1Dietary resveratrol improves antioxidant status of sows and piglets and regulates antioxidant gene expression in placenta by Keap1-Nrf2 pathway and Sirt1显示文摘Background: Resveratrol, a plant phenol, affords protection against inflammation and oxidative stress. The objective of this study was to investigate the effects of dietary resveratrol supplementation during pregnancy and lactation on the antioxidant status of sows and piglets and on antioxidant gene expression and pathway in placenta.Methods: Forty sows were allotted to 2 dietary treatments 20 d after breeding. Sows were fed a control diet and a control diet with 300 mg/kg resveratrol. Oxidative stress biomarkers and antioxidant enzymes were measured in the placenta, milk, and plasma of sows and piglets. Antioxidant gene expression and protein expression of Kelch-like ECH-associated protein 1-Nuclear factor E2-related factor 2(Keap1-Nrf2), nuclear factor kappa B-p65(NFκB-p65) and sirtuin1(Sirt1) were quantified in the placenta.Results: Dietary resveratrol increased the litter and piglets weaning weights. Antioxidant status in the milk, placenta and plasma of sows and piglets was partially improved by dietary resveratrol. In placenta, Nrf2 protein expression was increased and Keap1 protein expression was decreased by dietary resveratrol. The m RNA expression of antioxidant genes including catalase(CAT), glutathione peroxidase 1(GPX1), GPX4, superoxide dismutase 1(SOD1)and heme oxygenase 1(HO1), and phase 2 detoxification genes, including glutamate-cysteine ligase modifier(GCLM), microsomal glutathione S-transferase 1(MGST1) and UDP glucuronosyltransferase family 1 member A1(UGT1 A1), was increased by dietary resveratrol. Dietary resveratrol also increased Sirt1 and phosphorylated NFκB-p65 protein expression in the placenta. We failed to observe any influences of dietary resveratrol on pro-inflammatory cytokine levels, including those of interleukin 1β(IL-1β), IL-6, IL-8 and tumor necrosis factor α(TNF-α). However, we observed that the m RNA expression of IL-8 in placenta was reduced by maternal resveratrol. In addition, dietary resveratrol showed interactive effects with day of lactation on activities of SOD and CAT and levels of malonaldehyde(MDA) and hydrogen peroxide(H2 O2) in milk.Conclusions: Dietary resveratrol supplementation during pregnancy and lactation improves the antioxidant status of sows and piglets, which is beneficial to the reproductive performance of sows. 2Signaling pathways and targeted therapy for myocardial infarction显示文摘Although the treatment of myocardial infarction(MI)has improved considerably,it is still a worldwide disease with high morbidity and high mortality.Whilst there is still a long way to go for discovering ideal treatments,therapeutic strategies committed to cardioprotection and cardiac repair following cardiac ischemia are emerging.Evidence of pathological characteristics in MI illustrates cell signaling pathways that participate in the survival,proliferation,apoptosis,autophagy of cardiomyocytes,endothelial cells,fibroblasts,monocytes,and stem cells.These signaling pathways include the key players in inflammation response,e.g.,NLRP3/caspase-1 and TLR4/MyD88/NF-κB;the crucial mediators in oxidative stress and apoptosis,for instance,Notch,Hippo/YAP,RhoA/ROCK,Nrf2/HO-1,and Sonic hedgehog;the controller of myocardial fibrosis such as TGF-β/SMADs and Wnt/β-catenin;and the main regulator of angiogenesis,PI3K/Akt,MAPK,JAK/STAT,Sonic hedgehog,etc.Since signaling pathways play an important role in administering the process of MI,aiming at targeting these aberrant signaling pathways and improving the pathological manifestations in MI is indispensable and promising.Hence,drug therapy,gene therapy,protein therapy,cell therapy,and exosome therapy have been emerging and are known as novel therapies.In this review,we summarize the therapeutic strategies for MI by regulating these associated pathways,which contribute to inhibiting cardiomyocytes death,attenuating inflammation,enhancing angiogenesis,etc.so as to repair and re-functionalize damaged hearts.Qing Zhang Lu Wang Shiqi Wang Hongxin Cheng Lin Xu Gaiqin Pei Yang Wang Chenying Fu Yangfu Jiang Chengqi He Quan Wei 2022Signal Transduction and Targeted Therapy2022,7,4:11
