1Pharmacological effects of Astragaloside IV: a literature review显示文摘OBJECTIVE: To review the pharmacological effects and mechanisms of action of Astragaloside IV in Huangqi (Radix Astragali Mongolici). METHODS: Aticles focusing on Astragaloside IV in English and Chinese in databases were collected and reviewed in order to summarize the latest extraction separation, pharmacokinetics, and the pharmacological effects of astrageloside IV. RESULTS: Protective effects of Astrageloside IV on the cardiovascular system, immune, digestive, nervous system were identified, and the action mechanisms were associated with regulation of the calcium balance, anti-oxydant, antiapoptosis, antivirus, and so on. 3Prognostic value of M30/M65 for outcome of hepatitis B virus-related acute-on-chronic liver failure显示文摘AIM:To determine the prognostic value of circulating indicators of cell death in acute-on-chronic liver failure(ACLF)patients with chronic hepatitis B virus(HBV)infection as the single etiology.METHODS:Full length and caspase cleaved cytokeratin 18(detected as M65 and M30 antigens)represent circulating indicators of necrosis and apoptosis.M65and M30 were identified by enzyme-linked immunosorbent assay in 169 subjects including healthy controls(n=33),patients with chronic hepatitis B(CHB,n=55)and patients with ACLF(n=81).According to the 3-mo survival period,ACLF patients were defined as having spontaneous recovery(n=33)and non-spontaneous recovery which included deceased patients and those who required liver transplantation(n=48).RESULTS:Both biomarker levels significantly increased gradually as liver disease progressed(for M65:P<0.001 for all;for M30:control vs CHB,P=0.072;others:P<0.001 for all).In contrast,the M30/M65 ratio was significantly higher in controls compared with CHB patients(P=0.010)or ACLF patients(P<0.001).In addition,the area under receiver operating characteristic curve(AUC)analysis demonstrated that both biomarkers had diagnostic value(AUC≥0.80)in identifying ACLF from CHB patients.Interestingly,it is worth noting that the M30/M65 ratio was significantly different between spontaneous and non-spontaneous recovery in ACLF patients(P=0.032).The prognostic value of the M30/M65 ratio was compared with the Model for End-Stage Liver Disease(MELD)and Child-Pugh scores at the 3-mo survival period,the AUC of the M30/M65ratio was 0.66 with a sensitivity of 52.9%and the highest specificity of 92.6%(MELD:AUC=0.71;sensitivity,79.4%;specificity,63.0%;Child-Pugh:AUC=0.77;sensitivity,61.8%;specificity,88.9%).CONCLUSION:M65 and M30 are strongly associated with liver disease severity.The M30/M65 ratio may be a potential prognostic marker for spontaneous recovery in patients with HBV-related ACLF.Su-Jun Zheng Shuang Liu Mei Liu Malcolm A McCrae Jun-Feng Li Yuan-Ping Han Chun-Hui Xu Feng Ren Yu Chen Zhong-Ping Duan 2014World Journal of Gastroenterology2014,20,9:13

5Apatite Fission Track Evidence of Uplift Cooling in the Qiangtang Basin and Constraints on the Tibetan Plateau Uplift显示文摘The Qiangtang basin is located in the central Tibetan Plateau. This basin has an important structural position,and further study of its tectonic and thermal histories has great significance for understanding the evolution of the Tibetan Plateau and the hydrocarbon potential of marine carbonates in the basin. This study focuses on low temperature thermochronology and in particular conducted apatite fission track analysis. Under constraints provided by the geological background,the thermal history in different tectonic units is characterized by the degree of annealing of samples,and the timing of major(uplift-erosion related) cooling episodes is inferred. The cooling history in the Qiangtang basin can be divided into two distinct episodes. The first stage is mainly from the late Early Cretaceous to the Late Cretaceous(69.8 Ma to 108.7 Ma),while the second is mainly from the MiddleLate Eocene to the late Miocene(10.3 Ma to 44.4 Ma). The first cooling episode records the uplift of strata in the central Qiangtang basin caused by continued convergent extrusion after the BangongNujiang ocean closed. The second episode can be further divided into three periods,which are respectively 10.3 Ma,22.6–26.1 Ma and 30.8–44.4 Ma. The late Oligocene-early Miocene(22.6–26.1 Ma) is the main cooling period. The distribution and times of the earlier uplift-related cooling show that the effect of extrusion after the