2Blockade of cholecystokinin-2 receptor and cyclooxygenase-2 synergistically induces cell apoptosis, and inhibits the proliferation of human gastric cancer cells in vitro显示文摘Gastrin and cyclooxygenase-2(COX-2) playimportant roles in the carcinogenesis and progression ofgastric cancer.However,it remains unknown whether the combination of cholecystokinin-2(CCK-2) receptor antagonist plus COX-2 inhibitor exerts synergistic anti-tumor effects on human gastric cancer.Here,we demonstrated that the combination of AG-041R(a CCK-2 receptor antagonist) plus NS-398(a selective COX-2 inhibitor) treatment had synergistic effects on proliferation inhibition,apoptosis induction,down-regulation of Bcl-2 and up-regulation of Bax expression in MKN-45 cells.These results indicate that simultaneous targeting of CCK-2 receptor and COX-2 may inhibit gastric cancer development more effectively than targeting either molecule alone.(C)2008 Elsevier Ireland Ltd.All rights reserved.Sun, W. 5Myelin Oligodendrocyte Glycoprotein-IgG Contributes to Oligodendrocytopathy in the Presence of Complement, Distinct from Astrocytopathy Induced by AQP4-IgG显示文摘Immunoglobulin G against myelin oligodendrocyte glycoprotein(MOG-Ig G) is detectable in neuromyelitis optica spectrum disorder(NMOSD) without aquaporin-4 Ig G(AQP4-Ig G), but its pathogenicity remains unclear.In this study, we explored the pathogenic mechanisms of MOG-Ig G in vitro and in vivo and compared them with those of AQP4-Ig G. MOG-Ig G-positive serum induced complement activation and cell death in human embryonic kidney(HEK)-293 T cells transfected with human MOG. In C57 BL/6 mice and Sprague-Dawley rats, MOG-Ig G only caused lesions in the presence of complement. Interestingly, AQP4-Ig G induced astroglial damage, while MOGIg G mainly caused myelin loss. MOG-Ig G also induced astrocyte damage in mouse brains in the presence ofcomplement. Importantly, we also observed ultrastructural changes induced by MOG-Ig G and AQP4-Ig G. These findings suggest that MOG-Ig G directly mediates cell death by activating complement in vitro and producing NMOSDlike lesions in vivo. 6In Search of the Cretaceous-Tertiary Boundary in Heilongjiang River Area of China显示文摘Abstract A new geological section recognized in Baishantou of Jiayin, Heilongjiang Province, China, was studied preliminarily in searching for the K/T boundary. An early Paleocene florule characrerized by abundant Tiliaephyllum tsagajanicum leaves and its associated plants, was collected from the Baishantou Member (new member representing the lower part of the Paleocene Wuyun Formation) overlying the possible K/T boundary. A tuff bed was found from the upper part of the Furao Formation (uppermost Cretaceous) in this section. The new findings imply that the K/T boundary is probable lying in the interval between the base of the Baishantou Member and the tuff bed, based on the correlation of the present strata with the stratotype of the Tsagajan Group spanning the K/T boundary in 'Belaya Gara' of the Bureya Basin, Russia. The early Paleocene florule from the Baishantou Member and the early-middle Maastrichtian dinosaurs found from the Yuliangzi Formation underlying the Furao Formation suggest that the Baishantou section is more promising for searching for the K/T boundary in this area. Further studies of the section is needed, integrating paleontological, biostratigraphic, geochemical, paleomagnetic, and sedimentary research would benefit our understanding the mass extinction event and biotic recovery in the Heilongjiang River area, China, and even all of northeast Asia during the K/T boundary interval.Sun G. 7Late Cretaceous biota and the Cretaceous-Paleogene(K--Pg)Boundary in Jiayin of Heilongjiang,China显示文摘An international cooperative study of the Late Cretaceous biota and non-marine strata including the Cretaceous-Paleogene(K--Pg) boundary in Jiayin near the Heilongjiang River,China during 2002-2010,is summarized in this paper. The strata includes the Upper Cretaceous Yong'ancun-,Taipinglinchang-,Yuliang-zi-,and Furao formations,and the Paleocence Wuyun Formation consisting of Baoshantou Member and a Coalbearing Member. Seven palynological assemblages from Santonian to Paleocene are recognized,which is used for definition of the K--Pg boundary. Through the palynological analysis of three drilled boreholes (XHY-2005,2006,2008) in Xiaoheyan of western Jiayin,the K--Pg boundary is defined within 20. 00-20. 05 m in the borehole XHY-2006 (as a standard boundary) which is just between the top of Furao Formation (late Maasterichtian) and the base of Baishantou Member (early Danian) . These new research results are supported by paleo- magnetic,geochemical and other analyses,also. On the other hand,the presence of eight taxa of Maasterichtian dinosaurs (mostly hadrosaurids) have been recognized by the authors; and two mega-plant assemblages from the Yong'ancun Fm (Santonian) to Taipinglinchang Fm (Campanian) are confirmed. These research a-chievements indicate that the Upper Cretaceous-Paleocene in Jiayin can be well correlated to the strata in the neighboring Zeya-Bureja basin,Russia. The K--Pg boundary definited in Jiayin is the first well documented non-marine K--Pg boundary in China. 