| 1 | Dietary resveratrol improves antioxidant status of sows and piglets and regulates antioxidant gene expression in placenta by Keap1-Nrf2 pathway and Sirt1显示文摘Background: Resveratrol, a plant phenol, affords protection against inflammation and oxidative stress. The objective of this study was to investigate the effects of dietary resveratrol supplementation during pregnancy and lactation on the antioxidant status of sows and piglets and on antioxidant gene expression and pathway in placenta.Methods: Forty sows were allotted to 2 dietary treatments 20 d after breeding. Sows were fed a control diet and a control diet with 300 mg/kg resveratrol. Oxidative stress biomarkers and antioxidant enzymes were measured in the placenta, milk, and plasma of sows and piglets. Antioxidant gene expression and protein expression of Kelch-like ECH-associated protein 1-Nuclear factor E2-related factor 2(Keap1-Nrf2), nuclear factor kappa B-p65(NFκB-p65) and sirtuin1(Sirt1) were quantified in the placenta.Results: Dietary resveratrol increased the litter and piglets weaning weights. Antioxidant status in the milk, placenta and plasma of sows and piglets was partially improved by dietary resveratrol. In placenta, Nrf2 protein expression was increased and Keap1 protein expression was decreased by dietary resveratrol. The m RNA expression of antioxidant genes including catalase(CAT), glutathione peroxidase 1(GPX1), GPX4, superoxide dismutase 1(SOD1)and heme oxygenase 1(HO1), and phase 2 detoxification genes, including glutamate-cysteine ligase modifier(GCLM), microsomal glutathione S-transferase 1(MGST1) and UDP glucuronosyltransferase family 1 member A1(UGT1 A1), was increased by dietary resveratrol. Dietary resveratrol also increased Sirt1 and phosphorylated NFκB-p65 protein expression in the placenta. We failed to observe any influences of dietary resveratrol on pro-inflammatory cytokine levels, including those of interleukin 1β(IL-1β), IL-6, IL-8 and tumor necrosis factor α(TNF-α). However, we observed that the m RNA expression of IL-8 in placenta was reduced by maternal resveratrol. In addition, dietary resveratrol showed interactive effects with day of lactation on activities of SOD and CAT and levels of malonaldehyde(MDA) and hydrogen peroxide(H2 O2) in milk.Conclusions: Dietary resveratrol supplementation during pregnancy and lactation improves the antioxidant status of sows and piglets, which is beneficial to the reproductive performance of sows. | 2 | Transcriptome analysis of the effect of pyrroloquinoline quinone disodium(PQQ·Na2) on reproductive performance in sows during gestation and lactation显示文摘Background: Pyrroloquinoline quinone(PQQ), which is a water soluble, thermo-stable triglyceride-quinone, is widely distributed in nature and characterized as a mammalian vitamin-like redox cofactor. The objective of this study was to investigate the effects of pyrroloquinoline quinone disodium(PQQ·Na2) on reproductive performance in sows.Results: Dietary supplementation with PQQ·Na2 significantly increased the total number of piglets born, the number of piglets born alive and the born alive litter weight. It also increased the antioxidant status in the placenta, plasma and milk. The concentration of NO was significantly increased in the plasma and placenta. RNA-seq analysis showed that462 unigenes were differentially expressed between the control(Con) treatment and PQQ treatment groups.Among these unigenes, 199 were upregulated, while 263 unigenes were downregulated. The assigned functions of the unigenes covered a broad range of GO categories. Reproduction(27, 7.03%) and the reproduction process(27, 7.03%) were assigned to the biological process category. | 3 | Effects of Chinese herbs on lactation of rat and performance of offspring显示文摘An experiment was conducted to study the effects of different Chinese herbs on lactation in the rat. Seventy-two Sprague Dawley female rats at gestation day 18 were assigned to 9 groups randomly, with 8 rats each. Control rats were offered basal diets until the end of the weaning period. Rats in the other 8 groups were administered diets supplemented with 1% Medulla tetrapanacis (MT), Astragalus membranacens(AM), Vaccaria segetalis(VS), Leonurus heterophyllus sweet (LHS), Radix rehmanniae preparata (RRP), Squama manitis (SM), Schisandra chinensis (SC), Ligustrum lucidum (LL), respectively. After parturition, the litter size was culled to 8 pups per dam. Pup weight and milk yield were significantly higher in the VS, LHS, AM and SM groups than these in the control group during middle and late lactation (P<0.05 or P<0.01). | 4 | Modified halloysite nanotubes reduce the toxic