Design of Time-constrained Guidance Laws via Virtual Leader Approach显示文摘Guidance problems with flight time constraints are considered in this article.A new virtual leader scheme is used for design of guidance laws with time constraints. The core idea of this scheme is to adopt a virtual leader for real missiles to convert a guidance problem with time constraints to a nonlinear tracking problem, thereby making it possible to settle the problem with a variety of control methods.A novel time-constrained guidance(TCG) law, which can control the flight time of missiles to a prescribed time,is designed by using the virtual leader scheme and stability method. The TCG law is a combination of the well-known proportional navigation guidance(PNG) law and the feedback of flight time error. What's more, this law is free of singularities and hence yields better performances in comparison with optimal guidance laws with time constraints.Nonlinear simulations demonstrate the effectiveness of the proposed law.Zhao Shiyu Zhou Rui Wei Chen Ding Quanxin 2010Chinese Journal of Aeronautics2010,23,1:36
Toxicity of cadmium and its competition with mineral nutrients for uptake by plants:A review显示文摘Cadmium(Cd)is a toxic heavy metal occurring in the environment naturally and is also generated through various anthropogenic sources and acts as a pollutant.Human health is affected by Cd pollution in farmland soils because food is the main source of Cd intake in the non-smoking population. For crops,Cd toxicity may result from a disturbance in uptake and translocation of mineral nutrients and disturbance in plant metabolism, inhibiting plant growth and development. However, plants have Cd tolerance mechanisms, including restricted Cd uptake, decreased Cd root-to-shoot translocation, enhanced antioxidant enzyme activities, and increased production of phytochelatins.Furthermore, optimal supply of mineral nutrients is one of the strategies to alleviate the damaging effects of Cd on plants and to avoid its entry into the food chain. The emerging molecular knowledge contributes to understanding Cd uptake, translocation, and remobilization in plants. In this review,Cd toxicity and tolerance mechanisms, agricultural practices to minimize Cd accumulation,Cd competition with essential elements(calcium, copper, iron, zinc, and manganese),and genes associated with Cd uptake are discussed in detail, especially regarding how these mineral nutrients and genes play a role in decreasing Cd uptake and accumulation in crop plants.Shiyu QIN Hongen LIU Zhaojun NIE Zed RENGEL Wei GAO Chang LI Peng ZHAO 2020Pedosphere2020,30,2:20
Allele-aware chromosome-level genome assembly of Artemisia annua reveals the correlation between ADS expansion and artemisinin yield显示文摘Artemisia annua is the major natural source of artemisinin,an anti-malarial medicine commonly used worldwide. Here,we present chromosome-level haploid maps for two A.annua strains with different artemisinin contents to explore the relationships between genomic organization and artemisinin production.High-fidelity sequencing, optical mapping, and chromatin conformation capture sequencing were used to assemble the heterogeneous and repetitive genome and resolve the haplotypes of A.annua.Approximately 5o,ooo genes were annotated for each haplotype genome, and a triplication event that occurred approximately 58.12million years ago was examined for the first time in this species.A total of 3,903,467-5,193,414 variants(SNPs,indels, and structural variants)were identified in the 1.5-Gb genome during pairwise comparison between haplotypes, consistent with the high heterozygosity of this species.Genomic analyses revealed a correlation between artemisinin concents and the copy number of amorpha-4,11-dienes ynthasegenes. This correlation was further confirmed by resequencing of 36A.annua samples with varied artemisinin contents.Circular consensus sequencing of transcripts facilitated the detection of paralog expression.Collectively, our study provides chromosome-level allele-aware genome assemblies for two A.annua strains and new insights into the biosynthesis of artemisinin and its regulation, which will contribute to conquering malaria worldwide.Baosheng Liao Xiaofeng Shen Li Xiang Shuai Guo Shiyu Chen Ying Meng Yu Liang Dandan Ding Junqi Bai Dong Zhang Tomasz Czechowski Yi Li Hui Yao Tingyu Mai Caroline Howard Chao Sun Haitao Liu Jiushi Liu Jin Pei Jihai Gao Jigang Wang Xiaohui Qiu Zhihai Huang Hongyi Li Ling Yuan Jianhe Wei lan Graham Jiang Xu Boli Zhang Shilin Chen 2022Molecular Plant2022,15,8:9