3Assessing the effects of vegetation and precipitation on soil erosion in the Three-River Headwaters Region of the Qinghai-Tibet Plateau,China显示文摘Soil erosion in the Three-River Headwaters Region(TRHR)of the Qinghai-Tibet Plateau in China has a significant impact on local economic development and ecological environment.Vegetation and precipitation are considered to be the main factors for the variation in soil erosion.However,it is a big challenge to analyze the impacts of precipitation and vegetation respectively as well as their combined effects on soil erosion from the pixel scale.To assess the influences of vegetation and precipitation on the variation of soil erosion from 2005 to 2015,we employed the Revised Universal Soil Loss Equation(RUSLE)model to evaluate soil erosion in the TRHR,and then developed a method using the Logarithmic Mean Divisia Index model(LMDI)which can exponentially decompose the influencing factors,to calculate the contribution values of the vegetation cover factor(C factor)and the rainfall erosivity factor(R factor)to the variation of soil erosion from the pixel scale.In general,soil erosion in the TRHR was alleviated from 2005 to 2015,of which about 54.95%of the area where soil erosion decreased was caused by the combined effects of the C factor and the R factor,and 41.31%was caused by the change in the R factor.There were relatively few areas with increased soil erosion modulus,of which 64.10%of the area where soil erosion increased was caused by the change in the C factor,and 23.88%was caused by the combined effects of the C factor and the R factor.Therefore,the combined effects of the C factor and the R factor were regarded as the main driving force for the decrease of soil erosion,while the C factor was the dominant factor for the increase of soil erosion.The area with decreased soil erosion caused by the C factor(12.10×10^3 km^2)was larger than the area with increased soil erosion caused by the C factor(8.30×10^3 km^2),which indicated that vegetation had a positive effect on soil erosion.This study generally put forward a new method for quantitative assessment of the impacts of the influencing factors on soil erosion,and also provided a scientific basis for the regional control of soil erosion.HE Qian DAI Xiao'ai CHEN Shiqi 2020Journal of Arid Land2020,12,5:9
4Sensitivity-enhanced surface plasmon resonance sensor utilizing a tungsten disulfide (WS_2) nanosheets overlayer显示文摘Tungsten disulfide(WS_2), as a representative layered transition metal dichalcogenide(TMDC) material, possesses important potential for applications in highly sensitive sensors. Here, a sensitivity-enhanced surface plasmon resonance(SPR) sensor with a metal film modified by an overlayer of WS_2 nanosheets is proposed and demonstrated. The SPR sensitivity is related to the thickness of the WS_2 overlayer, which can be tailored by coating a WS_2 ethanol suspension with different concentrations or by the number of times of repeated post-coating.Benefitting from its large surface area, high refractive index, and unique optoelectronic properties, the WS_2 nanosheet overlayer coated on the gold film significantly improves the sensing sensitivity. The highest sensitivity(up to 2459.3 nm∕RIU) in the experiment is achieved by coating the WS_2 suspension once. Compared to the case without a WS_2 overlayer, this result shows a sensitivity enhancement of 26.6%. The influence of the WS_2 nanosheet overlayer on the sensing performance improvement is analyzed and discussed. Moreover, the proposed WS_2 SPR sensor has a linear correlation coefficient of 99.76% in refractive index range of 1.333 to 1.360. 5U-Pb Zircon and Re-Os Molybdenite Geochronology of the W-Mo Mineralized Region of South Qinling, China, and their Tectonic Implications显示文摘A W-Mo mineralized region is located along the northern margin of the South Qinling tectonic belt of China. WMo mineralization occurs mainly in Cambrian–Ordovician clastic and carbonate rocks, and the ore bodies are structurally controlled by NW–SE-and NNE–SSW-striking faults. Evidence for magmatism in the area is widespread and is dominated by intermediate–felsic intrusives or apophyses, such as the Dongjiangkou, Yanzhiba, Lanbandeng, and Sihaiping granitic bodies. Quartz-vein-type mineralization and fault-controlled skarn-type mineralization dominate the ore systems, with additional enrichment in residual deposits. At present, there are few or insufficient studies on(1) the age of mineralization,(2) the relationship between intermediate–felsic granite and W-Mo mineralization,(3) the source of ore-forming materials, and(4) the metallogenic and tectonic setting of the mineralized area. In this paper, we