Ru Ping Zhang Ru Gang Zhang Li Xia Guo Xing Wang Wang Dan Luo Yong Xie Hong Xie 2000World Journal of Gastroenterology2000,6,2:18
3Antibodies against the C-terminal peptide of rabbit oviductin inhibit mouse early embryo development to pass 2-cell stage显示文摘A full-length rabbit oviductin cDNA(1909bp) was cloned. It consists of a 5’-UTR of 52bp, an open reading frame (ORF) of 1374bp and a 3’-UTR of 483bp and has more than 80% homology with that of other mammal oviductins. N-terminal peptide (NTP) (384 residues) and C-terminal peptide (CTP) (73 residues) of deduced protein precursor has about 80% and 50% identity with that of other mammals respectively. Fusion proteins GST-NTP 368(1R-368N)and GST-CTP73 (369F-441A) were expressed and purified. NH2-terminal of CTP sequencing reveals that the purified protein is consistent with the deduced one. In order to study the function of NTP and CTP the mouse anti-NTP and rabbit anti-CTP antisera were prepared. Tissue-specific (skeleton muscle, oviduct, uterus, ovary, liver, heart and brain) analysis indicated that rabbit oviductin was only found in oviduct. The conditioned medium derived from the rabbit oviduct mucosa epithelial cells has a function of overcoming the early embryonic development block of Kunming mous e cultured in vitro. Anti-CTP antiserum could totally inhibit the early embryo development at 2-cell stage cultured in the conditioned culture medium, but anti-NTP antiserum couldn’t. There was a positive relationship between the ratio of early embryos at development block and the dosage of anti-CTP antiserum added in the conditioned culture medium. 4Generation of electricity from CO_2 mineralization: Principle and realization显示文摘Current CO2 reduction and utilization technologies suffer from high energy consuming. Thus, an energy favourable route is in urgent demanding. CO2 mineralization is theoretically an energy releasing process for CO2 reduction and utilization, but an approach to recovery this energy has so far remained elusive. For the first time, here we proposed the principle of harvesting electrical energy directly from CO2 mineralization, and realized an energy output strategy for CO2 utilization and reduction via a CO2-mineralization fuel cell(CMFC) system. In this system CO2 and industrial alkaline wastes were used as feedstock, and industrial valuable Na HCO3 was produced concomitantly during the electricity generation. The highest power density of this system reached 5.5 W/m2, higher than many microbial fuel cells. The maximum open circuit voltage reached 0.452 V. Moreover, this system was demonstrated viable to low concentration CO2(10%) and other carbonation process. 5Effects of cisplatin on telomerase activity and telomere length in BEL-7404 human hepatoma cells显示文摘Telomerase activity was inhibited in a dose and time-dependent manner with the treatment of cisplatin for 24, 48, or 72 h in a concentration ranged from 0.8 to 50 1uM in BEL-7404 human hepatoma cells. There were no changes in expression pattern of three telomerase subunits, its catalytic reverse transcriptase subunit (hTERT), its RNA component (hTR) or the associated protein subunit (TP1), after cisplatin treated for 72 h with indicated concentrations. Mean telomere lengths were decreased by the cisplatin treatment. 7Optimal Waist Circumference Cut-off values for Identifying Metabolic Risk Factors in Middle-aged and Elderly Subjects in Shandong Province of China显示文摘Objective To study the optimal waist circumference(WC) cut-off values for identifying metabolic risk factors in middle-aged and elderly subjects in Shandong Province of China. Methods A total of 2 873 men and 5 559 women were included in this cross-sectional study. Metabolic syndrome(MetS) was diagnosed according to the definition of Chinese Diabetes Society in 2004. The relation between WC and MetS was analyzed by multivariate logistic regression analysis. The optimal WC cut-off values were identified using the area under the ROC curve and the