collision between Eurasian plate and India plate obviously influenced the Qiangtang basin at 44.4 Ma. The Qiangtang basin underwent compression and started to be uplifted from the middle-late Eocene to the early Oligocene(45.0–30.8 Ma). Subsequently,a large-scale and intensive uplift process occurred during the late Oligocene to early Miocene(26.1–22.6 Ma) and the basin continued to undergo compression and uplift up to the late Miocene(10.3 Ma). Thus,uplift-erosion in the Qiangtang basin was intensive from 44.5 Ma to about 10 Ma. The timing of cooling in the second episode shows that the uplift of the Qiangtang basin was caused by the strong compression after the collision of the Indian plate and Eurasian plate. On the whole,the new apatite fission-track data from the Qiangtang basin show that the Tibetan Plateau started to extrude and uplift during 45–30.8 Ma. 6Experimental study on 830 MPa grade pipeline steel containing chromium显示文摘The diversity of microstructure and properties of 830 MPa grade pipeline steel containing chromium was investigated by optical microscope and transmission electron microscopy. The main microstructures were multiple configurations, containing lath bainite and granule bainite. Mechanical properties test results showed that the yield strength and tensile strength improved with increasing chromium content. The toughness and elongation decreased at the same time, so temper process was introduced. 7Protective effects of extracts of Schisandra chinensis stems against acetaminophen-induced hepatotoxicity via regulation of MAPK and caspase-3 signaling pathways显示文摘The present study was designed to evaluate protective activity of an ethanol extract of the stems of Schisandra chinensis(SCE) and explore its possible molecular mechanisms on acetaminophen(APAP) induced hepatotoxicity in a mouse model. The results of HPLC analysis showed that the main components of SCE included schisandrol A, schisandrol B, deoxyschisandrin, schisan--drin B, and schisandrin C and their contents were 5.83, 7.11, 2.13, 4.86, 0.42 mg·g^(-1), respectively. SCE extract was given for 7 con--secutive days before a single hepatotoxic dose of APAP(250 mg·kg^(-1)) was injected to mice. Our results showed that SCE pretreatment ameliorated liver dysfunction and oxidative stress, which was evidenced by significant decreases in aspartate transaminase(AST), alanine aminotransferase(ALT), malondialdehyde(MDA) contents and elevations in reduced glutathione(GSH) and superoxide dismutase(SOD) levels. These findings were associated with the result that the SCE pretreatment significantly decreased expression levels of 4-hydroxynonenal(4-HNE) and 3-nitrotyrosine(3-NT). SCE also significantly decreased the expression levels of Bax, mitogen-activated protein kinase(MAPK), and cleaved caspase-3 by APAP exposure. Furthermore, supplementation with SCE suppressed the expression levels of inducible nitric oxide synthase(iNOS) and cyclooxygenase-2(COX-2), suggesting alleviation of inflammatory response. 8Changes of Bacterial Community Structure in Copper Mine Tailings After Colonization of Reed (Phragmites communis)显示文摘Soil samples were collected from both bare and vegetated mine tailings to study the changes in bacterial communities and soil chemical properties of copper mine tailings due to reed (Phragmites communis) colonization. The structures of bacterial communities were investigated using culture-independent 16S rRNA gene sequencing method. The bacterial diversity in the bare mine tailing was lower than that of the vegetated mine tailing. The former was dominated by sulfur metabolizing bacteria, whereas the latter was by nitrogen fixing bacteria. The bare mine tailing was acidic (pH = 3.78), whereas the vegetated mine tailing was near neutral (pH = 7.28). The contents of organic matter, total nitrogen, and ammonium acetate-extractable potassium in vegetated mine tailings were significantly higher than those in the bare mine tailings (P < 0.01), whereas available phosphorus and electrical conductivity were significantly lower than those in the bare mine tailings (P < 0.01). The results demonstrated that 16S rRNA gene sequencing could be successfully used to study the bacterial diversity in mine tailings. The colonization of the mine tailings by reed significantly changed the bacterial community and the chemical properties of tailings. 