Impending HRT wave precursors to the Wenchuan M_s8.0 earthquake and methods of earthquake impending prediction by using HRT wave显示文摘We deployed four geo-electric monitoring stations in Sichuan and Yunnan provinces from 2004, using the new generation of equipment (PS-100) and technologies to capture the HRT wave earthquake precursor. Before the Wenchuan Ms8.0 earthquake, we recorded the HRT wave precursor at the only operating station in Hongge (HG, Δ=465 km) and found that significant impending signal had been recorded at the station in the early morning ( 0―5 am) of 12th of May, 2008. The precursor for this earthquake is consistent with precursors recorded for other strong earthquakes. The measured physical properties (geo-resistivity and telluric-current) show tidal wave period oscillations from several days to several months before the earthquakes and the amplitude of such HT oscillation increases significantly towards the occurrence of an earthquake. These HT and RT waves from the epicenter have a causal relationship with the earthquakes that happened several days later. The arrival time of two RT waves is proportional to the distance from the station to the epicenter. The estimated natural decay of the amplitude is correlated with the natural period (T0) of the earthquake fault, which is proportional to the fault length. From this relationship, we can predict the earthquake magnitude. For magnitude 6―9 earthquakes, the natural period is about 1―6 hours. Such oscillation comes from the epicenter area and they can propagate several thousand kilometers in the Earth's crust. Before a strong earthquake in the shallow crust, the conductive pore fluid will experience major changes before the fault rapture. Such fluid change will emit an oscillation in the pore fluid pressure. This is the mechanism for the HRT wave generation. Since the China Earthquake Administration funded the HRT wave short-term earth-quake prediction project in 2003, the first record of HRT precursor wave has been recorded from the 2004-12-26 Sumatra Mw9.0 earthquake with the largest epicentre distance Δ=2900 km. Thereafter, we have captured HRT waves from more than twenty strong earthquakes, which are well-matched and show repeatability, consistency and regularity. All our observation with the HRT waves demonstrate that HRT wave precursors to earthquakes indeed exist. Strong earthquakes can be predicted and short-term and impending earthquake prediction is achievable in the very near future. From all the observations, including the ones at HG station from Wenchuan Ms8.0 earthquake, we conclude that using HRT wave to predict earthquakes is feasible.QIAN FuYe ZHAO BiRu QIAN Wei ZHAO Jian HE ShiGen ZHANG HongKui LI ShiYu LI ShaoKun YAN GuLiang WANG ChengMin SUN ZhenKai ZHANG DongNing LU Jun ZHANG Ping YANG GuoJun SUN JiaLin GUO ChunSheng TANG YuXiong XU JianMing XIA KunTao JU Hang YIN BangHong LI Ming YANG DongSheng QI WeiLuo HE TaiMing GUAN HuaPing ZHAO YuLin 2009Science China Earth Sciences2009,52,10:8
Geological characteristics of shale rock system and shale oil exploration breakthrough in a lacustrine basin:A case study from the Paleogene 1st sub-member of Kong 2 Member in Cangdong sag,Bohai Bay Basin,China显示文摘A deep understanding of the basic geologic characteristics of the fine-grained shale layers in the Paleogene 1 st sub-member of Kong 2 Member(Ek_2~1) in Cangdong sag, Bohai Bay Basin, is achieved through observation of 140 m continuous cores and systematic analysis of over 1 000 core samples from two wells. Basic geological conditions for shale oil accumulation are proposed based on the unconventional geological theory of oil and gas. The shale rock system mainly developed interbedded formation of felsic shale, calcareous and dolomitic shale and carbonates; high quality hydrocarbon source rock formed in the stable and closed environment is the material base for shale oil enrichment; intergranular pores in analcite, intercrystalline pores in dolomite and interlayer micro-fractures make tight carbonate, calcareous and dolomitic shale and felsic shale effective reservoirs, with brittle mineral content of more than 70%; high abundance laminated shale rock in the lower section of Ek_2~1 is rich in shale oil, with a total thickness of 70 m, burial depth between 2 800 to 4 200 m, an average oil saturation of 50%, a sweet spot area of 260 km^2 and predicted resources of over 5×10~8 t. Therefore, this area is a key replacement domain for oil exploration in the Kongdian Formation of the Cangdong sag. At present, the KN9 vertical well has a daily oil production of 29.6 t after fracturing with a 2 mm choke. A breakthrough of continental shale oil exploration in a lacustrine basin is expected to be achieved by volume fracturing in horizontal wells.ZHAO Xianzheng ZHOU Lihong PU Xiugang JIN Fengming HAN Wenzhong XIAO Dunqing CHEN Shiyue SHI Zhannan ZHANG Wei YANG Fei 2018Petroleum Exploration and Development2018,45,3:8
