| 1 | Development of a multiplex one-step real-time RT-PCR assay for the simultaneous detection of eight viruses associated with febrile rash illnesses显示文摘Fever and rash illnesses(FRIs)are a series of common diseaseswith fever and rashes as clinicalmanifestations,most of which are caused by viral infection.The rashes of FRIs are generally nonspecific;therefore it is difficult to identify FRIassociated viruses solely based on clinical symptoms.To achieve rapid and accurate identification of FRI pathogens,a multiplex one-step real-time reverse transcription-polymerase chain reaction(RT-PCR)assay was developed and evaluated in this study.Primers and probes were selected for the detection of measles virus(MeV),rubella virus(RV),human enterovirus(EV),varicella-zoster virus(VZV),dengue virus(DENV),human parvovirus B19(B19),Epstein-Barr virus(EBV),and human herpes virus 6(HHV-6),which cover the most common pathogenic viruses of FRIs.Detection of the eight FRI-associated viruses,which was divided into two groups/tubes,was simultaneously performed under universal optimized reaction conditions in multiplex one-step real-time RT-PCR assay.The multiplex realtime RT-PCR showed high sensitivity and specificity in detecting the eight FRI-associated viruses.The limits of detection(LODs)for the eight viruses were in the range of 47–177 copies/reaction,and no cross reactions for the eight FRIassociated viruses were found in the multiplex assay.In addition,the results of the multiplex real-time RT-PCR assay were consistent with the results of a monoplex real-time RT-PCR assay and sequencing for clinical specimens obtained from FRI patients.With its advantages of high efficiency and rapid and accurate diagnosis,multiplex real-time RT-PCR was very feasible for the early diagnosis of FRI pathogenic viruses and would be of great help for the proper treatment,monitoring,and initiation of preventive measures for FRI cases. | Aili Cui Shulei Wang Qiang Zhang Huiling Wang Zhen Zhu Aqian Li Qinqin Song Yanzhe Hao Jilan He Wenbo Xu Yan Zhang | 2020 | Biosafety and Health2020,2,2: | 5 |
| 2 | Mechanical properties of irradiated multi-phase polycrystalline BCC materials显示文摘Structure materials under severe irradiations in nuclear environments are known to degrade because of irradiation hardening and loss of ductility,resulting from irradiation-induced defects such as vacancies,interstitials and dislocation loops,etc.In this paper,we develop an elastic-viscoplastic model for irradiated multi-phase polycrystalline BCC materials in which the mechanical behaviors of individual grains and polycrystalline aggregates are both explored.At the microscopic grain scale,we use the internal variable model and propose a new tensorial damage descriptor to represent the geometry character of the defect loop,which facilitates the analysis of the defect loop evolutions and dislocation-defect interactions.At the macroscopic polycrystal scale,the self-consistent scheme is extended to consider the multiphase problem and used to bridge the individual grain behavior to polycrystal properties.Based on the proposed model,we found that the work-hardening coefficient decreases with the increase of irradiation-induced defect loops,and the orientation/loading dependence of mechanical properties is mainly attributed to the different Schmid factors.At the polycrystalline scale,numerical results for pure Fe match well with the irradiation experiment data.The model is further extended to predict the hardening effect of dispersoids in oxide-dispersed strengthened steels by the considering the Orowan bowing.The influences of grain size and irradiation are found to compete to dominate the strengthening behaviors of materials. | Dingkun Song Xiazi Xiao Jianming Xue Haijian Chu Huiling Duan | 2015 | Acta Mechanica Sinica2015,31,2: | 4 |

| 7 | Maize ZmVPP5 is a truncated Vacuole H^+-PPase that confers hypersensitivity to salt stress显示文摘In plants, Vacuole H^+-PPases(VPPs) are important Aproton pumps and encoded by multiple genes. In addition to full-length VPPs, several truncated forms are expressed, but their biological functions are unknown. In this study, we functionally characterized maize vacuole H^+-PPase 5(ZmVPP5), a truncated VPP in the maize genome. Although ZmVPP5 shares high sequence similarity with ZmVPP1 ZmVPP5 lacks the complete structure of the conserved proton transport and the inorganic pyrophosphatase-related domain. Phylogenetic analysis suggests that ZmVPP5 might be derived from an