| 1 | Gene Cloning and Expression of the Pyrroline-5-carboxylate Reductase Gene of Perennial Ryegrass(Lolium perenne)显示文摘Salt and drought limit the range of applications of perennial ryegrass(Lolium perenne L.), which is one of the important turf and forage grasses. Previous studies have suggested that pyrroline-5-carboxylate reductase(P5CR) might play a central role in proline accumulation in plants that are responsive to stresses. In the present study, the Lolium perenne L. pyrroline-5-carboxylate reductase(LpP 5CR) gene was cloned from leaves of the cultivar ‘Derby' using the RACE technique. The full-length LpP 5CR gene was 1 047 bp in length, which comprised an open reading frame(ORF) of 840 bp in size. Sequence alignment revealed that the putative Lp P5 CR had a 94.3% similarity to Ta P5 CR. q RT-PCR displayed that the mR NA levels of the LpP 5CR gene were almost the same as that in the roots, stems, and leaves of perennial ryegrass seedlings subjected to normal condition or NaC l treatment for 1 h. Moreover, the transcription level of LpP 5CR was up- or down- regulated with Na Cl, polyethylene glycol(PEG), cold, or abscisic acid(ABA) treatment for 3 to 48 h. In addition, confocal microscopy localized the GFP-Lp P5 CR fusion protein to the cytoplasm of onion epidermal cells. | 2 | Stochastic Petri Net Based Modeling of Emergency Medical Rescue Processes During Earthquakes显示文摘The post-disaster emergency medical rescue(EMR) is critical for people’s lives. This paper presents a stochastic Petri net(SPN) model based on the process of the rescue structure and a Markov chain model(MC), which is applied to the optimization of the EMR process, with the aim of identifying the key activities of EMR. An isomorphic MC model is developed for measuring and evaluating the time performance of the EMR process during earthquakes with the data of the 2008 Wenchuan earthquake.This paper provides a mathematical approach to simulate the process and to evaluate the efficiency of EMR. Simultaneously, the expressions of the steady state probabilities of this system under various states are obtained based on the MC, and the variations of the probabilities are analyzed by changing the firing rates for every transition. Based on the concrete data of the event, the authors find the most time consuming and critical activities for EMR decisions. | 5 | Biodegradable magnesium alloy with eddy thermal effect for effective and accurate magnetic hyperthermia ablation of tumors显示文摘Magnetic hyperthermia therapy(MHT)is able to ablate tumors using an alternating magnetic field(AMF)to heat up magnetocaloric agents(e.g.magnetic nanoparticles)administered into the tumors.For clinical applications,there is still a demand to find new magnetocaloric agents with strong AMF-induced heating performance and excellent biocompatibility.As a kind of biocompatible and biodegradable material,magnesium(Mg)and its alloys have been extensively used in the clinic as an implant metal.Herein,we discovered that the eddy thermal effect of the magnesium alloy(MgA)could be employed for MHT to effectively ablate tumors.Under low-field-intensity AMFs,Mg A rods could be rapidly heated,resulting in a temperature increase in nearby tissues.Such AMF-induced eddy thermal heating of MgA could not only be used to kill tumor cells in vitro,but also be employed for effective and accurate ablation of tumors in vivo.In addition to killing tumors in mice,we further demonstrated that VX2 tumors of much larger sizes growing in rabbits after implantation of Mg A rods could also be eliminated after exposure to an AMF,illustrating the ability of MgA-based MHT to kill large-sized tumors.Moreover,the implanted MgA rods showed excellent biocompatibility and~20%of their mass was degraded within three months.Our work thus discovered for the first time that non-magnetic biodegradable Mg A,an extensively used implant metal in clinic,could be used for effective magnetic thermal ablation of tumors under a low-field-intensity AMF.Such a strategy could be readily translated into clinical use. | Nailin Yang Fei Gong Liang Cheng Huali Lei Wei Li Zongbin Sun Caifang Ni Zhanhui Wang Zhuang Liu | 2021 | National Science Review2021,8,1: | 1 |

| 13 | Tissue distribution of Lycium barbarum polysaccharides in rat tissue by fluorescein isothiocyanate labeling显示文摘To date, in vivo investigations of polysaccharide’s pharmacokinetics are significantly restricted by the difficulty in their detection. This study was conducted to establish the quantitative determination of Lycium barbarum polysaccharides(LBPs) based on fluorescein isothiocyanate(FITC) pre-labeling and to investigate their tissue distribution in rat. We obtained the calibration curves linear over the range of 0.0–25 μg/m L in rat tissue samples with correlation coefficients greater than 0.99. The inter-day and intra-day precisions(RSD, %) were within 15%, and the relative recovery ranged 95.2%–102.4%, with RSD range 1.48%–9.58%, indicating that this experiment was suitable for the determination of LBPs. The fluorescence intensity was measured after 24 h storage at room temperature, 3 times of freeze-cycle and cryopreservation at –20 ℃ for 15 day, these results indicated that the stability of the samples was good. LBP-FITC was mainly absorbed by the small intestine and stomach, and mainly excreted in the urine through the kidney;this distinct difference in the tissue distribution of LBPs could be attributed to the size of these LBPs in relation to the pore sizes of the vascular beds in the kidney and liver.