Micro-droplet Digital Polymerase Chain Reaction and Real-Time Quantitative Polymerase Chain Reaction Technologies Provide Highly Sensitive and Accurate Detection of Zika Virus显示文摘The establishment of highly sensitive diagnostic methods is critical in the early diagnosis and control of Zika virus(ZIKV)and in preventing serious neurological complications of ZIKV infection. In this study, we established micro-droplet digital polymerase chain reaction(ddPCR) and real-time quantitative PCR(RT-qPCR) protocols for the detection of ZIKV based on the amplification of the NS5 gene. For the ZIKV standard plasmid, the RT-qPCR results showed that the cycle threshold(Ct) value was linear from 10~1 to 10~8 copy/l L, with a standard curve R^2 of 0.999 and amplification efficiency of 92.203%;however, a concentration as low as 1 copy/l L could not be detected. In comparison with RT-qPCR, the dd PCR method resulted in a linear range of 10~1–10~4 copy/l L and was able to detect concentrations as low as 1 copy/l L. Thus, for detecting ZIKV from clinical samples, RT-qPCR is a better choice for high-concentration samples(above 10~1 copy/l L),while ddPCR has excellent accuracy and sensitivity for low-concentration samples. These results indicate that the ddPCR method should be of considerable use in the early diagnosis, laboratory study, and monitoring of ZIKV.Yuan Hui Zhiming Wu Zhiran Qin Li Zhu Junhe Liang Xujuan Li Hanmin Fu Shiyu Feng Jianhai Yu Xiaoen He Weizhi Lu Weiwei Xiao Qinghua Wu Bao Zhang Wei Zhao 2018Virologica Sinica2018,33,3:6
Reservoir Characteristics and Preservation Conditions of Longmaxi Shale in the Upper Yangtze Block, South China显示文摘The Upper Ordovician-Lower Silurian Longmaxi Shale in the Upper Yangtze block represents one of the most important shale gas plays in China. The shale composition, porosity, organic thermal maturity, and methane sorption were investigated at the Qilongcun section in the Dingshan area, southeastern Sichuan Basin. The results show that the Upper Ordovician-Lower Silurian Longmaxi Shale contains:(1) sapropelic I organic matter;(2) a 40-m thick bedded sequence where total organic carbon(TOC) content is > 2%;(3) a 30-m thick layer at the base of the Longmaxi Shale with a brittle mineral content higher than 50%; and(4) a mean methane adsorption capacity of 1.80 cm^3/g(7 MPa pressure). A positive correlation between TOC and sorbed gas indicates that organic matter content exerts an important control on methane storage capacity. Based on the analysis of the shale reservoir characteristics, the lower member of the Longmaxi Shale can thus be considered a favorable stratum for shale gas exploration and exploitation. It has similar reservoir characteristics with the Longmaxi Shale in the Jiaoshiba area tested with a high-yield industrial gas flow. However, based on tectonic analysis, differences in the level of industrial gas flow between the low-yield study area and the high-yield Jiaoshiba area may result from different tectonic preservation conditions. Evidence from these studies indicates the shale gas potential of the Longmaxi Shale is constrained by the reservoir and preservation conditions.RAN Bo LIU Shugen Luba JANSA SUN Wei YANG Di WANG Shiyu YE Yuehao Christopher XIAO ZHANHG Jian ZHAI Cangbo LUO Chao ZHANG Changjun 2016Acta Geologica Sinica(English Edition)2016,90,6:6