different diagnostic criteria for central obesity were compared. Results The WC was the risk factor for MetS independent of BMI, blood glucose, blood lipid, and blood pressure. The optimal WC cut-off value was 83.8 cm and 91.1 cm for identifying MetS in women and men, respectively. Compared with 80 cm and 85 cm for women and men, 85 cm and 90 cm had a higher Youden index for identifying all metabolic risk factors and MetS in women and men. 8Effect of long-term fertilization on soil nitrate distribution显示文摘A thirteen years long term field fertilizer experiment was conducted to monitor the effect of different fertilization on soil nitrate distribution. The results showed: (1) Applying relative excessive N fertilizer could result large quantities of NO - 3 residue and NO - 3 movement downward in soil profiles; amending phosphate fertilizer or organic manure with nitrogen fertilizer together could significantly improve the status of NO - 3 leaching downward due to the balanced uptake of nutrients by crops. (2) Appropriate amounts of nitrogen fertilizer which was equal or smaller than the optimal fertilization rate could not result in more NO - 3 leaching in Northern China. 11Serum Fetuin-A Levels and Thyroid Function in Middle-aged and Elderly Chinese显示文摘Serum fetuin-A levels are reportedly elevated in hyperthyroidism. However, there are few relevant epidemiologic studies. We conducted a cross-sectional study in Songnan community, China in 2009 to investigate the association between serum fetuin-A concentrations and thyroid function. A total of 2,984 participants aged 40 years and older were analyzed. Multivariable linear regression analysis revealed that serum fetuin-A concentrations were positively associated with log(free triiodothyronine) and were inversely associated with log(thyroid peroxidase antibody) after adjustment(both P < 0.05). Compared with the participants in the lowest tertile of free triiodothyronine and free thyroxine level, those in the highest tertile had higher fetuin-A concentrations. 14Efficacy of Moxifloxacin against Mycobacterium abscessus in Zebrafish Model in vivo显示文摘Objective Moxifloxacin(MFX)shows good in vitro activity against Mycobacterium abscessus and can be a possible antibiotic therapy to treat M.abscessus infection;however,other studies have shown a lower or no activity.We aimed to evaluate MFX activity against M.abscessus using zebrafish(ZF)model in vivo.Methods A formulation of M.abscessus labeled with CM-Dil was micro-injected into ZF.Survival curves were determined by recording dead ZF every day.ZF were lysed,and colony-forming units(CFUs)were enumerated.Bacteria dissemination and fluorescence intensity in ZF were analyzed.Inhibition rates of MFX and azithromycin(AZM,positive control)were determined and compared.Results Significantly increased survival rate was observed with different AZM concentrations.However,increasing MFX concentration did not result in a significant decrease in ZF survival curve.No significant differences in bacterial burdens by CFU loads were observed between AZM and MFX groups at various concentrations.Bacterial fluorescence intensity in ZF was significantly correlated with AZM concentration.However,with increasing MFX concentration,fluorescence intensity decreased slightly when observed under fluorescence microscope.Transferring rates at various concentrations were comparable between the MFX and AZM groups,with no significant difference.Conclusion MFX showed limited efficacy against M.abscessus in vivo using ZF model.Its activity in vivo needs to be confirmed.NIE Wen Juan XIE Zhong Yao GAO Shan TENG Tian Lu ZHOU Wen Qiang SHANG Yuan Yuan JING Wei SHI Wen Hui WANG Qing Feng HUANG Xue Rui CAI Bao Yun WANG Jun WANG Jing GUO Ru GE Qi Ping NIE Li Hui HAN Xi Qin DU Ya Dong CHU Nai Hui 2020Biomedical and Environmental Sciences2020,33,5:1

