1Transplantation of UC-MSCs on collagen scaffold activates follicles in dormant ovaries of POF patients with long history of infertility显示文摘Premature ovarian failure(POF) is a refractory disease for clinical treatment with the goal of restoring fertility. In this study,umbilical cord mesenchymal stem cells on a collagen scaffold(collagen/UC-MSCs) can activate primordial follicles in vitro via phosphorylation of FOXO3 a and FOXO1. Transplantation of collagen/UC-MSCs to the ovaries of POF patients rescued overall ovarian function, evidenced by elevated estradiol concentrations, improved follicular development, and increased number of antral follicles. Successful clinical pregnancy was achieved in women with POF after transplantation of collagen/UC-MSCs or UC-MSCs. 3Population-based cancer incidence analysis in Beijing, 2008-2012显示文摘Objective: To analyze the incidence of cancer during 2008-2012 in Beijing, China, and compare the cancer spectrum with that during 1998-1999.Methods: Data from the Beijing Cancer Registry(BCR), which covered 12 million residents and 16 administrative regions in Beijing, were checked and evaluated on basis of the criteria of data quality from the National Central Cancer Registry(NCCR) of China. Incidences were calculated stratified by cancer type, sex, areas(urban/rural), and age. The Chinese census population in 1982 and the world Segi's population were used for calculating the age-standardized incidences.Results: A total of 177,101 new cancer cases were diagnosed in Beijing between 2008 and 2012. The crude incidence rate(CR) of all cancers was 282.64/100,000(290.71/100,000 in males and 274.45/100,000 in females). The age-standardized rates by Chinese standard population(ASR-China) and by world standard population(ASR-world) were 124.46/100,000 and 161.18/100,000, respectively. Female breast cancer was the most common cancer, followed by lung cancer, colorectal cancer, liver cancer, and stomach cancer, with the CR of 59.87/100,000, 59.21/100,000, 32.49/100,000, 19.81/100,000 and 17.96/100,000, respectively. In urban areas, female breast cancer(68.50/100,000) was still the most common cancer, followed by lung cancer(61.23/100,000), colorectal cancer(37.23/100,000), prostate cancer(20.49/100,000) and stomach cancer(20.07/100,000). In rural areas, lung cancer(55.94/100,000) was the most common cancer, followed by female breast cancer(45.87/100,000), colorectal cancer(24.77/100,000), liver cancer(20.68/100,000) and stomach cancer(14.52/100,000). Great changes of the cancer spectrum were found from the period of 1998-1999 to the period of 2011-2012 in Beijing.Conclusions: The cancer burden in Beijing was heavier than the national average level. 4Characteristics and trends in incidence of childhood cancer in Beijing,China,2000-2009显示文摘Objective: To investigate the characteristics and incidence trends of childhood cancer in Beijing, China, from 2000 to 2009.Methods: A total of 1,274 cases with childhood cancer in Beijing from 2000 to 2009 were included in the study. All rates were age-standardized using the direct method to the world standard population and expressed per million person-years. Incidence trends were characterized by calculating annual percent change(APC) using Joinpoint Regression Program.Results: The crude incidence rate was 106.47 per million [age-standardized rate(ASR) 113.34] between 2000 and 2009 in Beijing with the most common diagnoses, leukemia(N=505, 39.64%, ASR 45.20), followed by central nervous system(CNS) tumors(N=228, 17.90%, ASR 19.28) and lymphoma(N=91, 7.14%, ASR 6.97). The incidence for all childhood cancers combined has increased during the study period, with an APC of 5.84% [95% confidence interval(95% CI): 1.0-10.9] after adjusted by world population. The ASR of all combined cancers in boys showed a slight, but no significant increase, with an APC of 5.33%(95% CI: –0.6-11.6); for girls, the trends increased significantly, with an APC of 6.54%(95% CI: 1.5-11.8).Conclusions: The incidence rate of childhood cancer in Beijing was higher than the average level of China and lower than that of western countries. The incidence trends of childhood cancer, especially leukemia among girls showed a significantly increase from 2000 to 2009. While among boys, no substantially change was seen during the observed time period. 