Ionomycin ameliorates hypophosphatasia via rescuing alkaline phosphatase deficiency-mediated L-type Ca^(2+) channel internalization in mesenchymal stem cells显示文摘The loss-of-function mutations in the ALPL result in hypophosphatasia(HPP), an inborn metabolic disorder that causes skeletal mineralization defects. In adults, the main clinical features are early loss of primary or secondary teeth, osteoporosis, bone pain, chondrocalcinosis, and fractures. However, guidelines for the treatment of adults with HPP are not available. Here, we show that ALPL deficiency caused a reduction in intracellular Ca2+ influx, resulting in an osteoporotic phenotype due to downregulated osteogenic differentiation and upregulated adipogenic differentiation in both human and mouse bone marrow mesenchymal stem cells(BMSCs). Increasing the intracellular level of calcium in BMSCs by ionomycin treatment rescued the osteoporotic phenotype in alpl+/- mice and BMSC-specific(Prrx1-alpl-/-) conditional alpl knockout mice. Mechanistically, ALPL was found to be required for the maintenance of intracellular Ca2+ influx, which it achieves by regulating L-type Ca2+ channel trafficking via binding to the α2δsubunits to regulate the internalization of the L-type Ca2+ channel. Decreased Ca2+ flux inactivates the Akt/GSK3β/β-catenin signaling pathway, which regulates lineage differentiation of BMSCs. This study identifies a previously unknown role of the ectoenzyme ALPL in the maintenance of calcium channel trafficking to regulate stem cell lineage differentiation and bone homeostasis. Accelerating Ca2+ flux through L-type Ca2+ channels by ionomycin treatment may be a promising therapeutic approach for adult patients with HPP.Bei Li Xiaoning He Zhiwei Dong Kun Xuan Wei Sun Li Gao Shiyu Liu Wenjia Liu Chenghu Hu Yimin Zhao Songtao Shi Yan Jin 2020Bone Research2020,8,2:4
Transcriptome analysis of purple pigment formation in Colocasia esculenta显示文摘Taro(Colocasia esculenta(L.)Schott)is an important crop in Africa,Southeast Asia, and subtropics and is used as a food and medicine. The purple color pigmentation is an appealing character in taro.We sampled taro corms of the cultivar‘Lipu Taro’at four developmental stages, including LPYS1(without purple pigment,50 days of development(DOD)),LPYS2(very few purple pigments,75 DOD),LPYS3(moderate purple pigments,115 DOD)and LPYS4(high purple pigments,205 DOD).The purpose of our study was to identify the key genes underpinning the purple pigmentation in taro based on RNA-sequencing.Through RNA-Seq,6453 differentially expressed transcripts(DETs)were identified between purple and non-purple pigmented samples. We identified 41 and 12 flavonoid and anthocyanin related DETs transcripts, respectively. These DETs were upregulated at LPYS2,LPYS3,and LPYS4 as compared to LPYS1,indicating their positive contribution to the color formation in taro. Moreover,we identified several DETs encoding for transcription factors, including MYB and bHLH,known to be major regulators of structural genes involved in the flavonoid-anthocyanin pathway. Finally,we reported several plant hormones(ethylene, auxin, gibberellin, jasmonic acid, and cytokinin)related DETs,which are predicted to play important roles in the corm coloration.Different regulation of transcripts representing the flavonoid-anthocyanin biosynthesis pathway, plant hormone transduction pathway, and transcription factors may have key roles in purple pigmentation in taro. Our findings will facilitate future research on improving the quality and appeal of taro.FANGLIAN HE WEIQING DONG SHAOLONG WEI ZUYANG QIU JINGLI HUANG HUIPING JIANG SHIYU HUANG LILI LIU 2021BIOCELL2021,45,3:2

Formation mechanisms, potentials and exploration practices of large lithologic gas reservoirs in and around an intracratonic rift:Taking the Sinian-Cambrian of Sichuan Basin as an example显示文摘By examining structures, sediments, reservoirs and accumulation assemblages in the Deyang-Anyue rift and its surrounding area, four new understandings are obtained.First, during the initiation period of Deyang-Anyue rift, multiple groups of faults developed in the rift due to the effect of tensile force, bringing about multiple mound and shoal belts controlled by horsts in the second member of the Sinian Dengying Formation;in the development stage of the rift, the boundary faults of the rift controlled the development of mound and shoal belts at the platform margin in the fourth member of Dengying Formation;during the shrinkage period of the rift, platform margin grain shoals of the Cambrian Canglangpu Formation developed in the rift margin.Second, four sets of large-scale mound and shoal reservoirs in the second member of Dengying Formation, the fourth member of Dengying Formation,Canglangpu Formation and Longwangmiao Formation overlap with several sets of source rocks such as Qiongzhusi Formation source rocks and Dengying Formation argillaceous limestone or dolomite developed inside and outside the rift, forming good source-reservoir-cap rock combinations;the sealing of tight rock layers in the lateral and updip direction results in the formation model of large lithologic gas reservoirs of oil pool