1Structure-function relationship of the mammarenavirus envelope glycoprotein显示文摘Mammarenaviruses, including lethal pathogens such as Lassa virus and Junín virus, can cause severe hemorrhagic fever in humans. Entry is a key step for virus infection, which starts with binding of the envelope glycoprotein(GP) to receptors on target cells and subsequent fusion of the virus with target cell membranes. The GP precursor is synthesized as a polypeptide, and maturation occurs by two cleavage events, yielding a tripartite GP complex(GPC) formed by a stable signal peptide(SSP), GP1 and GP2. The unique retained SSP interacts with GP2 and plays essential roles in virion maturation and infectivity. GP1 is responsible for binding to the cell receptor, and GP2 is a class I fusion protein. The native structure of the tripartite GPC is unknown.GPC is critical for the receptor binding, membrane fusion and neutralization antibody recognition.Elucidating the molecular mechanisms underlining the structure–function relationship of the three subunits is the key for understanding their function and can facilitate novel avenues for combating virus infections. This review summarizes the basic aspects and recent research of the structure–function relationship of the three subunits. We discuss the structural basis of the receptor-binding domain in GP1, the interaction between SSP and GP2 and its role in virion maturation and membrane fusion, as well as the mechanism by which glycosylation stabilizes the GPC structure and facilitates immune evasion. 2Cellular membrane cholesterol is required for porcine reproductive and respiratory syndrome virus entry and release in MARC-145 cells显示文摘Cholesterol represents one of the key constituents of small,dynamic,sterol-and sphingolipid-enriched domains on the plasma membrane.It has been reported that many viruses depend on plasma membrane cholesterol for efficient infection.In this study,the role of the plasma membrane cholesterol in porcine reproductive and respiratory syndrome virus(PRRSV) infection of MARC-145 cells was investigated.Pretreatment of MARC-145 cells with methyl-β-cyclodextrin(MβCD),a drug used to deplete cholesterol from cellular membrane,significantly reduced PRRSV infection in a dose-dependent manner.This inhibition was partially reversed by supplementing exogenous cholesterol following MβCD treatment,suggesting that the inhibition of PRRSV infection was specifically mediated by removal of cellular cholesterol.Further detailed studies showed that depletion of cellular membrane cholesterol significantly inhibited virus entry,especially virus attachment and release.These results indicate that the presence of cholesterol in the cellular membrane is a key component of PRRSV infection.SUN Ying XIAO ShaoBo WANG Dang LUO Rui LI Bin CHEN HuanChun FANG LiuRong 2011Science China(Life Sciences)2011,54,11:3

5Influence of Nano-Al_2O_3 on Properties of Magnesia Chrome Refractories显示文摘Magnesia chrome bricks were prepared with fused MgO-Cr2O3 synthesized material,fused magnesite and chrome ore as main starting materials. Nano-Al2O3 was added into refractories ( 2% ,4% and 6% in mass) substituting for Al2O3 micropowder. After mixing and shaping,the bricks were fired at 1 550 ℃ ,1 600 ℃ , 1 650 ℃ and 1 750 ℃ ,respectively. The microstruc-ture,sintering property,mechanical properties,thermal shock resistance and slag resistance of the specimens with the addition of nano-Al2O3 were investigated. 14Improvement of in situ LA-ICP-MS U-Pb dating method for carbonate minerals and its application in petroleum geology显示文摘In situ carbonate U-Pb dating is gaining popularity,and it has great potential for application in petroleum geology.However,the low U content(<10μg/g)and high common Pb content of carbonate minerals,along with the uneven distribution of U and Pb and the lack of matrix-matched reference material make carbonate U-Pb dating inaccurate and less successful,which limits the widespread application in geosciences.This study evaluated the limitations of in situ carbonate U-Pb dating and proposed a method to rationally determine the laser ablation parameters of samples by improving the experimental approach based on the laser ablation sector field inductively coupled plasma mass spectrometry(LA-SF-ICP-MS).By setting a different spot size and laser frequency for the reference material and unknown samples in the same session based on the U content of the sample,the ablation craters of the reference material and unknown samples were given the same depth/width ratio,avoiding systematic offset caused by differences in down-hole element fractionation and reducing the consume of reference material.Depending on the heterogeneous distribution of U and Pb contents in carbonate minerals,the method of grid screening and setting ablation spots during screening were used to quickly select domains with high U and low common Pb,which improves the efficiency of setting laser spots and the success rate of dating,as well as reduces the experimental time and economic cost.The accuracy and success rate of carbonate U-Pb dating were effectively improved by improving the experimental method,and the technique was applied to two carbonate samples with low U and high common Pb contents that were difficult to date by traditional methods.The two samples are the saddle dolomite in the central Sichuan Basin and the calcite cement in the sandstone reservoir of the Cretaceous Qingshuihe Formation in the South Junggar Basin.The robust ages have been obtained,which constrains the timing of the diagenetic and hydrocarbon accumulation process in the studied area.Xuesong Lu Lili GUI Weiyan CHEN Shaobo LIU Shitou WU Junjia FAN Qiang LIU Jing SUN Liangliang ZHANG Yang XIAO Wenxia YANG Rizhou CAO 2023Science China Earth Sciences2023,66,12:1