present geochronology results for numerous intrusive granitic bodies in the South Qinling tectonic belt. U-Pb zircon geochronology of the Lanbandeng monzogranite and Wangjiaping biotite monzogranite yields ages of 222.7 ± 2.3 and 201.9 ± 1.8 Ma, respectively. In contrast to the Late Triassic age of the Lanbandeng monzogranite, the age of the newly discovered Wangjiaping biotite monzogranite places it at the Triassic–Jurassic boundary. Re-Os molybdenite geochronology on the Qipangou W-Mo deposit yielded a model age of 199.7 ± 3.9 Ma, indicating the deposit formed in the early Yanshanian period of the Early Jurassic. Granitoid intrusions in the mineralized area are characterized by composite granite bodies that crystallized at ca. 240–190 Ma. While there were multiple stages of intrusion, most occurred at 210–220 Ma, with waning magmatic activity at 200–190 Ma. The Re-Os age of molybdenite in the region is ca. 200–190 Ma, which may represent a newly discovered period of W-Mo metallogenesis that occurred during the final stages of magmatism. The heat associated with this magmatism drove ore formation and might have provided additional ore-forming components for metallogenesis(represented by the Wangjiaping biotite monzogranite). Ore materials in the mineralized area were derived from mixed crustal and mantle sources. Enrichment of the region occurred during intracontinental orogenesis in the late Indosinian–Yanshanian, subsequent to the main Indosinian collision. 6Molecular basis of CONSTANS oligomerization in FLOWERING LOCUS T activation显示文摘The transcription factor CONSTANS(CO)integrates day-length information to induce the expression of florigen FLOWERING LOCUS T(FT)in Arabidopsis.We recently reported that the C-terminal CCT domain of CO forms a complex with NUCLEAR FACTOR-YB/YC to recognize multiple cis-elements in the FT promoter,and the N-terminal tandem B-box domains form a homomultimeric assembly.However,the mechanism and biological function of CO multimerization remained unclear.Here,we report that CO takes on a head-to-tail oligomeric configuration via its B-boxes to mediate FT activation in long days.The crystal structure of B-boxesCOreveals a closely connected tandem B-box fold forming a continuous head-to-tail assembly through unique CDHH zinc fingers.Mutating the key residues involved in CO oligomerization resulted in a non-functional CO,as evidenced by the inability to rescue co mutants.By contrast,a transgene encoding a human p53-derived tetrameric peptide in place of the B-boxesCOrescued co mutant,emphasizing the essential role of BboxesCO-mediated oligomerization.Furthermore,we found that the four TGTG-bearing cis-elements in FT proximal promoter are required for FT activation in long days.Our results suggest that CO forms a multimer to bind to the four TGTG motifs in the FT promoter to mediate FT activation.Xiaolin Zeng Xinchen Lv Rui Liu Hang He Shiqi Liang Lixian Chen Fan Zhang Liu Chen Yuehui He Jiamu Du 2022Journal of Integrative Plant Biology2022,64,3:2
7Artemisinin analogue SM934 protects against lupus-associated antiphospholipid syndrome via activation of Nrf2 and its targets显示文摘Kidney is a major target organ in both antiphospholipid syndrome(APS)and systemic lupus erythematosus(SLE).The etiology of antiphospholipid syndrome nephropathy associated lupus nephritis(APSN-LN)is intricate and remains largely unrevealed.We proposed in present work,that generation of antiphospholipid antibodies(aPLs),especially those directed towards the oxidized neoepitopes,are largely linked with the redox status along with disease progression.Moreover,we observed that compromised antioxidative capacity coincided with turbulence of inflammatory cytokine profile in the kidney of male NZW×BXSB F1 mice suffered from APSN-LN.SM934 is an artemisinin derivative that has been proved to have potent immunosuppressive properties.In current study,we elaborated the therapeutic benefits of SM934 in male NZW×BXSB F1 mice,a murine model develops syndrome resembled human APS associated with SLE,for the first time.SM934 treatment comprehensively impeded autoantibodies production,inflammatory cytokine accumulation and excessive oxidative stress in kidney.Among others,we interpreted in present work that both anti-inflammatory and antioxidative effects of SM934 is closely correlated with the enhancement of Nrf2 signaling and expression of its targets.Collectively,our finding confirmed that therapeutic strategy simultaneously exerting antioxidant and anti-inflammatory efficacy provide a novel feasible remedy for treating APSN-LN.Zemin Lin Yuting Liu Li Chen Shiqi Cao Yueteng Huang Xiaoqian Yang Fenghua Zhu Wei Tang Shijun He Jianping Zuo 2021Science China(Life Sciences)2021,64,10:2