before gas, continuous charging and independent preservation of each gas reservoir.Third, six favorable exploration zones of large-scale lithologic gas reservoirs have been sorted out through comprehensive evaluation, namely, mound and shoal complex controlled by horsts in the northern part of the rift in the second member of Dengying Formation, isolated karst mound and shoal complex of the fourth member of Dengying Formation in the south of the rift, the superimposed area of multi-stage platform margin mounds and shoals of the second and fourth members of Dengying Formation and Canglangpu Formation in the north slope area, the platform margin mounds and shoals of the second and fourth members of Dengying Formation in the west side of the rift, the platform margin mound and shoal bodies of the fourth member of Dengying Formation in the south slope area, etc.Fourth,Well Pengtan-1 drilled on the mound and shoal complex controlled by horsts of the second member of Dengying Formation in the rift and Well Jiaotan-1 drilled on the platform margin mound and shoal complex of the North Slope have obtained high-yield gas flows in multiple target layers, marking the discovery of a new gas province with reserves of(2-3)×10^(12) m^(3).This has proved the huge exploration potential of large lithologic gas reservoir group related to intracratonic rift.WEI Guoqi YANG Wei XIE Wuren SU NanX IE Zengye ZENG Fuying MA Shiyu JIN Hui WANG Zhihong ZHU Qiuying HAO Cuiguo WANG Xiaodan 2022Petroleum Exploration and Development2022,49,3:2
GTL or LNG:Which is the best way to monetize 'stranded' natural gas?显示文摘A large portion of world's natural gas reserves are 'stranded' resources, the drive to monetize these resources leads to the development of gas-to-liquids(GTL)and liquefied natural gas (LNG)technologies.LNG has the advantage of having been developed for the past 40 years and having an excellent safety record.GTL on the other hand is another option with substantial benefits, but its development stage and commercial viability are far behind LNG.This paper presents a techno-economic comparison of GTL with LNG,including technical development, plant efficiency, market potential for the products, and capital cost for the infrastructure. The aim is to give an overall view on both LNG and GTL and provide a perspective on the profitability of these two technologies.Dong Lichun Wei Shun'an Tan Shiyu Zhang Hongjing 2008Petroleum Science2008,5,4:2
Dynamic changes of postprandial plasma metabolites after intake of corn-soybean meal or casein-starch diets in growing pigs显示文摘Background: Postprandial nutrients utilization and metabolism of a certain diet is a complicated process. The metabolic feature of pigs after intake of corn-soybean meal or casein-starch diets are largely unknown. Therefore, this study was conducted to investigate the dynamic postprandial changes of plasma metabolic profile using growing pigs using metabolomics. Methods: Twenty-four growing pigs with average initial body weight(BW) about 30 kg were placed in metabolic cages and then fitted with precaval vein catheters. Pigs were fed daily 4% of initial body weight. Two experimental diets were included:(i) a starch-casein based purified diet(PD) and(ii) a common corn-soybean meal diet(CD).Plasma was collected before feeding and 0.5 h, 1 h, 2 h, 4 h, 8 h after feeding. Results: In both diets, compared to prior to feeding, the concentrations of glucose, most amino acids, metabolites such as 5-aminopentanoic acid, pipecolic acid, ornithine and 5-hydroxy-L-tryptophan were significantly increased in plasma during the first hour, whereas the concentrations of plasma triglycerides, glutamate, glycine, palmitelaidic acid, 13-HODE and oleic acid were decreased in the first hour. Compared with PD group, concentration of plasma leucine and isoleucine declined at 30 min in CD group. Plasma linoleic acid, sphingosine and many dipeptides were significantly higher in pigs fed CD.Conclusion: Most significant metabolic changes occurred during the first hour after feeding and then became relatively stable after 2 h in both diets. These results show a broad scope picture of postprandial changes in plasma metabolites after intake of PD and CD and could be a reference for further nutrition intervention as well as the design of nutritional studies.Tiantian Li Shimeng Huang Juntao Li Hu Liu Wei Wang Na Li Meng Shi Shiyu Tao Shuai Zhang Zhen Li Junjun Wang 2019Journal of Animal Science and Biotechnology2019,10,4:2
