| 1 | Electrochemical single nucleotide polymorphisms genotyping on surface immobilized three-dimensional branched DNA nanostructure显示文摘An electrochemical assay for single nucleotide polymorphisms (SNPs) genotyping is reported. Although electrochemical method is sensitive for DNA detection on surfaces, the ability of surface assay to precisely recognize DNA hybridization event is sacrificed to some extent due to the crowded confined surfaces environments that disfavor DNA hybridization. In the present study, we employed branched tetrahedron structure probes (TSPs) to replace regular linear single stranded DNA capture probes that were immobilized on solid surfaces. This three-dimensional DNA nanostructure lowers the density of immobilized DNA probes on confined surfaces, providing a hybridization environment that is similar to homogenous solution. | 2 | Production of β-1,3-glucanase and chitinase of two biocon-trol agents and their possible modes of action显示文摘Pichia membranefaciens Hansen and Candida guilliermondii (Cast) Langeronet Guerra are two antagonists of R. stolonifer on harvested nectarine and peach fruits. In this study, β-1,3-glucanase and chitinase activities of the antagonists were induced in vitro and in vivo. The highest β-1, 3-glucanase activity was detected in Lilly-Barnett minimal salt medium supplemented with glucose in combination with CWP of R. stolonifer as a carbon source. The β-1,3-glucanase activity of P. membranefaciens reached the maximum level, being 114.0 SU (specific activity unit), and that of C. guilliermondii reached 103.2 SU. The lowest β-1,3-glucanase activity was observed in the medium containing glucose as sole car-bon source. P. membranefaciens was able to produce signifi-cantly higher levels of chitinase (exochitinase and endochiti-nase) in vitro than C. guilliermondii grown in Czapeck mini-mal medium. | 5 | A nanoresonant gold-aptamer probe for rapid and sensitive detection of thrombin显示文摘Resonance light scattering (RLS) is a sensitive technique for monitoring scattered light induced by extended aggregates of chromophores. It has been widely used to study aggregations for its simple manipulation, high sensitivity and great versatility. Gold nanoparticles generate colorful light-scattering signals due to their unique surface plasmon resonances, hence extraordinary light scattering upon aggregation. In this paper we report a rapid and sensitive method based on gold nanoparticles and DNA aptamer to detect protein biomarkers by RLS. Thiol modified thrombin aptamer was covalently assembled to the surface of gold nanoparticles as nanobio probes. As thrombin has two specific binding sites for its aptamer, it can bridge the well dispersed nanoparticles and lead to a network of particle aggregations. The formation of aggregation ia measured by RLS, and the specific detection of thrombin at nM level is achieved. | 14 | Prognosis of gestational choriocarcinoma diagnosed incidentally during laparoscopy for a presumed cornual pregnancy:a report of five cases显示文摘The clinical presentations of gestational choriocarcinoma vary markedly, and a misdiagnosis could be made in atypical patients if simply relying on clinical features. Laparoscopic resection of uterine mass lesion is rarely used in gestational choriocarcinoma diagnosis because of the fear of heavy blood loss and distant metastasis. Five patients who were preoperatively diagnosed as having cornual pregnancy underwent laparoscopic resection of mass lesion and then proved to have gestational choriocarcinoma based on pathological examinations. Chemotherapy was started within two days after surgery, and the rate of complete remission was 100%. The mean follow-up time was 29.8±19.1 months, and no patient showed signs of relapse.