5Angiogenesis in hepatocellular carcinoma:mechanisms and anti-angiogenic therapies显示文摘Hepatocellular carcinoma(HCC)is the fourth leading cause of cancer-associated death worldwide.Angiogenesis,the process of formation of new blood vessels,is required for cancer cells to obtain nutrients and oxygen.HCC is a typical hypervascular solid tumor with an aberrant vascular network and angiogenesis that contribute to its growth,progression,invasion,and metastasis.Current anti-angiogenic therapies target mainly tyrosine kinases,vascular endothelial growth factor receptor(VEGFR),and plateletderived growth factor receptor(PDGFR),and are considered effective strategies for HCC,particularly advanced HCC.However,because the survival benefits conferred by these anti-angiogenic therapies are modest,new anti-angiogenic targets must be identified.Several recent studies have determined the underlying molecular mechanisms,including pro-angiogenic factors secreted by HCC cells,the tumor microenvironment,and cancer stem cells.In this review,we summarize the roles of pro-angiogenic factors;the involvement of endothelial cells,hepatic stellate cells,tumor-associated macrophages,and tumor-associated neutrophils present in the tumor microenvironment;and the regulatory influence of cancer stem cells on angiogenesis in HCC.Furthermore,we discuss some of the clinically approved anti-angiogenic therapies and potential novel therapeutic targets for angiogenesis in HCC.A better understanding of the mechanisms underlying angiogenesis may lead to the development of more optimized anti-angiogenic treatment modalities for HCC.Changyu Yao Shilun Wu Jian Kong Yiwen Sun Yannan Bai Ruhang Zhu Zhuxin Li Wenbing Sun Lemin Zheng 2023Cancer Biology & Medicine2023,20,1:3

7A non-metabolic function of hexokinase 2 in small cell lung cancer:promotes cancer cell stemness by increasing USP11-mediated CD133 stability显示文摘Background:Maintenance of cancer stem-like cell(CSC)stemness supported by aberrantly regulated cancer cell metabolism is critical for CSC self-renewal and tumor progression.As a key glycolytic enzyme,hexokinase 2(HK2)plays an instrumental role in aerobic glycolysis and tumor progression.However,whether HK2 directly contribute to CSC stemness maintenance in small cell lung cancer(SCLC)is largely unclear.In this study,we aimed to investgate whether HK2 independent of its glycolytic activity is directly involved in stemness maintenance of CSC in SCLC.Methods:Immunoblotting analyses were conducted to determine the expression of HK2 in SCLC CSCs and their differentiated counterparts.CSC-like properties and tumorigenesis of SCLC cells with or without HK2 depletion or overexpression were examined by sphere formation assay and xenograft mouse model.Immunoprecipitation and mass spectrometry analyses were performed to identify the binding proteins of CD133.The expression levels of CD133-associated and CSC-relevant proteins were evaluated by immunoblotting,immunoprecipitation,immunofluorescence,and immunohistochemistry assay.RNA expression levels of Nanog,POU5F1,Lin28,HK2,Prominin-1 were analyzed through quantitative reverse transcription PCR.Polyubiquitination of CD133 was examined by in vitro or in vivo ubiquitination assay.CD133+cells were sorted by flow cytometry using an anti-CD133 antibody.Results:We demonstrated that HK2 expression was much higher in CSCs of SCLC than in their differentiated counterparts.HK2 depletion inhibited CSC stemness and promoted CSC differentiation.Mechanistically,nonmitochondrial HK2 directly interacted with CD133 and enhanced CD133 expression without affecting CD133 mRNA levels.The interaction of HK2 and CD133 promoted the binding of the deubiquitinase ubiquitin-specific protease 11(USP11)to CD133,thereby inhibiting CD133 polyubiquitylation and degradation.HK2-mediated upregulation of CD133 expression enhanced the expression of cell renewal regulators,SCLC cell stemness,and tumor growth in mice.In addition,HK2 expression was positively correlated with CD133 expression in human SCLC specimens,and their expression levels were associated with poor prognosis of SCLC patients.Conclusions:These results revealed a critical non-metabolic function of HK2 in promotion of cancer cell stemness.Our findings provided new insights into the multifaceted roles of HK2 in tumor development.Juhong Wang Fei Shao Yannan Yang Wei Wang Xueying Yang Renda Li Hong Cheng Sijin Sun Xiaoli Feng Yibo Gao Jie He Zhimin Lu 2022Cancer Communications2022,42,10:2