10Sini powder ameliorates the inflammatory response in rats with stress-induced non-alcoholic fatty liver disease by inhibiting the nuclear factor kappa-B/pyrin domain-containing protein 3 pathway显示文摘OBJECTIVE: To evaluate the effectiveness of Sini powder for the treatment of non-alcoholic fatty liver disease(NAFLD) in rats and the molecular mechanisms involved.METHODS: A rat model of stress-induced NAFLD was established by a combination of long-term tethering and feeding of a high-fat, high-calorie diet. These rats were then intragastrically administered with either simvastatin, Sini powder, or vehicle for 1 week. The body mass and field test scores for each group were recorded weekly. Serum aspartate aminotransferase and alanine aminotransferase activities, and triglyceride, total cholesterol,and free fatty acid concentrations were measured.Liver tissue histopathology was examined on hematoxylin and eosin-stained paraffin sections and oil red O-stained frozen sections. The hepatic m RNA expression of nuclear factor kappa-B(NF-κB),NOD-, LRR-, and pyrin domain-containing protein 3(NLRP3), apoptosis-associated speck-like protein containing CARD(ASC), and caspase-1 were measured by reverse transcription-polymerase chain reaction(RT-PCR). The hepatic protein concentrations of NF-κB and NLRP3, ASC, caspase-1, interleukin-1β(IL-1β), interleukin-6(IL-6), and the serum concentrations of IL-1β and IL-6 were measured by enzyme-linked immunosorbent assay.RESULTS: Compared with the Blank group, rats in the Compound model group showed significant pathologic manifestations of NAFLD, and the expression of NF-κB, NLRP3, ASC, caspase-1, IL-1βand IL-6 were significantly higher(all P < 0.01). Both simvastatin and Sini powder significantly ameliorated the NAFLD pathology and the abnormal expression of NF-κB, NLRP3, ASC, caspase-1, IL-1β, and IL-6(all P < 0.01).CONCLUSION: Sini powder inhibits the inflamma-tory response in rats with NAFLD, which is mediated by NF-κB/NLRP3, IL-1β, and IL-6, reduces the effects of psychological stress, and improves lipid metabolism. 12Efficacy and safety of Xinglouchengqi decoction for acute ischemic stroke with constipation:study protocol for a randomized con-trolled trial显示文摘OBJECTIVE: To investigate the efficacy and safety of Xinglouchengqi(XLCQ) decoction in treatment of acute ischemic stroke with constipation.METHODS: In this prospective, multicenter, assessor-blinded, randomized controlled trial, 360 eligible participants will be randomized to the XLCQ group or the control group. Participants in the XLCQ group will receive XLCQ decoction, while those in the control group will undergo clysis therapy using glycerin enemas or oral administration of lactulose solution. Both groups will undergo a treatment period of(5 ± 2) d and a 1-month follow-up.The primary outcome measure will be the Constipation Scale score. The secondary outcome measures will include scores on the National Institutes of Health Stroke Scale, the Traditional Chinese Medicine(TCM) Stroke Scale, the Diagnostic Scale for TCM Syndromes of Ischemic Stroke and TCM Scale for Syndrome of Phlegm-heat and Fu-organ Excess.Therapeutic mechanism outcomes and safety outcomes will also be assessed. Assessments will be conducted at baseline, at the end of the treatment period, and at the follow-up. 13Genetic aberration in primary hepatocellular carcinoma:correlation between p53 gene mutation and loss-of-heterozygosity on chromosome 16q21-q23 and 9p21-p23显示文摘To elucidate the molecular pathology underlying the development of hepatocellular carcinoma (HCC), we used 41 highly polymorphic microsatellite markers to examine 55 HCC and corresponding non-tumor liver tissues on chromosome 9, 16 and 17. Loss-of-heterozygosity (LOH) is observed with high frequency on chromosomal region 17p13 (36/55, 65%), 9p21-p23 (28/55, 51%), 16q21-q23 (27/55, 49%) in tumors. Meanwhile, microsatellite instability is rarely found in these microsatellite loci. Direct sequencing was performed to detect the tentative mutation of tumor suppressor genes in these regions: p53, MTS1/p16, and CDH1/E-cadherin. Within exon 5-9 of p53 gene, 14 out of 55 HCC specimens (24%) have somatic mutations, and nucleotide deletion of this gene is reported in HCC for the first time. Mutation in MTS1/pl6 is found only in one tumor case. We do not find mutations in CDH1/E-cadherin. Furthermore, a statistically significant correlation is present between p53 gene mutation and loss of chromosome region 