9Nephrocyte-neurocyte interaction and cellular metabolic analysis on membrane-integrated microfluidic device显示文摘Cell-cell interaction and cell metabolic analysis provide new opportunities for better understanding of critical biochemical processes. Advanced microfluidic technologies enable to create more realistic in vitro microenvironment by spatial and temporal control of cell growth and co-culture. In this work, we design a microfluidic device to achieve the co-culture of PC12 cells and 293 cells, and study in vitro cell-cell interaction via cell metabolic analysis by mass spectrometry. The membraneintegrated microfluidic device was firstly used for cell co-culture, and the cellular metabolite was further investigated by mass spectrometer(MS). Our results showed that the differentiation of PC12 cells could be successfully induced by m NGF and also greatly influenced by the microchannel treatment of fetal bovine serum(FBS) solution. The identification of cell morphology, microtubule-associated protein 2(MAP-2) expression and viability of differentiated PC12 cells were conducted before 293 cells being introduced into the top microfluidic channels and stimulated to secrete cell metabolism products. 10Novel fibronectin-targeted nanodisk drug delivery system displayed superior efficacy against prostate cancer compared with nanospheres显示文摘Currently,prostate cancer is the most frequently diagnosed cancer in males and chemotherapy is often essential for treating advaneed prostate cancer.However,common chemotherapies for prostate cancer suffer from serious adverse effects due to poor drug targeting ability and tissue penetration,even with the help of conventional drug delivery systems.Here,encouraged by recent studies showing possible drug retention and tissue pen etration advan tages of unconventional non-spherical nan oparticles over conventional spherical nan oparticles,we design and construct a novel non-spherical nano disk drug delivery system for treating prostate cancer.norder to enhance tumor-targeting capability,these nanodisks are further modified with targeting peptide,Cys-Arg-Glu丄ys?Ala peptide with N-methylated Glu(CR(NMe)EKA),which recognizes extracellular matrix fibronectin and its complexes specifically expressed on the walls of tumor vessels and in tumor stroma.Compared with conventional nano spheres,the nano disks achieve much higher drug accumulati on at prostate tumor sites.When loaded with paclitaxel,the CR(NMe)EKA-modified nan odisks display superior an titumor efficacy to free paclitaxel,unmodified nan odisks and nano spheres.In summary,our study provides an attractive therapeutic strategy for targeted therapy against prostate cancer with simple preparation,high efficiency and low toxicity,and supplements a theoretical support for treatments realized by different shaped nano platforms.Our study also offers valuable data for understa nding biological effects of non-spherical nano disks and highlights the great pote ntial of unconventional nan oparticles in biomedical applicati ons.Luyao Wang Bingjie Zhou Shiqi Huang Mengke Qu Qing Lin Tao Gong Yuan Huang Xun Sun Qin He Zhirong Zhang Ling Zhang 2019Nano Research2019,12,10:1
11Flow and Temperature Fields in Slab Continuous Casting Molds显示文摘In order to develop super-board and super-thick slabs, the flow and temperatur fields were studied in slab continuous casting molds under different practical conditions, such as slab dimensions, with-drawing slab speed, design of nozzles, and superheat tempera-ture. The results showed that it is preferred to incline nozzle bores downwards and the submerged depth of the nozzles is best kept be-tween 250-300 mm. 12Screening of Sapindus germplasm resources in China based on agro-morphological traits显示文摘Sapindus L.species are widely cultivated for biodiesel,biomedical,and biochemical raw materials in southern China.However,yields fuctuate widely due to the lack of high-yielding,high-quality,stable cultivars.Therefore,the objectives of this study were to evaluate three species and one variety among 149 nationwide Sapindus species and screen for elite accessions which would serve as Sapindus breeding materials.Accessions were evaluated on 19 agro-morphological traits