10The spatial coupling characteristics between the construction of Qingzang National Park Cluster and the sustainable development of local communities显示文摘The development of Qingzang national park cluster is part of China’s efforts to establish a major ecological civi-lization project in the Qingzang Plateau,in line with global sustainable development goals.Based on preliminary scientific investigation and research in the Qingzang Plateau,the construction of the park cluster will include 21 national parks.To mitigate the conflict between conservation of the national park cluster and the develop-ment of local communities,this study proposes an analysis framework to identify the spatial coupling features between the national park cluster and sustainable development of communities.Four elements were selected to construct the analysis framework,including natural conditions,geographic location,cultural background,and national policies.This framework was applied to the 457 township communities within the 21 national parks.Results show the weak influence of the construction of the national park cluster for approximately 304 township communities,without significant spatial coupling traits,while the remaining 153 communities demonstrated significant spatial coupling features.These latter townships had developed four types of spatial coupling with na-tional parks,including eco-migrants,transportation hubs,characteristic cultures,and border development,which account for 17.4%,35.3%,19.8%,and 27.5%of the 153 townships respectively.A composite type with more than one spatial coupling feature was also found for 14 communities within the 153 townships.This provides a reference for policy making towards four major types of interactive modes between townships and the national park construction for the sustainable development of Qingzang Plateau.Rui Guo Dong Chen Daojing Zhou Baoyin Liu Hanchu Liu Yannan Zhao Yong Sun Jie Fan 2021Geography and Sustainability2021,2,1:1

16Observations on the Application of Nursing Risk Management in the Care of Critically Ill Patients in the Respiratory Unit显示文摘Objechive:Investigate the effectiveness of mursing risk management in the care of cntically ill patients in the respiratory umit.Methods:Among the cntically ill respiratory patients admitted to our hospital between May 2019 and April 2020,78 patients were randomly selected and divided into an observation group and a control group,each consisting of 39 patients.In the observation group.a mursing nisk management model was implemented,i.e,patients'clinical symptoms were observed at any time to monitor their treatment satisfaction and the effectiveness of their care and routine care was implemented for the control group.Results:The heart rate,respiratory rate,and pH of patients in the observation group were more stable than those in the control group,and their respiratory status was better,with differences in data.There was also sigmifcant statistical significance(P<0.05).The incidence of patient-provider disputes,unplanned extubation,and uplammed events were lower in the observation group conpared to the control group,and their data difference was satistically siguificant(P-0.05).The treatment satisfaction as well as the total effective rate of patients in the observation group was also much higher than that of the contol group,and there was also a statistically sigmificant difference in the data(P<0.05).Conclusion:The musing nisk management model has a significant therapeutic effect in the care of cnitically ill respiratory patients.Therefore,it is worth popularizing to use in the clinical mursing of respiratory cnitical patients.Yannan Sun 2020Journal of Clinical and Nursing Research2020,4,4:1