17The E3 ubiquitin ligase SINA1 and the protein kinase BIN2 cooperatively regulate PHR1 in apple anthocyanin biosynthesis显示文摘PHR1(PHOSPHATE STARVATION RESPONSE1)plays key roles in the inorganic phosphate(Pi)starvation response and in Pi deficiency-induced anthocyanin biosynthesis in plants. However, the post-translational regulation of PHR1 is unclear,and the molecular basis of PHR1-mediated anthocyanin biosynthesis remains elusive. In this study, we determined that MdPHR1 was essential for Pi deficiency-induced anthocyanin accumulation in apple(Malus × domestica). MdPHR1 interacted with MdWRKY75, a positive regulator of anthocyanin biosynthesis, to enhance the MdWRKY75-activated transcription of MdMYB1,leading to anthocyanin accumulation. In addition,the E3 ubiquitin ligase SEVEN IN ABSENTIA1(MdSINA1) negatively regulated MdPHR1-promoted anthocyanin biosynthesis via the ubiquitination-mediated degradation of MdPHR1.Moreover, the protein kinase apple BRASSINOSTEROID INSENSITIVE2(MdBIN2) phosphorylated MdPHR1 and positively regulated MdPHR1-mediated anthocyanin accumulation by attenuating the MdSINA1-mediated ubiquitination degradation of MdPHR1. 18The Association between LINC00511 Variants and Breast Cancer Susceptibility among the Han Chinese Population显示文摘Objective LINC00511 has been considered an oncogene for breast cancer,however,the association between LINC00511 Single nucleotide polymorphisms(SNPs)and breast cancer risk remains unclear.Methods A total of 504 breast cancer patients and 505 cancer-free controls were genotyped for 12 LINC00511 SNPs.A logistic regression analysis was conducted to explore the association between LINC00511 SNPs and breast cancer susceptibility,adjusting for reproductive factors.A haplotype analysis and interaction analysis were applied to assess the role of LINC00511 in breast cancer.Results LINC00511 SNPs were significantly correlated with breast cancer.The TT genotype of rs11657109 reduced the risk of breast cancer(OR:0.454,95%CI:0.212-0.970),while the AG and GG genotypes of rs17780195 played a protective role(OR:0.723,95%CI:0.539-0.970;OR:0.398,95%CI:0.184-0.860,respectively).The mutation of rs9906859 may also lead to a reduction in breast cancer susceptibility(OR:0.682,95%CI:0.520-0.894).Furthermore,LINC00511 SNPs(rs4432291,rs1558535,rs17780195 and rs9906859)are associated with the HER-2 status(P<0.05 for all).The interaction of rs9906859,breastfeeding and age increased the breast cancer risk by 4.318(OR:5.318,95%CI:2.214-12.774).Conclusions LINC00511 SNPs were associated with the risk of breast cancer.There was an association between LINC00511 SNPs and the HER-2 status.There were interactions between LINC00511 SNPs and reproductive factors that affected the risk of breast cancer.Fei Fei Chong Jing Jing Cao Yan Li Wang Qiu Yu Sun Meng Meng Song Xiao Ru Jiang Kai Juan Wang Lin Ping Xu Chun Hua Song 2020Journal of Nutritional Oncology2020,5,2:0

20Improving photosensitization for photochemical C02-to-CO conversion显示文摘Inspired by nature,improving photosensitization represents a vital direction for the development of artificial photosynthesis.The sensitization ability of photosensitizers(PSs)reflects in their electron-transfer ability,which highly depends on their excited-state lifetime and redox potential.Herein,for the first time,we put forward a facile strategy to improve sensitizing ability via finely tuning the excited state of Ru(Ⅱ)-PSs(Ru-1-Ru-4)for efficient CO2 reduction.Remarkably,[Ru(Phen)2(3-pyrenyl Phen)]^2+(Ru-3)exhibits the best sensitizing ability among Ru-1-Ru-4,over 17 times higher than that of typical Ru(Phen)3^2+.It can eficiently sensitize a dinuclear cobalt catalyst for CO2-to-CO conversion with a maximum turnover number of 66480.Systematic investigations demonstrate that its long-lived excited state and suitable redox driving force greatly contributed to this superior sensitizing ability.This work provides a newinsight into dramatically boosting photocatalytic CO2 reduction via improving photosensitization.Ping Wang Ru Dong Song Guo Jianzhang Zhao Zhi-Ming Zhang Tong-Bu Lu 2020National Science Review2020,7,9:0