incomplete gene duplication event. ZmVPP5 is expressed in multiple tissues, and ZmVPP5 was detected in the plasma membrane, vacuole membrane and nuclei of maize cells. The overexpression of ZmVPP5 in yeast cells caused a hypersensitivity to salt stress. Transgenic maize lines with overexpressed ZmVPP5 also exhibited the salt hypersensitivity phenotype. A yeast two-hybrid analysis identified the ZmBag6 like protein as a putative ZmVPP5-interacting protein. The results of bimolecular luminescence complementation(Bi LC)assay suggest an interaction between ZmBag6-like protein and ZmVPP5 in vivo. Overall, this study suggests that ZmVPP5 might act as a VPP antagonist and participate in the cellular response to salt stress. | 8 | Ensemble Mean Forecast Skill and Applications with the T213 Ensemble Prediction System显示文摘Ensemble forecasting has become the prevailing method in current operational weather forecasting. Although ensemble mean forecast skill has been studied for many ensemble prediction systems(EPSs) and different cases, theoretical analysis regarding ensemble mean forecast skill has rarely been investigated, especially quantitative analysis without any assumptions of ensemble members. This paper investigates fundamental questions about the ensemble mean, such as the advantage of the ensemble mean over individual members, the potential skill of the ensemble mean, and the skill gain of the ensemble mean with increasing ensemble size. The average error coefficient between each pair of ensemble members is the most important factor in ensemble mean forecast skill, which determines the mean-square error of ensemble mean forecasts and the skill gain with increasing ensemble size. More members are useful if the errors of the members have lower correlations with each other, and vice versa. The theoretical investigation in this study is verified by application with the T213 EPS. | 10 | Luminescence,Concentration Quenching and Thermal Stability of White Emitting Phosphor Ba_2Ca(BO_3)_2:Dy^(3+)显示文摘A white emitting phosphor Ba2Ca(BO3)2:Dy3+ was synthesized via a high temperature solid state reaction at 1000℃ for 5 h. The luminescence, mole fraction quenching and thermal stability of Ba2Ca(BO3)2:Dy3+ were investigated. According to the phase composition analyzed by X-ray powder diffraction, there is no crystalline phase except Ba2Ca(BO3)2 in the sample. Ba2Ca(BO3)2:Dy3+ can produce white emission under 348 nm excitation. The emission intensities of Ba2Ca(BO3)2:Dy3+ are affected by Dy3+ concentration. The concentration quenching effect was analyzed, and the concentration quenching mechanism was verified as dipole-dipole interaction. The critical distance(R c) obtained based on the crystal structure data is 2.911 nm. At 150℃, the emission intensity of Ba2Ca(BO3)2:Dy3+ is 68.0% of the initial value at room temperature. The activation energy for the thermal quenching calculated is 0.202 e V. | 11 | The efficacy and tolerability of lamotrigine adjunctive/monotherapy in patients with partial seizures refractory to poly-AEDs显示文摘Objective:This study was designed as an open-label trial to assess the effects of changing the antiepileptic drugs(AEDs) regi-men to lamotrigine(LTG) as adjunctive/monotherapy in patients with partial seizures who were dissatisfied with their drug regimen because of intractable seizures.Methods:The patients were recruited from multicenters using the following criteria:age≥18 years;at least 3 seizures per month during the last 16 weeks;previous use of at least 3 AEDs.The study involved a baseline phase and 2 experimental phases:LTG was first added to the regimen,and then patients could gradually change to LTG monotherapy if their seizures were reduced by at least 50 percent/month.Tolerability,the primary end point,was assessed using the Liverpool Adverse Experience Profile(LAEP).Secondary end points included quality of life,as measured with the Quality of Life in Epilepsy-31 inventory.Reductions in seizures from baseline throughout each phase were also analyzed.Results:One hundred and fourteen patients aged between 18 and 52 years(age 27.8±13.2 years;71 men and 43 women) were enrolled.After adding LTG,105 patients(92.11%) completed adjunctive therapy.Upon completion of the adjunctive phase,mean improvement from baseline was 2.6 points on the LAEP(p=0.037).The overall score on the QOLIE-31 improved by 8.49 points from baseline(p=0.023).At the end of the trial,26(22.81%) of patients completed LTG monotherapy,and 65 patients(57.02%) experienced