1Oncogenic AURKA-enhanced N6-methyladenosine modification increases DROSHA mRNA stability to transactivate STC1 in breast cancer stem-like cells显示文摘RNase III DROSHA is upregulated in multiple cancers and contributes to tumor progression by hitherto unclear mechanisms.Here,we demonstrate that DROSHA interacts withβ-Catenin to transactivate STC1 in an RNA cleavage-independent manner,contributing to breast cancer stem-like cell(BCSC)properties.DROSHA mRNA stability is enhanced by N6-methyladenosine(m^(6)A)modification which is activated by AURKA in BCSCs.AURKA stabilizes METTL14 by inhibiting its ubiquitylation and degradation to promote DROSHA mRNA methylation.Moreover,binding of AURKA to DROSHA transcript further strengthens the binding of the m^(6)A reader IGF2BP2 to stabilize m^(6)A-modified DROSHA.In addition,wild-type DROSHA,but not an m^(6)A methylation-deficient mutant,enhances BCSC stemness maintenance,while inhibition of DROSHA m^(6)A modification attenuates BCSC traits.Our study unveils the AURKA-induced oncogenic m^(6)A modification as a key regulator of DROSHA in breast cancer and identifies a novel DROSHA transcriptional function in promoting the BCSC phenotype.Fei Peng Jie Xu Bai Cui Qilan Liang Sai Zeng Bin He Hong Zou Manman Li Huan Zhao Yuting Meng Jin Chen Bing Liu Shasha Lv Peng Chu Fan An Zifeng Wang Junxiu Huang Yajing Zhan Yuwei Liao Jinxin Lu Lingzhi Xu Jin Zhang Zhaolin Su Zhiguang Li Fangjun Wang Eric W-FLam Quentin Liu 2021Cell Research2021,31,3:9

3Effects of cannabinoid receptor type 2 on endogenous myocardial regeneration by activating cardiac progenitor cells in mouse infarcted heart显示文摘Cannabinoid receptor type 2(CB2)activation is recently reported to promote proliferation of some types of resident stem cells(e.g.,hematopoietic stem/progenitor cell or neural progenitor cell).Resident cardiac progenitor cell(CPC)activation and proliferation are crucial for endogenous cardiac regeneration and cardiac repair after myocardial infarction(MI).This study aims to explore the role and possible mechanisms of CB2receptor activation in enhancing myocardial repair.Our results revealed that CB2receptor agonist AM1241 can significantly increase CPCs by c-kit and Runx1 staining in ischemic myocardium as well as improve cardiomyocyte proliferation.AM1241 also decreased serum levels of MDA,TNF-αand IL-6 after MI.In addition,AM1241 can ameliorate left ventricular ejection fraction and fractional shortening,and reduce fibrosis.Moreover,AM1241 treatment markedly increased p-Akt and HO-1 expression,and promoted Nrf-2 nuclear translocation.However,PI3K inhibitor wortmannin eliminated these cardioprotective roles of AM1241.In conclusion,AM1241 could induce myocardial regeneration and improve cardiac function,which might be associated with PI3K/Akt/Nrf2 signaling pathway activation.Our findings may provide a promising strategy for cardiac endogenous regeneration after MI.WANG YaBin MA Sai WANG Qiang HU WenXing WANG DongJuan LI XiuJuan SU Tao QIN Xing ZHANG XiaoTian MA Ke CHEN JiangWei XIONG LiZe CAO Feng 2014Science China(Life Sciences)2014,57,2:3
4The special shaped laser spot for driving indirect-drive hohlraum with multi-beam incidence显示文摘In indirect drive, reducing peak intensity of a single beam and controlling overlap of multi-beams are two opposite requirements for laser focal spot design. In this paper, an improved laser spot design technique for indirect drive built upon the geometric structures of laser propagation into hohlraum has been introduced. The proposed technique is able to generate appropriate continuous phase plate(CPP) producing a special shaped spot that can balance the opposite requirements. The corresponding CPP does not bring difficulties to the design and fabrication. 18Growth of nitrate contribution to aerosol pollution during wintertime in Xi'an,northwest China:Formation mechanism and effects of NH_(3)显示文摘With the strengthened controls on SO2 emissions and extensive increases in motor vehicles'exhaust,aerosol pollution shifts from sulfate-rich to nitrate-rich in recent years in Xi'an,China.To further gain insights into the factors on nitrate formation and efficiently mitigate air pollution,highly time-resolved observations of water-soluble inorganic ions(WSIIs)in PM_(2.5) were measured in a suburban area of Xi'an,China during wintertime.Hourly concentration of total WSIIs is 39.8μg m-3 on average,accounting for 50.3%of PM_(2.5) mass.In contrast to a slight decrease in the mass fraction of SO42-,NO_(3)-shows a sig-nificant increase of the PM_(2.5) contribution with the aggravation of aerosol pollution.This suggests the importance of NO_(3)-formation to haze evolution.Furthermore,homogeneous reactions govern the formation of NO_(3)-,while alkali metals such as calcium and sodium play an