by correlation analysis and principal component analysis.These displayed substantial diversity and a broad range of economic traits.In particular,accessions of Sapindus mukorossi and Sapindus rarak had more variation in economic traits than Sapindus delavayi and Sapindus rarak var.velutinus.Increased saponin accumulation may be achieved at the cost of seed oil production.Thirty elite accessions for oil and saponin production,and for comprehensive utilization were screened and accessions no.80,110,and 112 had significant potential to produce high yields.These elite accessions will facilitate the identification of genetic determinants of valuable traits and the effective utilization of trait variability in Sapindus breeding.Jiming Liu Shiqi Liu Yuanyuan Xu Caowen Sun Zhong Chen Xin Wang Lixian Wang Shilun Gao Guochun Zhao Qiuyang He Xuehuang Weng Liming Jia 2022Journal of Forestry Research2022,33,1:1
13Feasibility and physics potential of detecting ^(8)B solar neutrinos at JUNO显示文摘The Jiangmen Underground Neutrino Observatory(JUNO)features a 20 kt multi-purpose underground liquid scintillator sphere as its main detector.Some of JUNO's features make it an excellent location for^8B solar neutrino measurements,such as its low-energy threshold,high energy resolution compared with water Cherenkov detectors,and much larger target mass compared with previous liquid scintillator detectors.In this paper,we present a comprehensive assessment of JUNO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2 MeV threshold for the recoil electron energy is found to be achievable,assuming that the intrinsic radioactive background^(238)U and^(232)Th in the liquid scintillator can be controlled to 10^(-17)g/g.With ten years of data acquisition,approximately 60,000 signal and 30,000 background events are expected.This large sample will enable an examination of the distortion of the recoil electron spectrum that is dominated by the neutrino flavor transformation in the dense solar matter,which will shed new light on the inconsistency between the measured electron spectra and the predictions of the standard three-flavor neutrino oscillation framework.IfDelta m^(2)_(21)=4.8times10^(-5);(7.5times10^(-5))eV^(2),JUNO can provide evidence of neutrino oscillation in the Earth at approximately the 3sigma(2sigma)level by measuring the non-zero signal rate variation with respect to the solar zenith angle.Moreover,JUNO can simultaneously measureDelta m^2_(21)using^8B solar neutrinos to a precision of 20% or better,depending on the central value,and to sub-percent precision using reactor antineutrinos.A comparison of these two measurements from the same detector will help understand the current mild inconsistency between the value of Delta m^2_(21)reported by solar neutrino experiments and the KamLAND 14Study of Effect of Boundary Conditions on Patient-Specific Aortic Hemodynamics显示文摘Cardiovascular computational fluid dynamics(CFD)based on patient-specific modeling is increasingly used to predict changes in hemodynamic parameters before or after surgery/interventional treatment for aortic dissection(AD).This study investigated the effects of flow boundary conditions(BCs)on patient-specific aortic hemodynamics.We compared the changes in hemodynamic parameters in a type A dissection model and normal aortic model under different BCs:inflow from the auxiliary and truncated structures at aortic valve,pressure control and Windkessel model outflow conditions,and steady and unsteady inflow conditions.The auxiliary entrance remarkably enhanced the physiological authenticity of numerical simulations of flow in the ascending aortic cavity.Thus,the auxiliary entrance can well reproduce the injection flow fromthe aortic valve.In addition,simulations of the aortic model reconstructed with an auxiliary inflow structure and pressure control and the Windkessel model outflow conditions exhibited highly similar flow patterns and wall shear stress distribution in the ascending aorta under steady and unsteady inflow conditions.Therefore,the inflow structure at the valve plays a crucial role in the hemodynamics of the aorta.Under limited time and calculation cost,the steady-state study with an auxiliary inflow valve can reasonably reflect the blood flow state in the ascending aorta and aortic arch.With reasonable BC settings,cardiovascular CFD based on patient-specific ADmodels can aid physicians in noninvasive and rapid diagnosis.Qingzhuo Chi Huimin Chen Shiqi Yang Lizhong Mu Changjin Ji Ying He Yong Luan 2022Computer Modeling in Engineering & Sciences2022,,4:0