at least 50% reduction in seizure frequency compared to baseline,The mean improvement from baseline was 5.1 points on the LAEP(p=0.0059),and the overall score on the QOLIE-31 score improved by 12.72 points from baseline(p=0.0071).Twenty-two(19.30%) patients reported adverse effects and 9 patients discontinued participation in the trial because of adverse effects.Conclusion:For patients with partial seizures who were dissatisfied with their AED regimen because of intractable seizures,adding LTG to the drug regimen was well tolerated and effective in improving the quality of life and controlling seizures.Furthermore,switching to LTG monotherapy was associated with further improvement. | Chunjie Song Huiling Chen XiaoyongWang Hui Wang Qi Wan | 2009 | Journal of Nanjing Medical University2009,23,5: | 1 |
| 12 | DDR1 promotes LoVo cell proliferation by regulating energy metabolism显示文摘Cellular energy metabolism dysregulation is associated with colorectal cancer(CRC)development and progression.Discoidin domain receptor 1a(DDR1a),one of the five DDR1 isoforms,is closely related to cell proliferation,invasion,and apoptosis in various tumors.Whether it participates in cellular metabolic reprogramming and regulates CRC initiation and progression remains unclear.In this study,we compared the expression of DDR1 in CRC tissues and adjacent tissues from 126 postoperative CRC samples.Moreover,lentivirus-mediated DDR1a overexpression and knockdown were performed in LoVo cells,and cell viability and proliferation were determined by CCK-8 and BrdU assays,respectively.Oxygen consumption rate,extracellular acidification rate,and lactate production were used to determine the effect of DDR1a on metabolic reprogramming.Clinically,CRC patients with high DDR1 expression had poor differentiation and were at an advanced TNM stage.DDR1a promoted LoVo cell proliferation,mitochondrial function,and extracellular acidification.Moreover,DDR1a knockdown inhibited intracellular lactic acid production in LoVo cells,while a pyruvate kinase inhibitor(diamide,200μM)significantly reversed this progression.Taken together,our results reveal that DDR1 plays a crucial role in maintaining intracellular environment homeostasis through metabolic reprogramming. | Bin Xiong Zehui Xie Feixue Song Huiling Chen Xiaojuan Wang Zhengxu Jin Tiyun Han Yi Li Dekui Zhang | 2022 | Acta Biochimica et Biophysica Sinica2022,54,5: | 1 |

| 19 | Kindlin-2 regulates skeletal homeostasis by modulating PTH1R in mice显示文摘In vertebrates,the type 1 parathyroid hormone receptor(PTH1R)is a critical regulator of skeletal development and homeostasis;however,how it is modulated is incompletely understood.Here we report that deleting Kindlin-2 in osteoblastic cells using the mouse 10-kb Dmp1-Cre largely neutralizes the intermittent PTH-stimulated increasing of bone volume fraction and bone mineral density by impairing both osteoblast and osteoclast formation in murine adult bone.Single-cell profiling reveals that Kindlin-2 loss increases the proportion of osteoblasts,but not mesenchymal stem cells,chondrocytes and fibroblasts,in non-hematopoietic bone marrow cells,with concomitant depletion of osteoblasts on the bone surfaces,especially those stimulated by PTH.Furthermore,haploinsufficiency of Kindlin-2 and Pth1r genes,but not that of either gene,in mice significantly decreases basal and,to a larger extent,PTH-stimulated bone mass,supporting the notion that both factors function in the same genetic pathway.Mechanistically,Kindlin-2 interacts with the C-terminal cytoplasmic domain of PTH1R via aa 474–475 and Gsα.Kindlin-2 loss suppresses PTH induction of cAMP production and CREB phosphorylation in cultured osteoblasts and in bone.Interestingly,PTH promotes Kindlin-2 expression in vitro and in vivo,thus creating a positive feedback regulatory loop.Finally,estrogen deficiency induced by ovariectomy drastically decreases expression of Kindlin-2 protein in osteocytes embedded in the bone matrix and Kindlin-2 loss essentially abolishes the PTH anabolic activity in bone in ovariectomized mice.Thus,we demonstrate that Kindlin-2 functions as an intrinsic component of the PTH1R signaling pathway in osteoblastic cells to regulate bone mass accrual and homeostasis. | Xuekun Fu Bo Zhou Qinnan Yan Chu Tao Lei Qin Xiaohao Wu Sixiong Lin Sheng Chen Yumei Lai Xuenong Zou Zengwu Shao Meiqing Wang Di Chen Wenfei Jin Youqiang Song Huiling Cao Ge Zhang Guozhi Xiao | 2020 | Signal Transduction and Targeted Therapy2020,5,1: | 0 |