Performance evaluation of deep learning-based post-processing and diagnostic reporting system for coronary CT angiography: a clinical comparative study显示文摘To the Editor:Coronary computed tomography angiography(CCTA)has been increasingly, widely performed for diagnosing coronary artery, disease,lAnatomical diagnosis, that is,stenosis grading,is stillthe main diagnostic index provided'by most CCTA tests.Post-processing and interpretation of stenosis are 2 essential'steps that need to be performed bycardiovascular imaging professionals from scan completion to diagnosis conclusion, which is repetitive and time-consuming,taking an average of 30 minutes each case in China and becoming the bottleneck and gradually creating an imbalance between supply and demand. In ine with the rapid development of artificial intelligence(Al)technology in recent years,it has been expected to solve these specific problems. We developed an AI system for automating post-processing and diagnostic reporting of CCTA data using deep learning algorithms to establishanew1-clickworkflowforeverydayuse,namely,CCTA-AI(Figure 1).To further assess its capabilities, this study intends to answer 2 following questions:To what extent can it improve the efficiency of post-processing?To what extent can CCTA-AI detect and calculate coronary artery stenosis due to each atherosclerotic plaque?Nan Luo Yi He Jitao Fan Ning Guo Guang Yang Yuanyuan Kong Jianyong Wei Tao Bi Jie Zhou Jiaxin Cao Xianjun Han Fang Li Shiyu Zhang Rujing Sun Zhaozhao Wang Tian Ma Lixue Xu Hui Chen Hongwei Li Zhenchang Wang Zhenghan Yang 2022Chinese Medical Journal2022,135,19:2
Human-animal interactions and bat coronavirus spillover potential among rural residents in Southern China显示文摘Human interaction with animals has been implicated as a primary risk factor for several high impact zoonoses, including many bat-origin viral diseases. However the animal-to-human spillover events that lead to emerging diseases are rarely observed or clinically examined, and the link between specific interactions and spillover risk is poorly understood. To investigate this phenomenon,we conducted biological-behavioral surveillance among rural residents in Yunnan,Guangxi, and Guangdong districts of Southern China, where we have identified a number of SARS-related coronaviruses in bats.Serum samples were tested for four bat-borne coronaviruses using newly developed enzyme-linked immunosorbent assays(ELISA).Survey data were used to characterize associations between human-animal contact and bat coronavirus spillover risk.A total of 1,596 residents were enrolled in the study from 2015 to 2017.Nine participants(0.6%)tested positive for bat coronaviruses.265(17%)participants reported severe acute respiratory infections(SARI)and/or influenza-like illness(ILI)symptoms in the past year, which were associated with poultry,carnivore,rodent/shrew,or bat contact, with variability by family income and district of residence. This study provides serological evidence of bat coronavirus spillover in rural communities in Southern China. The low seroprevalence observed in this study suggests that bat coronavirus spillover is a rare event.Nonetheless, this study highlights associations between human-animal interaction and zoonotic spillover risk. These findings can be used to support targeted biological behavioral surveillance in high-risk geographic areas in order to reduce the risk of zoonotic disease emergence.Li Hongying Mendelsohn Emma Zong Chen Zhang Wei Hagan Emily Wang Ning Li Shiyue Yan Hong Huang Huimin Zhu Guangjian Ross Noam Chmura Aleksei Terry Philip Fielder Mark Miller Maureen Shi Zhengli Daszak Peter 2019Biosafety and Health2019,1,2:2

Tumor-repopulating cell-derived microparticles elicit cascade amplification of chemotherapy-induced antitumor immunity to boost anti-PD-1 therapy显示文摘Immune checkpoint blockade(ICB)therapy, particularly antibodies targeting the programmed death receptor 1(PD-1)and its ligand(PD-L1),has revolutionized cancer treatment. However, its efficacy as a standalone therapy remains limited.Although ICB therapy in combination with chemotherapy shows promising therapeutic responses, the challenge lies in amplifying chemotherapy-induced antitumor immunity effectively. This relies on efficient drug delivery to tumor cells and robust antigen presentation by dendritic cells(DCs).Here,we developed tumor-repopulating cell(TRC)-derived microparticles with exceptional tumor targeting to deliver doxorubicin(DOX@3D-MPs)for improve anti-PD-1 therapy.DOX@3D-MPs