effects of zearalenone in gestating sows on growth and muscle development of their offsprings显示文摘Background: Zearalenone(ZEN) is an estrogenic mycotoxin that is primarily produced by Fusarium fungi and has been proven to affect the reproductive capacity of many species to varying degrees. The present experiment was designed to study the maternal persistent effects of zearalenone toxicity in gestating sows on growth and muscle development of their offsprings, and the alleviation of zearalenone toxicity by modified halloysite nanotubes(MHNTs).Methods: Eighteen sows were fed with one of three dietary treatments that included the following:(1) a control diet,(2) a contaminated grain diet(with 50 % moldy corn, 2.77 mg/kg ZEN), and(3) a contaminated grain diet(with 50 %moldy corn, 2.76 mg/kg ZEN) + 1 % MHNTs. Each sow was exclusively fed its experimental diets from 35 to 70 d of gestation at a total of 2 kg daily. Muscle samples were collected from six piglets per treatment at birth, weaning and finishing.Results: The results showed that feeding the sows with the ZEN-contaminated diets from 35 to 70 d of gestation decreased the ADG, ADFI and G:F of their offsprings(P < 0.05). The muscle fiber numbers in the newborn, weaning and growing-finishing pigs and the muscle fiber diameters at birth and weaning were also decreased by maternal ZEN exposure(P < 0.05). The expressions of IGF-I, IGF-II, Myf-5 and Mstn at birth and IGF-II, Pax7, Myf-5 and Myo D1 at weaning were altered by feeding gestating sows with ZEN-contaminated diets(P < 0.05). The MHNTs reduced most of the ZEN-induced toxic effects: the ADG and ADFI on growth performance, the muscle fiber numbers at weaning and finishing and the muscle fiber diameters at weaning(P < 0.05). | 5 | Lactobacillus rhamnosus GG ameliorates DON-induced intestinal damage depending on the enrichment of beneficial bacteria in weaned piglets显示文摘Background: Deoxynivalenol(DON) is one of the most common environmental pollutants that induces intestinal inflammation and microbiota dysbiosis. Lactobacillus rhamnosus GG(LGG) is a probiotic that not only has anti-inflammatory effects, but also shows protective effect on the intestinal barrier. However, it is still unknown whether LGG exerts beneficial effects against DON-induced intestinal damage in piglets. In this work, a total of 36 weaned piglets were randomized to one of four treatment groups for 21 d. The treatment groups were CON(basal diet);LGG(basal diet supplemented with 1.77 × 10^(11)CFU/kg LGG);DON(DON-contaminated diet) and LGG + DON(DON-contaminated diet supplemented with 1.77 × 10^(11)CFU/kg LGG).Result: Supplementation of LGG can enhance growth performance of piglets exposed to DON by improving intestinal barrier function. LGG has a mitigating effect on intestinal inflammation induced by DON exposure, largely through repression of the TLR4/NF-κB signaling pathway. | 12 | Exopolysaccharides of Lactobacillus rhamnosus GG ameliorate Salmonella typhimurium-induced intestinal inflammation via the TLR4/NF-κB/MAPK pathway显示文摘Background Salmonella typhimurium(S.T),as an important foodborne bacterial pathogen,can cause diarrhea and gastroenteritis in humans and animals.Numerous studies have confirmed that exopolysaccharides(EPSs)have various biological functions,but the mechanism through which EPSs improve the immunity of animals against the invasion of pathogenic bacteria is unclear.Here,we explored the protective effect of EPSs of Lactobacillus rhamnosus GG(LGG)on the S.T-infected intestine.Methods Mice received adequate food and drinking water for one week before the start of the experiment.After 7 d of prefeeding,2×108 CFU/mL S.T solution and an equivalent volume of saline(control group)were given orally for 1 d.On the fourth day,the mice were treated with 0.5 mg/mL EPSs,1.0 mg/mL EPSs,2.0 mg/mL EPSs,or 2.0 mg/mL penicillin for 7 d.Finally,the body and relative organ weight,histological staining,and the levels of antioxidant enzyme activity and inflammatory cytokines were determined.Results The S.T-infected mice exhibited symptoms of decreased appetite,somnolence,diarrhea and flagging spirit.Treatment with EPSs and penicillin improved the weight loss of the mice,and the high dose of EPSs showed the best therapeutic effect.EPSs significantly ameliorated S.T-induced ileal injury in mice.High-dose EPSs were more effective than penicillin for alleviating ileal oxidative damage induced by S.T.The mRNA levels of inflammatory cytokines in the ileum of mice showed that the regulatory effects of EPSs on inflammatory cytokines were better than those of penicillin.EPSs could inhibit the expression and activation of key proteins of the TLR4/NF-κB/MAPK pathway and thereby suppress the level of S.T-induced ileal inflammation.Conclusions EPSs attenuate S.T-induced immune responses by inhibiting the expression of key proteins in the TLR4/NF-κB/MAPK signaling pathway.Moreover,EPSs could promote bacterial aggregation into clusters,which may be a potential strategy for reducing the bacterial invasion of intestinal epithelial cells. | Jinze Li Qiuke Li Qianhui Wu Nan Gao Zhihua Wang Yang Yang Anshan Shan | 2023 | Journal of Animal Science and Biotechnology2023,14,4: | 0 |