17High baseline tumor burden-associated macrophages promote an immunosuppressive microenvironment and reduce the efficacy of immune checkpoint inhibitors through the IGFBP2-STAT3-PD-L1 pathway显示文摘Background:Several clinical studies have uncovered a negative correlation between baseline tumor burden and the efficacy of immune checkpoint inhibitor(ICI)treatment.This study aimed to uncover the specific mechanisms underlying the difference in sensitivity to ICI treatment between tumors with high(HTB)and low(LTB)tumor burden.Methods:For in vivo studies,several mouse models of subcutaneous tumors were established,and transcriptome sequencing,immunohistochemistry,and flow cytometry assays were used to detect the immune status in these subcutaneous tumors.For in vitro experiments,co-culture models,cytokine antibody arrays,western blotting,flow cytometry,and enzyme-linked immunosorbent assays were used to explore the underlying molecular mechanisms Results:We found that MC38 or B16 subcutaneous tumors from the HTB group did not show any response to anti-programmed cell death protein-1(PD-1)therapy.Through flow cytometry assays,we found that the infiltration with CD8^(+)T cellswas significantly decreasedwhereasM2-like macrophageswere enriched in subcutaneous tumors of HTB groups compared with those of LTB group.These changes were not affected by the initial number of injected tumor cells or tumor age,nor could they be reversed by surgical tumor reduction.Intraperitoneal colony-stimulating factor 1 receptor(CSF-1R)inhibitor PLX3397 injection at different time points of tumor growth only had an effect when administered in the early tumor stage to maintain the“heat”of the tumor microenvironment during the process of tumor growth,thereby achieving a response to ICI treatment when the tumor grew to a large size.Mechanistically,we found that insulin-like growth factor binding protein 2(IGFBP2)expression levelswere significantly elevated in HTB tumor tissues.IGFBP2 promoted the programmed death-ligand 1(PD-L1)expression in M2-like macrophages by activating signal transducer and activator of transcription 3(STAT3),and PD-L1^(+)M2-likemacrophages exerted an immunosuppressive effect by inhibiting the proliferation and activation of CD8^(+)T cells in a PD-L1-dependent fashion.Conclusions:This study suggested that the low efficacy of ICI treatment in HTB tumors is mainly attributed to the intratumoral accumulation of PD-L1^(+)M2-like macrophages via the IGFBP2-STAT3-PD-L1 signaling pathway and their substantial inhibitory effects on T cell proliferation and activation.Zhaowei Wen Huiying Sun Zhihua Zhang Yannan Zheng Siting Zheng Jianping Bin Yulin Liao Min Shi Rui Zhou Wangjun Liao 2023Cancer Communications2023,43,5:0
18Understanding water conservation vs. prof ligation traits in vegetable legumes through a physio-transcriptomic-functional approach显示文摘Abstract Vegetable soybean and cowpea are related warm-season legumes showing contrasting leaf water use behaviors under similar root drought stresses,whose mechanisms are not well understood.Here we conducted an integrative phenomic-transcriptomic study on the two crops grown in a feedback irrigation system that enabled precise control of soil water contents.Continuous transpiration rate monitoring demonstrated that cowpea used water more conservatively under earlier soil drought stages,but tended to maintain higher transpiration under prolonged drought.Interestingly,we observed a soybean-specific transpiration rate increase accompanied by phase shift under moderate soil drought.Time-series transcriptomic analysis suggested a dehydration avoidance mechanism of cowpea at early soil drought stage,in which the VuHAI3 and VuTIP2;3 genes were suggested to be involved.Multifactorial gene clustering analysis revealed different responsiveness of genes to drought,time of day and their interactions between the two crops,which involved species-dependent regulation of the circadian clock genes.Gene network analysis identified two co-expression modules each associated with transpiration rate in cowpea and soybean,including a pair of negatively correlated modules between species.Module hub genes,including the ABA-degrading gene GmCYP707A4 and the trehalose-phosphatase/synthase gene VuTPS9 were identified.Intermodular network analysis revealed putative co-players of the hub genes.Transgenic analyses verified the role of VuTPS9 in regulating transpiration rate under osmotic stresses.These findings propose that species-specific transcriptomic reprograming in leaves of the two crops suffering similar soil drought was not only a result of the different drought resistance level,but a cause of it.Pingping Fang Ting Sun Arun Kumar Pandey Libo Jiang Xinyang Wu Yannan Hu Shiping Cheng Mingxuan Li Pei Xu 2023Horticulture Research2023,10,3:0