16q21q23 and 9p21-p23, which indicates that synergism between p53 inactivation and deletion of 16q21-q23 and 9p21-p23 may play a role in the pathogenesis of HCC. 14Development and utilization of a new chemically-induced soybean library with a high mutation density显示文摘Mutagenized populations have provided important materials for introducing variation and identifying gene function in plants. In this study, an ethyl methanesulfonate(EMS)-induced soybean(Glycine max) population,consisting of 21,600 independent M_2 lines, was developed.Over 1,000 M_(4(5))families, with diverse abnormal phenotypes for seed composition, seed shape, plant morphology and maturity that are stably expressed across different environments and generations were identified. Phenotypic analysis of the population led to the identification of a yellow pigmentation mutant, gyl, that displayed significantly decreased chlorophyll(Chl) content and abnormal chloroplast development. Sequence analysis showed that gyl is allelic to Minn Gold, where a different single nucleotide polymorphism variation in the Mg-chelatase subunit gene(ChlI1a) results in golden yellow leaves. A cleaved amplified polymorphic sequence marker was developed and may be applied to marker-assisted selection for the golden yellow phenotype in soybean breeding. 16Research advance in failure risk and local strength failure for high arch dams显示文摘To develop national economy and use the water resources and hydropower resources sufficiently,a lot of high arch dams,with the height of more than 200 m,have been and will be built in China.Although arch dams have good mechanical behavior,there is still failure possibility due to the huge water pressure and high stress level in dam,complex topographic and geologic conditions,formidable environment and high intensity earthquake.As one of the three main aspects concerning the safety of high arch dam,the study on global destruction,has been elaborated in the literatures,and research advance in the other two aspects,namely the failure risk and local damage of high arch dams,will be reviewed in this paper.In recent years,the failure risk of high arch dams has been investigated,and the model for identifying dam failure risk factors has been established.It is shown that the foundation deterioration and strong earthquake are the major risk sources for high dam failure.With the fault tree method,the failure mode and failure probability of high arch dams are studied,and the principle for determining failure mode and the method of calculating failure probability are proposed.Meanwhile,the determination principle of acceptable risk standard for high arch dam was proposed,and the acceptable risk rate and the acceptable standard value of various risk losses were given.For the local damage of arch dam,it is pointed out that the local damage belongs to the strength failure at material level.The study on local failure mechanism of arch dam is reviewed,based on the theories that from traditional strength theory to damage mechanics and meso-mechanics theory.Aiming at the cracking,the main pattern of local failure of high concrete dam,the research advances in the analysis methods and cracking criteria for smeared crack model and discrete crack model are summarized,and the research findings of preventive measures for local failure are shown.REN QingWen LI Qiang LIU Shuang 2012Chinese Science Bulletin2012,57,36:5

Based on the TCM Systems Pharmacology Analysis Platform(TCMSP database) and a text mining tool(GoPubMed database), we predicted and screened the active components of Bulao Elixir and its therapeutic targets for AD. Using the Database for Annotation, Visualization and Integrated Discovery(DAVID), we obtained the targets for AD. Cytoscape software was used to establish a network map of the active component-target and target-pathway of Bulao Elixir. Gene function, related biological processes and signaling pathways were analyzed using the DAVID database.RESULTS: Twelve active components were selected from 196 components of Bulao Elixir. Among 2209 targets, 102 effective targets were selected, and 30 important targets were identified via matching with the disease targets. After further analysis, 14 core targets were identified. Enrichment analysis revealed that most of these important targets were involved in multiple biological processes, including apoptosis, inflammatory reactions, and cell regulation cycles.