| 14 | Influence of Early Quantitative Feed Restriction on Performance and Carcass Characteristics of Broiler Chickens显示文摘An experiment was conducted to study the influence of early quantitative feed restriction on the performance and carcass characteristics of broiler chickens. Two hundred 8-day-old commercial Avian male broiler chicks were assigned to four groups randomly, with 50 broilers each. At the age of 8 days the broilers were restricted to 90%, 80% or 70% of the ad libitum intake for 7 days. When the restricion was finished, all birds received feed ad libitum again. The results showed that diet restriction resulted in a reduction in body weight and daily gain during the restriction period (P <0.05). The birds restricted to 90% of the ad libitum intake showed the occurring of growth compensation in the 3rd week. The birds restricted to 80% and 70% of the ad libitum intake had growth compensation in the 4th week. [Method]Whole-crop corn was stored in laboratory bag silos and outdoor bunker silos. Bag silos were stored for 3,7,14,28,56 and 120 d,respectively,and fermentation products were analyzed after opening; the fermentation products,which were from three 500 t above-ground outdoor bunker silos stored for 120 d,were detected. Eight sampling points were selected for each bunker silo. [Result]No 1,2-propanediol was detected out in laboratory bag silos,and lactic acid was dominant in fermentation products. The acetic acid content significantly increased( P < 0. 05) with the extension of storage time,whereas the acetic acid content was kept below one-third of the lactic acid content; 1,2-propanediol and a small amount of 1-propanol were detected out in three bunker silos,and lactic acid was dominant in fermentation products,whereas the content of acetic acid was slightly lower than that of lactic acid. The differences in fermentation products from different silos and different sites in silos presented heterogeneity; except to propanediol,lactic acid bacteria and yeast,there was no significant difference in fermentation products at the top and bottom layers of silos( P > 0. 05); there were significant differences in contents of lactic acid,acetic acid,1,2-propanediol,and yeast between the outer and inner parts of silos( P < 0. 05); except to p H values and acetic acid,there were significant differences in fermentation products between bunker silos( P < 0. 05). | 17 | Effect of supplemental Bacillus culture on rumen fermentation and performance in dairy cattle显示文摘Two parts were involved in this experiment.In experiment 1,32 Chinese Holstein cows with relatively similar body condition,lactation number and days in milk were selected.The cows were assigned in a randomized complete block design trial to determine the effect of supplemental Bacillus cultures to diet on production performance in dairy cattle.Four treatments,i.e., Bacillus licheniformis(strain number 1.813)group,Bacillus subtilis(strain number 1.1086)group,Bacillus cereus var.mvcoides (strain number 1.260)group and control group.Each treatment had eight replicates,each replicate had one cow,50 g per head per day.Results showed that Bacillus licheniformis group increased the milk yield(P<0.05).The other two groups didn’t have significant effect on milk yield than the control group(P>0.05).In experiment 2,3 Chinese Holstein cows with permanent fistulas were used.3×3 Latin squares were assigned to three diets:Bacillus lincheniformis culture,Bacillus subtilis culture and control.Bacillus licheniformis culture increased total rumen microorganism(P<0.05),decreased the ammonia N concentration at 2,4 h(P<0.05). | 18 | Study on HPLC method to determine contents of Schisandrin A and Schisandrin B in Schisandra chinensis extraction显示文摘The determination method of Schisandrin A and Schisandrin B in Schisandra chinensis was improved with the high performance liquid chromagraphy(HPLC).The sample was extracted exceedingly in the critical limit of CO 2 .The retention time of Schisandrin A and Schisandrin B was reduced,with methano/water(75:25)as mobile phase.The wavelength for detection was 254 nm.