9Id2 regulates the proliferation of squamous cell carcinoma in vitro via the NF-κB/Cyclin D1 pathway显示文摘Squamous cell carcinoma (SCC) is a significant cause of cancer morbidity and mortality worldwide, with an incidence of up to 166 cases per 100 000 population. It arises in the skin, upper aerodigestive tract, lung, and cervix and affects more than 200 000 Americans each year. We report here that a microarray experiment comparing 41 SCC and 13 normal tissue specimens showed that Id2, a gene that controls the cell cycle, was significantly up-regulated in SCC. Enforced expression of Id2 in vitro stimulated the proliferation of SCC cells and up-regulated the transcription of nuclear factor kappa B (NF-κB) and cyclin D1. Enhancement of the NF-κB activity with p65 significantly increased the cell proliferation and the transcription of cyclin D1, whereas inhibition of the NF-κB activity with I kappa B alpha mutant (IκBα M) and pyrroline dithiocarbamate (PDTC) abrogated cell proliferation and transcription of cyclin D1. Furthermore, a mutated NF-κB binding site in the cyclin D1 promoter fully abrogated the Id2-induced transcription of cyclin D1. 10Cyclocarya cf. paliurus (Batal.) Iljinskaja (Juglandaceae) from the Hunchun Formation (Eocene), Jilin Province, China显示文摘The leaflet architecture of Cyclocarya cf. paliurus (Batal.) Iljinskaja from the Hunchun Formation (Middle Eocene) shows similarity to that of modern C. paliurus (Batal.) Iljinskaja and the specimen is the oldest fossil record in Europe and Asia. The distributions of C. cf. paliurus and other fossil records,such as Glyptostrobus, Metasequoia, Nyssa, and Liquidambar, in Hunchun flora show that it would have been a warmer-temperature to subtropical climate in Hunchun District during the Eocene period.Tong-Xing SUN Albert G. 12K-ATP channel openers facilitate glutamate uptake by GluTs in rat primary cultured Astrocytes显示文摘Increasing evidence, including from our laboratory, has revealed that opening of ATP sensitive potassium channels(K-ATP channels) plays the neuronal protective roles both in vivo and in vitro. Thus K-ATP channel openers(KCOs) have been proposed as potential neuroprotectants. Our previous studies demonstrated that K-ATP channels could regulate glutamate uptake activity in PC12 cells as well as in synaptosomes of rats. Since glutamate transporters(GluTs) of astrocytes play crucial roles in glutamate uptake and KATP channels are also expressed in astrocytes, the present study showed whether and how KATP channels regulated the function of GluTs in primary cultured astrocytes. The results showed that nonselective KCO pinacidil, selective mitochondrial KCO diazoxide, novel, and blood-brain barrier permeable KCO iptakalim could enhance glutamate uptake, except for the sarcolemmal KCO P1075. Moreover pinacidil, diazoxide, and iptakalim reversed the inhibition of glutamate uptake induced by 1-methyl-4-phenylpyridinium(MPP+). These potentiated effects were completely abolished by mitochondrial K-ATP blocker 5-hydroxydecanoate. Furthermore, either diazoxide or iptakalim could inhibit MPP+-induced elevation of reactive oxygen species (ROS) and phosphorylation of protein kinases C(PKC). These findings are the first to demonstrate that activation of K-ATP channel, especially mitochondrial K-ATP channel, improves the function of GluTs in astrocytes due to reducing ROS production and downregulating PKC phosphorylation. 15The level of oncogene H-Ras correlates with tumorigenicity and malignancy显示文摘Normal bovine adrenocortical cells and some human fibroblasts can be transformed by SV40T and H-Ras in a Ras-dependent manner. We recently reported that high levels of Ras derived from 5' LTR of retrovirrus can induce highly malignant and fast growing tumors, while lower levels of Ras derived from internal ribosome entry site(IRES) promotes slower tumor growth and loss of malignancy. Ras derived from CMV promoters resulted in much lower Ras levels and loss of tumor malignancy and growth. Further studies showed that the tumors formed in the presence of lower levels of Ras and dominant negative P53(P53DD) had fewer apoptotic cells and grew faster than the tumors formed from cells with same level Ras and SV40T. Our studies suggest that low levels of Ras are insufficient to inhibit apoptosis induced by pRb inactivation. In contrast, high levels of Ras not only allow normal cells to exit senescence and form tumors, but also protect against pRb inhibition-induced cell apoptosis.Sun, B. C. Gao, Y. Deng, L. Li, G. Q.