additional role in retaining NO_(3)-in PM_(2.5) during clean periods.However,the heterogeneous hydrolysis reaction contributed more to NO_(3)-formation during the pollution periods under high relative humidity.Our investigation reveals that temperature,relative humidity,oxidant,and ammonia emissions facilitate rapid NO_(3)-formation.Using the random forest(RF)model,NO_(3)-concentrations were successfully simulated with measured variables for the training and testing datasets(R2>0.95).Among these variables,CO,NH_(3),and N02 were found to be the main factors affecting the NO_(3)-concentrations.Compared with the period without vehicle re-striction,the contributions of NO_(3)-and NH4+to PM_(2.5) mass decreased by 5.3%and 3.4%in traffic re-striction periods,respectively.The vehicle restriction leads to the decreases of precursor gases of NO2,SO2,and NH_(3) by 12.8%,5.9%,and 27.6%,respectively.The results demonstrate collaborative emission reduction of NOx and NH_(3) by vehicle restrictions,and using new energy vehicles(or electric vehicles)can effectively alleviate particulate matter pollution in northwest China.Hui Su Ting Zhang Suixin Liu Yao Qu Huan Li Jiamao Zhou Zhuzi Zhao Qiyuan Wang Lu Li Minxia Shen Shuoyuan Chen Steven Sai Hang Ho Junji Cao 2024Particuology2024,87,4:0
19Elaborations of the influencing factors on the formation of secondary inorganic aerosols in a heavily polluted urban area of China显示文摘In this study,online water-soluble inorganic ions were detected to deduce the formation mechanism of secondary inorganic aerosol in Xianyang,China during wintertime.The dominant inorganic ions of sulfate(SO_(4)^(2-)),nitrate(NO_(3)^(-)),and ammonium(NH_(4)^(+))(the sum of those is abbreviated as SNA)accounted for 17%,21%,and 12% of PM_(2.5)mass,respectively.While the air quality deteriorated from excellent to poor grades,the precursor gas sulfur dioxide(SO_(2))of SO_(4)^(2-)increased and then decreased with a fluctuation,while nitrogen dioxide(NO_(2))and ammonia(NH_(3)),precursors of NO_(3)^(-)and NH_(4)^(+),and SNA show increasing trends.Meteorological factors including boundary layer height(BLH),temperature,and wind speed also show decline trends,except relative humidity(RH).Meanwhile,the secondary conversion ratio shows a remarkable increasing trend,indicating that there was a strong secondary transformation.From the perspective of chemical mechanisms,RH is positively correlated with sulfur oxidation ratios(SOR),nitrogen oxidation ratios(NOR),and ammonia conversion ratios,representing that the increase of humidity could promote the generation of SNA.Notably,SOR and NOR were also positively related to the ammonia.On the one hand,the low wind speed and BLH led to the accumulation of pollutants.On the other hand,the increases of RH and ammonia promoted more formations of SNA and PM_(2.5).The results advance our identification of the contributors to the haze episodes and assist to establish more efficient emission controls in Xianyang,in addition to other cities with similar emission and geographical characteristics.Shuang Wang Qiyuan Wang Ting Zhang Suixin Liu Steven Sai Hang Ho Jie Tian Hui Su Yong Zhang Luyao Wang Tingting Wu Junji Cao 2024Journal of Environmental Sciences2024,,4:0
20Glutathione disrupts galectin-10 Charcot-Leyden crystal formation to possibly ameliorate eosinophil-based diseases such as asthma显示文摘Charcot-Leyden crystals(CLCs)are the hallmark of many eosinophilic-based diseases,such as asthma.Here,we report that reduced glutathione(GSH)disrupts CLCs and inhibits crystallization of human galectin-10(Gal-10).GSH has no effect on CLCs from monkeys(Macaca fascicularis or M.mulatta),even though monkey Gal-10s contain Cys29 and Cys32.Interestingly,human Gal-10 contains another cysteine residue(Cys57).Because GSH cannot disrupt CLCs formed by the human Gal-10 variant C57A or inhibit its crystallization,the effects of GSH on human Gal-10 or CLCs most likely occur by chemical modification of Cys57.We further report the crystal structures of Gal-10 from M.fascicularis and M.mulatta,along with their ability to bind to lactose and inhibit erythrocyte agglutination.Structural comparison with human Gal-10 shows that Cys57 and Gln75 within the ligand binding site are responsible for the loss of lactose binding.Pull-down experiments and mass spectrometry show that human Gal-10 interacts with tubulinα-1B,with GSH,GTP and Mg^(2+)stabilizing this interaction and colchicine inhibiting it.Overall,this study enhances our understanding of Gal-10 function and CLC formation and suggests that GSH may be used as a pharmaceutical agent to ameliorate CLC-induced diseases.Heya Na Hend Sayed Gabriela Jaramillo Ayala Xing Wang Yuhan Liu Jinyi Yu Tianhao Liu Kevin H.Mayo Jiyong Su 2023Acta Biochimica et Biophysica Sinica2023,55,4:0