18Metabolic reprogramming of proinflammatory macrophages by target delivered roburic acid effectively ameliorates rheumatoid arthritis symptoms显示文摘Rheumatoid arthritis(RA)is a common chronic inflammatory disorder that usually affects joints.It was found that roburic acid(RBA),an ingredient from anti-RA herb Gentiana macrophylla Pall.,displayed strong anti-inflammatory activity.However,its medical application is limited by its hydrophobicity,lack of targeting capability and unclear functional mechanism.Here,we constructed a pH responsive dual-target drug delivery system hitchhiking RBA(RBA-NPs)that targeted both CD44 and folate receptors,and investigated its pharmacological mechanism.In rat RA model,the nanocarriers effectively delivered RBA to inflammatory sites and significantly enhanced the therapeutic outcomes compared with free RBA,as well as strongly reducing inflammatory cytokine levels and promoting tissue repair.Following analysis revealed that M1 macrophages in the joints were reprogrammed to M2 phenotype by RBA.Since the balance of pro-and anti-inflammatory macrophages play important roles in maintaining immune homeostasis and preventing excessive inflammation in RA,this reprogramming is likely responsible for the anti-RA effect.Furthermore,we revealed that RBA-NPs drove M1-to-M2 phenotypic switch by down-regulating the glycolysis level via blocking ERK/HIF-1α/GLUT1 pathway.Thus,our work not only developed a targeting delivery system that remarkably improved the anti-RA efficiency of RBA,but also identified a potential molecular target to reversely reprogram macrophages though energy metabolism regulation.Na Jia Yunzhen Gao Min Li Yi Liang Yuwen Li Yunzhu Lin Shiqi Huang Qing Lin Xun Sun Qin He Yuqin Yao Ben Zhang Zhirong Zhang Ling Zhang 2023Signal Transduction and Targeted Therapy2023,8,8:0
19Do Geographically Isolated Grasslands Follow the Principle of Island Biogeography in a Landscape Scale? Taking Poyang Lake Grassland as an Example显示文摘As one of the basic theories of biodiversity conservation,island biogeography has been widely accepted in the past decades.Originally,island biogeography was put forward and applied in oceanic environments.But later on,it was found out that the application was not only limited to oceanic islands,but also in terrestrial environments with relatively isolated conditions.In terms of biodiversity level,island biogeography generally focuses on a small scale,such as species diversity and genetic diversity.The studies of biodiversity on a large-scale based on island biogeography,such as ecosystem and landscape scales,were seldomly conducted.Taking Poyang Lake,the largest fresh water lake in China as case study area,30 grasslands were randomly selected to study whether island biogeography can be applied to grasslands at a landscape level from three island attributes(area,distance and shape),and the most important ecological variable(flooding)in Poyang Lake.The results showed that in general,grasslands have the property of an island,and follow the basic principle of island biogeography.We found the area and flooding duration were the two most important determinants of landscape diversity.There was a significant positive correlation between the grassland area and the landscape diversity,which could be well expressed by logarithmic function model(R2=0.73).There was a negative correlation between flooding duration and landscape diversity,which could be described by an inverse model(R2=0.206).The distance to mainland and the shape of grassland were correlated with landscape diversity,but the fitting result of the models was not as good as expected.The possible reason could be that Poyang Lake is a seasonal lake,the water level varies with hydrological conditions,so that the grasslands are not strongly isolated and their shape is not stable enough required by island biogeography.Furthermore,it indicates that besides area,distance and shape attributes,flooding strongly affects the biodiversity of grassland vegetation,and should not be ignored when applying island biogeography theory to Poyang Lake.This study is expected to be a supplement for island biogeography in terrestrial environments,and the results are expected to benefit for the biodiversity conservation in Poyang Lake.Shiqi Luo Wenbo Chen Lei He Qiongbing Xiong 2023Phyton-International Journal of Experimental Botany2023,92,2:0