effectively elicit immunogenic tumor cell death to release sufficient tumor antigens.Heat shock protein 70(HSP70)overexpressed in DOX@3D-MPs contributes to capturing tumor antigens, promoting their phagocytosis by DCs,and facilitating DCs maturation, leading to the activation of CD8+T cells.DOX@3D-MPs significantly enhance the curative response of anti-PD-1 treatment in large subcutaneous H22 hepatoma, orthotopic 4T1 breast tumor and Panc02 pancreatic tumor models. These results demonstrate that DOX@3D-MPs hold promise as agents to improve the response rate to ICB therapy and generate long-lasting immune memory to prevent tumor relapse.Nana Bie Tuying Yong Zhaohan Wei Qingle Liang Xiaoqiong Zhang Shiyu Li Xin Li Jianye Li Lu Gan Xiangliang Yang 2023Signal Transduction and Targeted Therapy2023,8,11:1
Evaluation of the Mechanisms Underlying Amino Acid and Microbiota Interactions in Intestinal Infections Using Germ-Free Animals显示文摘Intestinal infectious diseases refer to the inflammatory changes in the intestinal tract caused by pathogens(including bacteria, viruses, fungi, protozoa,or parasites)or their toxic products.A large number of microorganisms colonize the intestinal tract of healthy people, which together with the intestinal epithelium constitute the biological barrier of the intestinal tract to resist infectious diseases. As an“invisible organ,”the intestinal flora is closely related to human nutrition metabolism and intestinal infections.A variety of intestinal flora participates in the nutritional metabolism of amino acids, and the small molecular substances produced by the amino acid metabolism through the intestinal flora can enhance intestinal immunity and resist bacterial infections. In turn, amino acids can also regulate the composition of the intestinal flora, maintain the steady-state of the intestinal flora, protect the intestinal barrier, and inhibit colonization by pathogenic bacteria. As a model animal with a clear microbial background,germ-free(GF)animals can clarify the mechanisms of interactions between intestinal microbes and amino acid metabolism in intestinal infections by combining genetic engineering technology and multi-omics studies. This article reviews related researches on the involvement of intestinal microbes in host amino acid metabolism and resistance to intestinal infections and discusses the advantages of GF animal models for studying the underlying mechanisms. The GF animal model is helpful to further study the intervention effects of amino acid metabolism of targeted intestinal flora on intestinal infections.Yapeng Yang Peng Bin Shiyu Tao Guoqiang Zhu Zhifeng Wu Wei Cheng Wenkai Ren Hong Wei 2021Infectious Microbes & Diseases2021,3,2:1

Mechanisms of allicin exposure for the sludge fermentation enhancement:Focusing on the fermentation processes and microbial metabolic traits显示文摘As a frequently used product with antimicrobial activity, consumed allicin might be dis-charged and concentrated in waste-activated sludge(WAS).However, the influence of al-licin(as an exogenous pollutant)on WAS fermentation has not been clearly revealed. This study aimed to disclose the impacts of allicin on volatile fatty acid(VFA)generation dur-ing WAS fermentation. The results showed that the appropriate presence of allicin(10 mg/g TSS)significantly enhanced the VFA yield(1894 versus 575 mg COD/L in the control)with increased acetate proportion(24.3%).Further exploration found that allicin promoted WAS solubilization, hydrolysis and acidification simultaneously.Metagenomic analysis revealed that the key genes involved in extracellular hydrolysis metabolism(i.e.,CAZymes),mem-brane transport(i.e.,gtsA and ytfT),substrate metabolism(i.e.,yhdR and pfkC)and fatty acid synthesis(i.e.,accA and accD)were all highly expressed.Allicin also induced the bacteria to produce more signalling molecules and regulate cellular functions, thereby enhancing the microbial adaptive and regulatory capacity to the unfavourable environment. Moreover, the variations in fermentative microbes and their contributions to the upregulation of func-tional genes(i.e.,ytfR,gltL,INV,iolD and pflD)for VFA generation were disclosed.Overall, the simultaneous stimulation of functional microbial abundances and metabolic activities contributed to VFA production in allicin-conditioned reactors.Feng Wang Jingyang Luo Shiyu Fang Wenxuan Huang Yunqi Zhang Le Zhang Xiaoshi Cheng Wei Du Fang Fang Jiashun Cao Yang Wu 2022Journal of Environmental Sciences2022,34,5:1