19Four-birds-with-one-stone:A multifunctional Ti-based material for solar-driven water evaporation显示文摘Sustainable,environmentally friendly and low-energy desalination materials have important research value for the increasing demand of freshwater year by year.However,it is a huge challenge to maintain high heat energy transfer efficiency without reducing the heat conversion capacity of specific solar photothermal conversion materials.Moreover,their efficiency and durability are greatly limited by the problems of seawater corrosion,oil,and bacteria pollutions.Till now,no related work has been reported to solve all the aforementioned problems via a simple four-birds-with-one-stone strategy.Herein,a class of multifunctional porous photothermal silver(Ag)modified Ti foams(Tf-TA/Ag series materials)is prepared for the development of advanced solar water evaporation devices,and provides alternative materials for alleviating freshwater crisis and treating sewage.The oil contact angle(OCA)changes from 41°to 180°,which significantly reduces the adhesion of oil.In addition,Tf-TA2/Ag sample also shows an excellent and sustained antibacterial effect,which maintains above 99.9%of antibacterial rate after repeated 5 times.The surface temperature of the Tf-TA2/Ag sample reaches 52.5℃ after simulated sun irradiation for 20 min,which is significantly higher than that of the contact groups(water:36.4℃,Ti foam:38.2℃ and Tf-TA2:40.9℃).The capacity of seawater evaporation and salt removal is enhanced due to the excellent photothermal properties,low reflectance,and uniform heat dissipation pores.The water production efficiency of Tf-TA2/Ag sample is 1.41 kg·m^(-2)·h^(-1) in artificial seawater and 0.76 kg·m^(-2)·h^(-1) in oily sewage under simulated sun irradiation.Furthermore,the hydrophilic and oleophobic properties of Tf-TA2/Ag are critical to extracting water from oil/water mixture in diverse water environments.Ultimately,this four-birds-with-one-stone approach provides a new perspective for the improvement of solar seawater desalination performance.Yannan Li Yaning Sun Dan Zhang Yu Xue Jie Wang Nan Zhang Jun Wang Jiangwei Zhang Yan Zhao Jun-Jie Liu 2024Nano Research2024,17,5:0
20Conserved termini and adjacent variable region of Twortlikevirus Staphylococcus phages显示文摘Methicillin-resistant Staphylococcus aureus(MRSA) is an increasing cause of serious infection,both in the community and hospital settings. Despite sophisticated strategies and efforts, the antibiotic options for treating MRSA infection are narrowing because of the limited number of newly developed antimicrobials. Here, four newly-isolated MRSA-virulent phages, IME-SA1, IMESA2, IME-SA118 and IME-SA119, were sequenced and analyzed. Their genome termini were identified using our previously proposed 'termini analysis theory'. We provide evidence that remarkable conserved terminus sequences are found in IME-SA1/2/118/119, and, moreover, are widespread throughout Twortlikevirus Staphylococcus phage G1 and K species. Results also suggested that each phage of the two species has conserved 5′ terminus while the 3′ terminus is variable. More importantly, a variable region with a specific pattern was found to be present near the conserved terminus of Twortlikevirus S. phage G1 species. The clone with the longest variable region had variable terminus lengths in successive generations, while the clones with the shortest variable region and with the average length variable region maintained the same terminal length as themselves during successive generations. IME-SA1 bacterial infection experiments showed that the variation is not derived from adaptation of the phage to different host strains.
